Robust literature exists to support the utility of CrossFit in improving metabolic and cardiovascular well-being. However, a metaphorical chasm exists not in the study of the physiological effects of CrossFit but in the psychological effects, namely quality of life (QOL). This thesis aimed to offer an installment into this gap by investigating trends among a population of 81 CrossFitters in south Mississippi. This paper discusses Richard Arena’s insightful and original contributions to interpreting the important interaction between Piero Sraffa and Ludwig Wittgenstein. It discusses this in terms of dilemmas they each encountered in transitions in their thinking in the 1930s, emphasizes the influence of Sraffa’s unpublished “Surplus Product” text, compares Sraffa’s critique of “natural science point of view” and Wittgenstein’s critique of logical form, and compares Sraffa’s later understanding of the relationship between production and distribution and Wittgenstein’s later understanding of forms of life and language-games. The relationship between wildlife and the environment they inhabit is dependent on both spatial and temporal scales. It is therefore crucial that biological investigations account for ecological scale when analyzing patterns and processes established, particularly when such investigations inform conservation management plans. To prevent early bridge failures, effective Structural Health Monitoring (SHM) is vital. Vibration-based damage assessment is a powerful tool in this regard, as it relies on changes in a structure’s dynamic characteristics as it degrades. The Native Hawaiian population is a marginalized group that has endured cultural trauma due, in part, to their history of colonization by Europe in 1778. This colonization has contributed to the Native Hawaiian’s loss of cultural identity, land, native language, and their way of life. As a result, many Native Hawaiians are left questioning their cultural identity. This project sought to better understand the impact of colonization on the Native Hawaiian people, specifically on their experience of homelessness, substance use, and overall disempowerment over time. The researcher evaluated middle school actors throughout the rehearsal process of a musical. The goal was to answer these three essential questions: Q1 Does feeling confident on stage as an actor/singer correlate with being a confident dancer? Nonalcoholic Steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) characterized by inflammation, fibrosis, steatosis, and oxidative stress, in addition to excessive fat accumulation in the liver. Cannabigerol (CBG), a precursor molecule of Cannabidiol (CBD), has demonstrated anti-inflammatory and anti-oxidative effects in various studies. Our previous data showed that a low dosage of CBG reduced inflammation and fibrosis in male NASH mice. However, the impact of CBG on female mice remains unknown. The mosquito family Culicidae is divided into 2 subfamilies named the Culicinae and Anophelinae. Nix, the dominant male-determining factor, has only been found in the culicines Aedes aegypti and Aedes albopictus, 2 important arboviral vectors that belong to the subgenus Stegomyia. Here we performed sex-specific whole-genome sequencing and RNAseq of divergent mosquito species and explored additional male-inclusive datasets to investigate the distribution of Nix. Except for the Culex genus, Nix homologs were found in all species surveyed from the Culicinae subfamily, including 12 additional species from 3 highly divergent tribes comprising 4 genera, suggesting Nix originated at least 133 to …

Genome-wide association studies (GWAS) of mood disorders in large case-control cohorts have identified numerous risk loci, yet pathophysiological mechanisms remain elusive, primarily due to the very small effects of common variants. We sought to discover risk variants with larger effects by conducting a genome-wide association study of mood disorders in a founder population, the Old Order Amish (OOA, n = 1,672). Our analysis revealed four genome-wide significant risk loci, all of which were associated with >2-fold relative risk. Quantitative behavioral and neurocognitive assessments (n = 314) revealed effects of risk variants on sub-clinical depressive symptoms and information processing speed. …

Anchorage-independent survival after intravasation of cancer cells from the primary tumor site represents a critical step in metastasis. Here, we reveal new insights into how MUC13-mediated anoikis resistance, coupled with survival of colorectal tumor cells, leads to distant metastasis. We found that MUC13 targets a potent transcriptional coactivator, YAP1, and drives its nuclear translocation via forming a novel survival complex, which in turn augments the levels of pro-survival and metastasis-associated genes. High expression of MUC13 is correlated well with extensive macrometastasis of colon cancer cells with elevated nuclear YAP1 in physiologically relevant whole animal model systems. Depression is a highly prevalent and debilitating condition, yet we still lack both in-depth knowledge concerning its etiopathology and sufficiently efficacious treatment options. With approximately one third of patients resistant to currently available antidepressants there is a pressing need for a better understanding of depression, identifying subgroups within the highly heterogeneous illness category and to understand the divergent underlying biology of such subtypes, to help develop and personalise treatments. Coronary artery calcification (CAC) is a measure of atherosclerosis and a well-established predictor of coronary artery disease (CAD) events. Here we describe a genome-wide association study of CAC in 22,400 participants from multiple ancestral groups. We confirmed associations with four known loci and identified two additional loci associated with CAC (ARSE and MMP16), with evidence of significant associations in replication analyses for both novel loci. Functional assays of ARSE and MMP16 in human vascular smooth muscle cells (VSMCs) demonstrate that ARSE is a promoter of VSMC calcification and VSMC phenotype switching from a contractile to a calcifying or osteogenic phenotype. …

Obesity is a risk factor for type 2 diabetes and cardiovascular disease. However, a substantial proportion of patients with these conditions have a seemingly normal body mass index (BMI). Conversely, not all obese individuals present with metabolic disorders giving rise to the concept of “metabolically healthy obese”. We use lipidomic-based models for BMI to calculate a metabolic BMI score (mBMI) as a measure of metabolic dysregulation associated with obesity. Using the difference between mBMI and BMI (mBMIΔ), we identify individuals with a similar BMI but differing in their metabolic health and disease risk profiles. After sampling the exoskeleton microbiomes of 225 ants from 3 different locations along a rural-urban gradient in the Chattanooga area, a photo library was collected and microbial diversity and morphospecies evenness of each location was determined. It was determined that there was no significant difference in either microbial diversity or morphospecies evenness between sites along the rural-urban gradient, even after adjusting data to be between only rural and urban areas. Urbanization was quantified using % impervious surface; however, a future study could quantify urbanization by other means, such as soil nutrient composition. We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. The efficient application of DFT and TD-DFT has been harnessed to study bond-breaking processes in some molecules which play a prominent role in enzymatic reactions. The first application includes Radical S-adenosyl methionine (SAM) enzymes which are fundamentally important sources of organic radicals to initiate diverse radical reactions. Recently a bio-organometallic intermediate (Ω) that contains an Fe‒C bond has been characterized and shown to be a common feature of radical SAM enzymes. Background: Gene therapy has recently emerged as an alternative for preventing and treating disease. Concerns have been expressed about the clinical value and the high cost of gene therapies.

Objective: This study assessed the authorization process of gene therapy products by the Food and Drug Administration (FDA) and the European Medicines Agency (EMA), evaluated the post-marketing safety signals reported to the FDA Adverse Event Reporting System (FAERS), and assessed the cost-effectiveness analysis of three gene therapies axicabtagene ciloleucel(axi-cel), lisocabtagene maraleucel (liso-cel), and tisagenlecleucel (tisa-cel).
