HIV/AIDS continues to be a significant public health problem in the US and globally. In the US, it disproportionately affects people of color. In 2012, Pre-exposure Prophylaxis (PrEP) was approved by the FDA for HIV prevention. PrEP reduces the chances of acquiring HIV by 99% among people at high risk. However, PrEP uptake among people of color remains low. Therefore, this study aimed to develop and test the predictability of an instrument to describe the intent to initiate and sustain PrEP use among people of color. Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. Genetics plays a critical role in understanding the molecular mechanisms underlying neurodegenerative disorders and pathogen evolution in infectious diseases. For example, identifying genetic variants associated with a disease phenotype uncovers functional pathways that could lead to potential drug targets and therapeutic interventions. In addition, tracking the genetic evolution of pathogens enables early detection and warning of infectious disease outbreaks. Direct Potable Reuse (DPR) is an innovative solution designed to address water scarcity by treating wastewater to directly produce safe drinking water. As droughts and water shortages become more frequent, DPR offers a sustainable and localized water supply by incorporating advanced treatment processes. DPR systems typically rely on a combination of technologies such as ozone followed by biological activated carbon (O3/BAC), reverse osmosis (RO), and advanced oxidation processes (AOPs) to remove a wide range of contaminants. However, low molecular weight organic compounds (LMWCs), particularly volatile organic compounds, can sometimes pass through RO membranes and AOP systems.

The relationship between BRAF-V600E mutations, mitochondrial DNA amplification and microsatellite instability-high (MSI-H) in colorectal cancer (CRC) has yet to be fully elucidated. The aim of the present study was to assess the association between the MSI status and BRAF-V600E gene mutations/clinicopathological features/mitochondrial DNA amplification in CRC. A non-interventional study analysis was performed using the clinicopathological features of 455 patients with CRC. Immunohistochemistry was used to evaluate four mismatch repair proteins (MutS homolog 2, MutS homolog 6, MutL homolog 1 and postmeiotic segregation increased 2), Ki-67 index, and programmed cell death protein 1 (PD-1) and programmed cell death-ligand 1 (PD-L1) expression. …

Soil contamination by cadmium (Cd), lead (Pb), and zinc (Zn) at the Tar Creek superfund site in northeast Oklahoma, United States, remains a threat to the environment and local ecosystem. Nearly all organisms experience heterogeneity in environmental conditions that can negatively impact fitness. A common adaptive strategy to reduce the impact of this stress is called facultative diapause, where development and reproduction are temporarily arrested to promote survival. Facultative diapause is characterized by three major criteria, the arrest must be (i) induced by an unfavorable environmental condition, (ii) reversible, and (iii) driven by genetically regulated mechanisms. BACKGROUND: Genome-wide association studies (GWAS) have identified more than 200 breast cancer risk-associated genetic loci, yet the causal genes and biological mechanisms for most loci remain elusive. Proteins, as final gene products, are pivotal in cellular function. In this study, we conducted a proteome-wide association study (PWAS) to identify proteins in breast tissue related to breast cancer risk.

METHODS: We profiled the proteome in fresh frozen breast tissue samples from 120 cancer-free European-ancestry women from the Susan G. Komen Tissue Bank (KTB). Access to genomic sequencing (GS) and resulting recommendations have not been well described in pediatric oncology. GS results may provide a cancer predisposition syndrome (CPS) diagnosis that warrants screening and specialist visits beyond cancer treatment, including testing or surveillance for family members. The Texas KidsCanSeq (KCS) Study evaluated implementation of GS in a diverse pediatric oncology population. We conducted semi-structured interviews (n = 20) to explore experiences of KCS patients' families around learning about a CPS diagnosis and following up on recommended care. The Cavity-Based X-ray Free Electron Laser (CBXFEL) is a possible future direction in the development of fully coherent hard X-ray sources of high spectral brilliance, a narrow spectral bandwidth of ≃ 1 − 100 meV, and a high repetition rate of ≃ 1 MHz. A diagnostic tool is required to measure CBXFEL spectra with a meV resolution on a shot-to-shot basis.

The proinflammatory cytokine interferon-gamma (IFNγ) is upregulated in a variety of infections and contributes to bone marrow failure through hematopoietic stem cell (HSC) activation and subsequent exhaustion. The cell surface protein, bone marrow stromal antigen 2 (BST2), is a key mediator of this process, as it is induced upon interferon stimulation and required for interferon-dependent HSC activation. To identify the mechanism by which BST2 promotes interferon-dependent HSC activation, we evaluated its role in niche localization, immune cell function, lipid raft formation, and intracellular signaling. Williams syndrome (WS) is a rare multisystemic disorder caused by recurrent microdeletions on 7q11.23, characterized by intellectual disability, distinctive craniofacial and dental features, and cardiovascular problems. Previous studies have explored the roles of individual genes within these microdeletions in contributing to WS phenotypes. Here, we report five patients with WS with 1.4 Mb-1.5 Mb microdeletions that include RFC2, as well as one patient with a 167-kb microdeletion involving RFC2 and six patients with intragenic variants within RFC2. We developed an ex vivo model system to analyze the influence of relevant environmental and mechanical factors potentially affecting the integrity of fetal membranes during fetoscopic surgery. The set-up exposes amniochorion membranes to insufflation at predefined levels of gas pressure, flow, humidity, and temperature. Change in fetal membranes stiffness is quantified during the phase mimicking surgery through measurement of membranes' strain in response to cyclic overpressure. The trocar induced perforation creates a mechanical weakness whose stability is assessed by increasing the insufflation pressure until membrane rupture. Adipose thermogenesis has been actively investigated as a therapeutic target for improving metabolic dysfunction in obesity. However, its applicability to middle-aged and older populations, which bear the highest obesity prevalence in the United States (approximately 40%), remains uncertain due to age-related decline in thermogenic responses. In this study, we investigated the effects of chronic thermogenic stimulation using the β3-adrenergic (AR) agonist CL316,243 (CL) on systemic metabolism and adipose function in aged (18-month-old) C57BL/6JN mice. Humans disturb the environment in specific places and at specific times. As wildlife learn to navigate these disturbances, they react as they would to the risk of predation. Wildlife biologists can identify where and when these disturbances will lead wildlife to congregate by comparing geospatial data, such as metrics that represent vegetation, topography, or other environmental conditions, to known locations of the animal(s) in question. In Chapter I, I discuss the impact of human disturbances, like urbanization and roads, on wildlife behavior. This dissertation analyzes Chesapeake Bay Watershed (CBW) agriculture by exploring residents' views on water quality issues, agriculture-related conflict, and farmers' adoption of nutrient management plans. Of all the attributes that define beef's eating quality, tenderness stands out as one of the most important. Previous studies have shown that consumers are willing to pay a premium for beef that is guaranteed to be more tender and are more likely to purchase that product repeatedly. Yet, due to the influence of numerous internal and external factors, controlling and predicting tenderness remains to be difficult. Wild pigs are an introduced species in the Americas that negatively affect ecosystems in many ways. Previous research has documented their negative effects on water quality and vegetation and their potential to spread disease among wildlife, domestic animals, and humans. However, how wild pigs affect predator-prey interactions between native wildlife has received little attention. This research explores multiple interactions between wild pigs, a native predator (cougars), and a native prey (black-tailed deer). COVID-19 harmed the resilience of the United States tourism industry, undermining its economic stability. Specifically, the drink tourism sector was negatively affected by the pandemic due to its heavy reliance on consumer demand. Enhancing the ability to withstand, recover, and adapt to changing conditions is fundamental to promoting financial development and economic growth. This study aims to explore the determinants of resilience within this industry, focusing on wineries, breweries, and distilleries in the western United States during 2020 and 2021. Most beef cattle in the US are feedlot-finished where intensive feeding of grains is used to increase weight and yield; however, a minority are raised in grass-fed operations that use exclusive pasture, rangeland, and hay for feeding, possibly leading to benefits for the land, soil, ecosystems, forages, and the meat itself. These operations use a variety of methods, and the goal of this was to recognize differences in the soil and forage metrics to help understand the resulting meat fatty acid composition and mineral content.
