Regulation Of Nonclassical Renin-Angiotensin System Receptor Gene Expression In The Adrenal Medulla By Acute And Repeated Immobilization Stress, Regina Nostramo, Lidia Serova, Marcela Laukova, Andrej Tillinger, Chandana Peddu, Esther L. Sabban

NYMC Faculty Publications

The involvement of the nonclassical renin-angiotensin system (RAS) in the adrenomedullary response to stress is unclear. Therefore, we examined basal and immobilization stress (IMO)-triggered changes in gene expression of the classical and nonclassical RAS receptors in the rat adrenal medulla, specifically the angiotensin II type 2 (AT2) and type 4 (AT4) receptors, (pro)renin receptor [(P)RR], and Mas receptor (MasR). All RAS receptors were identified, with AT2 receptor mRNA levels being the most abundant, followed by the (P)RR, AT1A receptor, AT4 receptor, and MasR. Following a single IMO, AT2 and AT4 receptor mRNA levels decreased by 90 and 50%, respectively. Their …

Anaplastic lymphoma kinase 1 (ALK-1) is a member of the insulin receptor tyrosine kinase family. ALK-1 was initially found in anaplastic large cell lymphoma (ALCL). ALK mutations have also been implicated in the pathogenesis of non-small cell lung cancer (NSCLC) and other solid tumors. Multiple small molecule inhibitors with activity against ALK and related oncoproteins are under clinical development. Two of them, crizotinib and ceritinib, have been approved by FDA for treatment of locally advanced and metastatic NSCLC. More agents (alectinib, ASP3026, X396) with improved safety, selectivity, and potency are in the pipeline. Dual inhibitors targeting ALK and EGFRm (AP26113), …

From Ivory To Babel To A New Foundation, Richard Boris Feb 2015

From Ivory To Babel To A New Foundation, Richard Boris

Journal of Collective Bargaining in the Academy

During my 12 years at the NationalCenter for Collective Bargaining in Higher Education and the Professions, I observed with increasing frustration the inability of administration and faculty leaders—union and governance—to fully grasp, analyze, and find pathways out of public higher education’s current existential crisis.

My many years of observing leaders of public higher education lead me to the inescapable conclusion that together the leaders share a culture that shorts strategic planning, thinking, and boldness and instead favors ad-hoc, incremental acceptance of the ever-changing, slimmed-down state of affairs. The rarified bubbles of presidential cabinets and union boards symbiotically promote policies that, …

Access Disparity And Health Inequality Of The Elderly: Unmet Needs And Delayed Healthcare, Tetsuji Yamada, Chia-Ching Chen, Chiyoe Murata, Hiroshi Hirai, Toshiyuki Ojima, Katsunori Kondo, Joseph R. Harris

NYMC Faculty Publications

The purpose of this study is to investigate healthcare access disparity that will cause delayed and unmet healthcare needs for the elderly, and to examine health inequality and healthcare cost burden for the elderly. To produce clear policy applications, this study adapts a modified PRECEDE-PROCEED model for framing theoretical and experimental approaches. Data were collected from a large collection of the Community Tracking Study Household Survey 2003-2004 of the USA. Reliability and construct validity are examined for internal consistency and estimation of disparity and inequality are analyzed by using probit/ols regressions. The results show that predisposing factors (e.g., attitude, beliefs, …

Induction Of Indoleamine 2,3-Dioxygenase By Borrelia Burgdorferi In Human Immune Cells Correlates With Pathogenic Potential, Andrea C. Love, Ira Schwartz, Mary M. Petzke Feb 2015

Induction Of Indoleamine 2,3-Dioxygenase By Borrelia Burgdorferi In Human Immune Cells Correlates With Pathogenic Potential, Andrea C. Love, Ira Schwartz, Mary M. Petzke

NYMC Faculty Publications

Borrelia burgdorferi, the bacterial agent of Lyme disease, induces the production of type I IFNs by human DCs through TLR7 and TLR9 signaling. This type I IFN response occurs in a genotype-dependent manner, with significantly higher levels of IFN-α elicited by B. burgdorferi strains that have a greater capacity for causing disseminated infection. A B. burgdorferi strain that was previously shown to induce IFN-α was found to elicit significantly higher levels of IDO1 protein and its downstream metabolite, kynurenine, compared with a B. burgdorferi mutant that lacks a single linear plasmid (lp36); this mutant is unable to induce IFN-α and …

Long-Term Assessment Of Fatigue In Patients With Culture-Confirmed Lyme Disease, Gary P. Wormser, Erica Weitzner, Donna Mckenna, Robert B. Nadelman, Carol Scavarda, John Nowakowski

NYMC Faculty Publications

BACKGROUND: Fatigue is a common symptom with numerous causes. Severe fatigue is thought to be an important manifestation of post-treatment Lyme disease syndrome. The frequency with which severe fatigue occurs as a long-term sequela in prospectively followed patients with Lyme disease is unknown. METHODS: Patients with culture-confirmed Lyme disease who originally presented with erythema migrans have been evaluated annually in a prospective study to determine their long-term outcome. In 2011-2013, subjects were evaluated for fatigue using an 11-item Fatigue Severity Scale (FSS-11) that has been used in studies of post-treatment Lyme disease syndrome. An FSS-11 score of ≥4.0 is indicative …

Microrna-146a And Microrna-146b Regulate Human Dendritic Cell Apoptosis And Cytokine Production By Targeting Traf6 And Irak1 Proteins, Haein Park, Xin Huang, Changming Lu, Mitchell S. Cairo, Xianzheng Zhou

NYMC Faculty Publications

We have previously reported 27 differentially expressed microRNAs (miRNAs) during human monocyte differentiation into immature dendritic cells (imDCs) and mature DCs (mDCs). However, their roles in DC differentiation and function remain largely elusive. Here, we report that microRNA (miR)-146a and miR-146b modulate DC apoptosis and cytokine production. Expression of miR-146a and miR-146b was significantly increased upon monocyte differentiation into imDCs and mDCs. Silencing of miR-146a and/or miR-146b in imDCs and mDCs significantly prevented DC apoptosis, whereas overexpressing miR-146a and/or miR-146b increased DC apoptosis. miR-146a and miR-146b expression in imDCs and mDCs was inversely correlated with TRAF6 and IRAK1 expression. Furthermore, …

Role Of Homocysteinylation Of Ace In Endothelial Dysfunction Of Arteries, An Huang, John T. Pinto, Ghezal Froogh, Sharath Kandhi, Jun Qin, Michael S. Wolin, Thomas H. Hintze, Dong Sun

NYMC Faculty Publications

The direct impact of de novo synthesis of homocysteine (Hcy) and its reactive metabolites, Hcy-S-S-Hcy and Hcy thiolactone (HCTL), on vascular function has not been fully elucidated. We hypothesized that Hcy synthesized within endothelial cells affects activity of angiotensin-converting enzyme (ACE) by direct homocysteinylation of its amino- and/or sulfhydryl moieties. This covalent modification enhances ACE reactivity toward angiotensin II (ANG II)-NADPH oxidase-superoxide-dependent endothelial dysfunction. Mesenteric and coronary arteries isolated from normal rats were incubated for 3 days with or without exogenous methionine (Met, 0.1-0.3 mM), a precursor to Hcy. Incubation of arteries in Met-free media resulted in time-dependent decreases in …

Faculty Development For Service-Learning: One Size Does Not Fit All, Lynn E. Pelco, Catherine Howard Jan 2015

Faculty Development For Service-Learning: One Size Does Not Fit All, Lynn E. Pelco, Catherine Howard

Division of Community Engagement Resources

This conference presentation describes the development of the professional development continuum currently being used at Virginia Commonwealth University to build the faculty’s capacity for teaching high-quality service-learning courses. Our professional development continuum was developed over the past 5 years by the staff of the VCU Service-Learning Office and was informed by program evaluation data, the principles of adult learning theory, and an “exposure-experience-expertise” developmental framework. The continuum targets instructors at varying levels of service-learning knowledge and experience so that faculty instructors with more service-learning experience participate in different professional development opportunities than do instructors who are new to the pedagogy.

Ranolazine was developed as an antianginal agent and was approved by the Food and Drug Administration in 2006 for use in chronic stable angina pectoris. Experimental and clinical studies have shown that it also has antiarrhythmic properties based on the frequency-dependent blockade of peak sodium channel current (peak INa) and rapidly activating delayed rectifier potassium current (IKr) in the atria and blockade of late phase of the inward sodium current (late INa) in the ventricles. Recent clinical studies have revealed the efficacy of ranolazine in prevention of atrial fibrillation in patients with acute coronary syndromes, prevention as well as conversion …

Heme Oxygenase-2 Deletion Impairs Macrophage Function: Implication In Wound Healing, Lars Bellner, Giuseppina Marrazzo, Nico Van Rooijen, Michael W. Dunn, Nader G. Abraham, Michal L. Schwartzman

NYMC Faculty Publications

Heme oxygenase (HO)-2 deficiency impairs wound healing and exacerbates inflammation following injury. We examine the impact of HO-2 deficiency on macrophage function and the contribution of macrophage HO-2 to inflammatory and repair responses to injury. Corneal epithelial debridement was performed in control and macrophage-depleted HO-2(-/-) and wild-type (WT) mice and in bone marrow chimeras. Peritoneal macrophages were collected for determination of phagocytic activity and classically activated macrophage (M1)-alternatively activated macrophage (M2) polarization. Depletion of macrophages delayed corneal healing (13.2%) and increased neutrophil infiltration (54.1%) by day 4 in WT mice, whereas in HO-2(-/-) mice, it did not worsen the already …

Assessing Values-Based Sourcing Strategies In Regional Food Supply Networks: An Agent-Based Approach, Caroline C. Krejci, Michael C. Dorneich, Richard T. Stone

Industrial, Manufacturing, and Systems Engineering Faculty Publications - Archive

The recent increase in demand for regionally produced food has resulted in a need for more efficient distribution methods. To connect regional food producers and consumers, intermediated regional food supply networks have developed. The intermediary, known as a regional food hub, serves as an aggregation point for products and information. It may also act as a filter to ensure that the requirements of both producers and consumers are consistently met. This paper describes an empirically based agent-based model of a regional food network in central Iowa that is intermediated by a food hub. The model was used to test a …

Towards Understanding When Service-Learning Fosters Efficacy Beliefs Of Preservice Teachers, Kathleen Tice, Larry Nelson

Curriculum and Instruction Faculty Publications - Archive

This research uses a mixed methods approach in focusing upon the role service-learning can play in enhancing preservice teachers’ sense of efficacy. Results of the quantitative component reveal significant gains in efficacy beliefs of preservice teachers who engaged in a service-learning project during a course in their teacher preparation. At the completion of a service-learning project, preservice teachers shared their perceptions of their participation through written reflections. Qualitative analysis of the preservice teachers’ written reflections and the characteristics of the service-learning experience uncover sources of efficacy, providing guidance for planning service-learning in teacher education. Additionally, findings document the importance of …
