Thin polymer coatings are used to improve the interface between biological species and functional materials. Their interaction is significantly influenced by the functional groups and roughness of the polymer film and prediction of the interaction is thus of great interest. However, for conventional polymer films, this cannot be examined independently because of the interplay of defects, residual solvent molecules, roughness, and functional groups. Solvent-free polymer films prepared by initiated chemical vapor deposition (iCVD) exhibit conformal, defect-free characteristics and enable precise tailoring of the functional groups. This facilitates to isolate the contribution of functional groups on the bio-interface performance. Consequently, in …

Pacific salmon, the signature species of the Pacific Northwest, have declined across their range for well over a century, due to a myriad of human-caused effects on their habitat and the fish themselves. Restoration efforts—some successful, some halting—began in earnest in the late 20th century, with considerable attention focused on the Columbia Basin, where historically salmon runs were crippled by a large interconnected hydroelectric system of federal and non-federal dams. In the 1980 Northwest Power Act, Congress created an interstate agency, the Northwest Power and Conservation Council, with access to a substantial amount of ratepayer dollars; the agency has chosen …

Hepatocellular carcinoma (HCC) represents one of the most common causes of cancer-related death worldwide, with few available treatment options. Cancer-testis antigens (CTAs), notably from the MAGE gene family, have opened new possibilities for improving immunotherapy in HCC due to their tumor-specific expression and high immunogenicity. However, the mechanisms regulating MAGE gene expression are largely unknown. This study explored mechanisms controlling the expression of MAGE-A3 and MAGE-C2. Exosomes, spherical vesicles secreted by cells containing diverse biomolecules, are gaining attention in veterinary medicine. Mesenchymal stem cell (MSC)-derived exosomes show promise for treating animal diseases. However, their safety profile, particularly the acute toxicity of canine MSC-derived exosomes, has yet to be adequately explored. This study evaluates the acute toxicity of canine MSC-derived exosomes on rat systemic health, adhering to ISO 10993-11 guidelines for systemic toxicity testing to support veterinary drug registration and development. Human interleukin-15 (hIL-15) is a cytokine that plays a crucial role in immune responses like T cell proliferation, activation, and prolongation of natural killer (NK) cells. This cytokine also impacts the cell proliferation of human adult low calcium temperature (HaCaT) keratinocytes which can lead to the wound-healing effect. This study aimed to optimize the expression and characterize the recombinant hIL-15 in Nicotiana benthamiana with the Agrobacterium-mediated transient expression system. The hIL-15 gene was fused with the Fc domain from the IgG1 antibody for purification. The expression cassette was optimized for sub-cellular targeting and agroinfiltration. Lusianthridin (LST), a bioactive compound isolated from Dendrobium venustum, exhibits notable vasoprotective properties. This study aimed to elucidate the underlying molecular mechanisms of LST by evaluating its effects on platelet function and endothelial cell activation in vitro. In human platelets, LST significantly inhibited collagen-induced aggregation, an effect associated with elevated phosphorylation of vasodilator-stimulated phosphoprotein (VASP) and suppression of Akt phosphorylation. In EA.hy926 endothelial cells, LST attenuated tumor necrosis factor-alpha (TNF-α)-induced endothelial dysfunction by reducing nitric oxide (NO) production and suppressing the expression of pro-inflammatory cytokines, including interleukin-6 (IL-6) and interleukin-1β (IL-1β). Objectives : (1) This study aims to predict the EQ-5D-5L utility scores from the impact of vision impairment (IVI) questionnaire in Thai patients using mapping techniques. Curcumin is widely recognized for its various pharmacological properties, including antioxidant, anti-inflammatory, and anti-tumor activities. Nevertheless, the development of curcumin as a therapeutic agent is impeded by its limited oral bioavailability, which stems from its chemical instability, poor aqueous solubility, and rapid degradation. This study aimed to develop granule formulations incorporating hyaluronic acid graft poly(N-isopropylacrylamide) or HA-g-pNIPAM to enhance dissolution and protect curcumin from degradation. Three formulations were developed: F10 (HA-g-pNIPAM physically mixed with curcumin), F10 Encap (curcumin encapsulated within HA-g-pNIPAM), and F11 (curcumin granules without HA-g-pNIPAM). Drug efflux by transporters of the ATP-binding cassette (ABC) family such as P-glycoprotein is the earliest and most well-studied multidrug resistant (MDR) mechanism. Overexpression of ATP-binding cassette transporters can decrease the effectiveness of chemotherapy by pumping anti-cancer drugs out of cancer cells. NSCLC severity is driven by dysregulation of the EGFR/PI3K/Akt pathway, promoting tumor growth and drug resistance. Hyperactive EGFR/PI3K/Akt signaling results in aggressive tumors and poor outcomes, further worsened by resistance to EGFR inhibitors. Targeting this pathway could inhibit tumor growth and resistance. artogomezianone, a compound derived from Artocarpus gomezianus, has shown considerable anti-cancer efficacy, mainly by inducing apoptosis and decreasing the proliferation of several cancers. A new vaccine technology based on the delivery of mRNA has gained widespread acceptance due to the COVID-19 pandemic. The delivery of mRNA formulated with lipid nanoparticles (LNPs) elicits a robust immune response, activating both humoral and cell-mediated response. Enterocytozoon hepatopenaei (EHP) is an intracellular microsporidian parasite causing hepatopancreatic microsporidiosis (HPM), a disease that impairs the digestion and nutrients absorption of shrimp and leads to growth retardation and size variation. Our studies aim to comprehensively investigate the host’s immune responses to the EHP infection and the pathogenesis of the parasite. It was found that genes in Toll and JAK/STAT pathways, as well as antimicrobial peptides, were highly upregulated upon the EHP infection. Importantly, suppression of the JAK/STAT pathways resulted in a significant reduction of the c-type lysozyme (LvLyz-c) expression and the elevation of EHP copy numbers. These indicate a …

This dissertation aims to develop two unconventional methods for the detection of a high-risk, cancer-causing human papillomavirus type 16 (HPV16) on miniaturized platforms for point-of-care testing (POCT). The first part focuses on a direct detection of HPV16 double-stranded DNA (dsDNA) using the non-self-pairing pyrrolidinyl peptide nucleic acid (acpcPNA) pair on a paper-based substrate coupled with fluorescence and electrochemical dual detections using a DNA-binding dye in a single device. Roxarsone (ROX), a growth enhancer in poultry and swine, has the potential to improve weight gain and prevent coccidiosis. Excessive use of ROX may lead to its accumulation in animal tissues, potentially contaminating food products and posing health risks to humans through consumption. This research aims to develop an electrochemical sensor based on a magnetic screen-printed graphene electrode (mSPGE) modified with a highly selective molecularly imprinted polymer (MIP) for the determination of ROX with enhanced sensitivity and selectivity. Drawing upon original research into the travaux préparatoires of the 1948 Genocide Convention, this Article advances several claims that complicate the standard account according to which genocide must entail a purpose to physically destroy at least a substantial part of a protected group. The core of the Article closely explores the words “intent,” “destroy,” and “in part,” showing how international authorities have settled on a received and largely uninterrogated wisdom regarding the meaning of these terms, one which is supported neither by the drafting history of the Genocide Convention, nor even by the actual results of the judicial decisions that …

Perceptions of multilingualism have an impact on the educational choices of urban Indians and larger policy decisions around language in education in the Indian state of Maharashtra. By surveying 150 speakers of Marathi, the official language of the state and mother tongue, or first language of survey respondents, perceptions of multilingualism reveal conceptual assumptions and biases that can be uncovered through cognitive-linguistic analysis. Unprompted, survey respondents drew connections between multilingualism and education, underscoring language ideologies, or beliefs about languages which motivate urban Maharashtrians' educational decisions. Transient Receptor Potential V1 (TRPV1) is a nonselective cation channel, which is expressed in nociceptors/ pain detecting receptors. Activated by a wide variety of exogenous and endogenous physical and chemical stimuli such as heat, TRPV1 is involved in temperature detection, body temperature regulation, and pain perception. It is generally accepted that TRPV1's conductance to cations, including Ca2+, depends on its conformational change from a closed to an open state following gating. Polycystin-2 (PC2) is an ion channel belonging to the polycystin subfamily of the transient receptor potential (TRP) cation channel superfamily. Mutations in PC2 are associated with autosomal dominant polycystic kidney disease (ADPKD), one of the most common genetic diseases in humans. Despite extensive studies over the past decades, the role of PC2 in normal and ADPKD kidneys remains largely unknown, and the activation and regulation of this ion channel are elusive. This dissertation argues that British children’s literature of the Golden Age addresses children and adults through their illustrations. It considers why the nineteenth century experienced a proliferation in fantasy and illustration, as well as why Golden Age children’s texts appeal to mixed-age audiences. Through an application of the term “cross-writing,” I argue that the Golden Age of children’s literature did not think of audiences in a binary manner, as some children’s literature scholars tend to assert. Instead, they used the dual mode of textual and visual narration to complicate the boundaries between childhood and adulthood. I also offer an original …

Pulmonary fibrosis (PF) poses a significant challenge in respiratory medicine due to its complex etiology and progression characterized by excessive lung scarring, leading to compromised pulmonary function. The multifaceted roles of Vascular Endothelial Growth Factor A (VEGF-A) and specifically its isoform VEGF-A165 are critical in this context, given their involvement in angiogenesis, lymphangiogenesis, and tissue repair. The dual nature of VEGF-A165, promoting angiogenesis and potentially exacerbating or mitigating fibrosis, underscores its significance in both the progression and recovery phases of PF. A Quality-By-Design Approach For The Development Of Inhalable Dry Powders For Tuberculosis (Tb) Treatment, Suyash M. Patil

Tuberculosis (TB), caused by Mycobacterium tuberculosis (M.tb), remains a pressing concern globally. The emergence of drug-resistant strains and the need for prolonged treatment regimens continue to complicate disease management. Innovative therapeutic strategies are urgently required to enhance treatment efficacy, improve patient adherence, and minimize adverse effects. Toward this, strategies involving use of drug combinations, reducing dosing frequencies, and targeting TB-infected cells are gaining attention. In my dissertation, we explored the combination therapy involving two FDA-approved anti-TB drugs, Bedaquiline (BDQ) and Pretomanid (PTD). Carbon offsets are widely criticized for lacking additionality. The public debate, how- ever, has failed to recognize that the problem of additionality goes far beyond offsets. It arises any time the government subsidizes an activity. Programs with longer histories than offsets and far more money at stake offer valuable lessons for managing lack of additionality in offsets. The article sets out the current efforts to prove offset additionality, explains why major government programs present comparable additionality challenges, and show why other programs’ problems of additionality are perceived as significantly different than those for offsets. Smallmouth bass Micropterus dolomieu are known as a popular sport fish for anglers. When a known smallmouth bass fishery appears to have a dwindling population, it raises questions about the population health. A common indicator of the health of the population is the mortality rate. By comparing mortality, conclusions can be made on whether smallmouth bass are dying at a different rate in the system over time. Northern pike Esox lucius and walleye Sander vitreus are both top predators in freshwater systems. Due to their predatorial nature, high populations of one species can prohibit populations of the other species from thriving, specifically northern pike foraging on walleye. This information is tracked by the Minnesota Department of Natural Resources through catch per unit effort (CPUE). CPUE data was gathered for all lakes in Beltrami County, Minnesota, and a regression analysis was done to test for a relationship between pike and walleye. It was found that the relationship between northern pike and walleye CPUE was significant (P = 0.02). …

Burbot Lota lota in north-central Minnesota lakes have the potential to reach large sizes and consume large volumes of prey. This caveat may make burbot susceptible to higher rates of biocontamination, bioaccumulation and biomagnification. The objective of this study was to determine how changes in age, length, gender, weight, and lake affect total mercury concentrations in burbot. In this experiment 28 burbot were angled from three lakes: Cass (n = 17), Winnibigoshish (n = 4), and Bad Medicine (n = 7). Then tissue samples were taken from each fish and were lyophilized and homogenized. Homogenized tissue samples were analyzed by …
