When the two eyes are presented with highly distinct stimuli, the resulting visual percept generally switches every few seconds between the two monocular images in an irregular fashion, giving rise to a phenomenon known as binocular rivalry. While a host of theoretical studies have explored potential mechanisms for binocular rivalry in the context of evoked model dynamics in response to simple stimuli, here we investigate binocular rivalry directly through complex stimulus reconstructions based on the activity of a two-layer neuronal network model with competing downstream pools driven by disparate monocular stimuli composed of image pixels. To estimate the dynamic percept, …

A principal cue for sound source localization is the difference in arrival times of sounds at an animal’s two ears (interaural time difference, ITD). Neurons that process ITDs are specialized to compare the timing of inputs with submillisecond precision. In the barn owl, ITD processing begins in the nucleus laminaris (NL) region of the auditory brain stem. Remarkably, NL neurons are sensitive to ITDs in high-frequency sounds (kilohertz-range). This contrasts with ITD-based sound localization in analogous regions in mammals where ITD sensitivity is typically restricted to lower-frequency sounds. Guided by previous experiments and modeling studies of tone-evoked responses of NL …

Groundwater Dynamics In An Unconfined Coastal Aquifer: Geophysical Investigations And Modeling, Jeeban Panthi
Groundwater Dynamics In An Unconfined Coastal Aquifer: Geophysical Investigations And Modeling, Jeeban Panthi
Open Access Dissertations
Coastal groundwater plays a vital role in the sustainability of coastal ecosystems and the freshwater supply for over one billion people worldwide. Understanding the movement and interaction of saltwater and freshwater in coastal aquifers is crucial for effective water resource management. The interface between these density-contrast waters is highly dynamic and sensitive to natural drivers, such as sea level rise, storm surges, and drought, as well as anthropogenic factors, including groundwater over-pumping. In recent years, the drivers of coastal groundwater dynamics have been increasingly well understood and quantified. However, there is a need for data-model fusion, including integrating different forms …

The remarkable biodiversity of the Earth fascinated natural scientists for centuries. Studying this biodiversity has been a major occupation of the life sciences because deciphering the underlying biological mechanisms of diversification provides fundamental knowledge for biological evolution. A significant body of evidence shows that today’s biodiversity originated from successive adaptive diversification periods (i.e.: evolutionary radiations) which are generally characterized by the rapid multiplication of lineages with a variety of adaptations and ecological roles. Some of the most well-known examples of evolutionary radiations include the striking wing pattern diversity of Heliconius butterflies, the great body form variety of Caribbean anole lizards, …

Prior to widespread COVID-19 vaccination, nursing homes were required to perform comprehensive weekly testing of staff. After the vaccine became available, testing became symptom-based for vaccinated staff despite continued waves of different severe acute respiratory coronavirus virus 2 (SARS-CoV-2) variants. Nursing homes need pragmatic, affordable strategies to identify SARS-CoV-2–positive staff because financial and job-related pressures drive reluctance to disclose mild symptoms. We sought to determine whether positive environmental samples for SARS-CoV-2 could serve as trigger for comprehensive staff testing in lieu of weekly testing of all staff.

Maternal pre-pregnancy (pregravid) obesity is associated with adverse outcomes for both mother and offspring. Amongst the complications for the offspring is increased susceptibility and severity of neonatal infections necessitating admission to the intensive care unit, notably bacterial sepsis and enterocolitis. Previous studies have reported aberrant responses to LPS and polyclonal stimulation by umbilical cord blood monocytes that were mediated by alterations in the epigenome. In this study, we show that pregravid obesity dysregulates umbilical cord blood monocyte responses to bacterial and viral pathogens. Specifically, interferon-stimulated gene expression and inflammatory responses to respiratory syncytial virus (RSV) and E. coli were significantly …

Objective: A 6-month longitudinal surveillance study of asymptomatic healthcare providers (HCP) was carried out at a large urban academic medical center in the United States to assess whether their job occupation with higher exposure risks to SARS-CoV-2 would equate with higher risk of contracting COVID-19 at the beginning of the pandemic before COVID-19 vaccines were available.
Methods: A longitudinal cohort study design was used to collect and analyze immunological and virological monitoring data and self-report survey assessments of personal protective equipment (PPE) availability, adherence to infection control guidelines, and time spent on COVID-19 wards.
Results: Among 289 eligible participants, SARS-CoV-2 …

Taï Forest virus (TAFV) is a lesser-known ebolavirus that causes lethal infections in chimpanzees and is responsible for a single human case. Limited research has been done on this human pathogen; however, with the recent emergence of filoviruses in West Africa, further investigation and countermeasure development against this virus is warranted. We developed a vesicular stomatitis virus (VSV)-based vaccine expressing the TAFV glycoprotein as the viral antigen and assessed it for protective efficacy in nonhuman primates (NHPs). Following a single high-dose vaccination, NHPs developed antigen-specific binding and neutralizing antibodies as well as modest T cell responses. Importantly, all vaccinated NHPs …

Infection with Marburg virus (MARV), the causative agent of Marburg virus disease (MVD), results in haemorrhagic disease and high case fatality rates (>40%) in humans. Despite its public health relevance, there are no licensed vaccines or therapeutics to prevent or treat MVD. A vesicular stomatitis virus (VSV)-based vaccine expressing the MARV glycoprotein (VSV-MARV) is currently in clinical development. Previously, a single 10 million PFU dose of VSV-MARV administered 1–5 weeks before lethal MARV challenge conferred uniform protection in nonhuman primates (NHPs), demonstrating fast-acting potential. Additionally, our group recently demonstrated that even a low dose VSV-MARV (1000 PFU) protected NHPs …

Platelet secretion requires Soluble N-ethylmaleimide Sensitive Attachment Protein Receptors (SNAREs). Vesicle SNAREs/Vesicle-Associated Membrane Proteins (v-SNAREs/ VAMPs) on granules and t-SNAREs in plasma membranes mediate granule release. Platelet VAMP heterogeneity has complicated the assessment of how/if each is used and affects hemostasis. To address the importance of VAMP-7 (V7), we analyzed mice with global deletions of V3 and V7 together or platelet-specific deletions of V2, V3, and global deletion of V7. We measured the kinetics of cargo release, and its effects on three injury models to define the context-specific roles of these VAMPs. Loss of V7 minimally affected dense and α …

Influenza-A virus (IAV) infects yearly an estimated one billion people worldwide, resulting in 300,000–650,000 deaths. Preventive vaccination programs and antiviral medications represent the mainstay of therapy, but with unacceptably high morbidity and mortality rates, new targeted therapeutic approaches are urgently needed. Since inflammatory processes are commonly associated with measurable changes in the cell membrane potential (Em), we investigated whether Em hyperpolarization via TREK-1 (K2P 2.1) K þ channel activation can protect against influenza-A virus (IAV)-induced pneumonia. We infected mice with IAV, which after 5 days caused 10–15% weight loss and a decrease in spontaneous activity, representing a clinically relevant infection. …

Platelets contribute to COVID-19 clinical manifestations, of which microclotting in the pulmonary vasculature has been a prominent symptom. To investigate the potential diagnostic contributions of overall platelet morphology and their α-granules and mitochondria to the understanding of platelet hyperactivation and micro-clotting, we undertook a 3D ultrastructural approach. Because differences might be small, we used the high-contrast, high-resolution technique of focused ion beam scanning EM (FIB-SEM) and employed deep learning computational methods to evaluate nearly 600 individual platelets and 30 000 included organelles within three healthy controls and three severely ill COVID-19 patients. Statistical analysis reveals that the α-granule/ mitochondrion-to-plateletvolume ratio …

Vascular smooth muscle cells (vSMCs) exert a critical role in sensing and maintaining vascular integrity. These cells abundantly express the low-density lipoprotein receptor–related protein 1 (LRP1), a large endocytic signaling receptor that recognizes numerous ligands, including apolipoprotein E–rich lipoproteins, proteases, and protease-inhibitor complexes. We observed the spontaneous formation of aneurysms in the superior mesenteric artery (SMA) of both male and female mice in which LRP1 was genetically deleted in vSMCs (smLRP1–/– mice). Quantitative proteomics revealed elevated abundance of several proteins in smLRP1–/– mice that are known to be induced by angiotensin II–mediated (AngII-mediated) signaling, suggesting that this pathway was dysregulated. …

The abrupt increase in surface air temperature over the last few decades has received abundant scholarly and popular attention. However, less attention has focused on the specific nature of the warming spatially and seasonally, using high-resolution reanalysis output based on historical temperature observations. This research uses the European Centre for Medium-range Weather Forecasts (ECMWF) Reanalysis Version 5 (ERA5) output to identify spatiotemporal features of daily mean surface air temperature, defined both as the mean of the maximum and minimum temperatures over the calendar day (“meanmaxmin”) and as the mean of the 24 hourly observations per day (“meanhourly”), across the terrestrial …

An accurate economic loss assessment for natural hazards is vital for planning, mitigation, and actuarial purposes. The widespread and costly nature of flood hazards, with the economically disadvantaged disproportionately victimized population, makes flood risk assessment particularly important. Here, flood risk is assessed as incurred by the homeowner vs. the flood insurer for insured U.S. properties through the derivation of average annual loss (AAL). AAL is estimated and partitioned using Monte Carlo simulation at the individual home scale, considering insurance coverage and deductible, and the first-floor height (i.e., height of the first floor above the ground), to determine the AAL proportion …

Flood risk to single-family rental housing remains poorly understood, leaving a large and increasing population underinformed to protect themselves, including regarding insurance. This research introduces a life-cycle benefit-cost analysis for the landlord, tenant, and insurer [i.e., (U.S.) National Flood Insurance Program (NFIP)] to optimize freeboard [i.e., additional first-floor height above the base flood elevation (BFE)] selection for a rental single-family home. Flood insurance premium; apportioned flood risk among the landlord, tenant, and NFIP by insurance coverage and deductible; rental loss; moving and displacement costs; freeboard construction cost; and rent increase upon freeboard implementation are considered in estimating net benefit (NB) …

Much of the U.S. petrochemical infrastructure is heavily concentrated along the western coast of the Gulf of Mexico within the impact zone of major tropical cyclone events. Flood impacts of recent tropical disturbances have been exacerbated by an overall lack of recognition of the vulnerabilities to process systems from water intrusion, as well as insufficient disaster mitigation planning. Vulnerability assessment methods currently call for the aggregation of qualitative data to survey the susceptibility of industrial systems to floodwater damage. A means to quantify these consequences is less often employed, resulting in a poor translation of the threat of flood hazards …
