Celebrating Leeuwenhoek’S Life 300 Years Later, Alan L. Gillen
Celebrating Leeuwenhoek’S Life 300 Years Later, Alan L. Gillen
Faculty Publications and Presentations
Antony van Leeuwenhoek entered eternity 300 years ago (August 26, 1723). He made hundreds of discoveries with his simple microscope. Perhaps his most notable discovery was of bacteria and its many shapes. He found great joy in God [http://answersingenesis.org/god/] ’s smallest creatures. I believe that his curiosity about the world, fueled by his belief in biblical creation [http://answersingenesis.org/creation/] , led to his discovery of bacteria. In a similar manner, his curiosity of blood led to his discovery of red corpuscles and capillaries. He remained a curious man until the last days of his life 300 years …

Glioblastoma (GBM) is a highly aggressive brain tumor, and its treatment is further complicated by the high selectivity of the blood–brain barrier (BBB). The scientific community is urgently seeking innovative and effective therapeutic solutions. Liposomes are a promising new tool that has shown potential in addressing the limitations of chemotherapy, such as poor bioavailability and toxicity to healthy cells. However, passive targeting strategies based solely on the physicochemical properties of liposomes have proven ineffective due to a lack of tissue specificity. Accordingly, the upregulation of transferrin receptors (TfRs) in brain tissue has led to the development of TfR-targeted anticancer therapeutics. …
The Influences Of Acculturative Stress And Gender Roles On Sexual Subjectivity In European, Asian, And Latinx Immigrant Women In The U.S., Silvia Re
Seton Hall University Dissertations and Theses (ETDs)
In the process of acculturation, cisgender immigrant women are at greater risk of experiencing acculturative stress, often entailing a reconsideration of their self-concepts and identities as members of new sociocultural contexts. Gender roles and sexual subjectivity are two identity features they can revise given their ties to culture and socialization. Results from previous studies suggest that cisgender immigrant women’s sociocultural contexts, related values, and attitudes may contribute to their levels of stress, sense of self-efficacy, self-esteem, and sexual subjectivity. This study aimed to fill gaps in the existing literature and raised awareness of the relationship between acculturative stress, gender role …

Daxx functions as a histone chaperone for the histone H3 variant, H3.3, and is essential for embryonic development. Daxx interacts with Atrx to form a protein complex that deposits H3.3 into heterochromatic regions of the genome, including centromeres, telomeres, and repeat loci. To advance our understanding of histone chaperone activity in vivo, we developed two Daxx mutant alleles in the mouse germline, which abolish the interactions between Daxx and Atrx (DaxxY130A), and Daxx and H3.3 (DaxxS226A). We found that the interaction between Daxx and Atrx is dispensable for viability; mice are born at the …

Unlike for advanced liver fibrosis, the practical rules for the early non-invasive diagnosis of cirrhosis in NAFLD remain not well defined. Here, we report the derivation and validation of a stepwise diagnostic algorithm in 1568 patients with NAFLD and liver biopsy coming from four independent cohorts. The study algorithm, using first the elastography-based tests Agile3+ and Agile4 and then the specialized blood tests FibroMeterV3G and CirrhoMeterV3G, provides stratification in four groups, the last of which is enriched in cirrhosis (71% prevalence in the validation set). A risk prediction chart is also derived to allow estimation of the individual probability of …

Orofacial clefts, including cleft lip and palate (CL/P) and neural tube defects (NTDs) are among the most common congenital anomalies, but knowledge of the genetic basis of these conditions remains incomplete. The extent to which genetic risk factors are shared between CL/P, NTDs and related anomalies is also unclear. While identification of causative genes has largely focused on coding and loss of function mutations, it is hypothesized that regulatory mutations account for a portion of the unidentified heritability. We found that excess expression of Grainyhead-like 2 (Grhl2) causes not only spinal NTDs in Axial defects (Axd) mice but also multiple …

Decision Making In Patients With Initial Positive Deep Margins On Diagnostic Biopsy Of Melanoma, Ana Eliza Souza Cunha
Decision Making In Patients With Initial Positive Deep Margins On Diagnostic Biopsy Of Melanoma, Ana Eliza Souza Cunha
Yale School of Medicine Physician Associate Program Theses
Background: Current guidelines recommend SLN biopsy (SLNB) among patients with initial Breslow Thickness (BRES) ≥0.8mm at initial screening. However, there is limited guidance regarding SLNB among patients who have deeper final depth after initial biopsy.
Methods: We conducted a retrospective cohort study of 62 thin melanoma patients with primary lesions with initial Breslow Thickness (BRES)
Results: Approximately 55% of patients (n = 34) included in our study underwent SLNB. After adjustment, there was no association between SLNB status and recurrence. Immunosuppressive drug use was associated with higher odds of local (OR= 28.56 CI 1.87- 435.53; p =0.016) and regional recurrence …

Yale School of Medicine Physician Associate Program Theses
Abstract/Summary:
Background: “The Society of Thoracic Surgeons mortality risk calculator”, which includes a race variable, is recommended for estimating Short- term (30 day) predicted risk of mortality after isolated coronary artery bypass graft, with a risk estimate of 3-4% stratifying patients into intermediate risk, a risk estimate above 5% stratifying patients into high risk for surgery and a risk estimate above 8% stratifying patients into the very high-risk category. We compared differences between Black and White individuals in Society of Thoracic Surgeons-predicted risk of mortality across various plausible risk factor combinations with the aim of evaluating if using the predictive …

Anterior chamber depth (ACD) is a quantitative trait associated with primary angle closure glaucoma (PACG). Although ACD is highly heritable, known genetic variations explain a small fraction of the phenotypic variability. The purpose of this study was to identify additional ACD-influencing loci using strains of mice. Cohorts of 86 N2 and 111 F2 mice were generated from crosses between recombinant inbred BXD24/TyJ and wild-derived CAST/EiJ mice. Using anterior chamber optical coherence tomography, mice were phenotyped at 10–12 weeks of age, genotyped based on 93 genome-wide SNPs, and subjected to quantitative trait locus (QTL) analysis. In an analysis of ACD among …

Short-Term Vehicle Speed Prediction With Spatiotemporal Convolution Fused With Variational Modal Decomposition, Kai Zhang, Haipeng Lu, Ying Han, Lingyun Zhang, Yujie Ding
Journal of System Simulation
Abstract: Accurate short-term vehicle speed prediction helps to resolve city traffic congestion problems. Focusing on the defect that CNN cannot process non-Euclidean geometric data, GCN and BiLSTM are combined to fully process the spatiotemporal characteristics of road network information, in which the advantages of GCN integrating global features and the ability of BiLSTM to extract temporal features are considered. In order to reduce the interference of noise to the data, variational modal decomposition (VMD) is introduced and short-term vehicle speed prediction model based on VMD-GCN-BiLSTM (VGBLSTM) is proposed . Simulation results show that the prediction accuracy of VGBLSTM model is …

Histamine from our diet or gut microbes can trigger gastrointestinal disturbances, and resistant potato starch (RPS) has previously been shown to alleviate these symptoms while increasing levels of health-associated bacteria such as Akkermansia through unknown mechanisms. Post hoc exploratory metabolomic analysis of serum amino acid, amine, and carnitine metabolites in participants consuming 3.5 g/day RPS or placebo (n = 48) was performed using liquid chromatography-mass spectrometry to determine whether RPS positively influences histamine metabolism and related parameters. Histamine levels were significantly reduced by RPS treatment, but histamine-degrading enzyme products were unaffected by RPS. RPS also reduced histamine-secreting Haemophilus and Lactobacillus …

The susceptibility of sensory cells to pathological conditions differs between the apical and basal regions of the cochlea, and the cochlear immune system may contribute to this location-dependent variability. Our previous study found morphological differences in basilar membrane macrophages between the apical and basal regions of the cochlea. However, the details of this site-dependent difference and its underlying structural and biological basis are not fully understood. In this study, we utilized scanning electron microscopy to examine the ultrastructure of macrophages and their surrounding supporting structures. Additionally, we examined the phagocytic activities of macrophages and the expression of immune molecules in …
