Pancreatic cancer is an aggressive malignancy, characterized by extensive desmoplasia and a hypoxic tumor microenvironment that contributes to therapy resistance. MYB, a proto-oncogene encoding a transcription factor, plays a crucial role in pancreatic tumor growth and metastasis. Recently, we also revealed a role of MYB in hypoxic survival of pancreatic cancer cells by promoting metabolic reprogramming through interaction with HIF1α, modulating its expression and binding to glycolytic gene promoters. In this study, we investigated how hypoxia influences the genome-wide occupancy of MYB using chromatin immunoprecipitation sequencing (ChIP-seq), and whether this effect is modulated by its interaction with HIF1α. Problem: Pediatric patients with congenital heart disease (CHD) are at an increased risk of constipation following cardiac surgery, which contributes to complications and delays in recovery. Standardized protocols are effective tools for prophylactic management of constipation. However, there is no current standardized protocol in place for postoperative bowel management in a pediatric cardiac intensive care unit (CICU).
Method: This quality improvement project (QI) used a descriptive design with pre- and post-implementation data collection to evaluate the effects of a postoperative bowel regimen protocol on constipation rates and compliance with ordering prophylactic bowel regimens. Teenagers' lack of digital sophistication and cyber hygiene makes them vulnerable to social engineering attacks, especially as they start using social media. Clickbait, one of such attacks, is primarily performed through social media to trick users into clicking on malicious links. With teenagers' increasing use of social media, clickbait poses a substantial threat to their online safety. The existing online safety measures for teens mainly focus on parental mediation, which can be perceived as restrictive and privacy-invasive. The German cockroach, Blattella germanica (Linnaeus) is a globally distributed indoor dwelling pest of medical concern. Insecticides are the preferred method of cockroach management due to their low cost, quick action, and availability. However, after continuous use, the development of insecticide resistance has emerged as a challenge for the pest management industry to effectively eliminate German cockroaches. While consequences of equine obesity are well defined, the extent to which persistent hyperleptinemia and accompanying leptin (Lep) resistance affect the fetal-maternal environment warrants further exploration. This study compared endocrine, fetal, placental, and uterine parameters during early pregnancy (through day 70) in mares with elevated leptin versus normal leptin (NL) concentrations. Fourteen mares were classified as either NL (n = 6; Lep 0.9 ± 0.3 ng/mL), moderate leptin (ML; n = 4; Lep 3.0 ± 0.3 ng/mL), or high leptin (HL; n = 4; Lep 5.8 ± 0.7 ng/mL) based on annual plasma leptin. Astronauts aboard the International Space Station (ISS) are chronically exposed to a complex space radiation environment composed of galactic cosmic rays (GCR), solar particles, and Earth's trapped radiation. These high-energy particles interact with spacecraft shielding, producing secondary particles that can penetrate inside and deposit energy in sensitive tissues. Although animal studies have been useful in assessing space radiation risks, anatomical and physiological differences limit their applicability to human anatomy especially neuroanatomy. Autism spectrum disorder (ASD) has been found to be frequently comorbid with many medical and psychological disorders, one of which is anxiety disorders. Research has shown that, when anxiety is present within ASD, various areas of the child’s functioning may be impacted, such as ASD symptom severity, emotional regulation skills, adaptability, social skills and daily living skills. Human impacts on carnivores are a persistent conservation challenge worldwide. We present a global analysis showing the overlap of conservation lands and the cumulative impact of humans on the distribution ranges of 257 terrestrial carnivore species. Our findings reveal that 64% of carnivore ranges overlap with areas characterized by high human pressures. We found that Indigenous peoples' lands emerge as crucial for carnivore population viability, potentially safeguarding 26% of carnivore ranges globally, while protected and wilderness areas cover roughly 10 and 16% of carnivore ranges, respectively. Hyperspectral image (HSI) band selection (BS) plays a crucial role in HSI dimensionality reduction, aiming to identify a representative subset of bands with minimal redundancy. However, conventional BS approaches primarily operate in the Euclidean domain, often overlooking the structural characteristics of pixels and spectral bands, such as spatial continuity and spectral dependencies. In addition, they handle each HSI as an integrated unit to harness implicit spatial information, disregarding spatial distribution variations across different homogeneous regions. Porous cranial lesions (cribra cranii and cribra orbitalia) are widely used by archaeologists as skeletal markers of poor child health. However, their use has not been validated with systematic data from contemporary populations, where there has been little evidence of these lesions or their health relevance. Using 375 in vivo computed tomography scans from a cohort-representative sample of adults aged 40+ years from the Bolivian Amazon, among food-limited, high-mortality forager-farmers, we identified cribra cranii on 46 (12.3%) and cribra orbitalia on 23 (6%). A new type of electrodes was prepared for determination of drugs depended on (TCH- PM-CuO NPs). This work recommended a novel CuO nanoparticles-depended on coated wire sensors for detected tetracycline hydrochloride (TCH) in pure and pharmaceutical formulations. Unsaturated aromatic ketones are important substrates for synthesizing various heterocyclic compounds. The reaction between triethylorthoformate and p-chlorophenyl resulted in formation of N,N'-bis-(p-chlorophenyl)formamidine ligand. Subsequently, this ligand interacted with the following metal ions (Co+2, Ni+2, Cu+2, Cr+3 and Ru+3) creating air-stable complexes with formula [M(L)2(H2O)n(Cl)x](X= 0, 1) with distinct octahedral morphologies. The formation of these compounds was identified using mass spectroscopy (MS), which validated the acquired geometries. Background: Human immunodeficiency virus (HIV) establishes persistent infection by integrating its proviral DNA into the host genome. While most integrated proviruses are defective, a small portion of proviruses are intact that represent a major obstacle to achieving an HIV cure. We have designed an approach for selective elimination of host cells harboring replication-competent HIV (SECH), through inhibition of autophagy and anti-apoptotic molecules during viral reactivation. However, the effects of SECH on the dynamics of intact and defective HIV provirus remain unclear.

Pathological events often impact tissue regions in a spatial variable manner, making it challenging to identify therapeutic targets. Spatial transcriptomics (ST) is a powerful technology to map spatially variable molecular mechanisms, yet suitable analytical methods have been lacking. We introduce SPaSE (Spatially-resolved Pathology ScorE), an optimal transport-based algorithm to compare ST data from diseased and control tissues. SPaSE computes a “pathology score” for each spot in the diseased sample, quantifying the pathological impact at that spot. In post-MI (myocardial infarction) mouse hearts, these scores delineated zones that matched independent expert annotations. The Genome in a Bottle Consortium (GIAB), hosted by the National Institute of Standards and Technology (NIST), is developing new matched tumor-normal samples, the first explicitly consented for public dissemination of genomic data and cell lines. Here, we describe a comprehensive genomic dataset from the first individual, HG008, including DNA from an adherent, epithelial-like pancreatic ductal adenocarcinoma (PDAC) tumor cell line and matched normal cells from duodenal and pancreatic tissues. In this study, a new, simple, and rapid method was developed for determining Bromhexine-HCl using silver nanoparticles. The solution of silver nanoparticles has a yellow color which has absorbency at a wavelength of 404 nm. Bromhexine was estimated through its reaction with silver nanoparticles in acidic environment at pH 5. This reaction causes the silver nanoparticles to aggregate and change the color of their colloidal solution from yellow to brown, leading to a decrease in their absorbance. Oxytocin is a neurohypophyseal hormone that plays a crucial role in regulating mother-infant bonding and maternal behavior. Oxytocin modulates sex-specific parental behaviors by binding to the oxytocin receptor (OXTR)-expressing neurons in the brain. This study investigates the sexually dimorphic OXTR neurons in the anteroventral periventricular nucleus (AVPV) to understand how oxytocin modulates behavior in females. Specifically, we explored the behavioral roles, neural projections, and intrinsic electrophysiological characteristics of these neurons in the female mice. Using immunohistochemistry revealed that the number of OXTR-expressing neurons was enhanced in postpartum females compared with virgin females, suggesting that these neurons are involved in female-specific …

The dynamics of neural circuits and their role in mediating cellular and organismal phenomena remain poorly understood, despite numerous efforts to dissect these processes through precise instantaneous measurements or longer-time averages and approximations. We use an alternative approach: we investigate these dynamics at the system's mesoscale by analyzing spike trains and waveforms. These extended activity patterns carry robust, tractable information, are highly responsive to physiological specifics, and enable detailed tracking of circuit behavior. In particular, this methodology allows for characterizing the functionality of tau-pathology-afflicted hippocampal circuits and identifying circuit-level abnormalities that are missed by through traditional analyses. Background: The macro-social and environmental conditions in which people live, such as the level of a country’s development or inequality, are associated with brain-related disorders. However, the relationship between these systemic environmental factors and the brain remains unclear. We here aimed to determine the association between the level of development and inequality of a country and the brain structure of healthy adults.

Nanoparticles of copper oxide (CuO) are synthesized using a photolysis process. x -ray diffraction (XRD]) is utilized to investigate CuO nanoparticle's crystal structure, and transmission electron microscopy (TEM) is employed to analyze the morphology and the average nanoparticle diameters. Results indicated that the CuO nanoparticles have a monoclinic shape with a mean size of 14 nm. The UV -- Visible measurement is shown that the bandgap of CuO nanoparticles is 1.61 eV. The CuO nanoparticles were applied in a photoanode section to fabricate photo organic sensitizer solar cells. This study dealt with the preparation and analytical study of a type of chelating resin. This type of resin was distinguished by containing a ligand of the type of aromatic Schiff bases (5A-T) by reacting the aromatic amine (5-aminoisopthalic acid) with terephthaldehyde. The prepared Schiff base was identified using infrared FT-IR, 1H-NMR, 13C-NMR, and precise element analysis technology. Detergents and lipid nanodiscs affect the cryo-EM structures of pentameric ligand-gated ion channels (pLGICs) including ELIC. To determine the structure of a pLGIC in a membrane environment that supports ion channel function, we performed single particle cryo-EM of ELIC in liposomes. ELIC activation and desensitization were confirmed in liposomes with a stopped-flow thallium flux assay. Using WT ELIC and a non-desensitizing mutant (ELIC5), we captured resting, activated, and desensitized structures at high resolution. In the desensitized structure, the ion conduction pore has a constriction at the 9' leucine of the pore-lining M2 helix, indicating that 9' is the desensitization gate …
