Crawfish processing by-products (CPBs) were hydrolyzed using protease to evaluate the potential for bioflavor production. Ten commercial proteases (neutral and alkaline) were tested at both 37$\sp\circ$C and their optimum temperatures using CPBs as substrate. Of these, APL-440 was selected for enzymatic hydrolysis of CPBs on the basis of activity per cost. Response surface methodology (RSM) was used to optimize the hydrolysis of CPBs using APL-440. Optimum hydrolysis conditions for enzymatic hydrolysis of CPBs with APL-440 were determined to be pH 8-9, 65$\sp\circ$C, 2.5 hr reaction time, 75% substrate concentration, and 0.3% APL-440. Blue crab (Callinectes sapidus) meat and its processing by-product (CPB) were characterized by instrumental, olfactometric, and sensory techniques to determine the usefulness of CPB as a feed stock for flavor recovery. Instrumental analysis with gas chromatography/mass spectrometry (GC/MS) and gas chromatography/Fourier transformed infrared spectroscopy (GC/FTIR) allowed for the identification of 77 and 80 compounds in claw meat and CPB, respectively. Fifty-five compounds were found common to both samples, suggesting that CPB could be a good source for flavor recovery. Quantitatively, trimethylamine (TMA), alkanes (C15-C17, C19), and indole were high ($>$50ng/g) in claw meat while TMA, carbon disulfide, dimethyltrisulfide, alkanes …

Race 1 isolates collected from tomato roots were used to develop an orange mutant of Fusarium oxysporum f. sp. lycopersici (FOL) for use in competitive infection tests against nonpathogenic F. oxysporum isolates. Eighty one color mutants were induced by exposure to ultraviolet light. The mutant ORA-22 retained pathogenicity and growth habit similar to the wild-type. Preliminary competitive infection tests indicated differences within the nonpathogenic population in the ability to suppress or enhance infection by FOL orange mutant ORA-22. Four vegetative compatibility groups (VCGs) were tested to determine if competitive infection ability was associated with VCG designation. Results from these tests …

This study was conducted to: (1) determine the effects of stocking density and dietary protein:energy ratio on survival, growth, feed consumption, feed conversion, lipo-somatic index, dress-out percentage, and productive protein value of common snapping turtles; and (2) determine the fatty acid composition of muscle, liver, and fat bodies of wild common snapping turtles and common snapping turtles fed a diet of known fatty acid composition. Hatchling common snapping turtles, Chelydra serpentina, were stocked at five or 10 animals per container (29 and 58 turtles/m$\sp2$, respectively), and fed one of seven prepared diets. In anticipation of the regional demand for tomatillo (Physalis ixocarpa Brot.) for the fresh market and sauce industry, four years of research trials (1990-1993) showed a significant adaptation of tomatillo to Louisiana planting conditions where, like tomato, it performed best in the cooler temperatures of spring and early fall. Field surveys indicated that virus diseases were major constraints on production. A foliar mosaic and yellow mottle found commonly affecting plants was caused by Physalis mosaic virus (PhyMV), identified by host reaction, electron microscopy, serology, and dsRNA analysis. Cucumber mosaic virus (CMV) and tomato spotted wilt virus (TSWV) were also found. …

In most of the United States, overwintering redclaw crayfish (Cherax quadricarinatus) is necessary because they cannot withstand winter temperatures. Consequently, hatcheries are required to produce juveniles for pond stocking. The objectives of this study were: (1) identify biological factors affecting production of juvenile redclaw, (2) determine production capabilities of a redclaw crayfish hatchery, and (3) determine the economic potential of redclaw hatcheries. Experiments were designed to assess fecundity and factors influencing spawning such as stocking density, male/female weight, diet, mortality, and cannibalism. The objective of the present study was to evaluate the effect of post-block mouse and bovine embryos on the development of pre-block embryos around the in vitro developmental block stage. The effect of the post-block embryos on the development of the pre-block embryos was evaluated in culture, in chimeras and in cytoplasmic hybrid embryos. The results indicate that post-block embryos were able to stimulate the development of the pre-block embryos in culture. The generation time of L. monocytogenes was calculated to be 72.2, 28.5, 17.01, and 6.9 h on crawfish tail meat held at 0, 4, 6, and 12$\sp\circ$C, respectively. Potassium sorbate (PS) (0.3%) spray treatments on crawfish held at 4$\sp\circ$C, extended the lag phase of the bacterium. However, once exponential growth began, a generation time not significantly different (P $>$ 0.05) than that of untreated crawfish was observed. Samples sprayed with 0.3% citric acid (CA) and held at 4$\sp\circ$C supported growth of the bacterium equally well as that of untreated samples. The agriculturally important soils of the lower Mississippi alluvial plain typically display surprisingly high levels of nitrogen (N) fertility given the low amounts of organic matter and readily available forms of N characteristically present in these soils. The principal goal of this research was to investigate N dynamics in these soils with special attention devoted to the possible role of clay-fixed NH$\sb4\sp+$ in N fertility. White flesh sweetpotatoes (Ipomoea batatas (L.) Lam) are a major food crop in developing countries. This study was to evaluate the protein and carotenoid content and quality of various white fleshed African sweetpotatoes and Apios americana tubers for use as a weaning food. The sweetpotato and Apios are not normally considered sources of carotenoids and proteins, but the wide use of sweetpotatoes as a source of carbohydrate and the renewed interest in Apios warrant the study of carotenoids and proteins in these crops. Among food-borne pathogens, Listeria monocytogenes is one of the more resistant to food processing methods, including irradiation. USFDA approval for irradiation of processed meat and seafood products likely to contain Listeria spp. is $<$3 kGy gamma radiation. The incidence of L. monocytogenes in food products varies between 5-30% and at levels $<10\sp4$ CFU/g. With the capacity of this bacterium to multiply at refrigeration temperatures and the zero tolerance level mandated by the USFDA it becomes imperative to maximize the efficiency of the irradiation process. The first purpose of this study was to determine if initial cell concentration and/or temperature at time of irradiation influences the radiosensitivity of L. monocytogenes. Secondly, to determine whether split dose application would effect the radiation response of this bacterium. Concentrations of 10$\sp3,$ 10$\sp6$ and 10$\sp9$ cfu/ml of L. monocytogenes Scott A, were suspended in tryptic soy broth and exposed to 0-5 kGy gamma (1.25MeV) radiation at 20, 4, and -80$\sp\circ{\rm C}.$ Split doses were applied in equal portions to initial cell concentrations of 10$\sp6$ and 10$\sp9$ at the same temperatures. Surviving cells under the various conditions were enumerated and irradiation D-values were calculated from linear regression curves. At all temperatures, cell concentrations of 10$\sp3{\rm CFU/ml}$ were reduced to non recoverable levels with 2 kGy gamma radiation. The irradiation D-value of 0.43 kGy for frozen (-80$\sp\circ{\rm C})$ cultures of 10$\sp6$ CFU/ml was significantly lower (p $<$ 0.05) than the D-values (0.58 and 0.62 kGy) for the same cell concentration at the other two temperatures (20$\sp\circ$ and 4$\sp\circ{\rm C},$ respectively). For higher cell concentration of 10$\sp9$ cfu/ml, a D-value of 0.42 kGy was obtained for both 4$\sp\circ{\rm C}$ and -80$\sp\circ{\rm C}$ these were significantly lower (p $<$ 0.05) than 0.50 kGy for 20$\sp\circ{\rm C}$ suspensions. With both cell concentrations, the lowest D-values were obtained when cells were irradiated in the frozen state indicating a that primary cell damage was due to direct effects of irradiation. Split doses of 1 and 2 hours time between fractions approached the generation times for this bacterium and were significantly reduced at 20$\sp\circ{\rm C}.$ However, there was no consistent trend in reductions at 4$\sp\circ$ or -80$\sp\circ{\rm C}.$ Split dose application of irradiation to food pathogens should be further explored.

Impedance technology is currently used as a measurement of microbial activity. The purpose of the current study was to determine the feasibility of utilizing impedance technology to evaluate the quality of white shrimp (marine species) and channel catfish (freshwater species). Impedance analysis measurements (I-values) of raw shrimp increased for samples stored at 5, 25, and 35$\sp\circ$C; however, control samples stored at $-$20$\sp\circ$C displayed no significant changes in I-values. I-values from thermally processed shrimp had no direct relationship with odor scores, increased storage time, or increased storage temperature. Mya arenaria (L.) is susceptible to the disease, hematopoietic neoplasia (Hn), in which atypical cells invade the blood and connective tissue of the clam's organs. In this study impacts of Hn on the reproductive and excretory systems were examined in clams collected from New Bedford Harbor and Little Buttermilk Bay, Buzzards Bay, MA.
The relationships between Hn seasonality and reproduction were evaluated from fresh blood samples and histological analysis of gonads. Both clam populations had biannual reproductive cycles with spawnings in the spring and fall. Hn prevalence exhibited some seasonality with maxima in the fall and late winter-early spring. Initially, various solvents were tested for their efficacy in isolating soluble nitrogen (SN) from annual ryegrass and berseem clover. Plant nitrogen (PN) solubilities were similar in.5 M NaCl, autoclaved rumen fluid, borate phosphate buffer and Burroughs' solution. The mean PN solubilities across forages ranged from 12% for.1 M acetic acid to 45% for.2% NaOH. At 4$\sp\circ$C, 15% TCA precipitated the most soluble plant protein nitrogen (SPPN). The objective of this study was to evaluate the antimicrobial properties of glycerol monolaurate (monolaurin) either alone or in combination with organic acids against Listeria monocytogenes in model broth or food. The minimal inhibitory concentration (MIC) of monolaurin was reduced by decreasing the pH value of the medium. The contribution of temperature to monolaurin effectiveness showed that lethal effects of monolaurin increased at higher temperatures and lower pH values, whereas, bacteriostatic effects on growth increased as temperature and pH decreased. Annual ryegrass (Lolium multiflorum Lam.) is frequently grown on acid soils with potentially toxic levels of aluminum (Al). Interaction effects of nitrogen (N) form and Al on growth and nutrient relationships in ryegrass require further definition for environmental, biological, and economic reasons. Nutrient solution and soil experiments were conducted in growth chambers to determine interactive effects of ammonium: nitrate (NH$\sb4$:NO$\sb3$) ratios and Al on growth, N absorption, nutrient composition, cation influx rate, and N use efficiency. Cattle lice may be the most underestimated livestock insect in terms of economic losses. The USDA estimates that U.S. livestock producers lose $125 million a year to cattle lice.
Heavy louse populations cause lowered milk production, loss of flesh, stunted growth, general unthriftiness and anemia. "Chronic" or "carrier" cows may abort due to louse-induced anemia. During severe winters, louse-infested animals are more susceptible to respiratory diseases.

A thematic unit for integrating social studies and science with other curriculum areas was developed for first grade students. The theme for the unit centered around farms. The unit was divided into five subunits: farms, farm animals, orchards/vineyards (to include fruits of the Yakima Valley and apples), crops (to include vegetables of the Yakima Valley, pumpkins, and wheat), and nutrition/four food groups. The literature was reviewed to establish a theoretical foundation for the unit and for activities that focused on reading, writing, and language development.

Four experiments were conducted to investigate the effects of sodium zeolite A (SZA) on calcium (Ca), phosphorus (P), and zinc (Zn) utilization in broiler chickens from 5 to 14 or 15 days of age. In Experiment 1, two levels each of SZA (0 and.75%) and (or) Ca (1.0 and 1.5%) were fed to both uninfected and Eimeria acervulina-infected chicks. Excess dietary Ca reduced (P $$.10) in infected chicks (Ca x coccidiosis, P $<$.05). Addition of SZA to diets with excess Ca further decreased (P $<$.05) gain and tibia ash. These results suggest that SZA may exacerbate the adverse effects of excess dietary Ca. In Experiment 2, four levels of dietary Ca (.6,.8, 1.0, and 1.2%) and (or) two levels of SZA (0 and.75%) were fed. Sodium zeolite A decreased (P $<$.05) plasma P and tibia magnesium (Mg) but increased (P $<$.05) tibia Ca, Zn, aluminum (Al), and manganese (Mn). Tibia ash and shearing force were increased in chicks fed SZA and inadequate dietary Ca, but were decreased in chicks fed SZA and 1.2% Ca (Ca x SZA, P $<$.05). Dietary SZA enhanced tibia ash, density, and shearing force when dietary Ca was low; however, SZA reduced many bone mineralization indices in chicks fed excess Ca. In Experiment 3, various levels of dietary P (.41,.55, and.69%), Ca (.6 and 1%), and (or) SZA (0 and.75%) were fed. The addition of Ca, SZA, or both exacerbated the adverse effects of feeding low-P diets, yet alleviated the adverse effects of feeding low-Ca, high-P diets. Dietary SZA reduced (P $<$.01) plasma P and increased (P $<$.02) tibia Mn, Zn, copper (Cu), and Al. The SZA-induced increase in tibia Al was most evident in chicks fed low levels of P (SZA x P, P $<$.02). These data demonstrate that the effects of SZA are influenced by dietary concentrations of Ca and P, and that the addition of SZA to diets low in P results in bone Al accumulation. In Experiment 4, the effects of SZA on chicks fed inadequate (35 ppm), marginal (40 ppm), adequate (85 ppm), and excess (4,000 ppm) levels of Zn were assessed. Sodium zeolite A increased (P $<$.05) tibia and pancreas Zn regardless of dietary Zn concentration. Dietary SZA tended to alleviate the adverse effects of feeding inadequate Zn and exacerbate the adverse effects of feeding excess Zn.

Size exclusion (SE) and hydrophobic interaction (HIC) high performance liquid chromatography (HPLC) methods were developed for determining hemoglobin, myoglobin, and total pigments in beef and chicken. Hemoglobin and myoglobin in beef, but not in chicken, were separated by HIC with minimum detectable levels of 0.187 and 0.085 $\mu$g/$\mu$l, respectively. Determination of total pigment by SE-HPLC (minimum detection of 0.001 $\mu$g/$\mu$l) was more sensitive than with spectrophotometric assay. Raw milk samples were analyzed to determine the bacteriological quality by the standard plate count (SPC), adenosine-5$\sp\prime$-triphosphate (ATP) assay, preliminary incubation counts, and the psychrotrophic bacteria count (PBC). In a comparison of SPC and ATP content, an overall correlation coefficient of 0.97 was found between the two methods. The correlations between ATP content and SPC of fresh raw milk samples at zero hour and after preliminary incubating these samples at 12.8 and 15.6$\sp\circ$C for 18 hours were.96,.97, and.97, respectively. The regression coefficients for all types of milk samples were highly significant ($P <$.01). High correlation coefficients demonstrated that the ATP measurement in combination with preliminary incubation practices could be a rapid technique for the determination of bacteriological quality of milk at receiving stations and milk processing plants. Median SPC for 175 fresh raw milk samples was 36,000/ml. The bioluminescence technique is reported previously to have a good correlation with SPC in the range of 1.0 $\times$ 10$\sp5$ to 1.0 $\times$ 10$\sp{10}$ bacteria/ml. The commingled milk coming at the receiving stations sometimes have a bacterial count that is less than 1.0 $\times$ 10$\sp{5}$ bacteria/ml. A correlation coefficient of.87 was calculated between the SPC and ATP values for the overall data while lowering the SPC range from 1.0 $\times$ 10$\sp5$ to 1.0 $\times$ 10$\sp4$ bacteria/ml. This range is more practical as a quick screening of raw milk at dairy processing facilities since 81 percent of all raw milk samples fell in this range. An equation (SPC = $-$1642213 + 6751(ATP)) was developed from the combined experimental data which could estimate the SPC of raw milk samples using their ATP content. Another phase of the experiment was to ascertain if the Petrifilm$\sp{\rm TM}$ technique could be effectively used in place of agar method to estimate PBC. The correlation coefficient between the Petrifilm$\sp{\rm TM}$ and the agar plate method for both the fresh and preliminary incubated raw milk samples was.99. A high linearity of relationship shows that one method can be substituted with the other. However, the Petrifilm$\sp{\rm TM}$ technique took longer than the usual time of 25 h to develop red colonies at 21$\sp\circ$C because of the slow reduction of the dye reagent triphenyl tetrazolium chloride (TTC) at this temperature.

Studies dealing with the availability and speciation of arsenic (As) as affected by soil redox potential and pH were initiated because of the lack of information on As chemistry in flooded soils. The chemistry of native and applied As was studied in a Crowley silt loam soil (Typic Albaqualf). Arsenic uptake and its toxic effect on two rice cultivars as affected by As chemical form and concentration were also studied. Mallards (Anas platyrhynchos; n = 339) and Blue-winged Teal (A. discors; n = 213; hereafter teal) were collected from western (1986) and eastern (1987) Saskatchewan grassland and parkland habitats during egg formation to examine body composition, diets and digestive organ size with respect to hypotheses concerning habitat (this study) and the use of body fat (Drobney 1980; Rohwer 1986a; Ankney and Afton 1988). From July to August of 1987 and 1988, a regional sampling of spruce-fir forests across New England was conducted. Forest floors and foliage were sampled from 161 sites, in 11 areas from Maine to New York. Potential net N mineralization, nitrification, pH and analysis of total concentration for selected elements were measured on forest floor samples. Foliar lignin, N, cellulose and elemental concentration were determined for red spruce and balsam fir. Calyptospora funduli is a pathogenic coccidium that infects the liver and pancreas of estuarine killifishes belonging to the genus Fundulus. Experimental infection of the atypical fish host Rivulus marmoratus with this intracellular protozoan parasite caused the development of hepatic macrophage aggregates (MA's) and granulomas. The Rivulus/Calyptospora host-parasite system thus represents a new laboratory model for investigations of MA biology.
