Serum amyloid A (SAA) is a family of proteins, the plasma levels of which may increase >1000-fold in acute inflammatory states. We investigated the role of SAA in sepsis using mice deficient in all three acute-phase SAA isoforms (SAA-TKO). SAA deficiency significantly increased mortality rates in the three experimental sepsis mouse models: cecal ligation and puncture (CLP), cecal slurry (CS) injection, and lipopolysaccharide (LPS) treatments. SAA-TKO mice had exacerbated lung pathology compared to wild-type (WT) mice after CLP. Background and aims: Genome and epigenome wide association studies identified variants in carnitine palmitoyltransferase 1a (CPT1a) that associate with lipid traits. The goal of this study was to determine the role of liver-specific CPT1a on hepatic lipid metabolism.

Approach and results: Male and female liver-specific knockout (LKO) and littermate controls were placed on a low-fat or high-fat diet (60% kcal fat) for 15 weeks. Mice were necropsied after a 16 h fast, and tissues were collected for lipidomics, matrix-assisted laser desorption ionization mass spectrometry imaging, kinome analysis, RNA-sequencing, and protein expression by immunoblotting. Objectives: To describe the increasing number of gastric cancer cases at Bahrain Defense Force Hospital with implementation of immunohistochemistry markers as prognostic factors. Methods: This study included histologically confirmed malignant gastric tumors diagnosed at Bahrain Defense Force Hospital from January 2009 to June 2019. Various epidemiological and pathological data were abstracted and recorded with immunohistochemical analysis of the proliferation marker Ki67 and cell-cycle regulator p53 as prognostic factors. An Exploration Into Health Equity Discourse In Mississippi: Organizational Commitments And Practitioner Perspectives, Thuy-Vy Lillian Pham

This thesis takes a sociological approach to examine healthcare workers' beliefs and medical organizations’ solidarity statements related to diversity and equity and how these discourses aligned and/or diverged. Through interviews with healthcare providers in Mississippi and content analysis of solidarity statements, the study finds that discussions on health disparities more frequently addressed socioeconomic inequality rather than systemic racism, diverging from organizational statements. This emphasis may partly stem from the demographics of the study participants, who were predominantly White—which also reflected the staff where they worked. Additionally, while healthcare workers recognized structural issues related to health disparities, they often showed hesitancy …

Robust literature exists to support the utility of CrossFit in improving metabolic and cardiovascular well-being. However, a metaphorical chasm exists not in the study of the physiological effects of CrossFit but in the psychological effects, namely quality of life (QOL). This paper discusses Richard Arena’s insightful and original contributions to interpreting the important interaction between Piero Sraffa and Ludwig Wittgenstein. It discusses this in terms of dilemmas they each encountered in transitions in their thinking in the 1930s, emphasizes the influence of Sraffa’s unpublished “Surplus Product” text, compares Sraffa’s critique of “natural science point of view” and Wittgenstein’s critique of logical form, and compares Sraffa’s later understanding of the relationship between production and distribution and Wittgenstein’s later understanding of forms of life and language-games. The Native Hawaiian population is a marginalized group that has endured cultural trauma due, in part, to their history of colonization by Europe in 1778. This colonization has contributed to the Native Hawaiian’s loss of cultural identity, land, native language, and their way of life. As a result, many Native Hawaiians are left questioning their cultural identity. Nonalcoholic Steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) characterized by inflammation, fibrosis, steatosis, and oxidative stress, in addition to excessive fat accumulation in the liver. Cannabigerol (CBG), a precursor molecule of Cannabidiol (CBD), has demonstrated anti-inflammatory and anti-oxidative effects in various studies. Our previous data showed that a low dosage of CBG reduced inflammation and fibrosis in male NASH mice. However, the impact of CBG on female mice remains unknown. The mosquito family Culicidae is divided into 2 subfamilies named the Culicinae and Anophelinae. Nix, the dominant male-determining factor, has only been found in the culicines Aedes aegypti and Aedes albopictus, 2 important arboviral vectors that belong to the subgenus Stegomyia. Here we performed sex-specific whole-genome sequencing and RNAseq of divergent mosquito species and explored additional male-inclusive datasets to investigate the distribution of Nix. Except for the Culex genus, Nix homologs were found in all species surveyed from the Culicinae subfamily, including 12 additional species from 3 highly divergent tribes comprising 4 genera, suggesting Nix originated at least 133 to …

Genome-wide association studies (GWAS) of mood disorders in large case-control cohorts have identified numerous risk loci, yet pathophysiological mechanisms remain elusive, primarily due to the very small effects of common variants. We sought to discover risk variants with larger effects by conducting a genome-wide association study of mood disorders in a founder population, the Old Order Amish (OOA, n = 1,672). Our analysis revealed four genome-wide significant risk loci, all of which were associated with >2-fold relative risk. Quantitative behavioral and neurocognitive assessments (n = 314) revealed effects of risk variants on sub-clinical depressive symptoms and information processing speed. …

Anchorage-independent survival after intravasation of cancer cells from the primary tumor site represents a critical step in metastasis. Here, we reveal new insights into how MUC13-mediated anoikis resistance, coupled with survival of colorectal tumor cells, leads to distant metastasis. We found that MUC13 targets a potent transcriptional coactivator, YAP1, and drives its nuclear translocation via forming a novel survival complex, which in turn augments the levels of pro-survival and metastasis-associated genes. High expression of MUC13 is correlated well with extensive macrometastasis of colon cancer cells with elevated nuclear YAP1 in physiologically relevant whole animal model systems. Depression is a highly prevalent and debilitating condition, yet we still lack both in-depth knowledge concerning its etiopathology and sufficiently efficacious treatment options. With approximately one third of patients resistant to currently available antidepressants there is a pressing need for a better understanding of depression, identifying subgroups within the highly heterogeneous illness category and to understand the divergent underlying biology of such subtypes, to help develop and personalise treatments. Coronary artery calcification (CAC) is a measure of atherosclerosis and a well-established predictor of coronary artery disease (CAD) events. Here we describe a genome-wide association study of CAC in 22,400 participants from multiple ancestral groups. We confirmed associations with four known loci and identified two additional loci associated with CAC (ARSE and MMP16), with evidence of significant associations in replication analyses for both novel loci. Functional assays of ARSE and MMP16 in human vascular smooth muscle cells (VSMCs) demonstrate that ARSE is a promoter of VSMC calcification and VSMC phenotype switching from a contractile to a calcifying or osteogenic phenotype. …

Obesity is a risk factor for type 2 diabetes and cardiovascular disease. However, a substantial proportion of patients with these conditions have a seemingly normal body mass index (BMI). Conversely, not all obese individuals present with metabolic disorders giving rise to the concept of “metabolically healthy obese”. We use lipidomic-based models for BMI to calculate a metabolic BMI score (mBMI) as a measure of metabolic dysregulation associated with obesity. Using the difference between mBMI and BMI (mBMIΔ), we identify individuals with a similar BMI but differing in their metabolic health and disease risk profiles. After sampling the exoskeleton microbiomes of 225 ants from 3 different locations along a rural-urban gradient in the Chattanooga area, a photo library was collected and microbial diversity and morphospecies evenness of each location was determined. It was determined that there was no significant difference in either microbial diversity or morphospecies evenness between sites along the rural-urban gradient, even after adjusting data to be between only rural and urban areas. Urbanization was quantified using % impervious surface; however, a future study could quantify urbanization by other means, such as soil nutrient composition. We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. We address this crisis using a ‘toxic aging coin’ approach; heads examines how age impacts toxicity, tails examines how chemicals accelerate aging. Virtual Reality (VR) technology has revolutionized the study of team effectiveness, yet many studies in this domain lack a comprehensive approach, focusing narrowly on performance metrics. This thesis investigates VR team effectiveness by integrating traditional Team Effectiveness frameworks with VR-specific dynamics. Utilizing Data Envelopment Analysis (DEA) and Group Style Inventory (GSI), this study assesses teams within a VR environment, specifically a multiplayer submarine simulation game, IronWolfVR. As healthcare becomes more complex, automated, and bureaucratic, patients often suffer from a lack of resources, agency, and visibility when seeking medical care. Rhetoric and Composition, specifically the subfield of Rhetoric of Health and Medicine (RHM), is interested in studying and intervening into such issues. One way to challenge our current understanding of healthcare is to consider how the rare disease patient experience reveals the gaps, limitations, and assumptions of illness and health.
