Structural And Functional Consequences Of Pde6 Prenylation In Rod And Cone Photoreceptors, Faezeh Moakedi

Phosphodiesterase-6 (PDE6) serves as a pivotal component in the phototransduction pathways of both cone and rod photoreceptors. In cones, PDE6 consists of tetrameric subunits: inhibitory (γ') and catalytic (α'). The catalytic subunit, PDE6α', contains a C-terminal prenylation motif. Deletion of this motif is associated with achromatopsia (ACHM), a form of color blindness. The mechanisms underlying the disease and the roles of PDE6 lipidation in vision remain elusive. Meanwhile, rod PDE6 is composed of α and β catalytic subunits and γ inhibitory subunits, with alterations in the C-terminal "prenylation motif" of PDE6β linked to retinitis pigmentosa (RP) pathology. Inflammatory Bowel Disease (IBD) is a prevalent disorder among youth in the United States. IBD is one of the leading causes of chronic pain in adolescence and symptoms can lead to reduced quality of life, social embarrassment, and lower self-esteem. Adherence to medical regimens for IBD is incredibly important to prevent morbidity, mortality, and impaired health-related quality of life (HRQOL). However, adolescents struggle generally with adherence and the regimen for IBD can be complicated and include aversive side effects. This thesis includes two main topics: 1) evaluation of probable maximum precipitation (PMP) for West Virginia to predict current and future PMP, and 2) utilizing acid mine drainage (AMD) sludge as a reclamation plan.

Probable maximum precipitation (PMP) is essential for dam design as well as for the inspection, operation, maintenance, and repairs of these facilities. West Virginia Department of Environmental Protection Dam Safety utilizes the 6-hr, 10-mi2 (26-km2) PMP established by Hydrometeorological Report (HMR) 51 based on the analysis of extreme rainfall. In this dissertation, I utilize the post-structural philosophy of Gilles Deleuze and Félix Guattari as a lens for investigating the proof process. Deleuze and Guattari were both post- structural philosophers who, like many in this tradition, troubled traditional notions related to stable identities, meaning, language, and mathematics. For Deleuze, sense and meaning is not the result of a sterile, transcendent effect or condition that is associated with propositions and states of affairs. Childhood adversity has long-lasting negative effects across the lifespan including increased mortality risk. The love and support individuals receive from others, also known as social support, has shown to be a protective factor against ACEs. However, little research has investigated the amplifying effects of social conflict and strain that often accompanies social relationships. Utilizing data from the Midlife Development in the U.S. (MIDUS) study, I tested whether higher levels of social support would buffer the negative effects of adverse childhood experiences on mortality risk, and whether higher levels of social strain would amplify these associations. Hodgkin’s lymphoma (HL) is a rare malignancy of lymphocytes that predominantly occurs in young adults aged 20-30 years or elderly individuals aged 65-75 years. Despite its low incidence, there were an estimated 223,512 HL survivors in the US in 2020. Hodgkin’s lymphoma shows a favorable prognosis among young adults, with a high cure rate of 85-90%; however, older adults experience poor prognosis, with a 5-year overall survival rate of 40-55% in patients over 60 years. HL survivors incur high total and out-of-pocket (OOP) healthcare expenditures, averaging $78,183 and $4,180 per patient in the first year after diagnosis, highlighting a considerable …

Research has investigated challenges that are created when one experiences a disaster within populations such as older adults and other vulnerable groups of people. However, there is little to no consideration given to how age in combination with trauma history are related to well-being after a disaster. Using two theories, socioemotional selectivity theory (SST, Carstensen, 2006) and the strength and vulnerability integration model (SAVI, Charles, 2010). An excretory/secretory (E/S) product, common to many helminths, is neutrophil inhibitory factor (NIF) which negatively affects neutrophil migration and activity. Due to differences in neutrophil accumulation between parasite resistant St. Croix STC and parasite susceptible Suffolk (SUF) sheep, we hypothesized that Hc-NIF may inhibit migration of SUF and not STC neutrophils. Previous studies have demonstrated that Hc produces NIF but is not commercially available. To test our hypothesis, neutrophils were cultured from STC and SUF sheep in the presence of Ancylostoma caninum-derived NIF (Ac-NIF) and measure chemotaxis to Interleukin-8 (IL-8), HcLA or H. contortus third-stage excretory/secretory (E/S) products. The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Previous data from our laboratory has shown that KNDy neurons may be responding to other inputs, including the melanocortin system, that will affect the timing of puberty. Child abuse is a significant public health concern that impacts children worldwide. Efforts to connect at-risk parents with prevention and intervention programs require the use of high-quality measures that evaluate child abuse potential. Researchers frequently use the Child Abuse Potential Inventory (Milner, 1986) and the Brief Child Abuse Potential Inventory (BCAP; Ondersma et al., 2005) which contain Validity indices that evaluate response patterns such as social desirability responding. Endodontic treatment aims to prevent or address apical periodontitis by reducing intracanal bacterial populations to promote tissue healing, requiring thorough debridement and disinfection of the canal space. This study aims to examine the presence of trace metal elements and chemical compounds in over-the-counter (OTC) and medical-grade sodium hypochlorite (NaOCl) solutions.

Method

Over-the-counter NaOCl brands (Clorox® and Great Value®) and medical grade 6% NaOCl solutions (CanalPro® and Chlor-Xtra®) were purchased, stored in amber glass bottles, and tested in triplicate. Each was analyzed using mass spectrometry, identifying 7881 chemical compounds with statistical comparisons via adjusted p-values, fold-change, and heat maps. …

Ecosystem services provided by dung-associated beetles, such as reducing pasture fouling, parasite suppression, supporting nutrient recycling, and increasing water infiltration, contribute to animal and pasture health. However, parasiticides used to manage cattle parasites, particularly flies and helminths, in West Virginia, can adversely affect these non-target beetles. Microtubules are ubiquitous cytoskeletal filaments essential in the function of eukaryotic cells. These polymers, comprised of αβ-tubulin heterodimers, are critical for cell division, intracellular transport, and formation of the axonemes of cilia and flagella. This functional diversity of microtubules is attributed to the incorporation of various α- and β-tubulin isotypes encoded by different genes. Indeed, mutations in human tubulin genes have been linked to various tissue-specific diseases, suggesting a unique function for each tubulin isotype. This doctorate dissertation consists of four individual studies with the overarching theme of the effect of feed manufacture on exogenous enzymes and antinutrients found in soybeans.

Study 1. Including exogenous enzymes in poultry diets can liberate nutrients that are typically inaccessible to monogastric animals. Phytase feeding research has traditionally been conducted by rearing broilers in raised wire cages, whereas commercially grown broilers are reared on built-up litter. Therefore, cage-based studies may not accurately represent commercial bird performance response to phytase. Multiple sclerosis (MS) is a cell-mediated, demyelinating autoimmune disease that affects the central nervous system (CNS). T helper (Th) 17 cells mediate the pathogenesis of MS and its animal model, experimental autoimmune encephalomyelitis (EAE), in part by infiltrating the CNS and interacting with astrocytes, a resident glial cell. Astrocytes are central to coordinating several inflammatory processes in the CNS and can respond to cytokines and chemokines produced by encephalitogenic Th17 cells. This dissertation discusses three instances of temporal prediction, applied to population dynamics and deep learning.

In population modeling, dynamic processes are frequently represented by systems of differential equations, allowing for the analysis of various phenomena. The first application explores modeling cloned hematopoiesis in chronic myeloid leukemia (CML) via a nonlinear system of differential equations. By tracking the evolution of different cell compartments, including cycling and quiescent stem cells, progenitor cells, differentiated cells, and terminally differentiated cells, the model captures the transition from normal hematopoiesis to the chronic and accelerated-acute phases of CML. The Barents Sea is a hotspot for environmental change due to global warming. These changes impact the structure and functioning of the marine ecosystem year-round, and it is therefore important to gain knowledge on trophic relationships and the energy flow from primary producers, i.e., ice algae (sympagic algae) and phytoplankton (pelagic algae) to consumers over the entire seasonal cycle. By using different lipid components as trophic markers, we provide seasonal coverage of the carbon and food-source composition of five of the most abundant and ecologically important zooplankton taxa inhabiting the Barents Sea: copepods, krill, amphipods, pteropods and chaetognaths. Although over 90 independent risk variants have been identified for Parkinson’s disease using genome-wide association studies, most studies have been performed in just one population at a time. Here we performed a large-scale multi-ancestry meta-analysis of Parkinson’s disease with 49,049 cases, 18,785 proxy cases and 2,458,063 controls including individuals of European, East Asian, Latin American and African ancestry. In a meta-analysis, we identified 78 independent genome-wide significant loci, including 12 potentially novel loci (MTF2, PIK3CA, ADD1, SYBU, IRS2, USP8, PIGL, FASN, MYLK2, USP25, EP300 and PPP6R2) and fine-mapped 6 putative causal variants at 6 known PD loci. Recognizing the vulnerabilities and complexities inherent in these communities, we emphasize here the necessity of ethical awareness throughout all research phases. Ethical obligations may extend to ensuring cultural sensitivity, safety, and equitable resource distribution. The core ethical principles of autonomy, beneficence, non-maleficence, and justice are promoted here to serve as a viable framework for ethical research.
