Dual specificity phosphatase 8 (DUSP8) is one of the phosphatases that oppose the activity of mitogen-activated protein kinases. Previous mouse studies have demonstrated the functional role of DUSP8 in regulating cardiomyocyte growth. However, it is still unclear how the activity of DUSP8 is regulated, and little is known about the regulation of DUSP8 at the gene level. The goal of this study is to determine whether the DUSP8 gene is immediately transcribed upon MAPK activation in the cells. The Eastern Diamondback Rattlesnake (Crotalus adamanteus, EDB) is the largest rattlesnake in the world and is thought to be declining in many portions of its range. Genetic venom studies in the Southeast United States have shown evidence of fine-scale differences within two main geographic groups on Jekyll Island. My objective is to expand on this finding to determine if epigenetic mechanisms, specifically DNA methylation, varies among different populations of EDB in similar environments, age classes, and sexes. Metabolic Dysfunction-Associated Fatty Liver Disease (MAFLD) is characterized by excessive fat accumulation in the liver, with metabolic dysfunction-associated steatohepatitis (MASH) representing a more severe form that can progress to cirrhosis or hepatocellular carcinoma (HCC). The sphingosine kinases and the S1P signaling pathway are key regulators of MASH and HCC progression. While much is known about the mechanisms contributing to MASH-HCC in males, less is known for females. Chronic obstructive pulmonary disease (COPD) is currently the fourth leading cause of death in the world, with an increasing prevalence as the world’s population ages. In the distal regions of the lungs, COPD is characterized by progressive emphysema due to an imbalance of proteases that continuously degrade essential extracellular matrix (ECM) proteins within the alveolar walls, causing fatal tissue impairment. Degradation of water quality is primarily driven by human activities, such as urbanization. Urban Stream Syndrome (USS) can be characterized by the negative physical, chemical, and biological changes to water quality as a result of urbanization. One key effect of USS is the input of excess nutrients, which can drive unwanted macrophyte and algal growth. New construction is prevalent in urbanizing areas, resulting in the influx of nutrients, sediments, and other materials into stream ecosystems. Physical forces drive many cellular processes such as migration and proliferation. A metastatic tumor will have invasive strands/chains that extend from the main tumor. These chains of cells collectively migrate away from the main tumor to establish new colonies elsewhere in the body. However, how physical forces drive this collective invasion and the required energy for this process is not well understood. Hepatitis, most importantly hepatitis B and hepatitis C, is a significant global health concern, requiring an accurate and early diagnosis to prevent severe liver damage and ensure effective treatment. The currently employed diagnostic methods, while effective, are often limited in their sensitivity, specificity, and rapidity, and the quest for improved diagnostic tools is ongoing. This review explores the innovative application of Raman spectroscopy combined with a chemometric analysis as a powerful diagnostic tool for hepatitis. This thesis began with a question: can diagrams create understanding, or only reflect it? Through paintings that layer automotive wiring schematics with family photographs, I explore how the human attachment system resists easy mapping. Using digital tools like Photoshop and scanning, alongside analog methods such as papier-collé and photo transfer, I examine how systems built for clarity—like wiring diagrams—contrast with the layered, often messy realities of human relationships. Drawing from attachment theory’s empirical foundation, rather than the metaphorical leanings of psychoanalysis, my work investigates how emotional bonds form, falter, and repair over time. Purpose: This study aimed to compare the intracanal smear layer removal ability of two adjunctive irrigation modalities: the AdvErL EVO laser, the GentleWave® Procedure, and their combination, after minimally invasive canal instrumentation.

Methods: Forty molars were accessed, the mesiobuccal canals of maxillary and mandibular molars were prepared up to apical size 25.04 using rotary instrumentation, then irrigated using: conventional needle irrigation (CNI), laser-assisted irrigation (LAI), the GentleWave® Procedure (GWP), and LAI followed by GWP (LAI+GWP). Acute respiratory distress syndrome (ARDS) is a prevalent, life-threatening lung condition, affecting nearly 200,000 Americans annually, with a 40% international mortality rate. There is no cure for ARDS, and current pharmacological treatments have limited effectiveness. Symptoms can be mitigated with mechanical ventilation, though this often leads to ventilator-induced lung injuries (VILI) and puts critically ill patients at risk of infections, including ventilator-associated pneumonia (VAP). A promising therapeutic is the extracellular matrix (ECM), a complex network of structural proteins and bioactive molecules that has been shown to have anti-inflammatory properties and prevent fibrosis. Warmer temperatures and increased temperature variability are predicted in the future due to climate change. Phenotypic outcomes of avian nestlings are heavily impacted by nest temperature. The objective of this study was to determine the influence of vibration during transportation on beef sirloin water holding capacity, color, and tenderness. USDA Choice center-cut sirloin top butts (n = 45 cases) were collected at a commercial processing facility and randomly allocated to one of three treatments 1) regular pallet driven 241 km (Reg241), 2) regular pallet driven 644 km (Reg644), and 3) vibration isolation pallet driven 644 km (Iso644). All subprimals were transported at -2.2 °C (d0) and stored for 10 days to mimic industry practices for product aging. Following storage (d11), one subprimal per case was evaluated …

Many scientific fields rely on the capture and modeling of light to extract underlying information about the world. Often, more information can be extracted by capturing more about the nature of the light, such as its spectral shape or polarization state. While polarization is a relatively unexplored topic in computer graphics, when used in tandem with other recent advancements in the field it has enormous potential to improve both forward and inverse models in other scientific disciplines. Sentence Determination As A Function Of Sex Offender Gender, Kyle Gamache, Matt Zaitchik, Judith Platania, Willem Pontbriand

The current study examines perceptions of dangerousness and sentencing recommendations for female sex offenders (FSOs) compared to male sex offenders (MSOs). Participants (N = 82) were randomly assigned to read one of three vignettes depicting a sexual offense committed by a male, female, or male-female co defendant pair. Key individual difference variables—Need for Cognition (NFC) and Gender Role Beliefs (GRB)—were assessed to examine their influence on sentencing and attributional judgments. NFC emerged as a significant moderator: participants with low NFC assigned longer sentences to FSOs, whereas high NFC participants gave longer sentences to MSOs. Educating more students about ecology and its beneficial applications to societal issues is urgent yet challenging. To address this challenge, diversifying ecology education is a key way to make ecology more inclusive, accessible, and interdisciplinary for more people than ever. Advancing this goal requires ecology educators to develop a more expansive view of (1) how to engage more diverse undergraduate students in ecology courses, especially those from historically underrepresented groups and non-majors, (2) the interdisciplinarity of content in those courses, and (3) the learner-centered pedagogies used to engage students. Introduction: Deaths due to opioid-related overdoses have increased across the United States. Specifically in the state of Kentucky, African Americans are the only population that continues to experience an increase in opioid-related deaths. Previous literature highlights African Americans having unique drug use patterns due to systematic barriers and how these barriers limit access to protective factors and substance use treatment. Traditional serological methods for body fluid detection are limited by poor specificity and sensitivity, lacking confirmatory capability and thereby increasing the risk of false positives that may compromise the integrity of forensic investigations. While molecular methods such as mRNA, miRNA, and DNA methylation offer improved specificity, they are often hindered by marker instability and operational complexity in routine forensic workflows. Meprobamate, carisoprodol, and naproxen are acidic/neutral drugs widely prescribed for muscle relaxation and inflammatory conditions, with frequent detection in forensic toxicology casework due to extensive clinical use. A method to qualitatively evaluate these compounds was developed to unify acid/neutral drug screening. This study incorporated blank blood, antemortem blood, postmortem blood, urine, and liver homogenate as representative biological matrices. Method validation was conducted in accordance with ANSI/ASB Standard 036 for Forensic Toxicology, encompassing key parameters including limit of detection (LOD), endogenous interferences, potential interferences, carryover, processed sample stability, and ionization suppression/enhancement. Dogfighting is a felony in all 50 states and is often tied to other organized crime activities including gambling, narcotics, and weapons trafficking. Because of the clandestine nature of dogfighting crimes, it may be necessary to determine the age of blood samples from suspected events, not just the source. In this study, flow cytometry was used to identify cellular autofluorescence signatures for estimating TSD (time-since-deposition) in canine blood deposits up to 6 months old. Determination of tissue source and the time at which a biological material is transferred from its source to another surface, known as the time-since-deposition (TSD), would assist forensic analysts by providing crucial context for DNA profiles generated from biological evidence. Many methods for cell identification have been explored using biomolecule markers such as mRNA, miRNA, and differential DNA methylation.
