Early Host Defense Against Virus Infections, Søren R. Paludan, Thomas Pradeu, Andreas Pichlmair, K. Brad Wray, Jacob Giehm Mikkelsen, David Olagnier, Trine H. Mogensen

Presidential Fellows Articles and Research

Early host defense eliminates many viruses before infections are established while clearing others so they remain subclinical or cause only mild disease. The field of immunology has been shaped by broad concepts, including the pattern recognition theory that currently dominates innate immunology. Focusing on early host responses to virus infections, we analyze the literature to build a working hypothesis for the principles that govern the early line of cellular antiviral defense. Aiming to ultimately arrive at a criteria-based theory with strong explanatory power, we propose that both controlling infection and limiting inflammation are key drivers for the early cellular antiviral …

Discrete Particle Dynamics Models: Computational Aspects And Applications, Yuxuan Wu Dec 2024

Discrete Particle Dynamics Models: Computational Aspects And Applications, Yuxuan Wu

Doctoral Dissertations and Master's Theses

Particle dynamics considers discrete particles as Newtonian point masses in diverse domains such as, pedestrian particles in pedestrian movement modeling and molecules in material science modeling. These dynamic modeling approaches share a commonality in computational approach where a series of ordinary differential equations are solved for the particle’s motion and equilibrium under active interactions and boundary conditions. In general, particle dynamics approaches are computationally intensive and address problems that require a vast parameter design space. In this dissertation, a computational paradigm is developed for large-scale particle dynamics modeling and demonstrated by three process design applications, (a) infection risk modeling for …

The Evidence Is In: Breast Implants Are Making People Sick Increasingly Women Find When They Have Their Implants Removed, Their Symptoms Improve—Or Disappear., Pamela J. Appea

Capstones

Over the years, breast implants have been associated with a wide range of health issues, including cancer and autoimmune diseases, but surprisingly these issues are not well known to the public. In 1992, health concerns led the FDA to halt the use of silicone gel breast implants for cosmetic surgeries, although implants containing saline remained available (it was during this period that the Oprah segment that Gmitro remembered ran on TV). In 2006, after more research, which some experts believe was insufficient, the FDA re-approved the use of silicone implants. In 2019, a novel “textured” breast implant manufactured …

Engineering Functionalized Nanomaterials For Targeting And Ablating Biofilms On Surfaces, Dhanush Lakmal Yapa Amarasekara Dec 2024

Engineering Functionalized Nanomaterials For Targeting And Ablating Biofilms On Surfaces, Dhanush Lakmal Yapa Amarasekara

Theses and Dissertations

Biofilms on both the surface and interior of medical devices pose major challenges, including hindering device performance and serving as infection sources. These biofilms form a protective layer around bacteria, shielding them from the body’s immune system and antibiotics. This makes infections difficult to treat, as the bacteria within the biofilm are resistant to both antibiotics and immune defenses. Additionally, biofilms can induce inflammation and irritation in nearby tissues, leading to further health complications. Antibiotic resistance raises concerns about the emergence of multi-drug resistant (MDR) pathogens. MDR infections are difficult to treat, leading to prolonged hospital stays and the need …

Elevated Ambient Temperature Associated With Reduced Infectious Disease Test Positivity Rates: Retrospective Observational Analysis Of Statewide Covid-19 Testing And Weather Across California Counties., Nicholas Wing-Ping Kwok, Joshua Pevnick, Keith Feldman

Manuscripts, Articles, Book Chapters and Other Papers

BACKGROUND: From medication usage to the time of day, a number of external factors are known to alter human body temperature (BT), even in the absence of underlying pathology. In select cases, clinical guidance already suggests the consideration of clinical and demographic factors when interpreting BT, such as a decreased threshold for fever as age increases. Recent work has indicated factors impacting BT extend to environmental conditions including ambient temperature. However, the effect sizes of these relationships are often small, and it remains unclear if such relationships result in a meaningful impact on real-world health care practices.

OBJECTIVE: Temperature remains …

Why Law "Beyond The State" Need Not Be A Threat To Democratic Values, Paul Schiff Berman Dec 2024

Why Law "Beyond The State" Need Not Be A Threat To Democratic Values, Paul Schiff Berman

Buffalo Law Review

Discussions of “law beyond the state” almost inevitably run into objections from those who believe that sub- or supranational legal orders necessarily subvert local democratic governance. Self-proclaimed populists and others express concern that the “will of the people” will be unduly subjected to the dictates of “cosmopolitan elites”1 or local factions, or corporate capture. These objections range across the political spectrum. Those on the right tend to focus on concerns that transnational orders will impose human rights or immigration rules on a national polity, while those on the left worry about trade regimes that might impose local labor or environmental …

Staphylococcus epidermidis, a common commensal bacterium, is a leading cause of nosocomial catheter-associated bloodstream infections. S. epidermidis sequence type 2 (ST2) is specifically recognized globally for causing invasive disease. In this study, we identified a novel putative integrated conjugative element, pICE-Sepi-ST2, unique to the genomes of S. epidermidis ST2. Our investigation identified pICE-Sepi-ST2 in all ST2 isolates from bloodstream infections. Meanwhile, ST2 isolates from other infection sources, such as catheters, prosthetic joints, and fracture fixations, showed variable pICE-Sepi-ST2 prevalence. pICE-Sepi-ST2 encodes two putative cell wall anchored proteins that we have designated SesX and SesY. Biochemical characterization of SesY revealed …

Medically important pathogenic fungi invade vertebrate tissue and are considered primary when part of their nature life cycle is associated with an animal host and are usually able to infect immunocompetent hosts. Opportunistic fungal pathogens complete their life cycle in environmental habitats or occur as commensals within or on the vertebrate body, but under certain conditions can thrive upon infecting humans. The extent of host damage in opportunistic infections largely depends on the portal and modality of entry as well as on the host's immune and metabolic status. Diseases caused by primary pathogens and common opportunists, causing the top approximately …
