Disease-modifying therapies are standard of care (SOC) for sickle cell disease (SCD), but hematopoietic cell transplantation (HCT) has curative potential. We compared outcomes prospectively through 2-years after biologic assignment to a Donor or No Donor (SOC) Arm based on the availability of an HLA-matched sibling or unrelated donor (BMTCTN 1503; NCT02766465). A donor search was commenced after eligibility confirmation. The primary endpoint was the comparison of survival 2 years after biologic assignment between treatment arms. Power calculations required 60 participants on the Donor Arm and 140 on the No Donor Arm to determine if early transplant-related mortality might be balanced …

Childhood obesity is a significant health problem in the United States with consequences presenting themselves in the short- and long-term. Physical activity is a modifiable factor associated with adiposity and can at least partially attenuate the consequences of childhood obesity. There are many factors which influence children’s participation in regular physical activity, some of which are also associated with pediatric obesity. The overarching purpose of this research is to investigate potential influences on the association of obesity and physical activity among typically developing children. Animal agriculture, the second-largest contributor to climate change, poses serious risks to human and global health. Animal/environmental activists work to reduce these risks but often face verbal ridicule and physical violence; however, little is known about the psychological suffering experienced by this group. This interpretative phenomenological analysis (IPA) aimed to explore the experience of psychological suffering in animal/environmental activists committed to ending exploitative animal agriculture and environmental activities. Ecopsychological theory and psychological constructs, such as eco-anxiety, eco-grief, and vicarious trauma, comprised the conceptual framework for this study. This study addresses the critical need to understand how mental health challenges and external stressors affect long-term recovery from an Opioid Use Disorder (OUD). Grounded in the Recovery Capital Framework, this study analyzed the relationship between depression, stressful life events, and recovery capital (RC) among individuals in recovery from severe OUD. Utilizing a quantitative nonexperimental survey design, data were collected from a purposive sample of 107 adults aged 18 and older who had been in recovery for at least 2 years. Dynamic Dysregulation Of Retrotransposons In Neurodegenerative Diseases At The Single-Cell Level\, Wankun Deng, Citu Citu, Andi Liu, Zhongming Zhao

Retrotransposable elements (RTEs) are common mobile genetic elements comprising ∼42% of the human genome. RTEs play critical roles in gene regulation and function, but how they are specifically involved in complex diseases is largely unknown. Here, we investigate the cellular heterogeneity of RTEs using 12 single-cell transcriptome profiles covering three neurodegenerative diseases, Alzheimer's disease (AD), Parkinson's disease, and multiple sclerosis. We identify cell type marker RTEs in neurons, astrocytes, oligodendrocytes, and oligodendrocyte precursor cells that are related to these diseases. The differential expression analysis reveals the landscape of dysregulated RTE expression, especially L1s, in excitatory neurons of multiple neurodegenerative diseases. …

Our understanding of brain circuit operations and disorders has rapidly outpaced our ability to intervene and restore them. Developing technologies that can precisely interface with any brain region and circuit may combine diagnostics with therapeutic intervention, expediting personalised brain medicine. Transcranial ultrasound stimulation (TUS) is a promising noninvasive solution to this challenge, offering focal precision and scalability. By exploiting the biomechanics of pressure waves on brain tissue, TUS enables multi-site targeted neuromodulation across distributed circuits in the cortex and deeper areas alike. We examined the zoographics, or the characteristics of nonhuman animal subjects, reported for the entirety of the Journal of the Experimental Analysis of Behavior (JEAB) through the most recent complete year (1958–2023). Animal sex in particular was evaluated to determine whether the lack of inclusion of female subjects in other disciplines extends to JEAB. Through systematic coding of all nonhuman empirical articles, we found consistent underreporting of most zoographics and a disproportionate use of male subjects relative to female subjects. Additionally, animal sex was commonly unreported and the inclusion of both male and female subjects was sparse. …

Introduction

Progressive aging, or senescence, of mesenchymal stem/stromal cells (MSCs) is a major obstacle faced when trying to culture potent stem cells for use in therapy. Senescent cells are irreversibly nondividing cells that cease performing critical functional effects. Elimination of senescent cells using biochemical means, such as the use of senolytic drugs like dasatinib, may be useful in retaining the viable and proliferating populations of the cells.

Nuclear receptors are important in normal physiology and disease. Physicians and scientists who study nuclear receptors organize and attend conferences and symposia devoted to foundational and translational nuclear receptor research, but the field lacks a platform for early-stage investigators and aspiring leaders. This review article explores the integration of thin window films and solar energy systems to enhance thermal comfort in luminous office buildings. Luminous office buildings, which prioritize natural lighting, often face challenges in maintaining thermal comfort for occupants. By integrating solar energy systems, such as photovoltaic (PV) and solar thermal systems, with thin window films, it is possible to optimize thermal comfort while harnessing sustainable energy sources. Objective: The object of this integrative review was to investigate the analgesic benefit and side effect profile of the coadministration of ketamine and magnesium. Secondary goals were to provide an evidence-based recommendation on the ideal dosing, frequency, and order of administration of the medications. Methods: Relevant articles published between January 2001 and May 2019 were examined for quality, strength, and generalizability. Results: Beginning evidence suggests ketamine may interact with magnesium to provide synergistic NMDA receptor antagonism and analgesia. Trauma induced mortality can often be attributed to hemorrhage in the early phase of trauma. Up to one-half of trauma deaths are due to uncontrolled bleeding within the first few hours of injury. Patient survival following a traumatic injury is reliant upon the trauma team's ability to gain early control of bleeding. Ways in which the trauma team can gain control of bleeding include rapid identification of the source of hemorrhage, applying direct pressure to the source of hemorrhage, and aggressive resuscitation. Prolonging the duration of analgesia in single-shot nerve blocks (SNBs) continues to be a problem in regional anesthesia. Both IV and perineural dexamethasone have proven to be effective in prolonging the duration of analgesia in SNBs.

Objective: The purpose of this integrative review was to determine how IV and perineural dexamethasone effect duration of analgesia, pain scores, opioid consumption, and complications in patients undergoing SNB.

Methods: An integrative review that included 12 peer-reviewed randomized-controlled trials (RCTs) and one meta-analysis was performed.

Objective: To compare the safety and efficacy of chlorhexidine versus Listerine in perioperative and intensive care settings in preventing ventilator-acquired pneumonia (VAP).

Methods: An integrative review using a keyword search was conducted in Cochrane, Cinahl, and Google Scholar databases. Data was collected regarding the efficacy of chlorhexidine and Listerine in preventing ventilator-acquired pneumonia, safety and side effect profiles, and in vivoin vitro secondary outcome measures.

Tacrolimus, commonly used for GvHD prophylaxis is usually administered via a dedicated CVL and trough levels drawn from the unexposed lumen. Being an oil-based medication, it may be adsorbed to the inside lumen of the CVL and result in falsely high levels drawn from an inadvertently exposed lumen. There is no treatment for decontamination of such CVLs and natural decay occurs over months before the CVL can be used to draw reliable trough levels.

This nursing best practice guideline will address the therapeutic relationship and its central importance to nursing practice. Effective nursing practice is dependent on an effective therapeutic relationship between the nurse and the client. The guideline addresses the qualities and capacities of an effective therapeutic relationship, the state of knowledge, and the knowledge needed to be effective in a therapeutic relationship. To implement a therapeutic relationship successfully, an organization's characteristics and workplace values must be supportive and in place (see Recommendations 5 to 14). The recommendations in this guideline, therefore, focus on practice, education, organization and policy

This article explores how emerging adults (18 to 29‐year‐olds) integrate pro‐environmental behaviors and spiritual practices of simplicity. First, this article synthesizes the research on emerging adulthood, including what it is, how they experience and express their spirituality, and their emotional and practical behaviors in response to the climate crisis. Then, this article explores how a small sample of Western Protestant emerging adults practice pro‐environmental simplicity in their daily lives.
