Journal of Dental Sciences
Oral ulcers are common inflammatory lesions of the oral mucosa that significantly affect patients’ quality of life through pain, eating difficulties, and psychological stress. Their development is multifactorial, involving mechanical trauma, infection, nutritional deficiencies, immune dysregulation, and oxidative stress. Conventional treatments including antiseptics, corticosteroids, and immunomodulators, provide symptomatic relief but are often limited by side effects, recurrence, and poor patient adherence. Increasing interest has therefore focused on herbal medicines as complementary or alternative therapies. Numerous single herbs such as Aloe vera, licorice, curcumin, chamomile, berberine, neem, sage, myrrh, and guava and traditional formulations have demonstrated anti-inflammatory, antioxidant, immunomodulatory, antimicrobial, and …

Runt-Related Transcription Factor 2 Transcriptionally Represses Sirtuin 1 To Regulate Osteoclastogenesis And Tooth Eruption, Jie Li, Changqing Yan, Xinyi Sun, Chenying Zhang, Xiaozhe Wang, Yang Liu, Shuguo Zheng
Journal of Dental Sciences
Background/purpose: Cleidocranial dysplasia (CCD), caused by Runt-related transcription factor 2 (RUNX2) haploinsufficiency, features delayed tooth eruption. Although impaired osteoclastogenesis contributes to eruption failure, the direct molecular mechanisms linking RUNX2 deficiency to osteoclast dysfunction remain elusive. Sirtuin 1 (SIRT1) is a critical regulator of osteoclastogenesis; however, whether it serves as a downstream effector of RUNX2 remains unexplored. This study aimed to elucidate the mechanistic link between RUNX2 and SIRT1 in regulating osteoclast differentiation and tooth eruption.
Materials and methods: Tooth eruption and osteoclast differentiation were assessed by micro-computed tomography and histological analysis in Runx2+/- mice. Chromatin immunoprecipitation quantitative polymerase chain …

Journal of Dental Sciences
Background/purpose: Root canal curvature is a complex anatomical variation that complicates endodontic treatments. Traditionally, dentists rely on two-dimensional periapical radiographs (PA) and manual methods to estimate curvature angles. However, this conventional approach is highly time-consuming and subjective, often leading to inconsistent measurements.
Materials and methods: This study aims to develop a fully automated, objective framework for precise angular measurement and severity grading of root canal curvature. The YOLO26n-seg model was employed to segment roots and classify single- or multi-rooted teeth from PA. Furthermore, we proposed an image processing algorithm that standardized orientation, extracted centerlines, and applied three-point circle fitting …

This study examined the effects of ischemic preconditioning (IPC) on physical performance, considering the influence of timing, warm-up (WU), participant characteristics, and IPC protocols structure. A total of 90 trials (1,439 participants) were retrieved from three databases and assessed using PICOS criteria. Multilevel meta-analyses with cluster-robust variance adjustments were conducted to calculate pooled effect sizes (Hedge’s g). Risk of bias and certainty of evidence were assessed using the RoB 2 tool and GRADE framework. IPC produced a trivial but significant improvement in physical performance (g = 0.13, P < 0.01), which persisted after excluding SHAM effects (g = 0.10; P < 0.01). Significant improvements were observed for maximum repetitions, time to failure, and power output, but not for jump ability, strength, or oxygen uptake. Comparable benefits were found for anaerobic (g = 0.15) and aerobic (g = 0.10) exercise, with greater effects in males and less-trained participants. Performance was further enhanced when IPC was followed by WU (g = 0.16), with the optimal IPC-to-WU interval being ~42 minutes. Without WU, the effective IPC window narrowed to 6–7 minutes. In practice, IPC can enhance physical performance, independently of sham effects, moderated by sex, training level, and WU. For competition or testing, the most effective strategy appears to be 3 or 4 × 5-min IPC protocol, followed by a 42-min interval, standardized warm-up, and endurance testing. For mechanistic studies, WU should be excluded, and testing conducted 6-7 minutes post-IPC. Future research should target women, middle-aged individuals, and elite athletes.

Secondary salinization is a significant global issue affecting up to 1 billion hectares of land, impacting biodiversity and particularly vegetation. One form of revegetation that can provide a return to landholders is saltbush (Atriplex spp.), which provides forage for a wide range of livestock. The effectiveness of saltbush plantings in providing useful habitat for fauna in agricultural landscapes suffering from secondary salinity is poorly understood. We aimed to address this knowledge gap by surveying bird, ant, and spider communities, and quantifying vegetation structure and floristic composition in saltbush and native woodlands in agricultural landscapes affected by secondary salinization in Western …

Background/purpose: The pH within infected root canals fluctuates dynamically under pathological conditions, yet its impact on stem cells from the apical papilla (SCAPs) in dental pulp regeneration remains unclear. This study investigated how potential of hydrogen (pH) variations govern SCAPs survival and the associated signaling to identify therapeutic targets for enhancing tissue regeneration.
Materials and methods: SCAPs were cultured at pH 5.4-10.4. Proliferation and apoptosis were assessed using cell counting kit-8 (CCK-8) assays, real-time live-cell analysis, and flow cytometry. RNA-sequencing (RNA-seq) identified differentially expressed genes, which were validated through reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR), Western blotting, and mechanistic …

Background/purpose: Niche for osteogenesis forms near specialized blood vessels that highly express both CD31 and endomucin (Emcn). However, localization of the vessels remains unknown during wound healing process after pulp revascularization. We investigated the localization of both CD31 and Emcn-expressing vessels and osterix (Osx)-expressing cells after pulp revascularization. Materials and methods: Five-week-old male C57BL/6J mice underwent pulp revascularization in the mesial root canal of the right first maxillary molar. The mice were sacrificed at 1 hour, 7 days, 14 days, and 21 days post-surgery, and immunofluorescence for CD31, Emcn, and Osx was performed. Additionally, calcein labeling was used to visualize …

Background/purpose: Hyperglycemia contributes to periodontal inflammation through oxidative stress and inflammatory mediator production in gingival fibroblasts. Sesamin, a major lignan from sesame, has antioxidant and anti-inflammatory properties. This study investigated whether sesamin attenuates high glucose (HG)-induced cyclooxygenase-2 (COX-2) expression and prostaglandin E2 (PGE2) production in human gingival fibroblasts (HGFs), and examined the involvement of Toll-like receptor 2 (TLR2)-dependent oxidative signaling.
Materials and methods: HGFs were exposed to HG in the presence or absence of sesamin. Cell viability, reactive oxygen species (ROS) generation, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity, COX-2 expression, and PGE2 production were evaluated. …

Background/purpose: In certain clinical bone grafting scenarios, bone substitutes frequently interface with the adjacent soft tissues. This study aimed to investigate whether deproteinized bovine bone mineral (DBBM) and coralline hydroxyapatite (CHA) induce paracrine responses in human gingival mesenchymal stem cells (GMSCs), thereby altering the pro-angiogenic capacity of their derived exosomes, and to profile specific cargo alterations.
Materials and methods: Exosomes were isolated from GMSCs co-cultured with DBBM (DBBM-G-EXO) or CHA (CHA-G-EXO), while native GMSCs-derived exosomes (G-EXO) were collected as controls. The angiogenic effects of these exosomes on human umbilical vein endothelial cells (HUVEC) were evaluated via cell proliferation, scratch wound …

Though promising in healthcare consultation applications, large language models (LLMs) face critical limitations in retaining and utilizing long-term memory across multiturn interactions. In particular, existing memory enhancing paradigms are constrained by limited context windows and embedding-based retrieval, often failing to maintain task relevance and still suffering from memory prototype collapse in multi-turn healthcare consultation. To address these challenges, we propose a cognitively-inspired memory framework named MemoryART, which is grounded in Adaptive Resonance Theory (ART)—a cognitive and learning theory of how humans and animals adapt to dynamic environments. MemoryART employs three memory modules—working memory, episodic memory, and semantic memory to support …

Current large language models (LLMs) exhibit significant deficiencies in episodic memory tasks including encoding, storing, and retrieving specific information from temporally dependent events over a long period of time. Recent approaches to handle memory tasks in LLMs, such as in-context learning, retrieval-augmented generation (RAG), and fine-tuning, may resolve the long-term retention issues, but are still inadequate to handle tasks requiring chronological awareness of the stored information. We introduce Agentic Retrieval with Temporal-Episodic Memory (ARTEM), a hybrid LLM-based agent architecture integrating LLMs with a self-organizing neural network named Spatial-Temporal Episodic Memory (STEM), designed to handle episodic memory tasks. Our approach employs …
