Establishment And Characterization Of Vincristine-Resistant S1 Colon Cancer Cells, Harsh Patel

Colorectal cancer (CRC) remains one of the most common causes of cancer-related mortality worldwide. Vincristine is frequently used to treat CRC, although the specific pathways leading to acquired resistance are still not fully understood. In this study, we established two novel vincristine-resistant human colon cell lines (S1/V1 and S1/V4) to explore the mechanisms underlying vincristine resistance, with a particular emphasis on the overexpression of ATP-binding cassette (ABC) transporters. The S1/V1 and S1/V4 sublines were derived by continuously exposing human S1 colon cancer cells to vincristine. An MTT cytotoxicity assay revealed that S1/V1 cells exhibit about 39-fold greater resistance to vincristine …

Gankyrin is an ankyrin-repeat protein overexpressed in certain cancers and is responsible for cell growth and proliferation. Gankyrin is important to the development of many types of cancer, including breast, lung, and liver cancer. Consequently, gankyrin and its various protein-protein interactions, are prospective therapeutic targets for inhibiting the progression of certain cancers. In 2016, the first small molecule inhibitor of gankyrin, cjoc42, was discovered and subsequently demonstrated modest antiproliferative activity against liver cancer cells. Previous work from our lab demonstrated that replacing the sulfonate ester of cjoc42 with an amide group significantly improved gankyrin binding while enhancing antiproliferative activity against …

The Attitudes and Beliefs Scale 2-Short Form (ABS2-SF) is one of the most widely used measures of irrational and rational beliefs central to Rational Emotive Behavior Therapy (REBT). While evidence supports its reliability and validity in English-speaking populations, little is known about its applicability across cultural and linguistic groups. This study investigated the psychometric properties and cultural validity of the Spanish version of the ABS2-SF in a Mexican sample (n = 579) collected from undergraduate students in Tampico, Mexico, and compared them with English-speaking American non-clinical samples (n = 1,167). Analysis of electroencephalogram (EEG) recordings in children with dyslexia has been commonly used to explore the neural mechanisms underlying reading disorders. Yet, challenges such as low signal-to-noise ratio (SNR), high inter-subject and inter-trial variability, and the inherently multivariate nature of EEG signals hinder the isolation of neural components elicited during reading diagnostic tests. To mitigate these challenges, the neural-congruency analysis framework was recently proposed, leveraging traditional machine learning optimization methods to incorporate domain knowledge about the congruency of neural responses across participants (i.e., consistent neural responses among proficient readers). Shifts between radial symmetry (actinomorphy) and bilateral symmetry (zygomorphy) in flowers have occurred multiple times independently within angiosperms (flowering plants). Evidence across angiosperms indicate the transcription factors RADIALIS (RAD), DIVARICATA (DIV), and CYCLOIDEA (CYC) play a major role in regulating floral symmetry and morphology. The CYC-like genes have been shown to shift their expression dorsally independently in nearly all shifts to bilaterally symmetrical flowers examined. Three core eudicot clades of CYC-like genes have been identified: CYC1, CYC2 and CYC3, with only CYC2 genes being shown to have a role in floral symmetry. This dissertation aims to investigate the racialization of my own family through the examination of migration and citizenship documents, primary sources, and oral histories as told by those who were present for or knew those present for events in question. Participants, including myself, were members of the same family, identified as either male or female, and ranged in age from 30 years to 80 years old. Participants were either monolingual English speakers or multilingual in English, Italian, and/or Sicilian. In the late 1950s and early 1960s, over 800 students from British East and Central Africa gained access to American education through Operation Airlift Africa (OAA), a visionary initiative led by Kenyan trade unionist and politician Tom Mboya. Funded by private American donors, including Senator John F. Kennedy, and supported by scholarships from U.S. institutions, the program enabled African students to attend high schools, colleges, and universities in North America, mostly in the United States. This thesis aims to elucidate the molecular mechanisms by which nucleotide binding dictates the intrinsic dynamics and to understand the functional conformational changes of Human Dynamin-1 during the GTP hydrolysis cycle through the use of Elastic Network Models (ENM). A comprehensive understanding of this process is crucial given dynamin's role in clathrin-mediated endocytosis (CME). The research utilized both experimentally derived crystal structures and AlphaFold-predicted structures of dynamin, specifically examining the GTPase and Bundle Signaling Element (BSE) domains across nucleotide-free, GTP-bound, GDP-bound, and transition states. This dissertation investigates podcasting and audio projects as multimodal practices in composition, analyzing how audio-based assignments both challenge and reproduce traditional writing norms. Drawing from personal teaching experience and a dataset of publicly accessible assignment prompts, rubrics, and instructional guides, the study examines how podcast projects are framed, assessed, and implemented in higher education. While often promoted as creative and accessible alternatives to essays, these assignments frequently replicate the same thesis-driven, rubric-centered, and exclusionary structures that define conventional academic writing. Poor nutrition, micronutrient deficiencies and malabsorption are common in individuals with SUD, potentially affecting recovery outcomes. Telomeres are conserved structures at the ends of eukaryotic chromosomes that promote genome stability. In most eukaryotes, the DNA found within these protective end caps consists of short, G-rich nucleotide repeats. Cells with critically short telomeres face a variety of potential adverse outcomes such as end-to-end chromosome fusion, apoptosis, etc. The length of telomeric DNA varies substantially between different plant species and even between genotypes of the same species. A previous study using T-DNA mutants of the model plant Arabidopsis thaliana with long or short telomeres suggested that telomere length may be an adaptive trait. Environmental factors can impact development for oviparous species during incubation. For sea turtles temperature can influence the sex ratio of the hatchlings and can decrease survival. The substrate the eggs are incubated in can differ between nests thus, impacting nests differently. This study examines the impact of sand characteristics, specifically grain size, color, calcium carbonate, change in sand surface temperature, and hydraulic conductivity, on overall nest viability in Sarasota County, Florida. This thesis aims to explore three applications of the field of marine optics. First, some marine optical principles are applied to satellite imagery to generate satellite-derived bathymetry (SDB) charts for Sarasota Bay, FL. SDB charts were successfully created using satellite images from July and August of 2023. Unique terrain features, such as the intercoastal waterway, can be seen in the SDB charts. Next, a multi-spectral light sensor system (MSLSs) was developed so it could be utilized to measure attenuation as a proxy for turbidity using marine optics concepts. Abstract

Introduction: Necrotizing enterocolitis (NEC) is a life-threatening disease in infants that causes direct damage to the intestines and requires emergent treatment. The purpose of this clinical review is to analyze the prevention of NEC in preterm neonates with breastfeeding.

Methods: PubMed was searched using a specific search strategy using the key terms “breast,” “feed,” “formula,” “necrotizing,” and “enterocolitis.” Operators and filters applied narrowed the results to 4 quality articles for review.

Results: One RCT, and two meta-analyses, reviewed had a reduction in the development of necrotizing enterocolitis among breastfed neonates. One randomized noninferiority trial totaling 123 neonates did not …

INTRODUCTION: Major depressive disorder is a debilitating mental health condition for many US adults. Traditional therapies may not be effective for many patients. The purpose of this paper is to evaluate the effectiveness of brexpiprazole, a second generation antipsychotic medication, as an adjunctive treatment for resistant MDD.

Understanding the needs of everyone in the food system is vital to solving the food waste problem. If interventions are misaligned with stakeholder needs, the resultant impact will be highly diminished and/or will result in unintended consequences.
