Enterococci have evolved resistance mechanisms to protect their cell envelopes against bacteriocins and host cationic antimicrobial peptides (CAMPs) produced in the gastrointestinal environment. Activation of the membrane stress response has also been tied to resistance to the lipopeptide antibiotic daptomycin. However, the actual effectors mediating resistance have not been elucidated. Here, we show that the MadRS (formerly YxdJK) membrane antimicrobial peptide defense system controls a network of genes, including a previously uncharacterized 3-gene operon (madEFG) that protects the Enterococcus faecalis cell envelope from antimicrobial peptides. Constitutive activation of the system confers protection against CAMPs and daptomycin in the absence of …

Zika virus (ZIKV) is unique among orthoflaviviruses in its vertical transmission capacity in humans, yet the underlying mechanisms remain incompletely understood. Here, we show that ZIKV induces tunneling nanotubes (TNTs) in placental trophoblasts which facilitate transfer of viral particles, proteins, mitochondria, and RNA to neighboring uninfected cells. TNT formation is driven exclusively via ZIKV non-structural protein 1 (NS1). Specifically, the N-terminal 1-50 amino acids of membrane-bound ZIKV NS1 are necessary for triggering TNT formation in host cells. Trophoblasts infected with TNT-deficient ZIKV

Effects Of Acute Pro-Inflammatory Stimulation And 25-Hydroxycholesterol On Hippocampal Plasticity And Learning Involve Nlrp3 Inflammasome And Cellular Stress Responses, Yukitoshi Izumi, Kazuko A O'Dell, Steven Mennerick, Charles F Zorumski

2020-Current year OA Pubs

Neuroinflammation is an increasingly important target for therapeutics in neuropsychiatry and contributes to cognitive dysfunction, disability and death across a range of illnesses. We previously found that acute effects of pro-inflammatory stimulation with lipopolysaccharide (LPS) on hippocampal long-term potentiation (LTP), a form of synaptic plasticity involved in learning and memory, requires synthesis of the oxysterol, 25-hydroxycholesterol (25HC) and exogenous 25HC mimics effects of LPS. However, downstream mechanisms engaged by LPS and 25HC remain uncertain. Here we use rat hippocampal slices and in vivo behavioral studies to provide evidence that acute modulation of synaptic plasticity by both LPS and 25HC requires …

The Important Role Of Pollinators In Sustainability Education, Bronwen Cowie, Paula Mildenhall

Research outputs 2022 to 2026

In this chapter we illustrate some of the ways the education sector can play a key role, not just in educating and activating students as advocates for and agents of sustainability but also in motivating and mobilising communities towards a sustainability agenda. Initially in this chapter it was important to look back and identify how important bees have been through the ages, noting that humans identified very early on that bees could enhance their lives. We then outline some of the contributions a focus on bees can make to sustainability education. One important contribution includes fostering a balanced perspective on …

An Attention-Based Model For Recognition Of Facial Expressions Using Cnn-Bilstm, Sushil Kumar Singh, Manish Kumar, Ikram Majeed Khan, A. Jayanthiladevi, Chirag Agarwal

Polytechnic Journal

In recent studies, computer vision and human-computer interaction have focused heavily on facial expression recognition (FER). Traditional deep learning algorithms have faced significant challenges when processing images with occlusion, uneven lighting, and positional inconsistencies and addressing dataset imbalances. These issues often result in low accuracy, unreliable recognition rates, and slow convergence. To address the challenges posed by non-frontal visual features, this study proposes a hybrid model that fusion of Convolutional Neural Networks (CNN) and Bidirectional Long Short-Term Memory (BiLSTM), augmented with a point-multiplication attention model and Linear Discriminant Analysis (LDA). Data preparation incorporates median filtering and global contrast normalization to …

Antibiotic (ABX)–induced microbiome dysbiosis is widespread in oncology, adversely affecting outcomes and side effects of various cancer treatments, including immune checkpoint inhibitors and chimeric antigen receptor T-cell (CAR-T) therapies. In this study, we observed that prior exposure to broad-spectrum ABXs with extended anaerobic coverage such as piperacillin-tazobactam and meropenem was associated with worse anti-CD19 CAR-T therapy survival outcomes in patients with large B-cell lymphoma (N = 422) than other ABX classes. In a discovery subset of these patients (n = 67), we found that the use of these ABXs was in turn associated with substantial dysbiosis of gut microbiome function, …

Genetic mutations in apoptosis-inducing factor (AIF) have a strong association with mitochondrial disorders; however, little is known about the aberrant splicing variants in affected patients and how these variants contribute to mitochondrial dysfunction and brain development defects. We identified pathologic AIF3/AIF3-like splicing variants in postmortem brain tissues of pediatric individuals with mitochondrial disorders. Mutations in AIFM1 exon-2/3 increase splicing risks. AIF3-splicing disrupts mitochondrial complexes, membrane potential, and respiration, causing brain development defects. Mechanistically, AIF is a mammalian NAD(P)H dehydrogenase and possesses glutathione reductase activity controlling respiratory chain functions and glutathione regeneration. Conversely, AIF3, lacking these activities, disassembles mitochondrial complexes, increases …

Future Climate-Driven Habitat Loss And Range Shift Of The Critically Endangered Whitefin Swellshark (Cephaloscyllium Albipinnum), Kerry Brown, Robert Puschendorf

School of Biological and Marine Sciences

Climate change is driving many species to shift their geographical ranges poleward to maintain their environmental niche. However, for endemic species with restricted ranges, like the Critically Endangered whitefin swellshark (Cephaloscyllium albipinnum), endemic to southeastern Australia, such dispersal may be limited. Nevertheless, there is a poor understanding of how C. albipinnum might spatially adjust its distribution in response to climate change or whether suitable refugia exist for this species in the future. Therefore, to address this gap, this study utilised maximum entropy (MaxEnt) modelling to determine the potential distribution of suitable habitat for C. albipinnum under present-day (2010–2020) climate conditions …

Shalom: A Judeo-Christian Response To Greed, Kwok Tung Cheung Feb 2025

Shalom: A Judeo-Christian Response To Greed, Kwok Tung Cheung

Journal of Religion and Business Ethics

In my recent conceptual analysis of greed, I argue that there are three different conceptual aspects, namely, a consuming desire to acquire more and more (the psychological), an inordinate love for material possession and power (the metaphysical), and an action of acquiring more than one’s share by taking what belongs to one’s fellow citizens, essentially with a calculating indifference toward others (the socio-historical). The aim of that analysis, as a foundational work, is to make sense of the vice or the moral mistake of greed in the contemporary world, especially by reviving the socio-historical aspect which is about justice. Building …

The Dirty Work Of Faith-Washing In A Faith-Friendly Business, Cathy Driscoll Feb 2025

The Dirty Work Of Faith-Washing In A Faith-Friendly Business, Cathy Driscoll

Journal of Religion and Business Ethics

Drawing on faith/spirituality at work and organizational legitimacy literatures, a case study of a large global meat company is used to deconstruct the concept of faith-friendly business and explore how faith-washing can provide a business with legitimacy and potentially distract stakeholders from irresponsible activities. In particular, I focus on the faith-friendly communications and initiatives in relation to treatment of non-salaried employees as a primary stakeholder of the company. I highlight a disconnect from religious social thought on meaningful and dignified work and many of the concepts found in the modern faith/spirituality at work literature, as well as societal expectations on …

A Long-Term Study Of Algal Blooms In Alexandria's Ecosystems: A Review, Samia K. Mikhail, Wagdy Labib

Blue Economy

Coastal ecosystems in Alexandria are facing significant stress, primarily due to the intrusion of discharged wastewaters of different origins. Over the past three decades, there has been a noticeable increase in the occurrence of harmful algal blooms (HABs) across Alexandria marine, brackish, and freshwater environments. Unfortunately, the situation is deteriorating, and HABs have become chronic issues characterized by frequent occurrences of various causative species. This phenomenon has spread widely affecting previously uninfected coastal areas with high frequency and diverse patterns of occurrence. This article provides an overview of the state of visible HABs associated with incidents of fish and/or invertebrate …

Shock Effects Including Maskelynite Formation In Three Shergottite Meteorites From Mars, George Ayeh Anim Feb 2025

Shock Effects Including Maskelynite Formation In Three Shergottite Meteorites From Mars, George Ayeh Anim

Dissertations and Theses

Martian meteorites result from large impact events on the surface of Mars that cause intense shock metamorphism. This study examines the shock metamorphic features of three shergottite meteorites, Dho 019, NWA 12241, and NWA 15628, to better understand shock impact events. The research uses petrographic analysis, scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and electron backscatter diffraction (EBSD) to analyze the meteorites. A key focus of this research is the formation of maskelynite, a glass with plagioclase composition, whose formation mechanism has been the substance of debate among researchers. This study proposes that maskelynite forms through a transient intermediary …

Commensality In The Eateries Of Roman Pompeii, Amaya Yolanda Eckersley Feb 2025

Commensality In The Eateries Of Roman Pompeii, Amaya Yolanda Eckersley

Dissertations and Theses

Despite a nearly 300-year excavation history at Pompeii and many identified eating and drinking establishments, social behavior in these public or semi-public spaces has eluded significant research. Elite Roman literature is the traditional guideline for understanding Roman mealtimes and what they meant for social behavior. This literature, however, fails to accurately represent its sub-elite inhabitants. These sub-elites of Rome largely had their meals prepared outside of their homes. Pompeii is no exception and provides a massive boon for researchers willing to consider archaeological evidence. The buildings and evidence buried at Pompeii have ultimately shown a wide array of tabernae related …

BACKGROUND: Chronic alcohol consumption leads to lipid accumulation, oxidative stress, cellular damage, and inflammation in the liver, collectively referred to as alcohol-associated liver disease (ALD). FAF2/UBXD8/ETEA (Fas-associated factor 2) is a ubiquitin ligase adaptor protein that plays a crucial role in the ubiquitin-mediated degradation of misfolded proteins in the endoplasmic reticulum. A recent genome-wide association study indicated an association between FAF2 and ALD; however, the exact contribution of FAF2 to ALD pathogenesis remains unclear. METHODS: FAF2 was knocked down using AAV-delivered shRNA in C57/BL6 mice. Mice were subjected to a chronic-plus-single binge ethanol feeding (NIAAA) model. Nine hours after gavage, …

Cornhusker Economics

The research aims to assess consumer preferences and attitudes toward CRISPR applications in the food sector and estimate consumer willingness to pay for a food product developed using CRISPR technology to enhance food safety and confer health benefits. Additionally, the research assesses the impact of information provision on perceptions, attitudes and willingness to pay. Specifically, we consider the potential effect of both the nature of information provided (i.e., whether it emphasizes the differences between GM and CRISPR technology or not) and the delivery format (i.e., text versus video) used to present content-equivalent information. In this context, we also assess consumer …

We measured the BASAL breathing frequency following an overnight fast in adult,non-pregnant/non-lactating, inactive mammals ranging in body mass from 15 to5520 kg. The data included results from 338 individual animals from 34 speciesthat were divided into terrestrial, semi-aquatic (Otariidae and Phocidae) and aquaticmammals. Following attempts to limit the collection of breathing frequency usinga basal definition and to correct the analysis phylogenetically, our results suggestthat there are differences in the allometric mass-exponent between terrestrial andaquatic/semi-aquatic mammals. An allometric regression model, whereby both bodymass and breathing frequency were transformed using log10 , suggested that theallometric mass exponent for terrestrial mammals (−0.303) was …

Recent studies have highlighted the crucial role of microglia (MG) and their interactions with the gut microbiome in post-stroke neuroinflammation. The activation of immunoregulatory pathways, including the aryl hydrocarbon receptor (AHR) pathway, is influenced by a dynamic balance of ligands derived from both the host and microbiota. This study aimed to investigate the association between stroke-induced dysbiosis and the resultant imbalance in AHR ligand sources (loss of microbiota-derived [indole-based] and increase of host-derived [kynurenine-based]) after stroke. Microbiota-derived AHR ligands decreased in human plasma and remained low for days following an ischemic stroke highlighting the translational significance. Transient-middle-cerebral-artery-occlusion was performed in …

Multispecies swards have been recognized as more sustainable than traditional ryegrass systems, in addition to providing a range of phytochemicals to the ruminant diet. In this regard, polyphenols may be linked to higher levels of bioactive fatty acids in milk and enhanced nutraceutical properties in dairy products. This study compares the fat composition and oxidative stability of milk produced by cows fed on multispecies swards (MULTI) with milk from two conventional swards [a perennial ryegrass monoculture (PRG) and a mixture of perennial ryegrass and white clover (PRG + WC)] during the Irish summer. Milk from cows fed on MULTI had …
