Sepsis remains a major challenge that necessitates improved approaches to enhance patient outcomes. This study explored the potential of machine learning (ML) techniques to bridge the gap between clinical data and gene expression information to better predict and understand sepsis. We discuss the application of ML algorithms, including neural networks, deep learning, and ensemble methods, to address key evidence gaps and overcome the challenges in sepsis research. The lack of a clear definition of sepsis is highlighted as a major hurdle, but ML models offer a workaround by focusing on endpoint prediction. We emphasize the significance of gene transcript information …

The challenge of managing aquatic connectivity in a changing climate is exacerbated in the presence of additional anthropogenic stressors, social factors, and economic drivers. Here we discuss these issues in the context of structural and functional connectivity for aquatic biodiversity, specifically fish, in both the freshwater and marine realms. We posit that adaptive management strategies that consider shifting baselines and the socio-ecological implications of climate change will be required to achieve management objectives. The role of renewable energy expansion, particularly hydropower, is critically examined for its impact on connectivity. We advocate for strategic spatial planning that incorporates nature-positive solutions, ensuring …

School of Biological and Marine Sciences

As a land-locked country in Southern Africa, Zambia is rich in natural resources suitable for fish production. However, aquaculture is still in its infancy with enormous potential as most of Zambia’s fish now comes from capture fisheries. The aim of this study was to review the recent progress, status, challenges, and opportunities in Zambia’s aquaculture industry while drawing some lessons from China’s aquaculture experience, focusing on policy instruments, external support, quality seeds, and the investment environment for possible adaptation. To achieve this, we reviewed the existing literature about aquaculture in Zambia. This paper starts by reviewing the overall fishery sector’s …

Sieving Silence: The Communal Question And The Archive Of Indian Constitutional History, Kanika Gauba Jan 2024

Sieving Silence: The Communal Question And The Archive Of Indian Constitutional History, Kanika Gauba

National Law School Journal

This paper shows how the archive of Indian constitutional history limits access to constitutional disagreement. It attempts an alternative framing of the issue of political safeguards for religious minorities (‘the communal question’) as a triangle of disagreement among the Sikhs, the Anglo‑Indians, and Sardar Vallabhbhai Jhaverbhai Patel. However, the task is complicated by the silence in the official archives on all three actors. If, as critical historians argue, silence is the very condition of the archive and the writing of history, this paper argues that the historian must ‘sieve’ the inevitably silent archives of Indian constitutional history for perturbations, follow …

Global food production intensification presents a major hurdle to ensuring food security amidst a growing world population. Widespread use of chemical fertilizers in recent decades has risked soil fertility, compounded by the challenges posed by climate change, particularly in arid regions. To address these issues, adopting plant growth-promoting (PGP) bacteria stands out as a promising solution, offering multifaceted benefits to arid agroecosystems. We isolated a bacterial strain, SW7, from mangrove sediment, characterised the entire genome followed by phylogenetic analyses, and evaluated its in-vitro PGP activity. Subsequently, we examined its impact on tomato seed germination and plant growth. The strain SW7 …

Cheating And The Effect Of Promises In Indian And German Children, Patricia Kanngiesser, Jahnavi Sunderarajan, Jan K. Woike Jan 2024

Cheating And The Effect Of Promises In Indian And German Children, Patricia Kanngiesser, Jahnavi Sunderarajan, Jan K. Woike

School of Psychology

Cheating is harmful to others and society at large. Promises have been shown to increase honesty in children, but their effectiveness has not been compared between different cultural contexts. In a study (2019) with 7- to 12-year-olds (N = 406, 48% female, middle-class), voluntary promises reduced cheating in Indian, but not in German children. Children in both contexts cheated, but cheating rates were lower in Germany than in India. In both contexts, cheating decreased with age in the (no-promise) control condition and was unaffected by age in the promise condition. These findings suggest that there may exist a threshold beyond …

Application And Development Of Conjugation Technology To Enhance Therapeutic Index Of Tlr7/8 Agonists For Vaccines, Noah Robert Powers Jan 2024

Application And Development Of Conjugation Technology To Enhance Therapeutic Index Of Tlr7/8 Agonists For Vaccines, Noah Robert Powers

Graduate Student Theses, Dissertations, & Professional Papers

Toll-like receptor 7/8 (TLR7/8) agonists represent a promising vaccine adjuvant target as they directly activate innate Th1-type proinflammatory responses inducing both humoral and cell mediated immunity. However, co-delivery of the small molecule agonists with subunit antigens to the target dendritic cells (DCs) is difficult due to their highly divergent physical and chemical properties resulting in rapid dissociation of the two components. Conjugation of adjuvant to antigen can co-deliver to DCs, mimicking the natural infection these cells have evolved to detect though successful formulation can be difficult due to poor aqueously soluble adjuvants and antigen instability. We have previously shown and …

Integrating Individual And Species Traits To Better Identify Risks To Persistence And Survival In Amphibian Populations, Ross Kenneth Hinderer

Graduate Student Theses, Dissertations, & Professional Papers

Amphibians have experienced major declines and are highly vulnerable to the impacts of climate change. Yet we still lack sufficient information to understand where amphibians are most vulnerable to changes in climate and how effects might manifest. In this dissertation, I worked to understand risks due to climate change and investigate effective indicators of survival in amphibians. To predict which species might be most affected by increasingly dry conditions, in Chapter 1, I extended the climate change vulnerability assessment framework to rank amphibian vulnerability based on projected climate change and species life history traits. Traits including clutch size, daily activity …

Graduate Student Theses, Dissertations, & Professional Papers

Agrobiodiversity, the diversity of crops that been co-produced through humanenvironment interactions over millennia, constitutes an important part of humanity’s cultural heritage. Agrobiodiversity is as much about the local ecosystem and biotic resources, such as soil, water, pollinators, and microbes, as it is about human processes like sociocultural and linguistic practices, including farming methods, knowledge, skills, and natural resource management. At the center of these dynamics are the smallholder farmers who engage in a range of stewardship practices that contribute to the diversification and evolution of the native crops that in turn support their food security. This research focuses on understanding …

Beyond The Participation Gap: Exploring How Affinity Groups Make Outdoor Recreation More Accessible And Inclusive For Underserved Communities, Jaclyn Fox Rushing

Graduate Student Theses, Dissertations, & Professional Papers

Outdoor recreation is associated with numerous health and wellness benefits for participants. Yet, there has been a long-term underrepresentation of underserved communities in outdoor recreation (i.e., people of color, LGBTQ+, and people with disabilities). This phenomenon has spurred substantial research since the 1970s, which has characterized the problem well and identified many of the underlying reasons. In recent years, there have been widespread efforts to increase relevance, diversity, equity, and inclusion (RDEI) in outdoor recreation and public land management, however participation among underserved communities remain relatively unchanged despite an increasingly diverse American public. This study seeks to move beyond describing …

Tarantula Toxin Transcripts And Toxin Structure Prediction By Alphafold, Christine Vega, Rebekkah Sepulveda, Ying Jia

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

Tarantula venom predominantly comprises peptides and small proteins (venom toxins). To explore this further, we generated a combined gland cDNA library from five different tarantula species. Subsequently we randomly sequenced 752 clones, which led to the identification of sixteen distinct toxin groups, including one novel group. Each group consists of a minimum of three identical sequences. A comprehensive analysis of these toxin groups was carried out, and we simulated the 3D structures of certain predicted toxins. These transcripts can be used to the production of venom toxins through recombinant techniques, offering promising applications in various biomedical applications.

The Development And Validation Of A Novel Patient-Derived 3d Meningioma Cell Culture Model, Laurien Lisette Van De Weijer

Peninsula Medical School Theses

Meningiomas are the most frequent intracranial brain tumours. Current treatment options of surgery and radiotherapy are sometimes insufficient and effective systemic therapies remain unestablished. The development of accurate in vitro systems to model the complexity of meningioma pathology is essential for predicting drug response and developing novel therapeutics. Therefore, in this study, I established an easy-to-use in vitro patient-derived meningioma spheroid model that maintained the morphological and molecular features of the parental tumours, including tissue histology, the tumour microenvironment, and the mutational profile. Comprehensive characterisation of the global transcriptomes of the novel patient-derived spheroids with traditional meningioma monolayer cultures and …

Pre-Clinical Targets For Ischemic Brain Injury, Daniel Morgan Jan 2024

Pre-Clinical Targets For Ischemic Brain Injury, Daniel Morgan

Peninsula Medical School Theses

Stroke is the second most common cause of disability worldwide, and 87% of strokes result from a blockage in the cerebral blood flow which results in ischemia. Injury to the myelinated axons located in white matter is observed in ischemic injuries but is generally underappreciated. Research has shown that there is extensive vesicular glutamate release during ischemia, causing excitotoxic injury to oligodendrocytes, resulting in demyelination. Glutamate receptor antagonists have been trialled in stroke patients but fall short due to adverse side effects from high dose preparations. Stroke is largely a disease of ageing, and may occur in the presence of …

Hermit Crabs As Model Species For Investigating The Behavioural Responses To Pollution, Mark Briffa, Gareth Arnott, Jörg D. Hardege Jan 2024

Hermit Crabs As Model Species For Investigating The Behavioural Responses To Pollution, Mark Briffa, Gareth Arnott, Jörg D. Hardege

School of Biological and Marine Sciences

Human impacts on the environment affect organisms at all levels of biological organisation and ultimately can change their phenotype. Over time, phenotypic change may arise due to selection but individual phenotypes are also subject to change via genotype × environment interactions. In animals, behaviour is the most flexible aspect of phenotype, and hence the most liable to change across environmental gradients including exposure to pollution. Here we review current knowledge on the impacts of pollution, broadly defined to include the release of substances, energy, and the effects of carbon emissions, on the behaviour of a highly studied group, the globally …
