Aquaporin-4 (AQP4) is a water channel protein that links the astrocytic endfeet to the blood-brain barrier (BBB) and regulates water and potassium homeostasis in the brain, as well as the glymphatic clearance of waste products that would otherwise potentiate neurological diseases. Recently, translational readthrough was shown to generate a C-terminally extended variant of AQP4, known as AQP4x, which preferentially localizes around the BBB through interaction with the scaffolding protein α-syntrophin, and loss of AQP4x disrupts waste clearance from the brain. Plastin-3 (PLS3) is a calcium-sensitive actin-bundling protein that has recently been linked to the development of childhood-onset osteoporosis. Clinical data suggest that PLS3 mutations lead to a defect in osteoblast function, however the underlying mechanism remains elusive. To investigate the role of PLS3 in bone mineralization, we generated MC3T3-E1 preosteoblast cells that are stably depleted of PLS3. Analysis of osteogenic differentiation of control and PLS3 knockdown (PLS3 KD) cells showed that depletion of PLS3 does not alter the first stage of osteoblast mineralization in which a collagen matrix is deposited, but severely affects the subsequent mineralization of that matrix. The bone is one of the most commonly affected organs in sickle cell disease (SCD). Repeated ischemia, oxidative stress and inflammation within the bone is largely responsible for promoting bone pain. As more individuals with SCD survive into adulthood, they are likely to experience a synergistic impact of both aging and SCD on their bone health. As bone health deteriorates, bone pain will likely exacerbate. Recent mechanistic and observational studies emphasize an intricate relationship between bone remodeling and the peripheral nervous system. Many preclinical and clinical studies support the use of antisense oligonucleotides (ASOs) as effective therapeutic strategies. However, acquiring ASOs for research purposes may be limited by partnerships with the pharmaceutical companies. Our lab previously developed an effective ASO strategy to lower human tau and reverse pathology in aged tauopathy model mice. Testing the efficacy of purchased tau lowering ASOs would provide support for these reagents as broad research tools. Purchased mouse and human tau lowering ASOs were infused or injected intracerebroventricularly into wildtype and tau transgenic mice. Vernal pools are small, ephemeral wetlands that become inundated each spring and provide many ecosystem services to the surrounding upland forests. They also provide critical habitat for amphibians and invertebrates, as their temporary nature keeps them free of fish and reduces predator populations. As part of a mapping project, we collected baseline field data on vernal pool characteristics throughout five Great Lakes National Parks: Pictured Rocks National Lakeshore, Sleeping Bear Dunes National Lakeshore, Apostle Islands National Lakeshore, Isle Royale National Park, and Voyagers National Parks. The Twin Lakes system, a series of lakes including Lake Gerald and Lake Roland, was historically categorized as oligotrophic but is now mesotrophic having experienced cultural eutrophication. A mass balance phosphorus budget model was constructed for the Twin Lakes system in Houghton, Co. Michigan for the 2022 stratified season. Additional spatial insights were gleaned through a survey of shoreline water quality, which corroborated the model results. The lakes currently experience substantial depletion of hypolimnetic dissolved oxygen during late summer stratification. A budget reveals approximately 22% (6.6 kg) of the current 29.5 kg total phosphorus inputs to Twin Lakes (during the …

The Keweenaw fault is perhaps the most significant geologic structure on the Keweenaw Peninsula, with an estimated 7-11 km of reverse slip juxtaposing volcanic strata of the ~1.1 Ga Portage Lake Volcanics above ~1.0 Ga Jacobsville Sandstone. The fault has been interpreted as a rift-bounding normal fault later inverted by compressional pulses of the Grenville Orogeny or, more recently, as part of a detached thrust fault system unrelated to an earlier normal fault. The fault is shown on published maps as a single continuous fault trace whose sinuosity implies multiple fault segments and complex slip dynamics. Mapping along Bête Grise …

This dissertation focuses on advancing Predictive Energy Management (PrEM) functions applied to modern connected and automated vehicles (CAV) cohorts. PrEM aims to utilize connectivity and ADAS functions to adaptively minimize vehicle energy consumption in a wide array of operations, extending the original control designed around a reduced set of test cycle procedures to adapt to real-world stochastic operating conditions. This research document is built upon three journal publications covering two PrEM schemes; the global cohort and local vehicle optimization paths. Both optimal control solutions are generated using various Neuroevolution centric processes.

The Southern Complex, located near Marquette, Michigan is in the southernmost portion of the Superior Province. This complex consists of granitoids and granitic gneiss that vary in composition. In twentieth century research, Hoffman (1987) and Tinkham (1997) concluded that the Southern Complex contained two units in which the granitoid was emplaced in the granitic gneiss. Recent research conducted by Petryk (2019) determined that the Southern Complex has a U-Pb zircon age of 2600 Ma. The Keweenaw area continues to be influenced by the century of copper mining that ended nearly 50 years ago. This project is focused on Torch Lake, an aquatic ecosystem that has been heavily impacted by mining waste disposal. The watershed has been impaired by mine discharge and tailings, smelter and smokestack plumes, and poor waste disposal practices. The lake is listed as a Great Lakes Area of Concern with beneficial use impairments of restrictions on fish consumption and a degraded benthic community. Enzymes are biological systems that aid in specific biochemical reactions. They lower the reaction barrier, thus speeding up the reaction rate. A detailed knowledge of enzymes will not be achievable without computational modeling as it offers insight into atomistic details and catalytic species, which are crucial to designing enzyme-specific inhibitors and impossible to gain experimentally. This dissertation employs advanced multiscale computational approaches to study the dynamics and reaction mechanisms of non-heme Fe(II) and 2-oxoglutarate (2OG) dependent oxygenases, including AlkB, AlkBH2, TET2, and KDM4E, involved in DNA and histone demethylation. For populations living with risk to rapid-onset environmental hazards, an effective early warning system (EWS) may be the most viable short- to mid-term solution for risk reduction. At Fuego volcano, Guatemala, more than 60,000 people distributed between more than 30 small communities live within the identified hazard zones for pyroclastic density currents (PDCS), highly lethal hot avalanches and surges of volcanic gases, rock, and ash. Despite ongoing risk reduction efforts by scientific and civil protection authorities, more than 400 people died during a paroxysmal eruption on 3 June 2018 when PDCs reached populated areas. Atmospheric clouds are crucial to weather and climate, and the rate at which droplets collide and coalesce to form precipitation is one of the fundamental controlling processes. The convection-cloud chamber allows the interactions between aerosols and cloud droplets produced by condensation to be investigated within a turbulent environment. Studying the full range of microphysical conditions in atmospheric clouds is not possible, however, unless conditions for droplet growth by collision and coalescence are also achieved. In this study, we explore the conditions favorable to collision-coalescence growth in convection-cloud chambers, extending previous work on steady-state droplet size distributions due to condensation alone. …

Renewable fuels from lignocellulosic biomass are an appealing option because they can seamlessly integrate into the existing fuel distribution infrastructure. Lignocellulosic biomass constitutes nonedible plant material obtained from plant cell walls. The natural recalcitrance of lignocellulosic biomass poses a challenge in accessing the cell wall carbohydrates during biochemical conversion. Despite various approaches, enzymatic hydrolysis of lignocellulosic biomass remains economically impractical due to incomplete knowledge about biomass recalcitrance and the influence of environmental factors on biomass quality.

The Archean Carney Lake Gneiss Complex (CLGC) is a suite of rocks on the southern margin of the Superior Province and within the Minnesota River Valley subprovince. It is one of many gneissic domes in Michigan, Wisconsin, and Minnesota whose complex geology is poorly understood due to a previous lack of economic interest in gneissic terranes and significant challenges to their investigation. Economic theory is often applied to plant strategies, arguing that trade-offs exist between maximizing quick returns on investment within short lifespans or producing longer-lived tissues with slower returns. While these trade-offs are well-studied aboveground, their influence on belowground traits, including root respiration, remains unclear. Resource heterogeneity and symbioses with mycorrhizal fungi may complicate root trade-off axes beyond those predicted by economics theory alone. Invasive emerald ash borer (EAB) (Agrilus planipennis Fairmaire) poses an imminent threat to the structure and function of North American hardwood forests, particularly black ash (Fraxinus nigra Marshall), and alters the hydrologic and ecological services of their wetlands. Black ash trees regularly grow in seasonally saturated soils and are responsible for hydrologic regulation and nutrient cycling. In this study, a gradient of black ash wetlands impacted by EAB were monitored to assess vegetation changes and near-surface soil nitrogen availability. Biomass combustion contributes a large amount of aerosol particles to the atmosphere impacting the climate system and human health. Identifying the molecular composition of biomass burning organic aerosol (BBOA) is challenging because of its complex nature. To predict the mobility of a vehicle over a terrain, one must understand the soil characteristics. In situ measurements performed by soldiers in the field are the current method of collecting this information, which is time-consuming, are only point measurements, and can put soldiers in harm's way. Therefore, this study investigates using remote sensing as an alternative approach to characterize terrain properties.

Cardiac resynchronization therapy (CRT) is a standard method of treating heart failure by coordinating the function of the left and right ventricles. However, up to 40% of CRT recipients do not experience clinical symptoms or cardiac function improvements. The main reasons for CRT non-response include: (1) suboptimal patient selection based on electrical dyssynchrony measured by electrocardiogram (ECG) in current guidelines; (2) mechanical dyssynchrony has been shown to be effective but has not been fully explored; and (3) inappropriate placement of the CRT left ventricular (LV) lead in a significant number of patients.

Woody biomass, particularly poplar, is a sought-after feedstock for lignocellulosic biofuels because of its year-round availability and high productivity. Because of cell wall components like lignin, poplar is inherently resistant to enzymatic saccharification. The experiment aims to determine whether the controlled overexpression of laccase genes alters the biomass composition and whether it can create poplar that is more digestible than the wildtype.
