One of the central roles in the design, development, and application advances in mediator-Based microbial electrochemical systems, such as microbial fuel cells (MFCs), is the establishment of efficient and successful communication between conductive electrode surfaces and microorganisms via modes of extracellular electron transfer (EET). Most microbial-Based systems require the use of artificial electroactive mediators in order to facilitate and/or enhance electron transfer. Our previous work established an exogenous phenazine-Based library as a mediator system to enable EET from the model microorganism Escherichia coli as a promising biotechnological host. Eukaryotic cells are building blocks to complex living systems, characterized by membrane-bound organelles. Studying how eukaryotic organelles react to cellular growth and size increase is crucial, but it demands biochemical and biophysical manipulations, as well as quantitative observation tools in microscopy. We developed a multi-color yeast strain with tagged fluorescent proteins, enabling systematic measurements of 6 organelles inside each cell using spectral confocal microscopy. These measurements provided insights into how organelle biogenesis is coordinated with cellular size and growth rate regulation via different signaling pathways. The adaptive immune system has inherent antitumor properties that are capable of inducing tumor-specific cell death. CD8+ and CD4+ T cells, two immune cell types critical to this process, recognize antigens bound by class I and II major histocompatibility complexes on the cell surface, respectively. After antigen recognition, T cells can signal growth arrest and cell death to tumor cells displaying the antigen. Neoantigens, or tumor-specific peptides, are targeted by cancer immunotherapies to induce T cell-mediated degradation. Treatments such as personalized vaccines and immune checkpoint blockade thus provide a unique avenue to selectively recognize and destroy tumor cells while keeping …

MYSM1 deficiency has been identified in patients with B cell lymphopenia, hypogammaglobulinemia, primary immune deficiency and bone marrow failure. MYSM1 is a known deubiquitinase that acts on histone H2A to promote gene expression. MYSM1 also functions in cellular responses to DNA injury and patients with loss of MYSM1 have increased sensitivity to DNA damaging agents. The specific role and function of MYSM1 in DNA damage responses has yet to be determined. Here, I have shown that MYSM1 does not mediate generation or repair of DSBs, but rather acts to attenuate and terminate DNA damage signaling after repair. Loss of MYSM1 …

The stomach contains gastric epithelial cells, including acid-pumping parietal cells (PCs), that are constantly renewed throughout life from a multipotent stem cell to maintain a balance between producing and protecting from the acid and enzymes needed to kill pathogens and initiate digestion. Despite this dynamic renewal, disorders of the gastric epithelium occur and are one of the most common etiologies of human diseases ranging from simple heartburn to reflux, peptic ulcers and gastric cancer. Transfer learning is a machine learning technique founded on the idea that knowledge acquired by a model during “pretraining” on a source task can be transferred to the learning of a target task. Successful transfer learning can result in improved performance, faster convergence, and reduced demand for data. This technique is particularly desirable for the task of brain decoding in the domain of functional magnetic resonance imaging (fMRI), wherein even the most modern machine learning methods can struggle to decode labelled features of brain images. Previous studies showed that West Antarctic Ice Sheet (WAIS) retreat from the Whales Deep Basin continental shelf edge in eastern Ross Sea was underway by 14.7 ± 0.4 cal kyr BP. Following an ice-shelf breakup at 12.3 ± 0.6 cal kyr BP, a calving cliff margin remained within 60 km of the shelf edge until 11.5 ± 0.3 cal kyr BP. McGlannan et al. (2017) inferred that an ice shelf reformed immediately during a subsequent rapid retreat. Soil salinity is an increasing problem facing agriculture in many parts of the world. Climate change and irrigation practices have led to decreased yields of large areas of farmland due to increased salt levels in the soil. Irrigation introduces salts to the soil that with time accumulate and threaten crop yield. In arid climates like Utah, the practice of irrigation is especially threatening to salt-sensitive crops including alfalfa (Medicago sativa). There are extensive amounts of psychological research that highlight the needs and experiences of racial ethnic minority populations in general and more specifically minority sexual identities and genders, also referred to as non-heteronormative populations (Alexeyeff & Besnier, 2014). Cattle grazing has been a historic use of rangelands in Utah since Pioneer settlement in the mid 1800's. Wright fishhook cactus is a small globose cactus endemic to an area of 280,000 ha in south-central Utah and was listed as endangered in October of 1979, by the U.S. Fish and Wildlife Service (USFWS). By 2010, concerns were expressed that soil compaction in proximity to the cactus posed a threat to this species, though there was no empirical data to support such concern. Cutthroat trout (Oncorhynchus clarkii) is a salmonid native to Western North America. For almost 200 years, the taxonomy of these species has fluctuated as ichthyologists and fisheries managers alike have tried to understand the diversity seen within these fishes. In the 1950s, Robert Behnke reexamined the cutthroat trout and identified 14 subspecies based on morphological observations, Pleistocene events, and modern geographic ranges. These designations were instrumental in preserving the remaining diversity of cutthroat trout. Over time, molecular techniques (i.e. karyotypes, allozymes, and mtDNA) have largely replicated Behnke's phylogenies. X-ray diffraction analysis (XRD) is an inexpensive method to quantify the relative proportions of mineral phases in a rock or soil sample. However, the analytical software available for XRD requires extensive user input to choose phases to include in the analysis. Engineers contribute to large-scale socio-technical challenges, and human-centered design offers a design thinking approach that helps engineers develop a thorough understanding of the socio-technical effects of their design work. Thus, effective strategies for assessing and teaching human-centered design are needed. Tumor mutational burden and heterogeneity has been suggested to fuel resistance to many targeted therapies. The cytosine deaminase APOBEC proteins have been implicated in the mutational signatures of more than 70% of human cancers. However, the mechanism underlying how cancer cells hijack the APOBEC mediated mutagenesis machinery to promote tumor heterogeneity, and thereby foster therapy resistance remains unclear. We identify SYNCRIP as an endogenous molecular brake which suppresses APOBEC-driven mutagenesis in prostate cancer (PCa). Overactivated APOBEC3B, in SYNCRIP-deficient PCa cells, is a key mutator, representing the molecular source of driver mutations in some frequently mutated genes in PCa, including FOXA1, …

Inflammatory bowel disease (IBD) is associated with gut dysbiosis and can lead to colitis-associated malignancies. Bacteroides uniformis (Bu) regulates animal intestinal homeostasis; however, the mechanism by which it alleviates colitis in mice remains unknown. We investigated the effects of B. uniformis JCM5828 and its metabolites on female C57BL/6J mice with dextran sulfate sodium salt (DSS) induced colitis. Treatment with Bu considerably alleviated colitis progression and restored the mechanical and immune barrier protein expression. Additionally, Bu increased the abundance of the symbiotic bacteria Bifidobacterium and Lactobacillus vaginalis while decreasing that of pathogenic Escherichia-Shigella, and modulated intestinal bile acid metabolism. The immune system plays an essential role in cancer evolution and cancer treatment. One of the major mechanisms of immune-mediated tumor rejection is the recognition of tumor antigens by T cells. Upon recognition of tumor antigens, naïve T cells become activated and subsequently infiltrate inside the tumor. Since the early discovery of tumor-specific T cells, many efforts have elucidated the molecular nature of their cognate antigens. Gender is a socially derived construct that is taught and understood at young ages. However, not every individual has the same social skills to understand these seen and unseen social cues about gender. The Bem Sex-Role Inventory was given to adult individuals with autism in the United States to better understand their masculine, feminine, and androgenous perceptions. These data were then compared to the original Bem Sex-Role Inventory general population study to see if there were any differences. The data show that males with autism had lower masculine scores and higher feminine scores than males in the general population. Females …

Employing a comprehensive review of literature across various databases, the study sheds light on the mechanisms through which environmental neurotoxicity influences health, highlighting key correlations between toxin exposure and the onset of neurodegenerative disorders. The research identifies significant gaps in current approaches to managing these risks, particularly concerning policy regulations and public awareness. Based on these findings, strategic recommendations are proposed to address and mitigate the identified challenges, with implications emphasizing the broader benefits for public health.

System stability deterioration in microgrids commonly occurs due to unpredictable faults and equipment malfunctions. Recently, robust control techniques have been used in microgrid systems to address these difficulties. In this paper, for DC-islanded microgrids that have sensors faults, a new passive fault-tolerant control strategy is developed. The suggested approach can be used to maintain system stability in the presence of flaws, such as faulty actuators and sensors, as well as component failures. The opportunistic fungal pathogen Cryptococcus neoformans causes lethal infections in immunocompromised patients. Macrophages are central to the host response to cryptococci; however, it is unclear how C. neoformans is recognised and phagocytosed by macrophages. Here we investigate the role of TLR4 in the non-opsonic phagocytosis of C. neoformans. We find that loss of TLR4 function unexpectedly increases phagocytosis of non-opsonised cryptococci by murine and human macrophages. The increased phagocytosis observed in Tlr4−/− cells was dampened by pre-treatment of macrophages with oxidised-LDL, a known ligand of scavenger receptors. The National Cancer Institute's Clinical Proteomic Tumor Analysis Consortium (CPTAC) investigates tumors from a proteogenomic perspective, creating rich multi-omics datasets connecting genomic aberrations to cancer phenotypes. To facilitate pan-cancer investigations, we have generated harmonized genomic, transcriptomic, proteomic, and clinical data for >1000 tumors in 10 cohorts to create a cohesive and powerful dataset for scientific discovery. We outline efforts by the CPTAC pan-cancer working group in data harmonization, data dissemination, and computational resources for aiding biological discoveries. Glioblastoma Multiforme (GBM) represents one of the most aggressive and metastatic brain tumors, with a dismal success rate of less than three percent after five years, particularly in tumors with active immune checkpoints. This necessitates the development of targeted endogenous agents for precise GBM treatment.
