The cloning of DNA fragments derived from the (beta)-like globin gene cluster of normal and (beta)('+)-thalassemic individuals is described. These and other cloned human DNA fragments are employed to define the strucure and transcription of a family of interspersed, repetitive RNA polymerase III transcription units in human DNA. These data allow unequivocal identification of these transcription units with the Alu family of interspersed, repetitive DNA sequences. Comparison of these sequences to those of other primate and rodent interspersed repeats reveals that the primate sequence resembles in many respects a dimer of the rodent sequence. Twenty-eight species of Euodynerus Dalla Torre are recognized from the Nearctic Region, of which one is Holarctic. Literature on the biology of fourteen species is reviewed and new biological information discussed for four species. The phylogenetic relationships of the genus Euodynerus to other genera and the classification and relationships of the species of Euodynerus are discussed. Seven subgenera are recognized of which four are new: Euodynerus, Pareuodynerus Bluethgen, Leucodynerus Bohart, Annodynerus new subgenus, Gonodynerus n. subgenus, Holodynerus n. subgenus and Plecodynerus n. subgenus. Previous studies indicated that a permeability barrier between blood and the uterine lumen might exist in rats. The abilities of selected radiolabelled test substances to enter the uterine lumen were assessed by comparing radioactivity concentrations in fluid (secreted or instilled) recovered from the uterine lumen with those in serum, at fixed times after single intravenous injections of a test substance.;Initial experiments conducted in immature, ovariectomised, estradiol-treated rats revealed that a barrier to {lcub}('3)H{rcub}-sucrose, {lcub}('3)H{rcub}-inulin, and {lcub}('125)I{rcub}-bovine serum albumin exists between blood and the uterine lumen which impeded the entry of these substances into the uterine lumen. A multivariate analysis was made of geographic variation in the cones of Pinus contorta as related to environmental factors. Variation between four previously defined subspecies was compared to variation in geographic location, elevation, topographic situation, climate and soil. The allocation of measurement effort was optimized through preliminary studies which objectively selected a set of characters for study, and determined the number of cones to measure to represent each site.;After choosing a resemblance function with desirable properties, data structure was investigated using three-dimensional stereograms of principal components, accompanied by a demonstration that almost no distortion resulted from this procedure. This has made progress toward an understanding of the initial events in this disease difficult and so a number of animal models which mimic various aspects of the disease are being studied widely. Asymptotic series expansions for the bias and mean square error of the logit estimator are developed. In addition, an estimator of the bias and asymptotic series for the expectation of this estimator and the estimator of the mean square error are derived. These formulations have been expressed in such a way that general coefficients can be calculated and then implemented for any choice of the parameters involved in the logit estimation problem. The formulas developed are applied, providing similar asymptotic series, for the estimation of the log odds ratio.;A general method for the linear combination of estimators is proposed. This …

Nephrocalcinosis has previously been detected in rats fed diets containing alkali-treated soyprotein. This is of some interest since this type of protein is used widely by the food industry and may be harmful to man.;Female weanling rats fed a diet containing 20% alpha protein (an alkali-treated soyprotein) in this study developed nephrocalcinosis of moderate severity after 9 weeks of feeding. It had previously been suggested that the inorganic phosphate content of the diet, rather than the protein, was responsible for renal calcification. Spontaneous mammary tumors in GR mice contain, in addition to the endogenous MMTV copies, acquired proviral sequences, which can be detected and characterized by the Southern transfer procedure. Hormone responsive and independent cell populations were selected from spontaneous GR tumors by transplanting tumor cell suspensions into pairs of hormone treated and untreated syngeneic males. Analysis of the acquired MMTV DNA copies revealed differences between hormone responsive and independent cell populations derived from the same tumor indicating that the original tumors were comprised of multiple cell populations. Poxviruses are the most complex eukaryotic viral agents when considered in terms of size, morphology, and replicative strategy. Much of the life cycle of the prototypic member of this family, vaccinia virus, has been studied using metabolic inhibitors and, more recently, conditional-lethal temperature-sensitive (ts) mutants. In this study, biochemical, electron microscopic, and genetic analysis utilizing these tools has lead to a greater understanding of the roles, first, of post-translational cleavage (PTC) and envelope assembly and, secondly, of host DNA-dependent RNA polymerase II (polymerase II) and ongoing host gene expression in vaccinia virus biogenesis.

A descriptive study of the cell envelope of Deinococcus radiodurans strain Sark and an exploration of the major components of the cell envelope was undertaken. The cell envelope consisted of six layers as seen in thin sections: a plasma membrane, 8.0nm thick; a peptidoglycan layer, 30nm thick; a compartmentalized layer, 20 - 75nm thick; an outer membrane, 6.5nm thick; an electron transparent zone, 10nm thick, and a hexagonal surface array, 2 - 3nm thick.;The outer cell envelope layers formed blebs throughout the growth cycle, which were shed as large vesicles (0.5(mu) to 3.5(mu) in diameter) from 5% of the cell …

The productivity of a new bush winter squash cultivar ('Autumn Pride') was compared to that of 'Blue Hubbard', a high-yielding vine cultivar. Yields of 'Autumn Pride' increased with increasing plant density and, at high plant densities, were greater than yields of 'Blue Hubbard' at its near optimum plant density.

Several factors could have contributed to the higher yield potential of 'Autumn Pride'. CO(,2) compensation points were determined to be 38 ppm and 77 ppm CO(,2) for 'Autumn Pride' and 'Blue Hubbard', respectively, at 30(DEGREES)C. A study was undertaken to determine if natural growth irradiance regimes influence the pigment content and/or photosynthesis-irradiance relationships of the tropical brown seaweed, Lobophora variegata. Pigment analyses were performed on samples collected from a series of depths between 9 and 49m. Chlorophyll a, chlorophyll c, and fucoxanthin were constant on a thallus weight basis, but showed small decreases when expressed in terms of thallus area. The beta-carotene content of L. variegata was much greater in shallow than in deep-water samples regardless of the pigment units employed. The degree to which seed crops in seed orchards reflect the genetic variability and superiority present among orchard parents (genetic efficiency) was examined in two loblolly pine seed orchards using seven allozyme loci as genetic markers.

The frequency of outcrossed progeny in the seed crop was estimated using unique markers, a single locus mixed mating model, and a multilocus model. These estimates were 0.982, 0.992, and 0.994, respectively. Thus, selfing did not contribute significantly to deviations from genetic efficiency in these orchards.

Pollen contamination, on the other hand, was found to have a significant impact on genetic efficiency. Unique markers …

Kinetic studies of the hydrogenation of carbon monoxide to methane and methanol were carried out over iron-titanium alloy catalysts, in a continuous flow fixed bed differential reactor. The catalysts studied included FeTi, Fe(,1.5)Ti and Fe(,2.5)Ti alloys with pure Fe used as a comparison. The catalysts used were all unsupported with an averaged initial particle size of 270 microns. The range of feed compositions was varied from 10 to 33% CO with the remainder being H(,2). Temperatures were varied from 520 to 590 K at a total pressure of 34 atm for all the catalysts studied. The ultrastructure of the lytic cycle of phage 41c in Bacillus subtilis 168 was examined by transmission electron microscopy. Thin sections of phage 41c-infected cells revealed that, prior to the appearance of phage structures, a marked rearrangement of the tightly packed, vesicular mesosomes was usually seen within the host cells. These mesosomes opened up into loosely arranged, lamellar forms. As phage accumulated, they appeared to be preferentially associated with the mesosomes. Towards the later portion of the lytic cycle, the mesosomes took on distinctive, loosely organized, spiral configurations. These ultrastructural changes occurred even in the presence of chloramphenicol, indicating that …

In a continuous fermentor with 100% recycle, growth of Escherichia coli and Bacillus polymyxa occurs in three sharply distinct phases: an initial phase of exponential growth that terminates when the glucose level in the fermentor falls below transport saturation levels; a second phase of linear growth whose rate is dependent on the rate of glucose provision to the fermentor, but whose length if fixed and independent of growth rate; and a third phase of slower glucose dependent linear growth. The specific growth rate falls in phase 2 and 3 while the mass doubling time lengthen, eventually reaching hundreds of hours. …

The MHC of the chicken (B complex) has been shown to exert a decisive influence on the fate of Rous sarcoma virus (RSV)-induced tumors. However, noninbred chickens with identical B genotypes and inbred lines indistinguishable at the MHC have demonstrated considerable variation in response to RSV challenge. The major objective of this research was to determine the cause of variation in anti-Rous sarcoma response observed among chickens with "identical" B genotypes.

Hypotrichs represent a highly diverse species ensemble displaying a wide range of form and function that reflects extensive radiation in a wide variety of habitats. Because of their constancy through time, particular habitats such as marine sands harbor forms that may have persisted with little change for hundreds of millions of years and, hence, may hold clues to help unravel mysteries of ciliate evolution. Growth rates, reproductive phenology, and longevity of Fucus distichus ssp. edentatus and F. distichus ssp. evanescens were evaluated in natural populations in New England. Both subspecies exhibited maximum growth during early summer, a slight decline in late summer, and a brief resurgence in the early fall. The maximum growth rates (in terms of elongation) for ssp. edentatus and ssp. evanescens was 3.5 cm/mo and 3.7 cm/mo, respectively. Populations of both subspecies showed a distinct bimodal reproductive periodicity, with maximums in the spring and fall. Individuals within natural populations were reproductive in either the spring or fall but never both seasons. …

Although membrane potentials have been measured in algal cells and in large cells of some higher plants, this investigation is the first to include such measurements in mesophyll cells of elm (Ulmus americana) leaves. The inquiry entailed the development of the technique for measuring elm leaf cell membrane potentials, the determination of membrane potentials expected of healthy, greenhouse elms, and the effects of various physical and chemical factors on the membrane potentials of elm leaf cells.

Genetic study on sweet potato resistance to the soil rot disease, Streptomyces ipomoea and other characters was conducted from 1979 through 1981. In control crosses, L(,3)-64 was incompatible with L(,4)-62 and L(,4)-112 with L(,4)-131. The average pod set for all progenies was 16.95%. Additive genetic effects were more important than non-additive in the inheritance of resistance. Resistant families were only produced from crosses between resistant parents and most progenies from crosses of moderatley resistant x resistant, resistant x susceptible and moderately resistant x susceptible parents produced moderately resistant segregates. Polycross progenies from resistant parents, L(,4)-131 and L(,6)-135, had families with …
