Make America Healthy: Reducing High Pharmaceutical Prices Without Reducing Innovation, Srividhya Ragavan

SMU Law Review

Cost of medication in the United States has historically remained one of the highest in the world. The conundrum that the United States has faced in balancing innovation with affordability of life-saving medications is palpable, especially when faced with the reality of patent monopolies conflicting with healthcare obligations. The result is an overflow of issues, which emphasize the need to delineate limits on innovation to balance pharmaceutical novelty with robust public health within the United States. This has increasingly shifted the focus of patent law into the realm of healthcare issues. Naturally, it leads to an inquiry about how best …

Rurality, Resilience, & Identity: A Soft Systems Methodology Approach To Understanding Self-Reported Issues In Rural America, Kayla M. Gabehart, Allegra H. Fullerton, Kristin Olofsson

Michigan Tech Publications

This study investigates how rural communities In Oklahoma conceive of their socioeconomic position in larger systems, as well as their resiliency and ability to withstand challenges. Utilizing systems thinking and polycentricity literature, we analyze interviews to construct an understanding of how rural communities perceive themselves, and how this impacts interactions with other communities and governments. Rural communities and their associated challenges are complex and impacted by a range of factors. We find that rural residents also feel this complexity, and understand their issues as products of overlapping systems and structures, and both internal and external factors. Additionally, we observe little …

Clots Reveal Anomalous Elastic Behavior Of Fiber Networks, Andrei Zakharov, Myra Awan, Terrence Cheng, Arvind Gopinath, Sang Joon John Lee, Anand K. Ramasubramanian, Kinjal Dasbiswas

Faculty Research, Scholarly, and Creative Activity

The adaptive mechanical properties of soft and fibrous biological materials are relevant to their functionality. The emergence of the macroscopic response of these materials to external stress and intrinsic cell traction from local deformations of their structural components is not well understood. Here, we investigate the nonlinear elastic behavior of blood clots by combining microscopy, rheology, and an elastic network model that incorporates the stretching, bending, and buckling of constituent fibrin fibers. By inhibiting fibrin cross-linking in blood clots, we observe an anomalous softening regime in the macroscopic shear response as well as a reduction in platelet-induced clot contractility. Our …

From Net Zero To Nature Positive, Simon J.D. Schillebeeckx, Ryan Merrill

Research Collection Lee Kong Chian School Of Business

The concept of Net Zero, once considered a significant step towards sustainability, is now seen as outdated and insufficient in addressing climate change and biodiversity loss. To heal our planet, businesses must collectively adopt Regeneration: a holistic, impact-focused, and much more effective Nature Strategy. It's crucial that we recognize other planetary boundaries beyond carbon and transcend a company’s responsibility beyond emissions culpability. By choosing a Nature Pledge over or in addition to a Net-Zero pledge, you will be making a profound commitment to real systemic change and planetary health.

To examine this issue, we tested experienced versus inexperienced RTS video gamers on multi-object tracking tasks (MOT) and dual-MOT tasks with visual or auditory secondary tasks (dMOT). We employed a street-crossing task with a visual working memory task as a secondary task in a virtual reality (VR) environment to examine any generalized attentional advantage. Background: Similar to action video games, RTS video games require players to switch attention between multiple visual objects and views. However, whether the attentional control advantage is limited by sensory modalities or generalizes to real-life …

Geographies Of Storage, Sayd Randle Jan 2024

Geographies Of Storage, Sayd Randle

Research Collection College of Integrative Studies

Resource storage has long played a key role in the production of socio-ecological arrangements and economic relations. Even so, storage as a concept has remained somewhat marginal within geographical scholarship, often obscured by an analytical focus on the dynamics of movement. Reviewing recent works from geography, science and technology studies, and anthropology that center sites and practices of storage, this essay elaborates the diverse ways in which storage arrangements mediate resource circulation and the production of space. This literature demonstrates that thinking systematically with storage can illuminate a range of novel temporal, material, and value entanglements in-the-making, pointing to potentially …

With the ongoing global anthropogenic climate change, a silent war rages in forests, the lungs of the earth. Invasive pathogens and insect pests are decimating native trees, compromising ecological function, biodiversity, and ecosystem services. Many of the processes surrounding successful biotic invasions are broad and have been previously analyzed (Hobbs et al., 2006; Blackburn et al., 2011). For invasive insect pests and pathogens, globalization, the rise in human population and associated environmental disturbances are recognized as fundamental drivers of introduction of propagules of non-native species to novel environments. Superior tolerance of extreme climate environments, in particular temperature, is also associated …

The Ubiquitin Specific Peptidase 22 (USP22), a component of the Spt-Ada-Gcn5 Acetyltransferase (SAGA) histone modifying complex, is overexpressed in multiple human cancers, but how USP22 impacts tumorigenesis is not clear. We reported previously that Usp22 loss in mice impacts execution of several signaling pathways driven by growth factor receptors such as erythroblastic oncogene B b2 (ERBB2). To determine whether changes in USP22 expression affects ERBB2-driven tumorigenesis, we introduced conditional overexpression or deletion alleles of Usp22 into mice bearing the Mouse mammary tumor virus-Neu-Ires-Cre (MMTV-NIC) transgene, which drives both rat ERBB2/NEU expression and Cre recombinase activity from the MMTV promoter resulting …

The Von-Hippel-Lindau (VHL) gene, acting as a tumor suppressor, plays a crucial role in the tumorigenesis of clear cell renal cell carcinoma (ccRCC). Approximately 90% of individuals with advanced ccRCC exhibit somatic mutations in the VHL gene. Belzutifan, orally administered small-molecule inhibitor of hypoxia-induced factor-2α, has demonstrated promising efficacy in solid tumors associated with germline loss-of-function mutations in VHL, including ccRCC. However, its impact on cases with somatic or sporadic VHL mutations remains unclear. Here, we present 2 cases where belzutifan monotherapy was employed in patients with advanced ccRCC and somatic loss-of-function mutations in VHL. Both patients exhibited a swift …

The human respiratory and circulatory systems collaborate intricately to ensure oxygen delivery to all cells, which is vital for ATP production and maintaining physiological functions and structures. During limited oxygen availability, hypoxia-inducible factors (HIFs) are stabilized and play a fundamental role in maintaining cellular processes for hypoxia adaptation. First discovered during investigations of erythropoietin production regulation, HIFs influence physiological and pathological processes, including development, inflammation, wound healing, and cancer. HIFs promote extracellular adenosine signaling by enhancing adenosine generation and receptor signaling, representing an endogenous feedback mechanism that curbs excessive inflammation, supports injury resolution, and enhances hypoxia tolerance. This is especially …

Plastic is a ubiquitous environmental contaminant, resulting in widespread exposure across terrestrial and marine spaces. In the environment, plastics can degrade into microparticles where exposure has been documented in a variety of fauna at all trophic levels. Human epidemiological studies have found relationships between inhaled microplastics and oxidative stress and inflammation. Previous studies of bottlenose dolphins (Tursiops truncatus) have reported prevalent exposure to plasticizing chemicals (e.g., phthalates) as well as particle loads in gastrointestinal tracts, but exposure from inhalation has not yet been studied. The objective of this study was to determine if inhalation is a viable route of microplastic …

OBJECTIVES: Some centers have recommended including concentrated fibrinogen replacement in massive transfusion protocols (MTPs). Given our center's policy of aggressive early balanced resuscitation (1:1:1), beginning prehospital, we hypothesized that our rates of hypofibrinogenemia may be lower than those previously reported.

METHODS: In this retrospective cohort study, patients presenting to our trauma center November 2017 to April 2021 were reviewed. Patients were defined as hypofibrinogenemic (HYPOFIB) if admission fibrinogen

RESULTS: Of 29 782 patients, 6618 level 1 activations, and 1948 patients receiving emergency release blood,

CONCLUSIONS: Low rates of hypofibrinogenemia were found in our center which treats hemorrhage with early, balanced …

The complex double-membrane organization of the envelope in Gram-negative bacteria places unique biosynthetic and topological constraints that can affect translocation of lipids and proteins synthesized on cytoplasm facing leaflet of cytoplasmic (inner) membrane (IM), across IM and between IM and outer membrane (OM). Uniformly oriented inside-out (ISO) vesicles became functional requisite for many biochemical reconstitution functional assays, vectorial proteomics, and vectorial lipidomics. Due to these demands, it is necessary to develop simple and reliable approaches for preparation of uniformly oriented IM membrane vesicles and validation of their sidedness. The uniformly ISO oriented membrane vesicles which have the cytoplasmic face of …

Exploring Uniform, Dual, And Dynamic Topologies Of Membrane Proteins By Substituted Cysteine Accessibility Method (Scam™), Mikhail Bogdanov

Faculty, Staff and Student Publications

A described simple and advanced protocol for Substituted Cysteine Accessibility Method as applied to transmembrane (TM) orientation (SCAM™) permits a topology analysis of proteins in their native state and can be universally adapted to any membrane system to either systematically map an uniform or identify and quantify the degree of mixed topology or establish transmembrane assembly dynamics from relatively static experimental data such as endpoint topologies of membrane proteins. In this approach, noncritical individual amino acids that are thought to reside in the putative extracellular or intracellular loops of a membrane protein are replaced one at the time by cysteine …
