Breast cancer became the most diagnosed cancer in the world in 2020. Chemotherapy is still the leading clinical strategy in breast cancer treatment, followed by hormone therapy (mostly used in hormone receptor-positive types). However, with our ever-expanding knowledge of signaling pathways in cancer biology, new molecular targets are identified for potential novel molecularly targeted drugs in breast cancer treatment. While this has resulted in the approval of a few molecularly targeted drugs by the FDA (including drugs targeting immune checkpoints), a wide array of signaling pathways seem to be still underexplored. Summer 2023, the Centers for Disease Control and Prevention reported several cases of locally acquired malaria in Florida, Texas, and Maryland, meaning that patients were infected within the US—the first such cases in over two decades (CDC 2023). The significance of the cases and how individuals and public health officials should respond to the situation is still unknown. The CDC is collaborating with state health departments on an investigation of seven locally acquired cases of Plasmodium vivax malaria in Sarasota County, Florida, and one case in Cameron County, Texas, and Maryland near Washington DC. Inborn errors of immunity (IEIs) are disorders caused by monogenic variants that lead to immune deficiency and dysregulation. Although rare, IEIs have shaped our understanding of how single genes function in the immune systems of both humans and mice. The identification of loss-of-function (LOF) and gain-of-function (GOF) variants in the highly conserved signal transducer and activator of transcription (STAT) family molecules have led to discoveries involving how STAT signaling affects immune cell development, differentiation, and the induction and regulation of immune responses. Germline GOF variants in STAT3 cause early-onset multi-organ autoimmunity and immune dysregulation. Patients with STAT3 GOF syndrome have …

Antimelanoma Activities Of Fused Thiazole Derivatives, Sanjay Adhikary

Melanoma, a fatal type of skin cancer, is still incurable. To develop a possible cure, we synthesized fused thiazole derivatives, and found them to be potent anti-melanoma agents both in vitro and in vivo. Thiazolo ethisterone derivatives were remarkably cytotoxic to melanoma cell lines and inhibited colonization of oncogenic cells. Compounds, in vitro, inhibited three key mechanisms of metastasis; cell migration, adhesion, and invasiveness. The compounds caused mild G2/M phase mitotic cell cycle arrest and induced moderate apoptosis. 3, 4-Dihydroxy substituted thiazolo ethisterone (ET32) upregulated beta-Parvin (PARVB) expression (both mRNA and protein expressions) with a log2 fold change value around …

Freshwater ecosystems are among the most modified and threatened environments, with increased anthropogenic activities causing habitat degradation, alteration to environmental function and structure, overexploitation of resources, and widespread distribution of non-native species leading to loss of biodiversity. A common conservation management strategy is to create protective areas that encompass the distribution of at-risk species. The objective of protected areas is to address issues that have been cited as the three primary threats to freshwater environments: land-use disturbances, altered hydrology, and introduction of non-native species. Temporal and spatial variations in crop performance are common in agricultural production fields in northeastern Arkansas. With the alluvial soils of the Mississippi Delta region, within-field soil texture can range from coarse sand to heavy clay. Such variability influences chemical and physical soil properties that affect yield potential of the region’s commercial cropping systems. With advances in precision management, delineating large commercial fields into zones of homogenous production potential could increase profitability and improve efficiency. This thesis examines husbands’ experiences and the role they play in breastfeeding families. The vast majority of research on this topic focuses solely on the mother’s viewpoint and experience. Through in-depth interviews with eight white males residing in the Midsouth, the lived experiences of husbands within the family context and the ways in which they are active participants in parenting decisions and practices are explored. Previous research indicates that breastfeeding initiation and duration can be contingent upon the husband’s approval. This study supported previous research by identifying three themes that highlighted the participants’ breastfeeding experience. The themes identified were the …

Triple-negative breast cancer (TNBC), the most fatal breast cancer subtype, lacks three main receptors (the human epidermal growth factor 2 receptor (ERBB2, formerly HER2), estrogen (ER) and progesterone (PR) receptors) and represents about 15% of all breast tumors. The most common treatment for TNBC is chemotherapy, being paclitaxel one of the most prescribed drugs. One of the main issues of paclitaxel is its high level of toxicity to non-cancerous cells and the development of paclitaxel resistance in the TNBC cells. Adjuvant therapy has been proposed as an alternative approach to increase the efficacy of a drug or circumvent drug resistance. …

Somapura Mahavihara, an 8th-century Buddhist monastery in Bangladesh, is a world heritage site. Measuring 22 hectares, the layout of the monastery is quadrangular shaped, reaching a height of approximately eighty feet above ground level. Physically this monastery provides an excellent example of the workmanship and design principles of the Pala Dynasty of 8th-century Bengal. The monastery’s architectural elements have been the center of academic studies on architecture, building morphology, ornamentation, and archeological excavation of this monastery. The opioid crisis has plagued the United States but disproportionately affects the often-overlooked Appalachian region. This area faces unique barriers preventing better access to quality Opioid Use Disorder (OUD) treatment facilities despite opioid-related deaths continuing to rise. An especially vulnerable population in this region are pregnant and postpartum women who face even more challenges acquiring proper drug treatment. Medically Assisted Treatment (MAT), also called Medications for Opioid Use Disorder (MOUD), is considered the standard of treatment for OUD and reduces the effects of NAS, yet it is heavily stigmatized and underutilized in populations who could benefit from the medication. This report presents over three years of systematically engaging, documenting and analyzing the diversity, equity and inclusion (DEI) needs/gaps and assets of state funded community mediation centers in Massachusetts. The report was compiled by researchers and an in-house DEI expert at the statutory state office of dispute resolution, the Massachusetts Office of Public Collaboration (MOPC) at the University of Massachusetts Boston. The office has been serving as a neutral forum and state-level resource for over 30 years.

An Improved Source Of Spin-Polarized Electrons Based On Spin Exchange In Optically Pumped Rubidium Vapor, K. J. Ahrendsen, Kenneth Wayne Trantham, D. Tupa, T. J. Gay

We have improved a polarized electron source in which unpolarized electrons undergo collisions with a mixture of buffer gas molecules and optically spin-polarized Rb atoms. With a nitrogen buffer gas, the source reliably provides spin polarization between 15% and 25% with beam currents >4 μA. Vacuum pump upgrades mitigate problems caused by denatured diffusion pump oil, leading to longer run times. A new differential pumping scheme allows the use of higher buffer gas pressures up to 800 mTorr. With a new optics layout, the Rb polarization is continuously monitored by a probe laser and improved pump laser power provides more …

In 1665 Robert Hooke, looking at cork through his microscope, discovered that plants are composed of elementary structures he named “cells”. Variation in the expression of a single genome in a complex eukaryotic organism guides the initiation, maturation, physiology, and biochemistry of cells with different shapes and sizes, playing different structural and functional roles in space and time. How many kinds of cells—”cell types”— an organism possesses of course depends on the organism’s cellular complexity, but the plasticity within a cell type fuels the emergence of the concept of cell state (Figure 1). Introduction
The PReferentially expressed Antigen in MElanoma (PRAME) protein has been shown to be an independent biomarker for increased risk of metastasis in Class 1 uveal melanomas (UM). Intrinsically disordered proteins and regions of proteins (IDPs/IDPRs) are proteins that do not have a well-defined three-dimensional structure and have been linked to neoplastic development. Our study aimed to evaluate the presence of intrinsic disorder in PRAME and the role these structureless regions have in PRAME( +) Class 1 UM.

Us-Russia Foreign Policy: Confronting Russia’S Geographic Anxieties, Caitlin P. Irby
The US Army War College Quarterly: Parameters
The United States must place Russia’s focus on geographic concerns at the center of future strategy development to build a constructive relationship with Russia and achieve US regional goals. This article analyzes Russia’s geography and historical impact on Russian foreign policy, outlines Moscow’s current foreign policy goals, and highlights underlying concerns for US policymakers and military practitioners. By pursuing policies that support Russian goals of economic integration, mitigation of demographic concerns, and security of national borders, the United States can set the foundation for productive engagement on critical issues.

Large colonies of two vulnerable, sympatric pinniped species, Peruvian fur seals (Arctocephalus australis Peruvian subpopulation; PFS) and South American sea lions (Otaria byronia; SASL), reside and reproduce in Punta San Juan, Peru (PSJ) where they are exposed to stressors such as contaminants and El Niño Southern Oscillation (ENSO) events. The use of cytokines as immune biomarkers can be indicative of an animal’s health and function by allowing for the assessment of an individual's cytokine balance as type I helper T cells (Th1) or type II helper T cells (Th2) dominant. Antibiotic-resistance gene elements (ARGEs) such as antibiotic-resistance genes (ARGs), integrons, and plasmids are key to the spread of antimicrobial resistance (AMR) in marine environments. Kuwait’s marine area is vulnerable to sewage contaminants introduced by numerous storm outlets and indiscriminate waste disposal near recreational beaches. Therefore, it has become a significant public health issue and warrants immediate investigation. Coliforms, especially Gram-negative Escherichia coli, have been regarded as significant indicators of recent fecal pollution and carriers of ARGEs. Deep eutectic solvents (DESs) have recently attracted much attention as potential green electrolyte solvents for redox flow batteries. DESs are considered not only as environmentally sustainable but also economically attractive electrolytes because they can be resourced from biological feedstock (alcohols, urea, choline) and are earth-abundant and of low toxicity. Despite these advantages, DESs still have limitations in important aspects such as reactant and ion transport, which is inhibited due to hydrogen-bonding-induced viscosity.
