Investigating Structural And Biochemical Characteristics Of A Peptidase Required For Staphylococcus Aureus Virulence, Sagar Bhusal Jan 2023

Investigating Structural And Biochemical Characteristics Of A Peptidase Required For Staphylococcus Aureus Virulence, Sagar Bhusal

Master's Theses

Staphylococcus aureus is an opportunistic Gram-positive pathogen that transiently colonizes 30% of the population at any given time. Upon breaching host skin or mucosal barriers, S. aureus can infect and cause disease in nearly all host tissues. Once inside the host, S. aureus expresses a plethora of virulence factors, mostly regulated by the Accessory Gene Regulatory (Agr) quorum sensing system, which facilitates the evasion of immune responses or directly promotes pathogenesis. The S. aureus Agr system relies on the production of a small peptide known as an auto-inducing peptide (AIP) to control virulence gene expression in response to cell density. …

Physical Activity, Self-Determination, And Self-Conscious Emotions In Adolescents And Young Adults With And Without Autism Symptomatology, Dakota Morales Jan 2023

Physical Activity, Self-Determination, And Self-Conscious Emotions In Adolescents And Young Adults With And Without Autism Symptomatology, Dakota Morales

Master's Theses

Previous literature suggests that autistic individuals engage in physical activity less frequently compared to typically developing peers (Bandini et al., 2013; Jones et al., 2017). This is noteworthy because exercise interventions for autistic individuals have found that engaging in physical activity can improve motor, social, and behavioral skills (Anderson-Hanley et al., 2011; Duffy et al., 2017; Nicholson et al., 2011; Oriel et al., 2011). Thus, the present study seeks to understand what factors may promote physical activity engagement, such as self-determination motivation regulations and body-related self-conscious emotions, in adolescents and young adults with and without autism symptomatology. Participants included 51 …

How To Build A Cortex: Coordinated Assembly Of Cortical Septins And Actomyosin In The Leader Bleb, Asia Marie Paguntalan Jan 2023

How To Build A Cortex: Coordinated Assembly Of Cortical Septins And Actomyosin In The Leader Bleb, Asia Marie Paguntalan

Master's Theses

In confined spaces, migrating cells can undergo mesenchymal-to-amoeboid transitions by altering their cortical dynamics and adhesion with the environment. Septins frequently associate with cortical actin and non-muscle myosin (NMII), but the functional nature of these interactions remains unclear. Upon non-adhesive confinement and NMII enrichment, fibroblasts can switch to a fast, leader bleb-based mode of motility, characterized by the absence of adhesions and stress fibers and formation of a single, elongated leader bleb. During this transition, cortical actin remodeling and polarized NMII contractility drive leader bleb stabilization by generating long-range cortical flows, in coordination with changes in septin localization and assembly …

Tracking The Legacy Of "Inner-City" Catholic Schools: An Analysis Of U.S. Elementary Catholic School Organizational And Demographic Data, Andrew F. Miller, Annie Smith, Kierstin M. Giunco, Audrey A. Friedman, Myra Rosen-Reynoso, Charles T. Cownie Iii Jan 2023

Tracking The Legacy Of "Inner-City" Catholic Schools: An Analysis Of U.S. Elementary Catholic School Organizational And Demographic Data, Andrew F. Miller, Annie Smith, Kierstin M. Giunco, Audrey A. Friedman, Myra Rosen-Reynoso, Charles T. Cownie Iii

Journal of Catholic Education

Over the past twenty years, Catholic elementary schools that self identify as “inner-city” have closed at a higher rate than Catholic schools in other locations. These schools have also long been associated with a legacy of effectively serving low-income students, students of color, and recent immigrant students, suggesting that the persistent closure of these schools may have a negative impact on these communities. In this paper, we set out to assess the extent to which there have been demographic or organizational changes over the past twenty years in these “inner-city” schools. We found that while these schools do still serve …

Leading In Liminality: Implications On Individual And Collective Identity, And Knowledge Creation For School Leaders, David Sorkin, Melodie Wyttenbach, John Reyes, Michael Warner Jan 2023

Leading In Liminality: Implications On Individual And Collective Identity, And Knowledge Creation For School Leaders, David Sorkin, Melodie Wyttenbach, John Reyes, Michael Warner

Journal of Catholic Education

In the spring of 2020, schools of all sectors across all nations were forced to close their doors as COVID-19 rippled through communities. Drawing upon the concept of liminality, which refers to a stage, state, or period of transition (Soderlund & Borg, 2017), this study investigated the intersections of the experience of liminality during the pandemic and functioning for Catholic school leaders. Interviews with urban school leaders were analyzed to understand key characteristics of liminality as experienced by school leaders within organizations. Findings indicate that school leaders responded to the liminal experience by sharpening the focus of work, which simultaneously …

The Positive Impacts Of A Professional Learning Community Model On Student Achievement In Small Schools, Christina Mariani-Petroze

Journal of Catholic Education

This study explores the impact of professional learning communities on student achievement in a small school setting. Aaron Hansen’s book, How to Develop PLCs for Singletons and Small Schools, offered a guide for arranging vertical, grade-level teams with one teacher per grade level at one private, K-8 school. The faculty engaged in high quality, effective professional development using PLC objectives and norms to analyze NEWA MAP data. They adapted instructional practices and implemented formative assessments to influence student growth in math and reading scores. Results indicate that the PLC training that took place between the Fall and Winter MAP testing …

Moral Formation In A Culture Of Relativism: Correlates Of Universalism And Relativism In The Moral Outlooks Of Emerging Adults, Steven Crawford Hayward Jan 2023

Moral Formation In A Culture Of Relativism: Correlates Of Universalism And Relativism In The Moral Outlooks Of Emerging Adults, Steven Crawford Hayward

Journal of Catholic Education

This study explores some of the demographic, personal, and experiential factors of emerging adults that correlate to a spectrum of moral outlooks ranging from moral universalism to moral relativism. Data was gathered from 466 volunteer undergraduate students. Respondents’ demographic, experiential, and personal factors served as independent variables. The relativism index score form Forsyth’s Ethics Position Questionnaire (1980) served as the dependent variable. Results suggest several conclusions: First, adults in the life of youth are a significant factor in moral outlook, exerting influence in both universalistic and relativistic directions. Second, religious practice and involvement contribute to a universal moral outlook. Third, …

The Effect Of Intrafamilial Racism On Biracial Identity Conflict, Lauren Tan Jan 2023

The Effect Of Intrafamilial Racism On Biracial Identity Conflict, Lauren Tan

Master's Theses

This study aimed to describe the relation between experiences of intrafamilial racism (i.e., racism perpetrated by family members against other family members), caregiver support, and identity conflict in biracial individuals. Identity conflict is defined as perceived incompatibility with two or more aspects of one’s identity (Sarno et. al., 2015) and is an experience likely to be common among biracial individuals. Regression analyses were conducted to determine the relation between intrafamilial racism, caregiver support, and identity conflict. Moderation analyses were performed to determine the impact of intrafamilial racism on identity conflict at different levels of caregiver support. We hypothesized that intrafamilial …

Identification Of New Substrates From Pseudomonas Aeruginosa Exos, Adam V. Thota Jan 2023

Identification Of New Substrates From Pseudomonas Aeruginosa Exos, Adam V. Thota

Master's Theses

Pseudomonas aeruginosa is a gram-negative environmental bacterium that uses its intrinsic and acquired antibiotic resistance to infect patients with compromised epithelium, such as cystic fibrosis and burn wounds, and corneal keratitis. In order to cause disease, most strains of P. aeruginosa use the type three secretion system (T3SS) to disrupt cellular signaling and integrity. However, P. aeruginosa may also secrete exotoxins after invading host cells, which prolongs host cell death and maintains an intracellular niche. Of the T3 secreted effector toxins, ExoS, specifically the ADP-ribosyltransferase activity of ExoS, was shown to delay the death of invaded corneal cells, but it …

Changing Vaccine Hesitant Attitudes Of Parents Using Moral Persuasion, Max Vitro Jan 2023

Changing Vaccine Hesitant Attitudes Of Parents Using Moral Persuasion, Max Vitro

Master's Theses

In the fight against Covid-19, overt, science-based messaging is not enough to persuade everyone to get vaccinated no matter how encouraging the data. Recent studies on attitudes toward vaccines and other health-promoting measures have provided clues as to why so many are still opposed, suggesting many who were resistant had reasons that were rooted along moral grounds. This process of moralization occurs when a belief becomes a moral matter of ‘right and wrong’ rather than a means to an end. Because moral beliefs are more entrenched, they’ve proven much more difficult to change. There is one approach that could increase …

Teachers’ Perceptions Of Ells By Racial Identity: A Pilot Study, Ha Tran Jan 2023

Teachers’ Perceptions Of Ells By Racial Identity: A Pilot Study, Ha Tran

Master's Theses

ABSTRACTAs racial and political tensions grow within the United States, it is not uncommon for negative teacher perceptions of English Language Learners (ELLs) to be documented in educational research. This observed phenomenon is especially worrisome for three reasons: (1) teachers’ perceptions of their students are known to affect their pedagogy, (2) teacher perception and pedagogy influences student outcomes, and (3) the ELL population continues to be the quickest growing subset within the American public-school student body. While extensive, previous research has not attempted to compare teacher perceptions of ELLs across varying racial identity groups of ELLs. Thus, this pilot study …

Characterization Of The Full-Length Bag3 Protein And Stress Induced Formation Of Bag3-Z, Ahmed Gamal Abdalla Zied Jan 2023

Characterization Of The Full-Length Bag3 Protein And Stress Induced Formation Of Bag3-Z, Ahmed Gamal Abdalla Zied

Master's Theses

Bcl2-associated athanogene-3 (BAG3) is a pro-autophagy co-chaperone that we havepreviously shown localizes to the cardiac sarcomere and is critical for proteostasis and maintenance of normal sarcomeric function. Indeed, BAG3 loss in heart failure (HF) results in accumulation of ubiquitinated sarcomeric proteins, and depressed maximum force generating capacity (Fmax). However, how BAG3 is regulated in the cell is not well understood, with uncertainty about its structure and proteoforms. During our analysis of human heart tissue, BAG3 appears as a “doublet”, with one band at 74 kDa (BAG3-Z) and a second at a higher 85 kDa (BAG3-FL). Previous studies hypothesized the full-length …

Incorporating Sex Chromosomes In Transcriptome Prediction Models And Improving Cross-Population Prediction Performance, Daniel S. Araujo Jan 2023

Incorporating Sex Chromosomes In Transcriptome Prediction Models And Improving Cross-Population Prediction Performance, Daniel S. Araujo

Master's Theses

Transcriptome prediction models built with data from European-descent individuals are less accurate when applied to different populations because of differences in linkage disequilibrium patterns and allele frequencies. We hypothesized multivariate adaptive shrinkage may improve cross-population transcriptome prediction, as it leverages effect size estimates across different conditions - in this case, different populations. To test this hypothesis, we made transcriptome prediction models for use in transcriptome-wide association studies (TWAS) using different methods (Elastic Net, Matrix eQTL and Multivariate Adaptive Shrinkage in R (MASHR)) and tested their out-of-sample transcriptome prediction accuracy in population-matched and cross-population scenarios. Additionally, to evaluate model applicability in …

Ecclesiological Sovereignty: Toward An Understanding Of The Sovereignty Of The Pope In The Context Of Luther's Responses, Erik Lee Grayvold Jan 2023

Ecclesiological Sovereignty: Toward An Understanding Of The Sovereignty Of The Pope In The Context Of Luther's Responses, Erik Lee Grayvold

Master's Theses

This thesis seeks to address the contention between the perceived sovereign power of the Papacy and the Protestant Reformers. It will first provide a historical survey of how the Pope came to understand himself through the history of the Church. It will then analyze responses from the Reformers regarding this topic, focusing mainly on confessional documents and responses to these documents from the Roman Catholic Church. It will conclude with a survey of Luther’s specific responses to the situation and how his response to Papal authority and sovereignty impacted this theological development. This survey will also acknowledge situations in which …

Comparison Of Taxonomy Assignment And Strain Detection In Urobiome Communities Using 16s Rrna Sequencing And Shotgun Metagenomic Sequencing, Delaney Sauer Jan 2023

Comparison Of Taxonomy Assignment And Strain Detection In Urobiome Communities Using 16s Rrna Sequencing And Shotgun Metagenomic Sequencing, Delaney Sauer

Master's Theses

Advancements in sequencing technologies have enabled scientists to gain insight into the microbes that inhabit the human body, including the urinary tract. Cataloging the bacteria that inhabit the urinary tract has primarily relied on amplification and sequencing of specific variable regions of the 16S rRNA gene enabling genus-level taxonomic identification. Recently, shotgun metagenomic sequencing has been employed such that bacterial taxonomy as well as the functionality that they encode can be inferred. In this study, we compare taxonomies assigned by 16S sequencing and shotgun metagenomic sequencing of the urinary microbiota (urobiome) of females with and without a clinical diagnosis of …

International Conflicts And Conflict Resolution: Periscoping The Role Of The United Nations, Chinyere Ecoma, Ejitu Ota Jan 2023

International Conflicts And Conflict Resolution: Periscoping The Role Of The United Nations, Chinyere Ecoma, Ejitu Ota

Khazar Journal of Humanities and Social Sciences

It is the contention of realist scholars and thinkers that states in the international system have a natural inclination or proclivity to war. This position is anchored on their belief that the world is inherently anarchic and that, therefore, it is in the interest of states in the international system to be prepared at all times to defend themselves. Relying on the historical-descriptive methodology of content analysis, this paper argues that though conflicts are inevitable in international relations, the international community has put in place certain mechanisms to avoid the occurrence of violent and destructive inter-state conflicts in order to …

Mathematics and System Engineering Faculty Publications

The increased complexity of modern systems is calling for an integrated and comprehensive approach to system design and development and, in particular, a shift toward Model-Based Systems Engineering (MBSE) approaches for system design. The requirements that serve as the foundation for these intricate systems are still primarily expressed in Natural Language (NL), which can contain ambiguities and inconsistencies and suffer from a lack of structure that hinders their direct translation into models. The colossal developments in the field of Natural Language Processing (NLP), in general, and Large Language Models (LLMs), in particular, can serve as an enabler for the conversion …

Butterfield, Gerald R. Johnston, Joseph C. Mitchell, Grover J. Brown, J. Brian Hauge, Andrew D. Walde, Eric C. Munscher

Research, Publications & Creative Work

Damage to a turtle’s shell can provide evidence of past events such as vehicle collisions, disease, predator encounters, or even a behavioural interaction between members of the same species. Documenting shell damage as part of long-term mark and recapture studies enables researchers to determine population trends, intraspecific interactions and identify potential issues within turtle populations. This paper analyses shell damage in populations of the Loggerhead Musk Turtle (Sternotherus minor) (Agassiz, 1857). We examined carapace shell damage frequency and severity in 2701 individual S. minor (1468 males and 1233 females) captured in spring-fed habitats in one state preserve and five state …

Functional Comparison Of Full-Length Palladin To Isolated Actin Binding Domain, Sharifah Albraiki, Oluwatosin Ajiboye, Rachel Sargent, Moriah R. Beck Jan 2023

Functional Comparison Of Full-Length Palladin To Isolated Actin Binding Domain, Sharifah Albraiki, Oluwatosin Ajiboye, Rachel Sargent, Moriah R. Beck

Research, Publications & Creative Work

Palladin is an actin binding protein that is specifically upregulated in metastatic cancer cells but also colocalizes with actin stress fibers in normal cells and is critical for embryonic development as well as wound healing. Of nine isoforms present in humans, only the 90 kDa isoform of palladin, comprising three immunoglobulin (Ig) domains and one proline-rich region, is ubiquitously expressed. Previous work has established that the Ig3 domain of palladin is the minimal binding site for F-actin. In this work, we compare functions of the 90 kDa isoform of palladin to the isolated actin binding domain. To understand the mechanism …

The Borders Of Theory: Towards An Artful Ontology Of Knowing In Qualitative Research, Cristina Valencia Mazzanti Jan 2023

The Borders Of Theory: Towards An Artful Ontology Of Knowing In Qualitative Research, Cristina Valencia Mazzanti

Education and Child Study: Faculty Publications

This article considers the ontology of theory as a shaping phenomenon in qualitative research by opening up its intersection with artistic dimensions of knowing and knowledge. We argue that the plurality of practices and perspectives for research hinges on what theory is understood to be. Thus, rather than approaching theory as something predefined, we draw on conceptualizations of the experience of art to articulate the being of theory as an aesthetic renewal of a shared human movement of becoming. We then illuminate theory’s potential as a multidimensional artful being by engaging with a subset of photographs of street art collected …

Dna Barcoding Indicates Multiple Invasions Of The Freshwater Snail Melanoides Tuberculata Sensu Lato In Florida, Lori Tolley-Jordan, Michael A. Chadwick, Jimmy K. Triplett

Research, Publications & Creative Work

Melanoides tuberculata sensu lato (Thiaridae) are polymorphic female-clonal snails of Asian and African origins that have invaded freshwaters worldwide, including those in Florida. Although the snails have been documented in Florida for at least 70 years, no studies have investigated whether the observed distribution is due to a single introduction or multiple independent invasions. Here, cytochrome oxidase I was used to measure genetic diversity within and among sites in Florida and compare genetic diversity between Florida and other regions of the world. We also examined the relationship between shell morphology and haplotype diversity to determine if shell morphs can serve …

Investigating The Viral Nature Of Neuronal Capsid Genes, Michael Manuel Long Jan 2023

Investigating The Viral Nature Of Neuronal Capsid Genes, Michael Manuel Long

Master's Theses

Memory formation is an essential part of everyday life. Neuronal communication is thought to be a key driver behind memory formation. There are two heavily studied methods of neuronal communication: chemical (neurotransmitter-mediated) and electrical (physically attached). When issues arise environmentally or genetically, people can experience one of many devastating memory disorders i.e. Alzheimer’s disease and other Dementias. Due to their prevalence, the treatments for disorders that hinder memory formation are a high priority for pharmaceutical companies. Currently, many of the treatments available focus on neurotransmitters or neuronal activity, and scientists are far from a cure. However, there may be an …

Characterizing The Dynamic Localization Of Cmi In Early Drosophila Development, Asra Habibullah Jan 2023

Characterizing The Dynamic Localization Of Cmi In Early Drosophila Development, Asra Habibullah

Master's Theses

The COMPASS-like family of lysine methyltransferases, MLR/MLX complexes, are epigenetic regulators that are essential for normal development through the methylation of the fourth lysine residue on histone 3 (H3K4), a universal epigenetic mark associated with active transcription. This family of complexes is highly conserved from yeast to mammals and the genes encoding the human MLR complexes have been associated with various developmental diseases and cancers (Dingwall and Fagan, 2019). In D. melanogaster, the enzymatic methyltransferase core of this complex is composed of two proteins: Cara Mitad (Cmi, also known as Lpt) and Trithorax-related (Trr). Although these proteins have been shown …
