Stored Blood Transfusion Cause Hyper Inflammatory Response In Monocytes Of Neonatal Mouse, Ashritha Chiguluri, Marie Amalie Balamurugan, Balamurugan Ramatchandirin, Mohan Krishnan

Posters: 2023 Summer Undergraduate Research Program

Background: Neonatal anemia is nearly universal in preterm infants and is associated with increased morbidity and mortality worldwide. When anemia is severe enough to be treated with RBC transfusion, clinicians must be aware of the risk of critical adverse effects such as necrotizing enterocolitis (NEC), an inflammatory bowel necrosis characterized by monocyte infiltration, and a leading cause of mortality in those born before 28-weeks gestation. We have recently elucidated the connection between anemia and NEC, specifically, the “leaky gut” phenotype that leads to monocytes infiltration and RBC transfusion-associated activation of these monocytes, and the resulting intestinal mucosal injury.

Three-Dimensional (3d) In Vitro Cell Culture Protocols To Enhance Glioblastoma Research, Janith Wanigasekara, Lara Carroll, Patrick J. Cullen, Brijesh K. Tiwari, James Curtin

Articles

Three-dimensional (3D) cell culture models can help bridge the gap between in vitro cell cultures and in vivo responses by more accurately simulating the natural in vivo environment, shape, tissue stiffness, stressors, gradients and cellular response while avoiding the costs and ethical concerns associated with animal models. The inclusion of the third dimension in 3D cell culture influences the spatial organization of cell surface receptors that interact with other cells and imposes physical restrictions on cells in compared to Two-dimensional (2D) cell cultures. Spheroids’ distinctive cyto-architecture mimics in vivo cellular structure, gene expression, metabolism, proliferation, oxygenation, nutrition absorption, waste excretion, …

Although gene therapy has made great achievements in both laboratory research and clinical translation, there are still challenges such as limited control of drug pharmacokinetics, acute toxicity, poor tissue retention, insufficient efficacy, and inconsistent clinical translation. Herein, a gene therapy gel is formulated by directly redispersing polyplex nanoparticles into granular hydrogels without any gelation pre-treatment, which provides great convenience for storage, dosing and administration. In vitro studies have shown that use of granular hydrogels can regulate the gene drug release, reduce dose dependent toxicity and help improve transfection efficacy. Moreover, the developed gene therapy gel is easy to operate and …

The current global outbreak of coronavirus (COVID-19) continues to be a severe threat to human health. Rapid, low-cost, and accurate antigen detection methods are very important for disease diagnosis. The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) nucleocapsid protein (N-Protein) is often used as the diagnostic and screening for coronavirus detection. To this end, we propose and experimentally validate a highly sensitive whispering gallery mode (WGM) optical cylindrical micro-resonator (CMR) for bio immunoassay detection. To study the biokinetic process of immunoassay, the surface of the WGM micro-resonator is functionalized with N-Protein monoclonal antibody (N-Protein-m Ab), which led to the specific detection …

Microfluidic Flowmeter Based On Liquid Crystal Filled Nested Capillary, Zhe Wang, Zhuochen Wang, Anuradha Rout, Qiang Wu, Yuliya Semenova Jan 2023

Microfluidic Flowmeter Based On Liquid Crystal Filled Nested Capillary, Zhe Wang, Zhuochen Wang, Anuradha Rout, Qiang Wu, Yuliya Semenova

Conference Papers

A novel flowmeter composed of a liquid crystal-filled nested capillary is proposed and experimentally demonstrated. Whispering gallery modes (WGMs) in the nested capillary are excited by a tapered fiber coupled perpendicularly to the nested capillary. The WGM transmission spectrum of the fiber taper was optimized to achieve the highest possible quality (Q) factor by moving the capillary along the axis of the fiber taper. The air flowing through the capillary cools it down, which leads to a temperature-induced change of the refractive index of the nematic liquid crystal. This change in turn leads to a spectral shift of the WGM …

Use Of Edna To Determine The Presence Of Cryptosporidium Spp. In Freshwater Sources In Harlan, Kentucky, Madison Hall

Honors Theses

Cryptosporidium spp. oocysts are transmitted via a fecal-oral route, e.g., water sources, and the parasite is a leading cause of waterborne disease in the United States. Research into Cryptosporidiosis in rural areas is lacking. This study utilized eDNA methods to determine if Cryptosporidium spp. was present in Harlan, Kentucky, an underserved area in Appalachia. We used a pump-powered commercially available Smith-Root Citizen Scientist eDNA (Vancouver, WA) system to filter freshwater sources and collect eDNA. Samples were collected from tap and spring water as controls and three times from five locations on the three tributaries of the Cumberland River: Martins Fork …

Theses, Dissertations and Capstones

This research aims to better comprehend how the environmental preservation issues of underfunding and overcrowding of America’s parks, recreation areas, and environmental surroundings are solved through quality leadership. These environmental issues are national, state, and community problems. Currently, there exists a wealth of literature on the environmental problems caused by overcrowding and underfunding. Unfortunately, the successful problem-solving ideas identified throughout these works often do not connect with an element of leadership. This research helps fill this identified gap in the currently available environmental preservation resources. This mixed methods study explored the perceptions, ideas, and successful solutions exhibited by environmental preservation …

Gut Microbial Metabolite Indole: A Stimulator Of Enteroendocrine Cell Differentiation Via Activation Of Aryl Hydrocarbon Receptor, James Hart Jan 2023

Gut Microbial Metabolite Indole: A Stimulator Of Enteroendocrine Cell Differentiation Via Activation Of Aryl Hydrocarbon Receptor, James Hart

Theses, Dissertations and Capstones

Enteroendocrine cells (EECs) regulate energy balance and glucose homeostasis by releasing hormones in response to food intake. Dysregulated EEC differentiation is observed in obesity, while gut microbiota metabolites influence this process. Here, we investigated the role of indole, a biologically active gut microbial metabolite, in EEC differentiation through aryl hydrocarbon receptor (AhR) activation. Human intestinal organoids derived from jejunal mucosal biopsies were exposed to indole. Indole treatment significantly increased mRNA levels of chromogranin A, an EEC marker. The effect was reversed by an AhR antagonist, indicating AhR involvement. Indole also upregulated AhR target gene mRNA levels. These findings highlight the …

Pandemic As Transboundary Harm: Lessons From The Trail Smelter Arbitration, Russell A. Miller Jan 2023

Pandemic As Transboundary Harm: Lessons From The Trail Smelter Arbitration, Russell A. Miller

Scholarly Articles

The COVID-19 pandemic has caused incalculable harm around the world. The fact that this immense harm can be traced back to a localized outbreak in or near Wuhan, China, raises questions about the responsibility China might bear for the pandemic under public international law. Famously applied in the seminal Trail Smelter Arbitration (1938/1941), the Transboundary Harm Principle provides that no state can use or allow the use of its territory in a manner that causes significant harm in the territory of other states. This article does not intend to tap into the unseemly, xenophobic spirit that animates much of the …

International Law In The Boardroom, Kishanthi Parella Jan 2023

International Law In The Boardroom, Kishanthi Parella

Scholarly Articles

Conventional wisdom expects that international law will proceed through a “state pathway” before regulating corporations: it binds national governments that then bind corporations. But recent corporate practices confound this story. American corporations complied with international laws even when the state pathway broke down. This unexpected compliance leads to three questions: How did corporations comply? Why did they do so? Who enforced international law? These questions are important for two reasons. First, many international laws depend on corporate cooperation in order to succeed. Second, the state pathway is not robust, then or now. It is therefore vital to identify alternatives to …

Carboxylic Acids And Light Interact To Affect Nanoceria Stability And Dissolution In Acidic Aqueous Environments, Matthew L. Hancock, Eric A. Grulke, Robert Yokel

Nanobiotechnology Center Faculty Publications

Cerium atoms on the surfaces of nanoceria (i.e., cerium oxide in the form of nanoparticles) can store or release oxygen, cycling between Ce3+ and Ce4+; therefore, they can cause or relieve oxidative stress within living systems. Nanoceria dissolution occurs in acidic environments. Nanoceria stabilization is a known problem even during its synthesis; in fact, a carboxylic acid, namely citric acid, is used in many synthesis protocols. Citric acid adsorbs onto nanoceria surfaces, limiting particle formation and creating stable dispersions with extended shelf life. To better understand factors influencing the fate of nanoceria, its dissolution and stabilization have …
