Adult hippocampal neurogenesis, the process of generating new neurons, relies on a rare population of neural stem and progenitor cells (NPCs) within the dentate gyrus complex microenvironment. Discovering the specific genes that define these cells is vital yet challenging due to overlapping expression patterns, limiting detection of rare cell populations using traditional approaches. By employing the computational digital sorting algorithm (DSA) that deconvolves complex gene expression data based on pattern recognition, we identified 129 genes enriched in murine NPCs. Background: Rare genetic variation provided by whole genome sequence datasets has been relatively less explored for its contributions to human traits. Meta-analysis of sequencing data offers advantages by integrating larger sample sizes from diverse cohorts, thereby increasing the likelihood of discovering novel insights into complex traits. Furthermore, emerging methods in genome-wide rare variant association testing further improve power and interpretability.

Peripheral sensory neurons regenerate their axons after injury to regain function, but this ability declines with age. The mechanisms behind this decline are not fully understood. While excessive production of endothelin 1 (ET-1), a potent vasoconstrictor, is linked to many diseases that increase with age, the role of ET-1 and its receptors in axon regeneration is unknown. Using single-cell RNA sequencing, we show that satellite glial cells (SGCs), which completely envelop the sensory neuron soma residing in the dorsal root ganglia (DRG), express the endothelin B receptor (ETBR), while ET-1 is expressed by endothelial cells. Though there has been substantial progress in the development of anti-human epidermal growth factor receptor 2 (HER2) therapies to treat HER2-positive metastatic breast cancer (MBC) within the past two decades, most patients still experience disease progression and cancer-related death. HER2-directed tyrosine kinase inhibitors can be highly effective therapies for patients with HER2-positive MBC; however, an understanding of resistance mechanisms is needed to better inform treatment approaches. We performed whole-exome sequencing on 111 patients with 73 tumor biopsies and 120 cell-free DNA samples to assess mechanisms of resistance. Methylmalonic acidemia (MMA) is a severe metabolic disorder affecting multiple organs because of a distal block in branched-chain amino acid (BCAA) catabolism. Standard of care is limited to protein restriction and supportive care during metabolic decompensation. Severe cases require liver/kidney transplantation, and there is a clear need for better therapy. Here, we investigated the effects of a small molecule branched-chain amino acid transaminase (BCAT) inhibitor in human MMA hepatocytes and an MMA mouse model. Background: The increased utilization of oral factor Xa inhibitors (FXaI) has led to a growing interest in the clinical utility of FXaI-specific anti-Xa concentrations. Critically ill populations are at risk of bleeding secondary to FXaI accumulation in the setting of end-organ dysfunction. To mitigate this risk, an FXaI anti-Xa concentration-guided approach to transitioning between oral and parenteral anticoagulation has been explored.

Hmongcentric Niche Charter Schools: The Role Of Social Capital On Students’ Perceived Academic Competence, Zha Blong Xiong, Ting Xu, Kyle Nickodem, Malina Her

Using Coleman’s (1988) social capital theory, this study investigated the connection between family and school social capital and the perceived academic competence of students enrolled in Hmongcentric niche charter schools. The study analyzed 124 participants consisting of third, fourth, and fifth-grade students residing in Minnesota, with 93% identifying as Hmong. Results of structural equation modeling showed that school social capital, as measured by school attachment and school connection, is significantly linked to students’ perceived academic competence. This study investigated awareness and perception of artificial intelligence in counselling practice among counsellors-in-training. Descriptive survey research design was adopted for this study. The population consisted of all counsellors-in-training (CITs) in Adeyemi Federal University of Education, Ondo. Two hundred (200) counsellors-in-training were randomly selected from Degree three (3) and four (4) as sample for the study. Mindfulness has emerged as a holistic approach that enhances psychological well-being and physiological functioning. This comprehensive review explores the relationship between mindfulness and major chronic conditions, including Diabetes Mellitus (DM), Hypertension (HTN), Insomnia, Obesity, Gastrointestinal Disorders (such as Irritable Bowel Syndrome and Inflammatory Bowel Disease), and Migraine. Adult obesity rates have increased from approximately 15% in 1980 to more than 40% of adults in 2018, and the rates continue to increase. White-collar workers are disproportionately impacted as a result of the sedentary work performed in a white-collar work environment. Increased levels of obesity result in elevated levels of psychological stress, chronic health conditions, and increased organizational costs. Vitamin D insufficiency (VDI) is primarily determined by serum levels of calcidiol, which serves as a biomarker for the less abundant but most potent bioactive metabolite, calcitriol. However, population studies often show discordance between calcidiol and calcitriol. Here, a genetically diverse population of 7 inbred mouse strains was used to investigate the role of interindividual genetic differences in driving calcidiol-to-calcitriol discordance under vitamin D sufficient (VDS) vs depleted (VDD) conditions. We found high interstrain variability in calcitriol that was discordant with calcidiol under VDS and VDD conditions. Peripheral nerve injuries (PNIs) have a significant impact on the quality of life for patients suffering from trauma or disease. Temporal lobe epilepsy (TLE) patients experience shifts between non-seizing and seizing brain states, but the structural networks underlying these transitions remain undefined and poorly characterized. We detected dynamic brain states in resting-state fMRI and constructed linked structural networks utilizing multi-shell diffusion-weighted MR data. Leveraging network control theory, we interrogated the structural data for all possible brain state transitions, identifying those requiring abnormal levels of transition energy (low or high) in TLE compared to matched healthy participants (n's = 25). Plant functional traits typically show strong covariation, e.g. as in the worldwide leaf economics spectrum (LES). Covariation is thought to arise from selective forces and physical constraints. However, processes shaping covariation at interspecific scales may differ from those at intraspecific scales. Potential sources of intraspecific trait variation include variation in genetics, abiotic environment, and biotic environment; any of these factors may cause divergence from interspecific patterns. Quaking aspen, Populus tremuloides (Salicaceae), is a widespread tree species ideal for assessing these processes' role in the intraspecific LES, due to its clonal growth along environmental gradients, intraspecific variation in cytotype (ploidy level), …

African Swine Fever In Indonesia: Insights From Southeast Asia’S Policy Response Through The One Health Approach, Niken Salindri, Wachyu Sulistiadi

Journal of Indonesian Health Policy and Administration

African Swine Fever (ASF) seriously threatens pig production systems, food security, and rural livelihoods across Southeast Asia. This study employs an analysis of secondary national data from Indonesia using a quantitative descriptive analysis approach, complemented by regional policy responses to ASF from 2019 to 2024. To contextualize regional findings, this study also integrates official statistics from Statistics Indonesia (BPS) covering provincial pork production trends (2021–2023), the number of ASF-affected pigs per province (2020–2024), and per capita pork consumption by district. Analyzing The Role Of A Dual-Function Diguanylate Cyclase / Phosphodiesterase For Attachment And Motility In Agrobacterium Tumefaciens, Priya Aryal

Cyclic diguanylate monophosphate (c-di-GMP) is a second messenger that is responsible for attachment and motility in many prokaryotic systems and controls the signaling pathway for the motile to sessile transition in many bacteria. Many c-di-GMP-metabolizing proteins are diguanylate cyclases (DGCs), responsible for the synthesis of c-di-GMP and characterized by a c-di-GMP synthesizing domain, e.g., GGDEF domain. Other c-di-GMP-metabolizing proteins contain c-di-GMP degradative domains, e.g., EAL, characterized by phosphodiesterase activity (PDE). In Agrobacterium tumefaciens, out of 31 predicted proteins regulating the c-di-GMP level, this project focuses on the predicted dual-function DGC/PDE, DcpB. Introduction: Meridianin C (MC) is a marine-derived indole alkaloid that has demonstrated kinase inhibitory and anti-tumor activities. Despite its diverse biological properties, no previous reports have systematically evaluated the in vivo quantitative analysis of MC and its metabolites. Methods: In this study, MC was synthesized following a previously reported procedure with slight modifications. A sensitive, accurate, and reliable ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS) method was developed to simultaneously detect MC and its five major metabolites (MC-1-N-O-GluA, MC-1-N-O-SO3H, MC-2′-N-O-GluA, MC-2′-N-O-SO3H, and MC-O-GluA-didehydration) in rat plasma. Nigerian immigrants represent one of the fastest-growing immigrant populations in the United States. Relocating to a new country can be a major source of stress, often increasing individuals’ vulnerability to mental health challenges. Background: Children born with a congenital anomaly have a higher risk of developing a brain tumor during childhood or adolescence, but the co-occurrence between specific types of congenital anomalies and specific types of childhood brain tumors (CBTs) is not well described. This study characterized the associations between specific congenital anomalies and CBTs.

Methods: We leveraged a population-based registry linkage study of births (1990-2018), congenital anomalies, and cancer from 9 states (n = 22,599,099 births). Congenital anomalies were classified as major structural without a known chromosomal or genetic syndrome, chromosomal, neurofibromatosis, and/or tuberous sclerosis complex. This commentary explores how micro- and nanoplastics, tiny plastic fragments present throughout the environment, pose emerging strategic challenges beyond environmental and health concerns. While micro- and nanoplastics are now found in water, soil, plants, animals, and even human bodies, their long-term effects on ecosystems, food and water safety, and human health remain poorly understood. These particles can infiltrate critical systems, potentially affecting troop readiness, infrastructure resilience, and national food security. Nanoplastics may also serve as vectors for biological or chemical contaminants, complicating detection and mitigation.
