The Impact Of Lactobacillus Supplementation On Gut Microbiome Composition And Phage Induction In Mice, Kimia Boreiri
Dissertations
The mammalian gastrointestinal (GI) tract harbors a dense and dynamic community of microbes known as the gut microbiota, which plays vital roles in digestion, immunity, and overall health. Among these, probiotic bacteria such as Lactobacillus strains have garnered attention for their ability to influence microbial community structure and host physiology. This study investigated the effects of five Lactobacillus strains, Lactobacillus casei R1A482, Lactobacillus casei O3, Lactobacillus rhamnosus GG, Lactobacillus gasseri ATCC 33323, and Lactobacillus paragasseri UMB1065, on the gut microbiome of Swiss Webster mice over a six-week feeding period. To reduce stress, mice were fed Lactobacillus strains using gelatin pellets …
Role Of Coronavirus Envelope Protein On Infectivity, Garrett E. Zaagman
Role Of Coronavirus Envelope Protein On Infectivity, Garrett E. Zaagman
Dissertations
Coronaviruses infect a wide range of hosts and have caused recent human pandemics. While much has been learned, gaps remain in understanding viral assembly and egress, particularly the role of the envelope (E) protein in specific infectivity. To study this, recombinant MHV viruses were created with E gene deletions and HiBiT-tagged spike (S) proteins to monitor virion release. In mouse cells, MHV∆E S-HiBiT secreted less spike and fewer infectious particles, indicating reduced specific infectivity. Comparisons between human HeLa-CEACAM and mouse DBT cells showed species-specific support for virus production. TMED10, a host membrane protein, was tested as a potential E-interacting partner …

Since European settlement, vast areas of U.S. land have been modified by humans. In the Midwest, an estimated 99% of native grasslands have been replaced by agriculture and urban development. Such alternations have replaced native habitat, redefined hydrology and introduced a wide array of chemicals into aquatic ecosystems. Modern agricultural practices utilize fertilizers and pesticides to maximize crop production. These chemicals frequently runoff into surrounding waters, potentially effecting aquatic organisms. One group of organism thought to be susceptible to agricultural contaminants are amphibians. Amphibians are vulnerable to pollution due, in part, to their permeable skin and biphasic life history. Global …

Investigation Of Multiple Factors Influencing The Vector-Borne Transmission Of Salmonella Typhimurium Via Blattella Germanica, Landen Sean Van Hulzen
Dissertations and Theses
The German cockroach (Blattella germanica) is a common urban pest that lives ubiquitously in human structures around the world. German cockroaches are a severe public health threat as they are known carriers and vectors of human pathogens including Salmonella enterica, Escherichia coli, Shigella spp, and more. German cockroach infestations have been associated with Salmonellosis outbreaks. While mechanical transmission by the German cockroach vector is well known, new research suggests that more nuanced biological transmission has a role in maintaining and transmitting Salmonella enterica serovar Typhimurium (S. Typhimurium). Despite this recent finding, many biological variables influencing vector-borne transmission of S. Typhimurium …

The progress in natural language processing (NLP) using large language models (LLMs) has greatly improved patient information extraction from clinical narratives. However, most methods based on the fine-tuning strategy have limited transfer learning ability for cross-domain applications. This study proposed a novel approach that employs a soft prompt-based learning architecture, which introduces trainable prompts to guide LLMs toward desired outputs. We examined two types of LLM architectures, including encoder-only GatorTron and decoder-only GatorTronGPT, and evaluated their performance for the extraction of social determinants of health (SDoH) using a cross-institution dataset from the 2022 n2c2 challenge and a cross-disease dataset from …

Background: Colorectal cancer (CRC) is a common, fatal cancer. Identifying subgroups who may benefit more from intervention is of critical public health importance. Previous studies have assessed multiplicative interaction between genetic risk scores and environmental factors, but few have assessed additive interaction, the relevant public health measure.
Methods: Using resources from CRC consortia, including 45,247 CRC cases and 52,671 controls, we assessed multiplicative and additive interaction (relative excess risk due to interaction, RERI) using logistic regression between 13 harmonized environmental factors and genetic risk score, including 141 variants associated with CRC risk.
Results: There was no evidence of multiplicative interaction …

BACKGROUND: Immune checkpoint inhibitors play an important role in the treatment of solid tumors, but the currently used immune checkpoint inhibitors targeting programmed cell death-1 (PD-1), programmed cell death ligand-1 (PD-L1), and cytotoxic T-lymphocyte antigen-4 (CTLA-4) show limited clinical efficacy in many breast cancers. B7H3 has been widely reported as an immunosuppressive molecule, but its immunological function in breast cancer patients remains unclear.
METHODS: We analyzed the expression of B7H3 in breast cancer samples using data from the Cancer Genome Atlas Program (TCGA) and the Gene Expression Omnibus (GEO) databases. MicroRNAs were selected using the TarBase, miRTarBase, and miRBase databases. …

Bacterial bloodstream infections (BSI) can be a substantial contributor to complications of GVHD treatment. The aim of this study was to determine the risk for BSI from neutrophil engraftment through day 100 post transplant in patients with acute GVHD (AGVHD) based on organ involvement and severity. Patients (n = 4064) who underwent an allogeneic hematopoietic stem cell transplant (HCT) reported to the CIBMTR registry were analyzed. Grade II-IV AGVHD occurred in 1607 (39.5%) patients and was associated with a greater day-100 incidence of post engraftment BSI than with grade 0/I (24.9 vs. 15.3%). Patients with grade III/IV AGVHD had the …

Chemotherapy treatment-related side effects are common and increase the risk of suboptimal outcomes. Exercise interventions during cancer treatment improve self-reported physical functioning, fatigue, anxiety, and depression, but it is unclear whether these interventions improve important clinical outcomes, such as chemotherapy relative dose intensity. The National Cancer Institute funded the Exercise and Nutrition to Improve Cancer Treatment-Related Outcomes (ENICTO) Consortium to address this knowledge gap. This article describes the mechanisms hypothesized to underpin intervention effects on clinically relevant treatment outcomes, briefly outlines each project's distinct research aims, summarizes the scope and organizational structure of ENICTO, and provides an overview of the …

Cell-based immune therapies ranging from CAR-T cells to tumor infiltrating lymphocytes (TILs) and endogenous T-cell products, have produced unprecedented clinical responses in hematologic malignancies and are currently under active investigation for solid tumors. Nevertheless, several key challenges continue to limit the durability and breadth of clinical benefit. IL-7 is a pleiotropic cytokine that increases both the number and function of lymphocytes. Although not yet clinically approved, IL-7 has been used in over 620 adult and pediatric patients for a variety of reasons including, for example, to hasten bone marrow recovery after allogenic stem cell transplantation, to reverse lymphopenia due to …

The production of broiler chickens in intensive systems struggles to maintain hygiene in production, despite contributing to combating malnutrition, generating employment, and increasing family income, especially in developing countries. Using a structured questionnaire survey, a cross-sectional study was carried out among 395 smallholder farmers to evaluate their knowledge, attitudes, and practices related to farm biosecurity in urban and peri-urban areas of Mozambique and determine the factors affecting KAP (knowledge, attitude, and practice) levels. Data were analyzed using Statistical Package for Social Science (SPSS) (IBM version 27). The findings indicate that education level and training significantly influence (p < 0.001) food safety in terms of biosecurity knowledge, attitudes, and practices. Smallholders who attended food safety training exhibited positive attitudes (95.5%, mean score of 89.71 ± 10.1) in contrast to those who did not participate in training (74.8%, mean score of 79.91 ± 9.68). Additionally, washing hands with soap before and after using the toilet and wearing protective clothing during chicken husbandry were significantly associated with education level (χ² = 15.345, P = 0.018); those with informal education were 34% less likely to have a negative attitude toward food safety compared to those with high education. A significant correlation (p < 0.01) was observed between residence area, training, and biosecurity knowledge, attitude, and practice. Because smallholders lack training, hygiene expertise, and biosecurity practices on their farms, this study concluded that they pose a risk to public health and food safety. To maintain consumer health and guarantee food safety, farmers should properly dispose of waste, including dead chickens, and receive continuous training in excellent biosecurity measures on the farm

Intensive production systems face challenges in treating individual chickens due to the large number of animals, resulting in the indiscriminate use of antibiotics as the only alternative. The use of antibiotics in poultry production has adverse effects on humans, as prolonged consumption of contaminated meat can lead to the development of antibiotic resistance in intestinal bacteria. A cross-sectional survey involving 395 smallholder farmers was conducted through face-to-face interviews to assess knowledge, attitudes, and practices (KAP) regarding antibiotic use in urban and peri-urban areas of Southern Mozambique. Data were analyzed using SPSS version 27. The findings revealed that 60% of respondents …

Rift Valley fever virus (RVFV) is an emerging mosquito-borne arbovirus of One Health importance that caused two large outbreaks in Rwanda in 2018 and 2022. Information on vector species with a role in RVFV eco-epidemiology in Rwanda is scarce. Here we sought to identify potential mosquito vectors of RVFV in Rwanda, their distribution and abundance, as well as their infection status. Since an outbreak of RVF occurred during the study period, data were obtained both during an interepidemic period and during the 2022 Rwanda RVF outbreak. Five districts of the eastern province of Rwanda were prospected using a combination of …

Irish potato (Solanum tuberosum) is a critical crop for food security and economic growth in Tsangano and Angónia Districts, Central Mozambique. Challenges like inconsistent yields and variable quality are often linked to suboptimal soil conditions, which limit production. This study aimed to classify and evaluate the suitability of soils for potato cultivation in Tete Province, where detailed soil assessments remain limited. Four pedons—TSA-P01 and TSA-P02 in Tsangano and ANGO-P01 and ANGO-P02 in Angónia—were examined for bulk density, texture, pH, organic carbon, and nutrient content using a combination of pedological methods and laboratory soil analysis. To determine each site’s potential for …

Dhanaan is a spontaneously fermented camel milk that is popular in the Somali regional state of Ethiopia and plays a sig- nificant role in the livelihoods of pastoral communities. This study aimed to investigate traditional fermentation methods of dhanaan in these communities. Four districts, namely, Gursum, Shabelle, Degehabur, and Birqot, were purposively selected on the basis of camel rearing, camel milk production, and consumption. A cross-sectional survey of traditional fermentation methods, handling, utilization, and the claimed therapeutic effects of dhanaan was conducted using both open- and closed- ended questions in the districts. The traditional preparation of dhanaan involves spontaneous fermentation …

[Introduction] The Transforming Growth Factor beta (TGF-beta) superfamily, encompassing molecules such as TGFs, activins, Bone Morphogenetic Proteins (BMPs), Growth/Differentiation Factor (GDFs) and Nodals, represents the largest family of growth and differentiation factors, playing crucial roles in developmental and physiological processes across animal species (1). These molecules are integral to tissue homeostasis and cell fate determination. Among them, TGF-beta is particularly noted for its regulatory influence on immune responses and tissue fibrosis (2, 3). Recent research has expanded our understanding of the immune functions of other superfamily members, including activin A and BMPs (4). These molecules signal through receptor complexes composed …

Background: Podcasts are a widely available and popular educational medium for healthcare providers, residents, students, and patients. Podcasts are convenient, accessible, and may be effective education tools for listeners. While some studies have attempted to measure the effectiveness of podcasting to enhance knowledge, there are few studies evaluating podcasts in pharmacy education. The purpose of this project was to provide students with the opportunity to create a podcast episode through an in-class, semester-long assignment and to evaluate student perceptions of knowledge, confidence, and professional identity.
Educational activity: Over the course of a semester-long elective class, students worked in groups to …
