Accurate assessment of drug receptor occupancy (RO) holds significant importance in both drug development and personalized medicine, as it facilitates quantitative characterization of dose-response relationship for a given drug. This information not only aids appropriate dose selection for clinical trials but also guides personalized dose optimization in precision medicine.
Molecular imaging, utilizing receptor-specific imaging agents for tissue visualization, has emerged as the major option for in vivo measurement of tissue RO. Cell and gene therapies, a ground-breaking class of drugs that can heal genetic diseases and cure cancer, often have treatment costs over $1 million. Most cell and gene therapies are delivered using virus-based delivery vectors, which are incredibly expensive to manufacture. Liquid chromatography is the favored method of viral vector purification since it is easily scalable and can be functionalized to purify by biospecificity, charge, hydrophobicity, or size. However, chromatography-based purification processes are highly expensive and usually only recover 20-50% of the viral vectors. Biomaterials that integrate multiple functionalities, mimic the extracellular matrix (ECM) microenvironment to support cellular growth, and adhere robustly to damaged tissues are highly needed to advance tissue engineering. Protein-based biomaterials are promising due to their inherent biocompatibility, biomimicry, biodegradation, and cell-supportive properties. Herein, by leveraging the unique ability of dithiolanes to generate on-demand in-situ thiols, we developed a new class of dithiolane-modified, protein-based biomaterial that combines unique, seemingly opposing functions for tissue engineering. Dithiolane-modified gelatin, a model protein used herein, enabled photoinitiator-free photo-crosslinking to form multi-functional gelatin-dithiolane (GelDT) hydrogels, which displayed exceptional long-term stability in cell culture media (>28 …

Greenhouse grafting and field trials were conducted to examine the contribution of the RED NAC transcription factor on low nitrogen phenotypes in hybrid poplar (P. tremula x P. alba). Reciprocal grafting trials showed both root and shoot expression contribute to phenotype under low nitrogen conditions, and shoot expression has a leading role in contribution to low-nitrogen phenotype. The impact of RED NAC expression level on auxin polar transport was investigated using a DR5::GUS auxin-responsive reporter gene line as rootstock for RED NAC modified scions. With global biodiversity in decline and many species isolated in fragmented habitats, conservation efforts increasingly rely on natural recolonization and translocation to restore ecosystems. These strategies face challenges such as genetic erosion and habitat limitations, highlighting the need for effective monitoring practices. This research assesses the success of wolf recovery initiatives by examining long-term genetic and genomic health using both traditional and advanced molecular methods. First, we investigated the genetic health of 19 gray wolves (Canis lupus) relocated to Isle Royale National Park to restore ecosystem balance. The climate-altering effects of atmospheric CO2 make it necessary to develop innovative technologies that can remove it from the atmosphere. Mineral carbonation is a scalable technology to sequestrate and store CO2 in a stable carbonate mineral form. The efficiency of the mineral carbonation process depends on both feed mineralogy and process parameters. Comprehensive modeling and prediction of mineral carbonation efficiency for different CO2 – reactive rocks has not been achieved in the past. Hypertrophic cardiomyopathy (HCM) is the most common genetic heart disorder. HCM is characterized by cardiac hypertrophy, fibrosis, and an increased risk of fatal arrhythmias and sudden cardiac death. Previous studies in humans with HCM have demonstrated increased cardiac norepinephrine spillover and reduced beta-1 receptor (β1R) expression in the left ventricle (LV). We have previously demonstrated that cardiac sympathetic tone is elevated in an alpha-tropomyosin mutant mouse model of HCM. We hypothesized that HCM mice would demonstrate reduced β1R expression in the LV and attenuated heart rate (HR) and dP/dt max responses to ramped infusion of the β1R agonist, dobutamine. …

Associative learning, a key cognitive process seen across the animal kingdom, enables organisms to form connections between stimuli and adapt their behaviors based on past experiences. A particularly powerful example is fear conditioning, where animals learn to associate a neutral stimulus with an aversive one, allowing them to predict and avoid potential threats. Inspired by this mechanism, this project implements associative learning on an unmanned ground vehicle (UGV) to develop adaptive behavior through neuromorphic principles. Despite the critical nature which microbial communities play in the natural world and human civilization, the breadth of understanding remains shallow. Challenged by scale, high variability between environments, and extensive diversity, microbial ecologists strive to understand connections between a community’s structure and function, as well as the ecological and evolutionary mechanisms underlying observed natural patterning of communities. Organisms are distributed differentially across world’s surface based on many environmental characteristics. What part do physical characteristics play in demarcating the distribution of species? Using several international fiddler crab collections and GIS-linked sets of habitat factors, I sought an answer to this question. Geographic distribution data for 23 species from over 1700 collecting sites situated along the coasts of Europe, Africa, North America, Central America, the Caribbean Islands, and South America were used to characterize coastal biogeographic biomes around the perimeter of the North and South Atlantic Ocean. With growing concern over environmental sustainability and dwindling fossil resources, it is crucial to prioritise the development of alternative feedstocks to replace fossil resources. Spent coffee grounds (SCGs) are an environmental burden with an estimated six million tons being generated on a wet basis annually, globally. SCGs are rich in cellulose, lignin, protein, lipids, polyphenols and other bioactive compounds which are important raw materials for use in industries including pharmaceuticals and cosmetics. Furthermore, the energy sector has the potential to capitalize on the high calorific value of SCGs for biofuel and biogas production, offering a sustainable alternative to fossil fuels. …

This study investigated the potential of soy-based materials as a sustainable alternative in fashion design, focusing on creating textiles using batik techniques. The process involved producing soymilk, bonding pigments, repurposing soy pulp, employing batik methods, and enhancing textures with soy fibers. The resulting sample showcased a visually captivating moth motif on soy textile, demonstrating soy materials' aesthetic and environmental potential. The findings affirm the viability of soy textiles in fashion, offering a biodegradable, renewable option with reduced water and chemical usage compared to traditional materials. Antipatharians, or black corals, form a key component of the benthic fauna found throughout the Gulf of Mexico (GOM). In the northwestern GOM, they are a common member of the communities found on the natural relic coral—algal banks known as the South Texas Banks as well as on artificial reefs. They are known to provide important habitat for many ecologically and economically important species in the area but are relatively understudied. This paper presents a two-stage feature selection scheme using machine learning techniques. In the first stage a wrapper method is adopted to select various combinations of subsets of features from the original dataset. The performance of the model is evaluated by three classifiers: K-Nearest Neighbor (KNN), Support Vector Machines (SVM), and Random Forest (RF). In the second and final stage, a sequential backward feature selection Method is applied. The proposed method is demonstrated on eighteen datasets and the average classification accuracy of eighteen datasets achieved is 89.81%, 87.55%, and 89.82% using the KNN, SVM, and RF classifiers, respectively with a …

This article presents an academic review of the collection of articles “on translation” and entitled river in an ocean. It engages with the foreword by Françoise Vergès and the introduction by editor Nuzhat Abbas, which set the tone of ‘decolonial feminism’ that permeates the essays. The reviews of the eleven essays—by women from southeast Asia, Africa, Palestine and Saudi Arabia—come from the perspective of feminist translation studies but pay careful attention to their very specific concerns around exile and life in diaspora.

Climate change remains an urgent, ongoing global issue that requires critical examination of institutional polluters. This includes the world’s largest institutional consumer of petroleum: the United States military. The Department of Defense (DoD) is a massive institution with little oversight, a carbon footprint spanning the globe, a budget greater than the next ten largest nations combined, and overly generous exemptions to environmental regulations and carbon reduction targets. Global right-wing anti-gender movements have developed a transnational pattern in their uses of language as a means of subjugation and of control, and this article examines manifestations of this pattern in two specific states: Brazil and Russia. By exploring the anti-gender languaging, rhetoric, and legislation advanced by Jair Bolsonaro (President of Brazil 2019–2022) and Vladimir Putin (President of Russia 2000–2008 and 2012 to the present) and their supporters, we find that, in both contexts, anti-gender campaigns are reactions to and also drivers for increasingly dramatic political and social conflicts, and in each case, anti-gender ideologies are embedded in larger right-wing …

The mainstream adoption of cryptocurrencies has led to a surge in wallet-related issues reported by ordinary users on social media platforms. In parallel, there is an increase in an emerging fraud trend called cryptocurrency-based technical support scam, in which fraudsters offer fake wallet recovery services and target users experiencing wallet-related issues.In this paper, we perform a comprehensive study of cryptocurrency-based technical support scams. We present an analysis apparatus called HoneyTweet to analyze this kind of scam. Through HoneyTweet, we lure over 9K scammers by posting 25K fake wallet support tweets (so-called honey tweets). Jacques Maritain’s ideas about democratic renewal remain as important and inspirational as ever. A less well-known element of Maritain’s political thinking is his support for grassroots community organizing. This support, peppered throughout his writings, comes into stark relief when examining his longtime friendship and correspondence with Saul Alinsky, the dean of community organizing in the US. This article argues, first, that we must understand Maritain and Alinsky’s friendship to properly appreciate the legacy of Maritain’s political thought and, second, that understanding the complementarity of their approaches allows us to see that together these two friends present a powerful strategy for …

The idea of average is utilized in a variety of scenarios, yet the literature has indicated that students have multiple disconnected understandings for the concept of average. In this study, I provide an account of two students who reconciled their meanings for average by considering an average as a replacement with a constant value. This report discusses an intervention that teachers can leverage to help their students make their meanings for average coherent and conceptually based.
