Structural Variant Allelic Heterogeneity In Mecp2 Duplication Syndrome Provides Insight Into Clinical Severity And Variability Of Disease Expression, Davut Pehlivan, Marwan S Shinawi, Et Al.

2020-Current year OA Pubs

BACKGROUND: MECP2 Duplication Syndrome, also known as X-linked intellectual developmental disorder Lubs type (MRXSL; MIM: 300260), is a neurodevelopmental disorder caused by copy number gains spanning MECP2. Despite varying genomic rearrangement structures, including duplications and triplications, and a wide range of duplication sizes, no clear correlation exists between DNA rearrangement and clinical features. We had previously demonstrated that up to 38% of MRXSL families are characterized by complex genomic rearrangements (CGRs) of intermediate complexity (2 ≤ copy number variant breakpoints <  5), yet the impact of these genomic structures on regulation of gene expression and phenotypic manifestations have not been investigated.

METHODS: To study the role of the genomic rearrangement structures on an individual's clinical phenotypic variability, we employed a comprehensive …

Microglial Cd2ap Deficiency Exerts Protection In An Alzheimer's Disease Model Of Amyloidosis, Lingliang Zhang, David M. Holtzman, Et Al.

2020-Current year OA Pubs

BACKGROUND: The CD2-associated protein (CD2AP) was initially identified in peripheral immune cells and regulates cytoskeleton and protein trafficking. Single nucleotide polymorphisms (SNPs) in the CD2AP gene have been associated with Alzheimer's disease (AD). However, the functional role of CD2AP, especially its role in microglia during AD onset, remains elusive.

METHODS: CD2AP protein levels in cultured primary cells and in 5xFAD mice was studied. Microglial CD2AP-deficient mice were crossed with 5xFAD mice and the offspring were subjected to neuropathological assessment, behavioral tests, electrophysiology, RNA-seq, Golgi staining, and biochemistry analysis. Primary microglia were also isolated for assessing their uptake and morphology changes. …

Primary Anorectal Mammary-Like Adenocarcinoma: A Potential Diagnostic Pitfall With Conventional Colorectal Adenocarcinoma, Margaret L Axelrod, Xiuli Liu, Pooja Navale

2020-Current year OA Pubs

Anogenital mammary-like glands (AGMLGs) are present in the anogenital region that bear striking morphologic and protein-expression similarities to mammary glands in the breast. AGMLGs can give rise to both benign and malignant lesions which mimic primary breast lesions. Herein, we report two mammary-type adenocarcinomas arising from AGMLGs, including one in the previously unreported site of the rectum. Recognition of mammary-type adenocarcinoma in the rectal and anogenital regions is crucial as clinical management options may differ compared to conventional colorectal adenocarcinomas.

A Topological Route To Engineering Robust And Bright Supersymmetric Laser Arrays, Soujanya Datta, Mohammad Alizadeh Fard, Ramy El-Ganainy, Krishanu Roychowdhury

Michigan Tech Publications

In recent years, several proposals that leverage principles from condensed matter and high-energy physics for engineering laser arrays have been put forward. The most important among these concepts are topology, which enables the construction of robust zero-mode laser devices, and supersymmetry (SUSY), which holds the potential for achieving phase locking in laser arrays. In this work, we show that the relation between supersymmetric coupled bosonic and fermionic oscillators on one side, and bipartite networks (and hence chiral symmetry) on another side can be exploited together with non-Hermitian engineering for building one- and two-dimensional laser arrays with in-phase synchronization. To demonstrate …

Social Learning And Cultural Enrichment For Fish Welfare, Chiawen Chiang, Becca Franks Dec 2024

Social Learning And Cultural Enrichment For Fish Welfare, Chiawen Chiang, Becca Franks

Animal Sentience

This commentary examines how understanding animal culture—shared behaviors and knowledge transmitted through social learning—plays a role in advancing welfare practices for captive fishes. Despite extensive evidence of fishes' complex sociocultural capacities, their psycho-social needs remain underestimated and unmet in zoos and aquaria. We explore what considering fish culture might mean for welfare practices: diverse social learning opportunities, cultural development, and thoughtful group management. More knowledge about wild fishes' lives will help bridge the gap between their full, complex lives and current captive welfare practices.

Beyond The Surface: Understanding Temperature Polarisation In Dcmd Membranes Through Varied Operational Parameters, Hiras Ahamed Hijaz, Masoumeh Zargar, Abdellah Shafieian, Amir Razmjou, Mehdi Khiadani

Research outputs 2022 to 2026

Conventionally, a single Temperature Polarisation Coefficient (TPC) value is calculated to quantify Temperature Polarisation (TP). In this research, the extent of polarisation is investigated by capturing temperature profiles at specific points along a MD membrane using miniature thermocouples, eliminating the need for TPC calculations. The extent of polarisation at a point is affected by two contributory factors, namely the proximity of flow inlets and the difference in vapour pressure across the membrane at that point. Under this direction, this work examined the influence of permeate temperature, feed salinity, and flow direction on the development of the temperature profiles. Our analysis …

Pyrophosphate-Free Glycolysis In Clostridium Thermocellum Increases Both Thermodynamic Driving Force And Ethanol Titers, Bishal Dev Sharma, Shuen Hon, Eashant Thusoo, David M. Stevenson, Daniel Amador-Noguez, Adam M. Guss, Lee R. Lynd, Daniel G. Olson

Dartmouth Scholarship

Background

Clostridium thermocellum is a promising candidate for production of cellulosic biofuels, however, its final product titer is too low for commercial application, and this may be due to thermodynamic limitations in glycolysis. Previous studies in this organism have revealed a metabolic bottleneck at the phosphofructokinase (PFK) reaction in glycolysis. In the wild-type organism, this reaction uses pyrophosphate (PPi) as an energy cofactor, which is thermodynamically less favorable compared to reactions that use ATP as a cofactor. Previously we showed that replacing the PPi-linked PFK reaction with an ATP-linked reaction increased the thermodynamic driving force of glycolysis, but only had …
