Hall bar devices fabricated on single quantum wells revealed that surface preparation can tune transport properties while maintaining peak mobilities around 105 cm2V−1s−1. Manganese-germanide spintronic contacts were integrated via solid-state reaction, with contact quality assessed through Schottky diodes, transfer length …

Hypoxia-Inducible Factor 3Α1'S Role In Promoting Iron Loading, Tumorigenesis, And Metastasis In Colorectal Cancer, Luke B. Villareal Dec 2024

Hypoxia-Inducible Factor 3Α1'S Role In Promoting Iron Loading, Tumorigenesis, And Metastasis In Colorectal Cancer, Luke B. Villareal

Biomedical Sciences ETDs

The interplay between hypoxia and iron metabolism drives colorectal cancer (CRC) progression. This dissertation investigates hypoxia-inducible factor 3α1 (HIF-3α1), a protein linked to poor CRC outcomes, focusing on its roles in metastasis, epithelial-to-mesenchymal transition (EMT), and iron loading via transferrin receptor (TFRC). HIF-3α1 promotes CRC cell migration by upregulating zinc finger E-box binding homeobox 2 (ZEB2) and suppressing N-myc downstream-regulated gene 1 (NDRG1), while TFRC-mediated iron accumulation supports tumor growth and survival. Iron chelation reduced EMT markers, migration, and tumor growth, highlighting iron's critical role. Additionally, HIF-3α1-induced TFRC expression altered nucleotide synthesis enzymes carbamoyl-phosphate synthetase-aspartate transcarbamoylase-dihydroorotase (CAD) and phosphoribosyl pyrophosphate …

Synthesis Of Glycan Antigens For Therapeutic Antibodies And Glycoconjugate Vaccines, Synthesis Of An Inflammatory Pentasaccharide, And Design And Synthesis Of Ceragenins For Use In Medical Devices And Osteomyelitis, Shawn Gubler

Theses and Dissertations

A promising approach in combatting infections from antibiotic resistant bacteria/fungi is using therapeutic monoclonal antibodies (mAbs) that can bind the outer surfaces of microbes. We have developed a synthetic vaccine platform that can rapidly generate mAbs against surface glycans from desired strains, such as lipopolysaccharides and capsular polysaccharides. To create therapeutic antibodies against Ruminococcus gnavus, Burkholderia multivorans, and Staphylococcus aureus, we synthesize glycan antigens from each strain that are necessary for use in our glycoconjugate vaccine platform. Blooms of the Gram-positive bacterium Ruminococcus gnavus have been correlated with inflammatory bowel disease, and recently a polysaccharide produced by this organism was …

Exploring The Accuracy Of Interferometric Quantum Measurements Under Conservation Laws, Nicolò Piccione, Maria Maffei, Andrew N. Jordan, Kater W. Murch, Alexia Auffèves

Mathematics, Physics, and Computer Science Faculty Articles and Research

A (target) quantum system is often measured through observations performed on a second (meter) system to which the target is coupled. In the presence of global conservation laws holding on the joint meter-target system, the Wigner-Araki-Yanase theorem and its generalizations predict a lower bound on the measurement’s error (Ozawa’s bound). While practically negligible for macroscopic meters, it becomes relevant for microscopic ones. Here, we propose a simple interferometric setup, arguably within reach of present technology, in which a flying particle (a microscopic quantum meter) is used to measure a qubit by interacting with it in one arm of the interferometer. …

Closed Domain Question Answering With Language Models: Application Of Retrieval-Augmented Generation And Parameter Efficient Fine-Tuning In Healthcare, Aaron Cummings Dec 2024

Closed Domain Question Answering With Language Models: Application Of Retrieval-Augmented Generation And Parameter Efficient Fine-Tuning In Healthcare, Aaron Cummings

Master's Theses

Dementia care presents significant challenges for informal caregivers, particularly in managing behavioral symptoms that affect over 90% of individuals with Alzheimer’s Disease and Related Dementias (ADRD) during the moderate-to-severe stages. These symptoms, including agitation, wandering, and repetitive activities, impose emotional and physical burdens on caregivers, often exacerbated by a lack of reliable, accessible, and personalized resources. Non-pharmacological interventions, while evidence-based, are underutilized due to knowledge gaps and the inefficiency of traditional training and information retrieval methods.

This research explores the adaptation of large language models (LLMs) to address these challenges by developing a framework for closed-domain Question Answering (QA) systems, …

Analysis Of Natural Structures And Chemical Mapping Data Reveals Local Stability Compensation In Rna, Robert L. Cornwell-Arquitt, Riley Nigh, Michael T. Hathaway, Joseph D. Yesselman, David A. Hendrix

Department of Biochemistry: Faculty Publications

RNA molecules adopt complex structures that perform essential biological functions across all forms of life, making them promising candidates for therapeutic applications. However, our ability to design new RNA structures remains limited by an incomplete understanding of their folding principles. While global metrics such as the minimum free energy are widely used, they are at odds with naturally occurring structures and incompatible with established design rules. Here, we introduce local stability compensation (LSC), a principle that RNA folding is governed by the local balance between destabilizing loops and their stabilizing adjacent stems, challenging the focus on global energetic optimization. Analysis …

Stellar Properties From A Self-Consistent Analysis, Pius Privatus, Pulapa V Kanaka Rao, Cirino Pappalardo

Tanzania Journal of Science

Galaxies’ spectral energy distribution (SED) has been explored through the use of spectral synthesis codes. Most of the codes so far are solely stellar and assume a negligible nebular contribution. Fitting Analysis using Differential Evolution Optimisation (FADO) is the first spectral synthesis tool to consider the nebular contribution. The study examines how the primary stellar attributes are affected when FADO is applied to the Max Planck Institute for Astrophysics and Johns Hopkins University (MPA-JHU) data, namely the star-formation rate, metallicity, and stellar mass. The findings show that nebular emission results in the change of the slope of the equation of …
