Journal of Dental Sciences
Abstract Background/purpose Nucleotide-binding oligomerization domain-like receptor family caspase recruitment domain containing protein 5 (NLRC5) plays a regulatory role in innate and adaptive immunity. However, its role in periodontitis remains unclear. This study investigated the effects of NLRC5 on periodontitis and the underlying mechanism. Materials and methods Experimental periodontitis models of wild-type and Nlrc5 knockout mice were established to detect alveolar bone loss. The inflammatory environment was established with Porphyromonas. gingivalis lipopolysaccharide ( P. gingivalis LPS). The expression of NLRC5 in periodontal ligament stem cells (PDLSCs) were detected with P. gingivalis LPS stimulated. After knocking-down or overexpressing the NLRC5 expression level, …

Journal of Dental Sciences
Abstract Background/purpose Orthognathic surgery addresses craniofacial and occlusal deformities caused by skeletal discrepancies. This study compares the aesthetic outcomes of maxilla-only, mandible-only, and bimaxillary surgeries, focusing on facial angles, proportions, and soft tissue aesthetics. It also identifies parameters most influencing postoperative aesthetics. Materials and methods Sixty Taiwanese patients (30 males, 30 females) were divided into three groups: maxilla-only, mandible-only, and bimaxillary surgeries (20 patients per group). Pre- and postoperative cephalograms were analyzed using six cephalometric methods. Paired and independent t-tests (P < 0.05) were conducted to evaluate changes and deviations from reference norms. Results The results of the study showed that maxilla-only surgery could lead to significant improvements in midface aesthetics, especially a notable increase in the nasolabial angle ( P < 0.05) and a reduction in UL-E line distances ( P < 0.05). The mandible-only surgery could enhance the lower facial symmetry with significant changes observed in the H angle ( P < 0.05), SN-MP angle ( P < 0.05), and LL-E line distances ( P < 0.05). Bimaxillary surgery resulted in the most comprehensive improvements, including a significant increase in the lower anterior facial height (LAFH, P < 0.05), an enhanced nasolabial angle ( P < 0.05), and an overall improvement in facial balance. However, a decrease in postoperative nasal prominence was observed across all surgical types when compared to reference norms, suggesting that secondary adjustments may be required. Conclusion Maxilla-only surgery improves midface proportions, while mandible-only surgery enhances lower facial balance. Bimaxillary surgery provides the most comprehensive outcomes. Individualized surgical planning and potential nasal refinements are crucial for optimal results.

Journal of Dental Sciences
Abstract The transition from pre-clinical to clinical operative dentistry requires effective training tools; however traditional typodont teeth often fall short in simulating clinical scenarios such as caries preparation. This study evaluated the perspectives of 71 third-year dental students from the classes of 2024 and 2025 at Harvard School of Dental Medicine on the use of 3D-printed caries teeth as a preparatory tool. Students participated in case-based collaborative learning and operative exercises using these models, with pre- and post-surveys capturing feedback on realism, usability, and educational impact. Statistical analysis revealed significantly increased confidence in modifying cavity preparations, understanding caries removal principles, …

Extreme hypertriglyceridemia, defined as triglyceride (TG) levels ≥1000 mg/dL, is almost always indicative of chylomicronemia. The current diagnostic approach categorizes individuals with chylomicronemia into familial chylomicronemia syndrome (FCS; prevalence 1-10 per million), caused by the biallelic combination of pathogenic variants that impair the lipolytic action of lipoprotein lipase (LPL), or multifactorial chylomicronemia syndrome (MCS, 1 in 500). A pragmatic framework should emphasize the severity of the phenotype and the risk of complications. Therefore, we endorse the term "persistent chylomicronemia (PC)" defined as TG ≥1000 mg/dL in more than half of the measurements to encompass patients with the highest risk for …

Background: Chylomicronemia is characterized by fasting triglyceride (TG) ≥1000 mg/dL; its longitudinal course is not well studied.
Methods: Using National Health and Nutrition Examination Survey (NHANES) data (1999-2018; n = 21,998), we determined chylomicronemia prevalence and temporal trend. Using Mayo Clinic data (4,524,506 TG measurements for 1,294,044 individuals), we studied the longitudinal course and ascertained persistent chylomicronemia (PC), defined as TG ≥1000 mg/dL in more than half the measurements for individuals with ≥3 measurements. We used logistic regression to assess factors associated with PC.
Results: In NHANES, the prevalence of chylomicronemia was 0.20% overall, with higher prevalence in men (0.32%) …

Introduction: Pretreatment levels of low-density lipoprotein cholesterol (LDL-C) are a primary consideration for the clinical diagnosis of homozygous familial hypercholesterolemia (HoFH). However, determination of pretreatment LDL-C can be challenging in this population because many individuals begin lipid-lowering therapy (LLT) early in life before diagnosis of HoFH.
Methods: A previously developed simulation model was adapted to estimate pretreatment LDL-C in people living with HoFH, based on their treated LDL-C value and current LLT (statin, ezetimibe, alirocumab, evolocumab, and/or lipoprotein apheresis).
Results: The tool includes an interface for users to provide relevant details for estimating non-clinical pretreatment LDL-C, specifically for patients suspected …

BACKGROUND: Anti-racism efforts are imperative for campus communities, yet little is known about whether perceiving their presence on campuses relates to a range of mental health outcomes among students.
METHODS: We analyzed data from the Healthy Minds Study 2020-2021 (
RESULTS: Individuals who disagreed that their schools combatted racism in their campus communities had significantly greater odds of mental health problems (depression, anxiety, psychotic experiences, suicidal ideation, suicide plan, suicide attempt, perceived need for help, and loneliness), and lower odds of flourishing when compared with those who strongly agreed with the statement. For most outcomes, we observed an apparent dose-response …

BACKGROUND: Evidence-based interventions (EBI) for cardiovascular disease (CVD) in low- and middle-income countries (LMIC) may face feasibility challenges due to the inadequacy of existing instruments. To address this, researchers developed the Contextual Index of Feasibility on Early-Stage Implementation in LMIC (CATALYTIC) tool, which integrates contextual factors into the assessment of feasibility.
METHODS: The tool's items were developed through a systematic review and key informant interviews, and were later assessed for relevance and importance by 13 LMIC researchers and implementers employing a Delphi technique. The survey was then tested for usability by five individuals with CVD experience in LMIC. The CATALYTIC …

AIM: This study aimed to optimize non-invasive echocardiographic evaluation of myocardial function in a mouse model of diastolic heart failure, emphasizing the methodological challenges in assessing diastolic and left atrial (LA) function. Recognizing that clinical human studies frequently assess cardiac performance in non-sedated subjects, this investigation compared systolic and diastolic functional outcomes in mice subjected to heart failure with preserved ejection fraction (HFpEF) using two anesthetic agents: Avertin (mild sedation) and isoflurane (deep sedation). Additionally, we present a histological and echocardiographic correlation of the LA changes in established HFpEF mouse model.
RESULTS: Mice received angiotensin II and phenylephrine (AngII/PE) infusions …

Objective: Autoimmune or inflammatory rheumatic diseases (AIRDs) increase the risk for poor COVID-19 outcomes. Although rurality is associated with higher post-COVID-19 mortality in the general population, whether rurality elevates this risk among people with AIRD is unknown. We assessed associations between rurality and post-COVID-19 all-cause mortality, up to two years post infection, among people with AIRD using a large nationally sampled US cohort.
Methods: This retrospective study used the National COVID Cohort Collaborative, a medical records repository containing COVID-19 patient data. We included adults with two or more AIRD diagnostic codes and a COVID-19 diagnosis documented between April 2020 and …
