Background/Objectives: Peptide-based inorganic nanoparticles (PINPs) have emerged as promising candidates for intracellular delivery due to their unique structural and functional attributes. These hybrid nanostructures combine the high surface area and tunable optical/magnetic properties of metal cores (e.g., Au, Ag, Fe3O4) with the biocompatibility, targeting specificity, and responsive behavior of peptides. In particular, peptides with amphipathic or cell-penetrating features could facilitate efficient transport of molecular cargos across cellular membranes while enabling stimulus-responsive drug release in target tissues. Aims: An intrinsic feature of gene transcription is the formation of DNA superhelices near the transcription bubble, which are resolved upon induction of transient double-stranded breaks (DSBs) by topoisomerases. Unrepaired DSBs are pathogenic as they lead to cell cycle arrest, senescence, inflammation, and organ dysfunction. We posit that DSBs would be more prevalent at the genomic sites that are associated with gene expression. The objectives were to identify and characterize genome-wide DSBs at the nucleotide resolution and determine the association of DSBs with transcription in cardiac myocytes.

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy characterized by the clonal proliferation of myeloid precursors and rapid progression. Historically consisting of intensive chemotherapy, AML management has evolved significantly due to advances in molecular diagnostics and risk stratification. This review discusses current therapeutic paradigms in AML, emphasizing the growing role of personalized medicine across age and risk groups. For younger, fit patients, intensive regimens such as the "7 + 3" protocol remain the standard, often enhanced by targeted agents like FMS-like tyrosine kinase 3 (FLT3) and IDH inhibitors. Patients with myeloid neoplasms with loss-of-function TP53 mutations and erythroid differentiation have poor outcomes, and a better understanding of disease biology is required. Upregulation of interferon-γ (IFN-γ) signaling has been associated with acute myeloid leukemia (AML) progression, selection of TP53 mutated clones and chemotherapy resistance, but its drivers remain unclear. In this study, we found that the surface receptor C-C motif chemokine receptor-like 2 (CCRL2) is overexpressed in AML with erythroid differentiation and TP53 mutations compared to other AML subtypes and healthy hematopoietic cells. Acute Lymphoblastic Leukemia (ALL) is a hematologic malignancy that primarily affects children and young adults. While advancements in treatment—including chemotherapy and cranial irradiation—have significantly improved 5-year survival rates in children to nearly 90%, radiation-induced cognitive impairment (RICI) remains a persistent and debilitating side effect. The brain, particularly the hippocampus, is highly susceptible to radiation-induced oxidative stress and inflammation. In the treatment of ALL, the combined use of chemotherapy and radiotherapy contributes to a heightened oxidative burden, exacerbating the risk of cognitive dysfunction. This two-part investigation explores the dynamic behavior of braided pneumatic actuators (BPAs) under bio-inspired pulse modulation, with the aim of improving their biomimetic force output and control. The first study examines the effect of pulse length and inter-pulse timing on BPA performance, revealing that force output is highly sensitive to the temporal structure of input pulses mirroring biological muscle behavior. Using dual-pulse actuation schemes, the results demonstrate that force responses exceed the additive contributions of individual pulses, with peak amplification occurring consistently at a 27 ms inter-pulse gap. Between the 2016 and 2024 presidential elections, support for Republican candidate Donald Trump among Latine voters increased from 29% to 42%. This rise has attracted interest given Trump's nativist rhetoric and policies that target Latine immigrants. Previous literature has examined the growing Republican partisanship among Latine Americans, focusing on the processes of assimilation and the effects of racial ideology on the partisanship of the Latine electorate. Although this research is gaining traction, the political attitudes of Latine Republicans remain understudied. Ratios between two metabolites are sensitive indicators of metabolic changes. Lipidomic profiling studies have revealed that plasma ether lipids, a class of glycero- and glycerophospho-lipids with reported health benefits, are negatively associated with obesity. Here, we utilized lipid ratios as surrogate markers of lipid metabolism to explore the processes underlying the inverse relationship between ether lipid metabolism and obesity. Plasma lipidomics data from two independent human cohorts (n = 10,339 and n = 4,492) were integrated to assess the associations between 82 lipid ratios and obesity-related markers in males and females. Cannabinoid receptor 1 (CB1) has been implicated in multiple inflammatory diseases by regulating pro-inflammatory mediators or altering immune cell polarization. However, the expression and direct functional role of CB1 in T cells remain largely unexplored. Here, we demonstrate that primary murine T cells express CB1 and that its novel agonist, BI-5756, directly increases the frequencies of regulatory T cells (Tregs) in primary murine pan T cells after activation. In addition, BI-5756 exhibits an in vivo protective effect against graft-versus-host disease (GvHD), an allogeneic T cell-mediated inflammatory complication after allogeneic hematopoietic cell transplantation (allo-HCT), resulting in an improved overall survival with …

This applied research explores the accessibility, visibility, and comprehensiveness of sexual and reproductive health (SRH) education and services at universities, with a specific focus on contraceptive risk awareness. Recognizing college students as particularly vulnerable to misinformation regarding hormonal contraceptives, unintended pregnancy, and STI exposure, the study examines the role of university-provided SRH resources in mitigating these risks.

Subarachnoid hemorrhage (SAH) is a life-threatening hemorrhagic stroke caused by bleeding in the arachnoid space of the brain, often resulting in long-term disabilities and poor quality of life (QOL). Individuals living in more advantaged neighborhoods report higher levels of QOL post-stroke, but little has been reported about QOL and neighborhood data after SAH.
This study sought to examine the relationship between measures of socioeconomic status (SES) and outcomes after SAH. SES encompasses a variety of factors. To investigate the impacts of dietary Bacillus-based probiotics and yeast-derived prebiotics on the hepatic transcriptome profile, 500 Hisex White laying hens were randomly allotted into five dietary treatments from 37 to 52 weeks of age: control; control + Bacillus subtilis; control + Bacillus subtilis and Bacillus licheniformis; control + Bacillus coagulans; and control + Saccharomyces cerevisiae yeast cell wall. Transcriptome analysis revealed a substantial number of differentially expressed genes exclusively between the control and prebiotic groups, identifying 2221 genes (FDR ≤ 0.05), with 980 genes upregulated (log2 fold change 0.69 to 24.62) and 1241 downregulated (log2 fold change −0.74 to …

The fusion energy research and development landscape has seen significant advances in recent years, with important scientific and technological breakthroughs and a rapid rise of investment in the private sector. The concept of internal models dominates contemporary theories of sensorimotor control, with researchers across neurosciences, specifically motor control, routinely explaining observed behaviors through computational representations that supposedly exist within the nervous system. In this perspective, I present a critical examination of internal model frameworks in sensorimotor control. I argue that representational approaches mischaracterize biological systems for several fundamental reasons: (1) Internal models require homuncular interpreters, creating infinite regress problems; (2) The purported neural implementations of internal models remain empirically elusive despite decades of research; (3) Biological movement systems exhibit multiscale, nonlinear, and non-Gaussian dynamics that fundamentally defy reduction to conventional …

Neurofibromin/NF1 is a RAS (rat sarcoma virus) GTPase activating protein and estrogen receptor (ER) transcriptional corepressor. NF1low status, identified by copy number loss or low mRNA/protein expression is associated with endocrine therapy resistance in approximately 20% of ER+/HER2− (human epidermal growth factor receptor 2) early-stage breast cancers. The identification of targeted treatments for NF1low ER+/HER2− breast cancer is therefore a priority. In this study proteogenomic analysis of ER+/HER2− breast cancer demonstrated that NF1low tumors exhibited elevated cyclin-dependent kinase 4/6 (CDK4/6) activity. BACKGROUND: Opioid use and misuse during pregnancy rose from 1.5 to 6.5 per 1000 deliveries between 1999 and 2014 and continues as a significant public health concern. A fivefold increase in neonatal opioid withdrawal syndrome (NOWS) has accompanied the increase in opioid use. The Eating, Sleeping, Consoling care approach (ESC) has been shown to improve outcomes for infants with NOWS and is quickly becoming the standard of care for infants affected by opioid use disorder. Introduction: Research indicates a strong correlation between obesity and the risk of dementia, both are linked to steroid receptor coactivator-1 (SRC-1), a transcriptional coactivator.
Methods: We used RNA sequencing analysis (RNA-Seq) to investigate the transcriptome of SRC-1-KO mice, and identified S100 calcium-binding protein A6 (S100A6), an AD associated gene, as one target of SRC-1. We tested cognitive behaviors in SRC-1-KO mice and mice with a humanized SRC-1 mutation (SRC-1L1376P), and performed promoter luciferase assays on S100A6.
Results: Loss of SRC-1 caused alterations in gene signatures that are commonly associated with neurodegenerative diseases, including AD, and diminished the neural plasticity of …

This study explores the aerodynamic benefits of bio-inspired design modifications for autonomous drones using advanced Computational Fluid Dynamics (CFD) simulations. Four bio-inspired configurations—leading-edge serrations, winglets, riblet surfaces, and curved wings—were assessed and compared against a baseline drone model to evaluate their impact on aerodynamic performance. The results indicated that all bio-inspired designs significantly enhanced lift, reduced drag, and improved overall aerodynamic efficiency. The leading-edge serration configuration achieved the highest performance gains, with a 33.6% increase in maximum lift coefficient (CL) and a 29.5% improvement in lift-to-drag ratio (CL/CD), primarily due to delayed flow separation and reduced turbulence. Lafora disease (LD) is a fatal childhood progressive myoclonus epilepsy and glycogen storage disease that is caused by recessive mutations in either EPM2A or EPM2B. The hallmarks of LD are cytoplasmic, aberrant glycogen-like aggregates, called Lafora bodies (LBs), that drive disease progression. The 9th Annual Lafora Science Symposium was held in San Diego, California and brought together over 70 researchers, clinicians, academic trainees, and friends and family members of patients with LD and 80 attendees joined virtually. Community wildfire adaptation is happening at multiple scales across the western United States. A range of collaboratively formed groups have addressed compounding issues related to wildfire risk, forest health and land use. In Southwestern Idaho, place-based collaboratives emerging in the early 2000s have considered trade-offs between localized social and ecological needs as they approach wildfire adaptation from the bottom-up. More recently, regional partnerships have emerged that emphasize cross-boundary wildfire risk reduction to cover larger geographic areas for risk mitigation. In between these micro- and macro-scales of activity exist bridging agencies such as county and state-level programs that account for gaps …
