Single-cell RNA sequencing (scRNA-seq) has transformed our understanding of cellular heterogeneity and gene expression dynamics. Despite its potential, the inherent noise and sparsity of scRNA-seq data pose significant challenges in clustering cells into biologically meaningful groups. This dissertation addresses these challenges through two novel methodologies aimed at enhancing the accuracy and robustness of scRNA-seq data analysis.
First, we introduce the Differential Feature Selection (DIFS) framework, designed to improve the identification of differential features in scRNA-seq data. Climate change presents an unprecedented challenge to global agriculture and food security. Small farms are especially vulnerable to the local impacts of large-scale drivers of change. Effective adaptation in agriculture requires working across scales, and geographic, political, and disciplinary boundaries to address barriers. I use elements of case study, agent-based modeling and serious games, to design a model of farmer decision-making using the sociocognitive framework of climate change adaptation. I examine how adaptation functions as a process, how complex dynamics influence farmer behavior, and how individual decisions influence collective behavior in response to climate change. Abstract Background/purpose Additive manufacturing (AM) technology, such as selective laser melting (SLM), has been used to fabricate medical devices of Ti-6wt.% Al-4wt.%V (Ti6Al4V) alloys in dentistry. Strontium (Sr) has been shown to have the potential to treat osteoporosis. The aim of this study was to investigate the physicochemical and biological properties of strontium-containing coatings on selective laser melted Ti6Al4V (SLM-Ti6Al4V) substrate. Abstract Background/purpose Oral lichen planus (OLP) is a chronic inflammatory disease with unknown mechanisms of pathogenesis. Keratin 17 (KRT17) is a protein that regulates numerous cellular processes. This study aimed to explore the expression of KRT17 in OLP and its correlation with the severity of OLP. Materials and methods RNA sequencing using epithelium from 5 OLP patients and 5 health control (HC) was performed, followed by functional analysis. Traumatic Brain Injury (TBI) significantly affects social interactions and emotional well-being. Following COVID-19, there has been growing interest in how individuals with TBI use online social media groups for support. This study examined engagement patterns in four Facebook support groups: two for TBI and two control groups. A deductive approach was used to categorize 4,133 posts and comments into a framework of support discourse, including seeking and giving support, and non-social support. Each category was further subdivided to analyze post content in more detail. Comments on posts were coded for helpfulness/appropriateness. Results revealed that posts in all groups fell into …

Vitamins are essential organic compounds that vary widely in chemical structure and are vital in small quantities for numerous biochemical and biological functions. They are critical for metabolism, growth, development and maintaining overall health. Vitamins are categorised into two groups: hydrophilic and lipophilic. Vitamin K (VK), a lipophilic vitamin, occurs naturally in two primary forms: phylloquinone (VK1), found in green leafy vegetables and algae, and Menaquinones (VK2), present in certain fermented and animal foods and widely formulated in VK supplements. Chitosan, a biopolymer obtained from chitin, is known for its remarkable adsorption abilities for dyes, drugs, and fats, and its diverse array of antibacterial characteristics. This study explores the extraction and characterization of chitosan from the mycelium of Amanita phalloides. The moisture content, ash content, water binding capacity, fat binding capacity, and degree of deacetylation of the extracted chitosan were determined. The chitosan exhibited a high yield of 70%, crystallinity of 49.07%, a degree of deacetylation of 86%, and potent antimicrobial properties against both Gram-negative and Gram-positive bacteria. Competition for mating opportunities among individuals of the same sex is a well-documented phenomenon, yet the extent of cooperation in such contexts remains underexplored. This study aims to investigate the occurrence of cooperative bargaining between two males in scenarios of limited and equal female availability, using non-wild descendants of the Trinidad guppy (Poecilia reticulata) as a model species. Ranunculus triternatus, a rare endemic species of the Columbia River Gorge, faces conservation challenges due to its limited range and fragmented populations. This study assesses the genetic diversity of R. triternatus and compares it with its widespread congener, Ranunculus glaberrimus, using inter-primer binding site (iPBS) markers. Genetic data from 80 individuals across eight populations were analyzed using Nei’s genetic diversity, Shannon’s diversity index, and AMOVA. Results showed R. triternatus had higher genetic diversity than western populations of R. glaberrimus, with eastern populations of R. glaberrimus displaying greater diversity than western ones. There is a complex of cutworm species that are pests in western Canada, with important species including Agrotis orthogonia (pale western cutworm), Euxoa auxiliaris (army cutworm), and Euxoa ochrogaster (redbacked cutworm). Cutworm damage is sporadic and patchy, and as a result is difficult to predict. During outbreak years, insecticide applications and/or reseeding are necessary and yield losses can be significant. Recently, field-collected cutworm larvae were reared in the lab to estimate levels of parasitism, which ranged from 2% to 25%, depending on the province and the year of sampling, and to identify the parasitoid complex attacking cutworms. Research suggests children who prefer internal over external explanations of inequality (e.g., innate ability differences vs. structural racism/sexism) are more likely to develop social biases. Adults with greater cognitive reflection (i.e., analytic thinking) and socioeconomic status (SES) are sometimes less likely to engage in internal reasoning about social groups. We investigated whether cognitive reflection and SES similarly predict children’s explanations of inequality. Five to 12-year-old children (N = 162) completed Rizzo et al.’s (2021) explanations for inequalities task for the domains of race and gender. Indonesia, one of the world’s most biodiverse countries, is undergoing mass deforestation, exacerbating climate change and leading to accelerated loss of species. This project addressed the urgent need to conserve endangered Indonesian biodiversity. Specifically, the potentially life-saving bioactive compounds harbored within its plants. A group of Indonesian researchers from Universitas Nasional (UNAS) in Jakarta received training in RApid Metabolome Extraction and Storage (RAMES) technology, an ethical, low-impact, field-deployable and cost-effective methodology developed by Rutgers University. This instrumental case study describes the steps taken to establish and implement a university–school partnership to improve rural science teachers’ content knowledge and teaching practices and students’ achievement in elementary science and high school biology courses. Our research questions explored the impact of professional development and place-based learning on student outcomes, with the study’s methodology involving needs assessments, teacher training, and continuous support through modeling and coaching. The findings include gains in achievement and substantial gains in science education. In response to Ashley Todd-Diaz and Alex H. Poole’s 2022 article published in American Archivist, “Who Is This Godot? the Academy of Certified Archivists and Graduate Archival Education,” the Academy of Certified Archivists (ACA) affirms the continuing relevance of archival certification and its place in the profession. Non-bonded interactions are fundamental forces that govern molecular relationships between two or more molecules. These interactions contribute to the stability of complex biological structures like DNA, RNA, and proteins, and control various biological processes. Almost all of these processes are significantly influenced by protein-protein and protein-ligand intermolecular interactions. Here, the interactions of various proteins with other proteins, peptides, and/or ligands were quantified computationally to tackle human health-related problems. For estimating the intermolecular interactions, a number of computational approaches including protein structure modeling, molecular dynamics simulations, molecular docking, ensemble docking, semi-empirical methods, etc., were used. With the rise of population placing strain on food supplies and the failure of current prevention strategies to mitigate effects of agricultural pollution, research is needed to develop new technologies for the removal and recovery of nutrients from agricultural runoff and drainage. Geopolymer, an alkali-silicate material polymerized within an acidic or alkaline medium, is promising as an adsorptive substrate due to its multifaceted customizability, cost effectiveness, and adsorptive properties.
