In the future, sustainability in the dairy industry will require creative and scale-able solutions for better utilization of the primary solid component in milk: lactose. One promising avenue of “up-cycling” underutilized lactose is converting it to valuable rare sugars. Rare sugars like D-tagatose and D-allulose are increasing in popularity extremely quickly. D-tagatose and D-allulose are hexose sugars, D-allulose being an epimer of D-fructose and D-tagatose an isomer of D-galactose. They have excellent functional properties in food systems with clean sweetness profiles similar to sucrose. Critically, they also have excellent nutritional characteristics, such as low caloric values and low glycemic indices. …

Diffuse Large B-cell Lymphoma (DLBCL) is the most common aggressive non-Hodgkin lymphoma (NHL). Despite the use of newer agents, such as polatuzumab vedotin, more than one-third of patients have ultimately relapsed or experienced refractory disease. MiRNAs are single-stranded, ~22-nucleotide-long RNAs that interact with their target RNA. They are significant regulators of post-transcriptional gene expression. One significant miRNA, miR-155, is involved in the pathophysiology of DLBCL and it is a critical modulator of hematopoiesis, inflammation, and immune responses. Targets of miR-155, such as histone deacetylase 4 (HDAC4), suppressor of cytokine signaling-1 (SOCS1) and immune cells, play a crucial role in DLBCL …

Background Despite the high rates of firearm ownership and firearm-related injuries and mortalities in Southern US states, understandings on the factors contributing to these are lacking.
Methods Using wave 10 (2021) data from a longitudinal study, we examined firearm-related experiences among 636 ethnically diverse young adults (mean age=26 years; 62% female) in Texas, USA.
Results Just over half of participants had ready access to firearms, with 22.3% having carried a firearm outside of their home, 4.9% having been threatened with a firearm by a romantic partner and 4.4% by a non-romantic partner. More firearm access and carriage were reported in …

Subunit Kir6.1 and SUR2B form a subtype of ATP sensitive potassium channel (KATP) which is mainly expressed in vascular, gastrointestinal and lymphatic smooth muscles controlling the muscle contractility tones. Gain-of-function mutations of this channel lead to Cantu Syndrome, featured with hypertrichosis, coarse facial expression, cardiac hypertrophy and sometimes lymphedema. Conventional patch-clamp assays are available for studying the consequences of Cantu Syndrome, however, the throughput is low. In this thesis, two higher throughput fluorescence -based assays, thallium flux assay and DiBAC4(3) assay with unique capabilities of large and small current detections respectively were developed for more comprehensive mutation characterizations together with …

Police officers in Trinidad and Tobago face highly stressful environments due to increasing violent crimes and threats to their lives. Organizational stress contributes to emotional exhaustion, highlighting the need for research on stress and coping among these officers. The purpose of this basic qualitative study was to explore job-related stressors and coping strategies that male police officers in Trinidad and Tobago use to deal with their everyday job demands. Han Selye’s theory of stress and Lazarus’ transactional model of stress and coping were used as theoretical frameworks for this study. Plasmodium, the causative agent of malaria, infects hepatocytes prior to establishing a symptomatic blood stage infection. During this liver stage development, parasites reside in a parasitophorous vacuole (PV), whose membrane acts as the critical interface between the parasite and the host cell. It is well-established that host cell autophagy-related processes significantly impact the development of Plasmodium liver stages. Expression of genes related to autophagy and lysosomal biogenesis is orchestrated by transcription factor EB (TFEB). Although various joint injuries result in post-traumatic osteoarthritis (PTOA), differences in chondrocyte response to specific injuries, such as blunt compression or fracture, are unclear. Furthermore, the role of underlying joint inflammation, or synovitis, is often not considered. We investigated how injury mechanisms and underlying synovitis affect chondrocyte gene expression using osteochondral injury models with synovial co-culture. We hypothesized that the state of synovitis as well as the mechanism of biomechanical cartilage injury differentially alter the gene expression of chondrocytes and that these responses are regulated by the pro-inflammatory cytokine interleukin 1 (IL-1). Through their involvement in various processes such as phagocytosis of foreign materials and damaged cellular debris, regulatory factor release, and surveillance, the macrophages of the brain, microglia, maintain homeostasis within the central nervous system. These diverse microglial functions therefore require transition to a range of phenotypic states. The epigenetic regulation of this activity, especially through trimethylation of lysine 4 on histone H3, and its regulator, lysine demethylase (KDM)5B, is an emerging research area. The sexual abuse and prostitution of Black women is a system of oppression that has its foundation in slavery and is still embedded in the culture of the U.S. This ongoing practice of domination, racism, sexism, and class exploitation is at work daily in the lives of Black women. Human noroviruses (HuNoVs) are a significant cause of epidemic and sporadic acute gastroenteritis worldwide. The lack of a reproducible culture system hindered the study of HuNoV replication and pathogenesis for almost a half-century. This barrier was overcome with our successful cultivation of multiple HuNoV strains in human intestinal enteroids (HIEs), which has significantly advanced HuNoV research. We optimized culture media conditions and generated genetically modified HIE cultures to enhance HuNoV replication in HIEs. Building upon these achievements, we now present new insights into this culture system, which involve testing different media, unique HIE lines, and additional virus strains. HuNoV infectivity …

In addition to modulating serotonin transport, selective serotonin reuptake inhibitors (SSRIs) have multiple other mechanisms that may contribute to clinical effects, and some of these latter actions prompt repurposing of SSRIs for non-psychiatric indications. In a recent study of the SSRIs fluvoxamine, fluoxetine and sertraline we found that, unlike the other two SSRIs, sertraline acutely inhibited LTP at a low micromolar concentration through inverse agonism of sigma 1 receptors (S1Rs). In the present studies, we pursued mechanisms contributing to sertraline modulation of LTP in rat hippocampal slices. For example, an AR head-mounted display (HMD) may include and/or represent circuitry, such as a driver unit, that controls and/or drives the brightness and/or luminance of a perceptual quantizer (PQ) and/or gamma display. In one example, the circuitry may implement 8-bit counters to control the brightness and/or luminance of pixels within the PQ or gamma display. Objectives: Mitral valve surgery is the reference treatment for severe symptomatic mitral regurgitation (MR). Percutaneous mitral valve interventions, such as the MitraClip procedure, offer an alternative, particularly for high-risk patients. The aim of this systematic review and meta-analysis was to analyze the safety and effectiveness of transcatheter mitral valve repair (TMVR) compared to surgical mitral valve repair or replacement (SMVR) in elderly patients with mitral regurgitation.
