Drug resistance and a dire lack of transmission-blocking antimalarials hamper malaria elimination. Here, we present the pantothenamide MMV693183 as a first-in-class acetyl-CoA synthetase (AcAS) inhibitor to enter preclinical development. Our studies demonstrate attractive drug-like properties and in vivo efficacy in a humanized mouse model of Plasmodium falciparum infection. The compound shows single digit nanomolar in vitro activity against P. falciparum and P. vivax clinical isolates, and potently blocks P. falciparum transmission to Anopheles mosquitoes. Genetic and biochemical studies identify AcAS as the target of the MMV693183-derived antimetabolite, CoA-MMV693183. Pharmacokinetic-pharmacodynamic modelling predict that a single 30 mg oral dose is sufficient …

Communities Of Water: An Examination Of Cooperation And Conflict In Water Management Practices In The Central Peruvian Andes, Sylvie D. Littledale
Communities Of Water: An Examination Of Cooperation And Conflict In Water Management Practices In The Central Peruvian Andes, Sylvie D. Littledale
Theses and Dissertations
This thesis explores two examples of water management in the highland Peruvian villages, San Pedro de Llancha and San Antonio de Chinchina. One example is of cooperation and union between the two communities. The other is of conflict between the same two villages just 40 years prior. I examine ethnographic and ethnohistorical data from both the collaborative period and the conflictive period of these two communities’ relations over water. The data suggest that, while the outcomes of these two periods were drastically different, the processes through which these communities came into relation with one another were quite similar. Their communal …

Ragdoll Recovery: Manipulating Virtual Mannequins To Aid Action Sequence Proficiency, P Watson, S Gaudl
School of Engineering, Computing and Mathematics
In this paper, we present a Virtual Reality (VR) prototype to support the demonstration and practice of the First Aid recovery position. When someone is unconscious and awaiting medical attention, they are placed in the recovery position to keep their airways clear. The recovery position is a commonly taught action sequence for medical professionals and trained first-aiders across industries. VR is a potential pathway for recovery position training as it can deliver spatial information of a demonstrated action for a subsequent copy. However, due to limits of physical interaction with virtual avatars, the practice of this motor sequence is normally …

Nfat Factors Are Dispensable For The Development But Are Critical For The Maintenance Of Foxp3+ Regulatory T Cells, De Brito C Barahona, Amiya Kumar Patra
Peninsula Medical School
The transcription factors of the nuclear factor of activated T cell (NFAT) family play a crucial role in multiple aspects of T cell function. It has recently been reported that NFATs play an important role in the suppressive function of CD4+CD25+Foxp3+ regulatory T (Treg) cells. In this study, we have investigated the role of NFATs in the thymic development of Treg cells in mice. We show that NFAT factors are dispensable for the development of Foxp3+ Treg cells in the thymus but are critical for the maintenance of both the phenotype and survival of Treg cells in the thymus as …

Making Invisible Rna Visible: Discriminative Sequencing Methods For Rna Molecules With Specific Terminal Formations, Megumi Shigematsu, Yohei Kirino
Computational Medicine Center Faculty Papers
Next generation sequencing of RNA molecules (RNA-seq) has become a common tool to characterize the expression profiles of RNAs and their regulations in normal physiological processes and diseases. Although increasingly accumulating RNA-seq data are widely available through publicly accessible sites, most of the data for short non-coding RNAs (sncRNAs) have been obtained for microRNA (miRNA) analyses by standard RNA-seq, which only capture the sncRNAs with 5′-phosphate (5′-P) and 3′-hydroxyl (3′-OH) ends. The sncRNAs with other terminal formations such as those with a 5′-hydroxyl end (5′-OH), a 3′-phosphate (3′-P) end, or a 2′,3′-cyclic phosphate end (2′,3′-cP) cannot be efficiently amplified and …
Bio-Ionic Liquid Functionalized Hydrogels Towards Smart Tissue Regeneration, Vaishali Krishnadoss
Bio-Ionic Liquid Functionalized Hydrogels Towards Smart Tissue Regeneration, Vaishali Krishnadoss
Theses and Dissertations
A blend of scaffolds, biologically active molecules, and cells are required to assemble functional constructs to repair and regenerate damaged tissue or organ via tissue engineering. The scaffold supports cell growth and proliferation and acts as a medium for diverse cellular activities. Even though hydrogel's high-water content and flexible nature make it a pronounced applicant as a scaffold, they exhibit significant technical limitations such as the absence of cell-binding motifs, lack of oxygen, conductivity, adhesive properties, growth of cells in a 3-dimensional (3D) microenvironment. In this thesis, a novel material platform is evaluated and studied to address the concerns mentioned …

Stability Analysis Of Delay-Driven Coupled Cantilevers Using The Lambert W-Function, Daniel Siebel-Cortopassi
Stability Analysis Of Delay-Driven Coupled Cantilevers Using The Lambert W-Function, Daniel Siebel-Cortopassi
USF Tampa Graduate Theses and Dissertations
A coupled delay-feedback system of two cantilevers can yield greater sensitivity than that of asingle cantilever system, with potential applications in atomic force microscopy. The Lambert W-function analysis concept for delay differential equations is used to more accurately model the behavior of specific configurations of these cantilever systems. We also use this analysis concept to find parameters which yield stability for greater parameter ranges, of the delay differential equations. The Q factor, or quality factor, is the ratio of energy stored in the system, to the energy lost per fixed oscillation/movement cycle. Having stability of the cantilevers corresponds to the …

Alzheimer’s disease (AD) is the most common neurodegenerative disorder. Pathologically, it is characterized by the deposition of amyloid beta (Aβ) plaques and presence of neurofibrillary tangles. These drive microglia neuroinflammation and consequent neurodegeneration. While the means to affect Aβ plaque accumulation was achieved how it affects disease outcomes remains uncertain. Cerium oxide (CeO2) reduces Aβ plaques, oxidative stress, inflammation, and Alzheimer’s disease (AD) signs and symptoms. Specifically, CeO2 nanoparticles (CeO2NPs) induces free radical scavenging and cell protective intracellular signaling. This can ameliorate the pathobiology of an AD-affected brain. In order to investigate, CeO2NPs …

Technological Approaches To Enhance Ecosystem Carbon Sink In China: Nature-Based Solutions, Guirui Yu, Jianxing Zhu, Li Xu, Nianpeng He
Bulletin of Chinese Academy of Sciences (Chinese Version)
Carbon peak in 2030 and carbon neutralization in 2060 has been established as one of the important strategic goals of China's economic and social development. During 2010-2020, the capacity of terrestrial ecosystems in China to sequester carbon dioxide (CO2) varied from 1.0 to 1.3 Gt/a, with high uncertainty. To enhance the carbon sink capacity of Chinese terrestrial ecosystems, the combination with land space planning and ecological protection is essential. It is necessary to first stabilize the existing carbon storage in these ecosystems, such as forest, grassland, wetland, coast, and others, and then implement some important ecological protection and ecological restoration …
