Immobilization Of P. Digitatum And Bioconversion Of Limonene To Alpha-Terpineol., Qiang Tan Jan 1996

Immobilization Of P. Digitatum And Bioconversion Of Limonene To Alpha-Terpineol., Qiang Tan

LSU Historical Dissertations and Theses

The parameters of bioconversion of limonene to $\alpha$-terpineol by Penicillium digitatum were determined. This bioconversion was carried out stereo-specifically and selectively by this fungus. (R)-(+)-limonene was selectively converted to (R)-(+)-$\alpha$-terpineol by mycelia of this fungus during early growth phase. Immobilization of the fungal mycelia for use in continuous production of this product was possible. Aeration was critical to this bioconversion. The optimum aeration was about 1.6 mg/L for growing cells and immobilized cells. Substrate inhibition of the bioconversion was not observed when the substrate concentration was below 4% (v/v). End product inhibition was observed. The $\rm Ki\sb{app}$ was 12.08 mM. …

Reduction And Management Of Vibrio Vulnificus In Gulf Coast Oyster (Crassostrea Virginica)., Yu-Pin Chen Jan 1996

Reduction And Management Of Vibrio Vulnificus In Gulf Coast Oyster (Crassostrea Virginica)., Yu-Pin Chen

LSU Historical Dissertations and Theses

The AmeriPure Process (AmeriPure Oyster Company, New Orleans, Louisiana), which is a mild heat treatment for a specific time period, proved effective in eliminating Vibrio vulnificus from the Gulf Coast oysters (Crassostrea virginica), thus reducing the risk of possible V. vulnificus infections. The treatment was effective in the reduction of V. vulnificus in both $1.5\times10\sp5$ MPN/g (high artificially contaminated level) and $2.4\times10\sp3$ MPN/g (low artificially contaminated level) to undetectable level (MPN $<$ 3/g). The mild heat treatment also reduced naturally occurring V. vulnificus $(8.9\pm3.7\times10\sp4$ MPN/g) present in shellstock oysters harvested in warmer months to undetectable levels. The D value for V. vulnificus present in shellstock oyster averaged 45.6 $\pm$ 3.3 seconds at the AmeriPure Process temperature. Mild heat treatment can also control aerobic plate count numbers during refrigerated storage (0-2$\sp\circ$C) and extend the shelf-life of raw oysters for at least 7 days over unprocessed samples. Mild heat treated oysters need to be refrigerated (${<}4\sp\circ$C) to ensure the safety and quality, as well as shelf-life. Oysters treated according to the AmeriPure Process when exposed to an average room temperature of 27$\sp\circ$C were found unacceptable after 24 hours due to the high number of aerobic plate count (10$\sp7$ CFU/g) and were considered not safe to eat. Although there was a slight lightening in color, sensory panels, composed of both traditional raw oyster consumers and infrequent consumers, found the treated product to be comparable in flavor, texture and smell to untreated oysters. According to the result of sensory evaluation, the good oyster flavor of treated samples increased and the off-flavor and off-odor of the treated oysters were significantly lower than the control samples after 7 days of storage on ice. Possible temperature variations, which could occur during the transportation and storage were studied in a temperature abuse study. The result showed that no Vibrio vulnificus was recovered and no significant microbiological quality change was observed after 10 hours of exposure to the room temperature of $22\pm2\sp\circ$C.

Chemical Effects On Microbial And Sensory Qualities Of Catfish In Planktonic And Dynamic Systems., Michael Adrain Land

LSU Historical Dissertations and Theses

Listeria monocytogenes has the potential to be a serious pathogen for the compromised health segment of our population (immunocompromised, young, elderly and the pregnant). While the infectious dose is generally believed to be high 1.0 $\times$ 10$\sp6$, infectious dosages of 1.0 $\times$ 10$\sp3$ have been reported. Listeria monocytogenes poses a threat because of several factors: ubiquity in nature, isolation from fresh/salt waters, soil, ruminates and 17% of the human population. Listeria monocytogenes can colonize surfaces within 20 minutes, evade sanitizing, and is capable of growth from 37$\sp\circ$C to below refrigeration temperature. Fish and seafoods are high in water activity and …

Analysis Of The Influence Of Genotype On Cayenne Pepper Fruit-Receptacle Detachment., Kay P. Gersch

LSU Historical Dissertations and Theses

Two genotypes of cayenne pepper, Capsicum annuum L., were identified that differ significantly in ease of fruit detachment. Both greenhouse and field-grown plants of these genotypes, Cajun 1-9027 and Cap-9004, were investigated for differences in cell type and organization at the fruit-receptacle junction. Scanning electron microscopy revealed that mature fruit of Cajun 1-9027, which did not separate, exhibited a distinct region of sclerified cells that extended from the periphery of the fruit into the receptacle for at least 15 cell layers. In contrast, mature fruit of the more readily detachable Cap-9004 had fewer sclerified cells at the region of detachment. …

A Physiological Study Of Common Carpetgrass (Axonopus Affinis) Subjected To Cultural And Environmental Stress., Edward Wayne Bush

LSU Historical Dissertations and Theses

Common carpetgrass (Axonopus affinis Chase) is a low maintenance stoloniferous grass widely distributed throughout the southern coastal plains of the United States. This study investigated seed germination enhancement, seedhead suppression, waterlogging tolerance, winter hardiness, and maintenance practices for common carpetgrass. Pre-soaking common carpetgrass and centipedegrass (Eremochloa ophiuroides (Munro.) Hack.) seed was beneficial at the 30C germination temperature. Priming seed in KNO$\sb3$ solutions significantly accelerated seed germination of both centipedegrass and carpetgrass at 30C, and increased total germination percentage of centipedegrass. At 20 and 25C, seed priming significantly improved mean time of germination for both grass species and total germination percentage …

Immunocytochemical Localization Of Gonadotropins, Growth Hormone, And Prolactin In Equine Pituitary., Mohammad Saeed Rahmanian

LSU Historical Dissertations and Theses

The present research was performed to characterize immunoreactive (ir) pituitary cells in the equine species. Two experiments were performed to measure the prevalence of single- and double-hormone labeled gonadotropes, lactotropes, somatotropes, and mammosomatotropes. In Experiments 1 and 2, pituitaries from three intact pony mares were processed for ultrastructural evaluation of cells stained for luteinizing hormone (LH), follicle-stimulating hormone (FSH), prolactin (PRL), and growth hormone (GH). Primary antibodies and gold-conjugated antibodies were used to double-stain ir-cells. The percentages of pituitary cells stained for each hormone in intact mares were: total gonadotropes, 15 to 32%; LH-only, 2 to 16%; FSH-only, 1 to …

Dairy Herd Improvement Records As Replacement Of Technician Breeding Receipt Database For Routine Estimates Of Non-Return Rates For Al Bulls In Technician-Devoid Areas., Rick A. Baron Jan 1996

Dairy Herd Improvement Records As Replacement Of Technician Breeding Receipt Database For Routine Estimates Of Non-Return Rates For Al Bulls In Technician-Devoid Areas., Rick A. Baron

LSU Historical Dissertations and Theses

The objective of this study was to find whether DHI data could be used to estimate sire non-return rates to replace current technician data estimates. Bull weighted least squares means for non-return rates were calculated separately for five overlapping 60-90 day service periods from each data source. Models included stud, sire, service number, and linear and quadratic form of breeding month for both data sources, service unit for technician and lactation for DHI data. Sire and lactation were not significant $(P > .10).$ Technician differences $(P < .05)$ were in service unit, stud, service number, and linear and quadratic service month in all but one service period. DHI differed $(P < .05)$ for service number, month (linear and quadratic), and stud in two service periods. Technician R-square values were 0.23 to 0.28 versus 0.94 to 0.96 for DHI. Sire estimated non-return rates were weighted using the inverted estimator standard error squared and compared. Sire, stud, data source, service period, and appropriate interactions were modeled. Weighted bull non-returns differed $(P < .01)$ in magnitude across data sources. Stud, data source by stud, and sire within stud by data source were significant $(P .10).$ Four fertility categories based on mean and standard deviation of the weighted estimates were formed within the data sources across service periods. These categories were correlated (0.5 $>$ r $>$ 0.9) and 52.9 to 87.4% congruent within data source for adjacent …

Industry advisors representing various facets of the pork industry were recruited to review and challenge the concepts incorporated in this publication. Also, in situations where “gray areas” existed, these industry …

A Proposed Acrosomal System For Identifying Stages Of The Cycle Of The Seminiferous Epithelium And A Model For The Kinetics Of Spermatogenesis In The Rabbit, Antonio Carlos S Castro Jan 1995

A Proposed Acrosomal System For Identifying Stages Of The Cycle Of The Seminiferous Epithelium And A Model For The Kinetics Of Spermatogenesis In The Rabbit, Antonio Carlos S Castro

Doctoral Dissertations

Most quantitative investigations of spermatogenesis requites identification of stages of the cycle of the seminiferous epithelium. The current "tubular morphology" system of staging for the rabbit permits identification of only eight stages and is not applicable to all experimental conditions. A more advantageous staging system, based on changes in the acrosome and morphology of the developing spermatids, had not been developed for the rabbit. Knowledge of spermatogonial kinetics, including the identity of spermatogonial subtypes and the number and timing of spermatogonial divisions, is also important, but had not been fully elucidated in this species. This investigation was undertaken to develop …

Changes Of Antioxidants And Fatty Acid Composition In Stabilized Rice Bran During Storage., Taisun Shin Jan 1995

Changes Of Antioxidants And Fatty Acid Composition In Stabilized Rice Bran During Storage., Taisun Shin

LSU Historical Dissertations and Theses

The effects of extrusion temperature and post extrusion holding time, microwave heating, and gamma irradiation on antioxidants and lipid composition in rice bran were studied. Post extrusion holding time had no apparent effect on rice bran stability. Increased holding times reduced (p $<$ 0.05) total vitamin E. Oryzanol concentration was lower (p $<$ 0.05) only with 6 min holding time. Inactivation of lipolytic enzymes was obtained by extrusion temperatures of 120, 130, and 140$\sp\circ$C. Increased extrusion temperatures reduced the retention of vitamin E and oryzanol during storage. An extrusion temperature of 110$\sp\circ$C provided the highest retention of vitamin E and oryzanol during storage although FFA level was higher (p $<$ 0.05) than that of the other extrusion temperatures from 105 to 375 days. There were no significant (p $>$ 0.05) decreases in individual vitamin E vitamers until 4 min microwave heating. After 8 min heating, individual E vitamers decreased significantly (p $$ 0.05) in FFA level by 24 weeks of storage with microwave heating for 12 min. Microwave heating for 4, 8, and 12 min did not provided adequate inactivation of lipolytic enzymes. Total vitamin …

The fish were fed six isonitrogenous (38% crude protein) and isocaloric (1.36 $\times$ 10$\sp4$ kJ/kg), purified diets differing in lipid content, for nine weeks, to investigate the effects of selected dietary essential fatty acids (EFA) on growth performance and tissue fatty acid composition. Each diet, except the low-fat (LF) diet, contained 8% total lipid from (1) menhaden fish oil (MFO), (2) oleate (18:1n-9) supplemented with linoleate (18:2n-6), (3) linolenate (18:3n-3), (4) a …

Genotypic Variation In Sweetpotato Ipomoea Batatas (L.) Lam. Clones., Arthur Quejada Villordon Jan 1995

Genotypic Variation In Sweetpotato Ipomoea Batatas (L.) Lam. Clones., Arthur Quejada Villordon

LSU Historical Dissertations and Theses

Arbitrarily-primed PCR-based assays established the presence of sweetpotato intra-clonal genetic variability. These DNA polymorphism assays provided benchmark information regarding cultivar genetic uniformity in sweetpotato foundation seed programs. Arbitrarily-primed markers were also used to compare the genetic uniformity among sweetpotato clones derived conventionally, i.e., through adventitious sprouts, and nodally-based propagation systems. Initially, 38 primers generated 110 scorable DNA fragments using two virus-indexed plants from each clone source. Twenty-one bands (18.6%) were scored as putative polymorphic markers based on the presence or absence of amplified products. A subset of 14 marker loci generated by four selected primers was used to further assay …

Aqueous Processing Influences On Composition And Functionality Of Mechanically Separated Broiler Meat., Henri Khalil Salman Jan 1995

Aqueous Processing Influences On Composition And Functionality Of Mechanically Separated Broiler Meat., Henri Khalil Salman

LSU Historical Dissertations and Theses

The demand for breast meat has caused a surplus of broiler legquarters, which cannot be easily or efficiently deboned by hand. Oxidative stability and texture of mince recovered with two mechanical deboners, a belt/drum separator with 5 mm (BD5), 2 mm (BD2), and 5 mm followed by 2 mm (BD5/2) drum or auger/sieve (SD) deboner with slit openings of 0.25 mm, were evaluated. Mechanical deboned chicken meat (MDCM) had higher fat, ash and lower moisture than hand deboned meat. BD2 meat contained calcium at levels below those allowed by USDA, but had higher fat that decreased oxidative stability. Washing of …

Dietary Chromium Supplementation For Pigs And Chickens., Terry Lynn Ward Jan 1995

Dietary Chromium Supplementation For Pigs And Chickens., Terry Lynn Ward

LSU Historical Dissertations and Theses

Three experiments were conducted with pigs and four experiments were conducted with chickens to determine the effects of supplemental dietary Cr on growth performance, carcass characteristics, tissue minerals and plasma metabolite and hormone concentrations. In pigs (Exp. 1) 200 ppb Cr from seven different sources tended to decrease performance during the grower phase and increase growth performance during the finisher phase. Dietary Cr had minimal effects on plasma metabolite and hormone concentrations and Cr did not affect carcass characteristics. In general, dietary Cr increased tissue Cr concentration, except for longissimus muscle (LM). In Exp. 2, pigs were fed 400 ppb …

The Production Of Triploid Oyster Larvae (Crassostrea Virginica (Gmelin)) In Louisiana., John Eric Supan

LSU Historical Dissertations and Theses

Gonadal recycling of oyster broodstock, salinity effects during induction, and cytochalasin B (CB) dosage were investigated as factors affecting the production of triploid oyster larvae in Louisiana. These factors were studied at an oyster hatchery on Grand Isle, LA during 1992-1994 to help transfer triploid oyster technology to the Gulf region. Oyster broodstock were held nearshore in Caminada Bay, Louisiana and histologically analyzed between induced spawnings and during the winter of 1992-1993. Gametogenesis was analyzed qualitatively by developmental staging and quantitatively using mean gonad/body ratios (GBR) per section. The monthly percent occurrence of the developmental stages, as well as histological …

Changes Of Amylases And Carbohydrates In Sweetpotatoes During Storage And Their Effects On Viscosity Of Sweetpotato Puree., Xiangyong Liu Jan 1995

Changes Of Amylases And Carbohydrates In Sweetpotatoes During Storage And Their Effects On Viscosity Of Sweetpotato Puree., Xiangyong Liu

LSU Historical Dissertations and Theses

A critical problem associated with the production of sweetpotato puree is the inconsistency of final product. Two possible factors, amylase activity and carbohydrate content in sweetpotatoes during storage, were investigated. It was found that $\alpha$- and $\beta$-amylase activities do not significantly change during storage, and have no significant effects on viscosity of sweetpotato puree. The inconsistent products in sweetpotato puree processing are mostly due to the change of alcohol insoluble solids (AIS) in sweetpotatoes during storage. The decrease of AIS is partially due to respiration that converts starch into CO$\sb2$ and H$\sb2$O. A new bio-processing method was proposed to improve …

Five, multiparous, Holstein cows fitted with ruminal and duodenal cannulas were assigned to 5 x 5 Latin squares at peak, early, and mid lactation to determine the required level of Met in duodenal digesta for maximum synthesis of milk protein. Cows were fed corn-based diets, formulated to be most limiting in Met and Lys, and were infused with incremental amounts of Met and a basal amount of Lys. The calculated required contributions of Met to total essential AA passing to the duodenum for maximum milk protein synthesis exceeded 5.5 and 5.2% of essential AA for the peak and early lactation …

G94-1230 The Foaling Mare, Kathleen P. Anderson Jan 1994

G94-1230 The Foaling Mare, Kathleen P. Anderson

University of Nebraska-Lincoln Extension: Historical Materials

This NebGuide outlines the foaling process and appropriate actions to take during both normal and abnormal foaling.

The ultimate goal of any breeding operation should be both maximal foaling and a high survival rate of foals. Most mares will have a normal parturition if left unattended, however, the value of broodmares and their progeny can make leaving parturition to nature an expensive gamble. As the value of mares and foals rise, it becomes more desirable to have someone present during foaling, allowing for immediate assistance to both mare and foal if needed.

Being present when a mare foals can be …

Development Of Aluminum Tolerance In Rice (Oryza Sativa L.) Through Tissue Culture Techniques., Herry S. Utomo Jan 1994

Development Of Aluminum Tolerance In Rice (Oryza Sativa L.) Through Tissue Culture Techniques., Herry S. Utomo

LSU Historical Dissertations and Theses

Three tissue culture techniques, somaclone evaluation, in vitro screening of suspension cells, and in vitro screening of protoplasts were evaluated for their potential to produce aluminum (Al) tolerant rice. For somaclone evaluation, a collection of 9,000 rice somaclones was screened. After 11 days of germination in 200 uM Al, the length of the primary root was measured and used as the criterion for selection since a principal effect of Al toxicity is inhibition of root growth. Four superior lines were identified. Root length of the most tolerant line was 50% greater than its control following germination in screening solution. A …
