Abstract: Exercise is a potent stimulus for combatting skeletal muscle ageing. To study the effects of exercise on muscle in a preclinical setting, we developed a combined endurance–resistance training stimulus for mice called progressive weighted wheel running (PoWeR). PoWeR improves molecular, biochemical, cellular and functional characteristics of skeletal muscle and promotes aspects of partial epigenetic reprogramming when performed late in life (22–24 months of age). Chronic spinal cord injury (SCI) lesions retain increased densities of microglia and macrophages. In acute SCI, macrophages induce growth cone collapse and facilitate axon retraction away from lesion boundaries. Little is known about the role of sustained inflammation in chronic SCI or whether chronic inflammation affects regeneration. We used the colony-stimulating factor-1 receptor inhibitor, PLX-5622, to deplete microglia and macrophages months after complete crush SCI in female mice. Transcriptional analyses revealed a significant inflammatory depletion within chronic SCI lesions after PLX-5622 treatment. Significance: Cerebral blood flow (CBF) imaging is crucial for diagnosing cerebrovascular diseases. However, existing large neuroimaging techniques with high cost, low sampling rate, and poor mobility make them unsuitable for continuous and longitudinal CBF monitoring at the bedside. Aim: We aimed to develop a low-cost, portable, programmable scanning diffuse speckle contrast imaging (PS-DSCI) technology for fast, high-density, and depth-sensitive imaging of CBF in rodents. The second-generation myosin activator danicamtiv (DN) has shown improved function compared with the first-generation myosin activator omecamtiv mecarbil (OM) in nonfailing myocardium by enhancing cardiac force generation but attenuating slowed relaxation. However, whether the functional improvement with DN compared with OM persists in remodeled failing myocardium remains unknown. Therefore, this study aimed to investigate the differential contractile responses to myosin activators in nonfailing and failing myocardium. Background: Most tumors initially respond to treatment, yet refractory clones subsequently develop owing to resistance mechanisms associated with cancer cell plasticity and heterogeneity. Methods: We used a chemical biology approach to identify protein targets in cancer cells exhibiting diverse driver mutations and representing models of tumor lineage plasticity and therapy resistance. An unbiased screen of a drug library was performed against cancer cells followed by synthesis of chemical analogs of the most effective drug. Introduction: Moderate-to-severe traumatic brain injury (TBI) results in an early loss of immature hippocampal granule cells and the activation of typically quiescent neural stem cells (NSCs) in the dentate gyrus. Activation of NSCs leads to a robust increase in proliferation and generation of neural progenitor cells (NPCs), supporting restoration of the immature neuron population of over a period of 1–2 weeks. However, it is unclear if neurons born early after injury develop normally, survive long-term and functionally integrate into the hippocampal network. The fusion of skeletal muscle stem cell (MuSC) to myofibers during hypertrophy has exclusively focused on the transfer of the MuSC nucleus, leaving the fate of other MuSC organelles, such as mitochondria, largely unexplored. The objective of this study was to determine whether MuSCs transfer their mitochondria upon myofiber fusion in response to a hypertrophic stimulus. To achieve this goal, we specifically labeled MuSC mitochondria with Dendra2 fluorescence by crossing the MuSC-specific CreER (Pax7CreER/CreER) mouse with the Rosa26-Dendra2 mouse to generate the Pax7-Dendra2 mouse. Dominant notions of pregnancy frame gestation as a solely female process. However, the experiences of transmasculine birthers challenge this assumption, demonstrating that pregnancy is not inherently tied to womanhood but is instead shaped by social and cultural narratives and representations. This paper explores the unique pregnancy and parental exercises of transmasculine birthers by examining public social media posts under the hashtag #seahorsedad on Instagram. Utilizing metaphorical criticism, I aim to uncover how transmasculine birthers, known within the community as “seahorse dads,” both uphold and resist traditional notions of gender, pregnancy, and parenting through a combination of textual and visual metaphor. …

Research suggests that nature connection is positively correlated with pro-environmental behaviors, concern for the climate, general wellbeing, and subjective levels of climate change distress. Few research studies have examined how to increase nature connectedness without increasing psychological distress. Such research is especially important for those who are disproportionately affected by climate change such as Indigenous Peoples. This project investigates how Indigenous Data Sovereignty protocols apply to environmental data collected on Tribal lands by federal and state entities. The 2011 and 2014 flooding of the Missouri River and a need for further soil moisture data led the United States Army Corps of Engineers Omaha District to implement a Mesonet weather station monitoring network across the Upper Missouri River Basin (UMRB). Post-mortem interval (PMI) estimation is a central component of forensic anthropological investigation, but traditional methods for estimating time since death lose accuracy after ~72 hours. Recent research indicates that some steroid hormones may undergo predictable degradation patterns after death, offering potential use as biochemical clocks for PMI estimation beyond this threshold. Dehydroepiandrosterone (DHEA), and its sulfate form (DHEA-S) are of particular interest due to their metabolic stability, systemic influence, and association with trauma. Single-walled carbon nanotubes (SWCNTs) possess unique physicochemical and optical properties that make them ideal candidates for biomedical imaging, biosensing, and disease diagnostics. This thesis explores the potential of SWCNT-based spectral fingerprinting combined with ML (Machine Learning) algorithms to optimize bioimaging, disease detection and prediction, with a specific focus on differentiating between healthy and amyotrophic lateral sclerosis (ALS) lymphoblastic patient samples. By functionalizing SWCNTs with single-stranded DNA, we enhance their stability and target specificity, enabling their application in serum patient samples.

Attention-Deficit/Hyperactivity Disorder (ADHD) is a widespread psychological disorder in children, poorly impacting executive functioning domains of inhibitory control and working memory. Event-related potentials (ERPs) provide neuroelectric insight into these cognitive processes. However, most ERP research focuses on one task type, limiting direct comparisons of cognitive demands across different task paradigms. This study examined P3 amplitude and latency in ADHD children using a visual NoGo task (response inhibition) and an auditory oddball task (working memory), and associations between the P3 ERP and behavioral measures of executive functions.

In the United States, turfgrass has replaced native landscapes in over 40 million acres, with 500 square miles added each year (Tallamy, 2019). Fecal bacterial contamination shapes how communities manage and use estuarine waters, with consequences for ecosystem health and public access. In Rhode Island's Narrow River, elevated concentrations of fecal coliforms and enterococci have restricted shellfishing for decades despite ongoing management efforts. From 2004 to 2018, local organizations implemented Best Management Practices (BMPs) to address water quality challenges, yet their effectiveness remains incompletely understood. This includes a system for manipulating coral fragments and affixing them to a cement base plug. images mimicking real life or even particular art styles. Yet for all their capability, casually
training a GAN on an average machine can be infeasible as GANs require an enormous amount

The incidence of obesity related glomerulopathy (ORG) is rising worldwide with very limited treatment methods. Paralleled with the gut-kidney axis theory, beneficial effects of butyrate, one of short-chain fatty acids produced by gut microbiota, on metabolism and certain kidney diseases have gained growing attention. Ensuring accurate traffic perception and road safety in complex urban environments remains a significant challenge. Advanced traffic monitoring increasingly relies on deep learning, which requires large data volumes. However, existing datasets are often limited to CCTV video footage or focus on dynamic scenarios captured by sensors mounted on ego vehicles. This study shows that populations of CAFs have distinct effects on pancreatic cancer progression and shows that depletion of CAFs expressing adipose markers potentiates tumor/metastasis suppression effects of immune checkpoint blockade.

Presented here is the case of a 68-year-old woman with blastic plasmacytoid dendritic cell neoplasm (BPDCN) treated with tagraxofusp (TAG) maintenance therapy post-allogeneic hematopoietic stem cell transplantation (allo-HCT). Prior to allo-HCT, the patient was treated with hydroxyurea and mini-CVD (cyclophosphamide, vincristine, and dexamethasone alternating with methotrexate (Methotrexate) and cytarabine) + venetoclax + TAG for 5 cycles, which induced morphologic complete remission with minimal residual disease. After allo-HCT, the patient had persistent cytogenic abnormalities 45,XX,der(7)add(7)(p13)del(7)(q11.2q22)add(7)(q32),add(12)(p13),-15,del(16)(q23),-17,+22,+2mar[1]/46,XX[19], and was then treated with TAG maintenance therapy at 9 mg/kg on a 28-day cycle for 16 cycles. Testing The Waters: Expanding Chapter 9 Bankruptcy To Encourage Treatment Of Chemically Contaminated Drinking Water, Alexandra Zimmer

Chemical contamination of drinking water supplies has become a significant issue across the globe with serious health and safety impacts. While the true extent of the impact is still being determined, costs associated with remediation efforts to clean up are astonishing. Municipalities, in particular cities, towns, and counties, suffer significant damages both through costs incurred directly for treatment of chemically contaminated drinking water supplies and through lost revenues resulting from municipal residents’ exposure.
