Lower extremity loss affects nearly 150,000 individuals in the United States yearly yielding extreme health care costs annually. Typically, prosthetic devices are prescribed for 5-8 years despite known changes to an individual’s gait as time progresses. Patients can expect to undergo approximately 8-12 weeks of gait retraining to improve their comfort and efficiency. Biomechanical changes occur when running with a specific running prosthetic including considerable biomechanical and neuromuscular interlimb asymmetries and alterations in loading patterns. This phenomenological study exposes the complex cognitive and organizational processes school leaders navigate as they translate high-stakes state mandates into localized action. By examining the implementation of Virginia’s revised Standards of Accreditation and Accountability (SOA), this study challenges the prevailing managerial paradigm that views policy implementation as a linear, mechanical execution of directives. Instead, grounded in sensemaking theory and SDT, this research conceptualizes implementation as a situated and interpretive act shaped by leaders’ prior experiences, beliefs, and psychological needs for autonomy, competence, and relatedness.

Upon hatching, squid paralarvae must become proficient at locomotion and prey capture before their yolk reserves are depleted, while also contending with the physical constraints associated with low/intermediate Reynolds number (Re) environments. Although some aspects of behavior and jet dynamics have been examined in squids during early ontogeny, little is known about routine behaviors in paralarvae, whether repeatable patterns are employed, or how these behaviors are achieved dynamically. The Lafayette River, a shallow, eutrophic tributary of the Chesapeake Bay, experiences seasonal harmful algal blooms (HABs). While management efforts focus on reducing external nutrient inputs, internal loading from benthic sediments remains poorly constrained. This study quantifies spatial and seasonal variability in nutrient and radium isotope fluxes across the sediment–water interface at three sites where HABs are known to initiate and represent contrasting sediment textures and hydrodynamic conditions. Sediment cores and porewater samples were collected in summer 2023 and winter 2024 to measure grain size, porosity, dissolved nutrients (NH₄⁺, NOₓ⁻, PO₄³⁻), and radionuclide activities (²²⁴Ra and ²²⁸Th). United States citizens rely on state government employees to provide essential public services. During the COVID-19 pandemic, these employees responded by administering both routine and novel procedures to protect public health, often placing them on the frontlines and at increased risk of contagion. Concurrently, the pandemic-driven economic downturn created budget shortfalls that threatened the job and financial security of the state workforce. Together, these conditions substantially increased job demands for state government employees.

Pancreatic ductal adenocarcinoma is a highly lethal malignancy characterized by poor prognosis and resistance to conventional therapies. Novel treatment strategies are needed to overcome this resistance. Non-thermal physical modalities, including nanosecond pulsed electric field (nsPEF) and cold atmospheric pressure plasma discharge (CAP), have emerged as potential anticancer approaches; however, the mechanisms underlying their combined effects remain unclear.

This study evaluated whether nsPEF enhances CAP-induced cytotoxicity and investigated the associated molecular mechanisms using the murine Pan02 pancreatic cancer cell line. I argue that Harebrained Schemes’ video game series Shadowrun critiques transhumanism in three distinct yet related ways: in terms of religion, political control, and identity capitalism. In doing so, Shadowrun attempts to counter claims that video games are an inherently transhumanist medium and provides an opportunity for players to resist a potential transhuman future. This further elucidates the trends and perceptions of transhumanism within American culture around the time period. Listeria monocytogenes is a facultative intracellular foodborne pathogen capable of crossing the placental barrier, leading to miscarriage, stillbirth, and neonatal sepsis, and has a fetal/neonatal fatality rate of ~20–30%. Although the importance of canonical virulence factors such as Internalin A (InlA) and Internalin B (InlB) in L. monocytogenes crossing of the placental barrier has been demonstrated, clinical strains lacking functional canonical internalins have been reported to cause severe neonatal disease, suggesting additional mechanisms of placental translocation.

Altered mental status (AMS) in advanced chronic kidney disease (CKD) frequently reflects overlapping metabolic, cardiopulmonary, and neurologic contributors rather than a single unifying diagnosis. We present a 59-year-old woman with autosomal dominant polycystic kidney disease (ADPKD) progressing to end-stage renal disease (ESRD), congestive heart failure (CHF) with preserved ejection fraction (HFpEF), and stiff person syndrome (SPS) who developed recurrent encephalopathy accompanied by equivocal pulmonary findings and elevated cardiac troponin without electrocardiographic evidence of acute ischemia. Her hospital course was marked by severe renal dysfunction, metabolic acidosis, anemia, and intermittent electrolyte abnormalities. INTRODUCTION: Recent advances in osteoimmunology have illuminated the significant role of neuroimmune interactions in bone and joint health. The neuroimmune system critically regulates bone cell function through a complex network of molecular mediators - including signals from the sympathetic and parasympathetic nervous systems, pro- and anti-inflammatory cytokines, cortisol and other hormones, and growth factors. These mediators interact within a nonlinear network where each component can influence others reciprocally, with the brain serving as a central coordinator.

Genetic variants near LYPLAL1 are associated with Metabolic dysfunction-Associated Steatotic Liver Disease (MASLD) in humans, but their impact on LYPLAL1 function is unknown. We identified LYPLAL1 loss-of-function variants from UK BioBank (UKBB) whole-exome sequencing data that had AlphaMissense or GPN-MSA scores in the top 20% of LYPLAL1 variants for being disruptive. We aggregated these variants and carried out burden analysis for effects on MRI Proton Density-Fat Fraction (MRI-PDFF) and ICD-based MASLD in UKBB. Rare loss-of-function LYPLAL1 variants were associated with reduced MRI-PDFF and ICD diagnosed MASLD across sexes. We used CRISPR to knockout and overexpress LYPLAL1 in human hepatoma cells …

AbstractKoalas (Phascolarctos cinereus) give birth to highly altricial young that rely on maternal milk for nutritional support and immune protection in the first year of life. This study applied proteomics to analyse stage-specific shifts in the koala milk proteome across early-, mid-, and late-lactation. Proteins were extracted from seven samples and analysed using liquid chromatography-tandem mass spectrometry. Quantitative profiling identified 874, 1220, and 498 proteins in early-, mid- and late-lactation stages, respectively. The most abundant proteins—including β-lactoglobulin, caseins, clusterin, trichosurin, albumin and lactotransferrin—were consistently detected across all stages, reflecting crucial roles in nutrition, immune function, and detoxification. Human-driven habitat fragmentation is known to disrupt gene flow in a wide range of species. However, its consequences for endemic plant species that depend on specialised invertebrate pollinators remain poorly understood. Although honeybees are capable of travelling considerable distances while foraging, many small-bodied native bee species lack the capacity to expand their foraging ranges or cross anthropogenic barriers, rendering habitat fragmentation the most significant threat to these ecological interactions. The escalation of ecological degradation, marked by climatic instability, precipitous biodiversity loss, and systemic chemical infiltration, necessitates a shift toward integrated biotechnological intervention. This analysis considers how the trajectory of diverse advanced methodologies, including environmental DNA (eDNA) surveillance, CRISPR-based genetic restitution, and synthetic biology-driven bioremediation, might be synthesized into a unified Planetary Health Framework. Glutamate dehydrogenase (GDH) is a hexameric enzyme. GDH is involved in several pathways and cellular processes such as oxidation-reduction homeostasis, ammonia metabolism, lipid biosynthesis, insulin and lactate production, and acid-base equilibrium. The main objective of this thesis is to understand the structural and biochemical properties of this enzyme and why some organisms, for e.g., Halobacterium salinarum, have more than one GDH with different coenzyme specificities. The catabolism of glutamate is linked to NAD+ specific GDHs, meanwhile, NADP+ specific GDHs play an anabolic role in ammonia assimilation. Accurately predicting functional epigenomic events from DNA sequences is critical to understanding gene regulation and the functional impact of non-coding variants. Despite considerable progress, critical challenges hamper the effectiveness and efficiency of existing deep learning approaches. These challenges include negative transfer in multi-task learning (MTL), suboptimal network architectures, and pervasive label noise, particularly the positive-unlabeled problem arising from data sparsity in single-cell assays. Background

Genome-wide distributions of Alu elements contribute to a broad range of structural variants (SVs) through Alu/Alu-mediated genomic rearrangement (AAMR). Yet, the prevalence and characteristics of AAMR on the human genome and its scale in generating pathogenic SVs remain poorly understood.

Methods

We established a disease-focused, AAMR-SV dataset and a control dataset to comprehensively delineate the genomic landscape of Alu mutagenesis. The disease-focused dataset included 407 published pathogenic AAMR-SV alleles in 115 known genes for Mendelian disorders or traits through a literature survey. ΔFOSB, an unusually stable member of the AP-1 family of transcription factors, mediates long-term maladaptations that play a key role in the pathogenesis of drug addiction, cognitive decline, dyskinesia, and several other chronic neurological and psychiatric conditions. We have recently identified that 2-phenoxybenzenesulfonic acid-containing compounds disrupt the binding of ΔFOSB to DNA in vitro in cell-based assays, and one such compound, JPC0661, disrupts ΔFOSB binding to genomic DNA in vivo in the mouse brain with partial efficiency. Purpose: To evaluate the effects of rexinoid NEt-3IB on desiccating stress-induced dry eye, as well as monocyte/macrophage gene expression and cellular trajectory.

Methods: Eyes were topically treated with rexinoid NEt-3IB (5 µM) or vehicle three times a day for 5 days of desiccating stress-induced dry eye. Single-cell RNA sequencing (RNA-seq) profiled gene expression in conjunctival immune cells. RNA-seq was also used to evaluate gene expression in lipopolysaccharide (LPS)-stimulated, dexamethasone-treated (Dex, 1 µM), or NEt-3IB-treated (1-1000 nM) cultured monocytes. Background: Preterm infants requiring prolonged hospitalization in the neonatal intensive care unit (NICU) are often exposed to early-life stress, which may contribute to long-term health consequences. Immune checkpoint inhibitors (ICIs) have emerged as a promising treatment for various cancers, including advanced hepatocellular carcinoma (HCC). However, a significant proportion of patients with HCC, particularly those with metabolic dysfunction-associated liver disease (MASLD), exhibit resistance to ICI therapy. Studies have revealed that the presence of specific gut bacteria, such as Akkermansia, Bifidobacterium, and Lachnoclostridium, is associated with improved outcomes with ICI-treated HCC patients. Conversely, the overgrowth of bacteria like Enterobacteriaceae is linked to resistance to therapy. Background

Untargeted metabolomics has emerged as a powerful approach to uncover metabolic dysregulation associated with cancer progression. When integrated with a machine learning strategy it facilitates the discovery of key metabolic pathways and predictive biomarkers with high diagnostic and prognostic value.

Methods

In this study, we employed liquid chromatography coupled to high-resolution Tribrid Orbitrap mass spectrometry to perform comprehensive metabolic profiling of bladder cancer (BLCA) as well as predict invasiveness of the disease.

Background: Meningiomas are the most common primary intracranial tumors in the United States, comprising nearly 41% of central nervous system tumors. While the majority are benign, atypical (World Health Organization Grade II) and malignant (Grade III) subtypes are clinically significant due to their aggressive biology, higher recurrence, and increased mortality risk. Objective: Cognitive dysfunction (CD) is a prevalent symptom in childhood-onset systemic lupus erythematosus (cSLE). This study aimed to investigate the neurobehavioral basis of CD in cSLE.

Methods: Patients with cSLE (N=20) and age- and sex-matched healthy controls (HCs, N=20) completed questionnaires and multiple neurocognitive tests. The Systemic Lupus Erythematosus Disease Activity Index 2000 and laboratory markers were used to monitor patients' clinical status. Neuroimaging assessments included functional near-infrared spectroscopy (fNIRS), functional magnetic resonance imaging (fMRI), and structural MRI.

Importance: Current pretransplant clinical scoring systems fail to accurately estimate post-liver transplant (LT) survival, making recipient selection challenging. Persistent immune dysfunction contributes to early LT recipient mortality but is not captured by existing models.

Objective: To identify plasma biomarkers of pretransplant immune dysfunction and to develop a biomarker-based pre-LT risk stratification tool to predict post-LT mortality.

The lymphatic system is a highly branched endothelial tubular network that facilitates the migration of immune cells from the peripheral tissues to lymph nodes (LNs) and other lymphoid organs. Complement factors are essential for innate and adaptive immune functions. Complement anaphylatoxin C5a is crucial in vascular endothelial cell activation and lymphocyte polarization. Understanding the impact of C5a and its cognate receptor C5ar1 signaling on lymphatic function could provide new insights into the mechanisms of immune dysregulation observed in chronic inflammatory diseases. We demonstrate that acute C5a challenge in wildtype C57B6/J mice significantly reduced lymph propulsion compared to their C5ar1-deficient counterparts. …
