Obesity is a serious global health problem because of its increasing prevalence and comorbidities, but its treatments are limited. The serotonin 2C receptor (5-HT2CR), a G-protein-coupled receptor, activates proopiomelanocortin (POMC) neurons in the arcuate nucleus of hypothalamus (ARH) to reduce appetite and weight gain. However, several 5-HT analogs targeting this receptor, e.g., lorcaserin (Lor), suffer from diminished efficacy to reduce weight after prolonged administration. Here, we show that barbadin (Bar), a novel b-arrestin/b2-adaptin inhibitor, can prevent 5-HT2CR internalization in cells and potentiate long-term effects of Lor to reduce appetite and body weight in male mice. Mechanistically, we demonstrate that Bar …

BACKGROUND: The oral glucose tolerance test (OGTT)-glucose response curves (GRCs; incessant increase, monophasic, and biphasic) reflect insulin sensitivity and β-cell function, being worse in the former and superior in the latter. Here, we examined if the OGTT-GRC pattern is worse in obese antibody (glutamic acid decarboxylase 65-kDa [GAD65] and insulinoma-associated protein-2 [IA-2])-positive (Ab
METHODS: Forty-seven obese youth, 15 Ab
RESULTS: Incessant increase OGTT-GRC is the most frequent curve type and is three-fold higher in Ab
CONCLUSION: Severe insulin deficiency, a characteristic of type 1 diabetes, seems to be related to higher prevalence of incessant increase in Ab

Rural communities in Appalachia are displaying increased obesity prevalence, yet traditional interventions have not provided a broad enough impact to improve dietary consumption patterns. Therefore, expanding efforts that address the food environment and incorporate behavioral nudges through community-developed marketing strategies may be a viable mechanism to improve food and beverage choices within this unique population. This study installed shelf-wobblers across n = 5 gas stations in one rural Appalachian county in Kentucky. Smart Snacks were identified from store inventory lists utilizing the CDC Food Service Guideline for Federal Facilities calculator and were categorized into high-protein snacks, low-fat carbohydrate snacks, meal …

Obesity is a growing problem globally and domestically. Obesity is related to several chronic and noncommunicable diseases such as hypertension, type II diabetes, coronary heart disease, and some cancers. Both obesity and its related diseases are forms of overnutrition, which is the excess intake of nutrients that causes increased body fat to the point of impaired health. Using data from the Medical Expenditure Panel Survey (MEPS) from 2008-2016, this paper is the first to estimate the causal effect of overnutrition on medical expenditure by examining the effect of overnutrition diseases on health care costs, correcting for the endogeneity bias of …

Contrasting to the established role of the hypothalamic agouti-related protein (AgRP) neurons in feeding regulation, the neural circuit and signaling mechanisms by which they control energy expenditure remains unclear. Here, we report that energy expenditure is regulated by a subgroup of AgRP neurons that send non-collateral projections to neurons within the dorsal lateral part of dorsal raphe nucleus (dlDRN) expressing the melanocortin 4 receptor (MC4R), which in turn innervate nearby serotonergic (5-HT) neurons. Genetic manipulations reveal a bi-directional control of energy expenditure by this circuit without affecting food intake. Fiber photometry and electrophysiological results indicate that the thermo-sensing MC4RdlDRN neurons …

The hypothalamus is a critical regulator of glucose metabolism and is capable of correcting diabetes conditions independently of an effect on energy balance. The small GTPase Rap1 in the forebrain is implicated in high-fat diet-induced (HFD-induced) obesity and glucose imbalance. Here, we report that increasing Rap1 activity selectively in the medial hypothalamus elevated blood glucose without increasing the body weight of HFD-fed mice. In contrast, decreasing hypothalamic Rap1 activity protected mice from diet-induced hyperglycemia but did not prevent weight gain. The remarkable glycemic effect of Rap1 was reproduced when Rap1 was specifically deleted in steroidogenic factor-1-positive (SF-1-positive) neurons in the …

The principal sensing of dietary methionine restriction (MR) occurs in the liver, where it activates multiple transcriptional programs that mediate various biological components of the response. Hepatic Fgf21 is a key target and essential endocrine mediator of the metabolic phenotype produced by dietary MR. The transcription factor, Nfe2l2, is also activated by MR and functions in tandem with hepatic Atf4 to transactivate multiple, antioxidative components of the integrated stress response. However, it is unclear whether the transcriptional responses linked to Nfe2l2 activation by dietary MR are essential to the biological efficacy of the diet. Using mice with liver-specific deletion of …
Guilt By Obesity: A Closer Look At Obesity Stigmas, Loriana Goulding
Guilt By Obesity: A Closer Look At Obesity Stigmas, Loriana Goulding
Intuition: The BYU Undergraduate Journal of Psychology
Obesity stigmas have led to an increased interest in the body-positivity movement in recent years. However, despite the decreasing discrimination and shaming of the obese, many people still consciously and unconsciously adhere to obesity stigmas and myths. These false beliefs are harmful not only to obese individuals but also to society, given that over 1/3 of the U.S. adult population is now obese (Centers for Disease Control and Prevention, 2017). These stigmas can be especially harmful to obese individuals who are victims of wrongdoings and to obese individuals who are accused of a wrongdoing (Yamawaki, Riley, Rasmussen, & Cook, 2018). …

Art Therapy With Children And Adolescents Experiencing Obesity: Embracing An Object Relations Approach, Rohan Roberts Goldman
Expressive Therapies Capstone Theses
This thesis looked at how art therapy can help identify problems in attachment between children and adolescents living with obese bodies and their parents. In this literature review, I examined the dynamics of child-parent/caretaker relationships and the way object relations and attachment play a significant role in the child’s mental health development. Incorporating art therapy into the treatment for this population has shown some signs of improving the dynamic between children and their caretaker. Through this research, I concluded that children need a consistent caregiver to be attuned to their needs. If they lack attuned and consistent care, the child …

The study aim was to test hypotheses informed by self-determination theory (SDT) regarding associations of adolescent motivators for weight loss and family feeding practices on understanding adolescent weight management and dietary behaviors. Adolescents (n = 71) with obesity were recruited from a large medical center in the Midwest USA and completed questionnaire assessments via an online survey. Results supported hypotheses that endorsement of health motivators for weight loss, conceptualized as autonomous (intrinsic) motivation, and positive family support would be associated with healthier weight management practices and dietary behaviors. Nuanced findings related to social- and self-esteem-related motivators for weight loss indicated …

Effect Of High Intensity Interval Training Vs Low Intensity Training On Fat Loss In Patients With Obesity, Jonathan Aaron Deck, Adarsh Gupta
Rowan-Virtua Research Day
Nowadays, there are several exercise modalities to lose weight, retain, and or build lean body mass. A widely known training philosophy that has gained increased popularity is high intensity interval training (HIIT). HIIT is based on exercising at submaximal or maximal intensity for a brief period then reducing work to minimal intensity for a longer duration in a series of repeated bouts. The benefit of HIIT is to increase fat loss through the scientific principle of EPOC (excess post exercise oxygen consumption). Low intensity training (LIT) also has its benefits in that it is more sustainable but with the caveat …

Working Together? A Situational Analysis Of Combining Prevention Efforts Targeting Obesity And Eating Disorders In Schools, Alana Ireland, Shelly Russell-Mayhew
The Qualitative Report
The serious consequences and difficulties with treatment of obesity and eating disorders have prompted many to suggest focusing on prevention. Although most often considered distinct conditions with competing needs, some have advocated for an integrated approach to the prevention of a spectrum of weight-related issues including obesity and eating disorders. Despite a strong rationale for focusing prevention on the spectrum of weight-related issues, tensions exist with regard to whether this is feasible or best practice. The current study used situational analysis to explore the tensions associated with the broader situation of preventing weight-related issues in schools. Semi-structed interviews and document …

Leptin Activation Of Metabolically-Related Hypothalamic Neurons, Kailee Hixon
Leptin Activation Of Metabolically-Related Hypothalamic Neurons, Kailee Hixon
Undergraduate Honors Theses
Obesity is often associated with multiple other clinical conditions, including hypertension and cardiovascular disease. Currently, more than one-third of the world’s population is classified as obese. Leptin is a neuropeptide that is released from adipose cells and is responsible for reducing appetite and increasing metabolism. Leptin also has a role in the activation of cardiovascular and metabolic pre-sympathetic neurons and has been reported to increase blood pressure and heart rate. Thus, understanding the activation of the autonomic nervous system by leptin has implications in the development and safety of drugs to avoid activation of cardiovascular pre-sympathetic neurons. This is important …
