Research in Drosophila melanogaster (D. melanogaster) has been growing in order to identify the fundamental processes of human disorders of the central nervous system such as Alzheimer’s disease, diabetes, and other neurological disorders at a molecular level. Altered insulin signaling itself has been linked to widespread nervous system dysfunction including cognitive dysfunction, neuropathies, and susceptibility to neurodegenerative disease. However, knowledge of the cellular mechanisms underlying the effects of insulin on nervous system function is still incomplete. Antibody-drug conjugates (ADCs) are one of fastest growing classes of oncology drugs in modern drug development. By harnessing the powers of both cytotoxic chemotherapy and targeted therapy, ADCs are unique in offering the potential to deliver highly potent cytotoxic agents to cancer cells which express a pre-defined cell surface target. In lung cancer, the treatment paradigm has shifted dramatically in recent years, and now ADCs are now joining the list as potential options for lung cancer patients. Single-cell DNA sequencing (scDNA-seq) methods are powerful tools for profiling mutations in cancer cells; however, most genomic regions sequenced in single cells are non-informative. To overcome this issue, we developed a multi-patient-targeted (MPT) scDNA-seq method. MPT involves first performing bulk exome sequencing across a cohort of cancer patients to identify somatic mutations, which are then pooled together to develop a single custom targeted panel for high-throughput scDNA-seq using a microfluidics platform. We applied MPT to profile 330 mutations across 23,500 cells from 5 patients with triple negative-breast cancer (TNBC), which showed that 3 tumors were monoclonal and 2 tumors were …

The use of immune checkpoint inhibitors (ICIs) has evolved rapidly with unprecedented treatment benefits being obtained for cancer patients, including improved patient survival. However, over half of the patients experience immune related adverse events (irAEs) or toxicities, which can be fatal, affect the quality of life of patients and potentially cause treatment interruption or cessation. Complications from these toxicities can also cause long term irreversible organ damage and other chronic health conditions. Toxicities can occur in various organ systems, with common observations in the skin, rheumatologic, gastrointestinal, hepatic, endocrine system and the lungs. Introduction: Do externalities work and matter differently in a world of scarcity vs. a world of abundance? In this article, we critically examine the economic phenomena of externalities. The concept of externality, an important idea in economics and law, is useful in exploring the complex and dynamic relationships between resource supply and human flourishing within various sociotechnical systems.

Assessment and prediction of vulnerable plaque progression and rupture risk are of utmost importance for diagnosis, management and treatment of cardiovascular diseases and possible prevention of acute cardiovascular events such as heart attack and stroke. However, accurate assessment of plaque vulnerability assessment and prediction of its future changes require accurate plaque cap thickness, tissue component and structure quantifications and mechanical stress/strain calculations. Multi-modality intravascular ultrasound (IVUS), optical coherence tomography (OCT) and angiography image data with follow-up were acquired from ten patients to obtain accurate and reliable plaque morphology for model construction. We used network analysis to identify subtypes of relapsing-remitting multiple sclerosis subjects based on their cumulative signs and symptoms. The electronic medical records of 113 subjects with relapsing-remitting multiple sclerosis were reviewed, signs and symptoms were mapped to classes in a neuro-ontology, and classes were collapsed into sixteen superclasses by subsumption. After normalization and vectorization of the data, bipartite (subject-feature) and unipartite (subject-subject) network graphs were created using NetworkX and visualized in Gephi. Degree and weighted degree were calculated for each node. Graphs were partitioned into communities using the modularity score. Feature maps visualized differences in features by community. Network …

Small cell undifferentiated (SCU) histology and alpha-fetoprotein (AFP) levels below 100 ng/mL have been reported as poor prognostic factors in hepatoblastoma (HB); subsequent studies reported SMARCB1 mutations in some SCU HBs confirming the diagnosis of rhabdoid tumor. The Children's Hepatic tumors International Collaboration (CHIC) database was queried for patients with HB who had AFP levels less than 100 ng/mL at diagnosis or were historically diagnosed as SCU HBs. Seventy-three of 1605 patients in the CHIC database were originally identified as SCU HB, HB with SCU component, or HB with low AFP levels. The serine/threonine protein kinase calcium/calmodulin-dependent protein kinase kinase 2 (CAMKK2) plays critical roles in a range of biological processes. Despite its importance, only a handful of inhibitors of CAMKK2 have been disclosed. Having a selective small molecule tool to interrogate this kinase will help demonstrate that CAMKK2 inhibition can be therapeutically beneficial. Herein, we disclose SGC-CAMKK2-1, a selective chemical probe that targets CAMKK2.

Honey bee colonies have perished at an alarming rate for over 15 years. Among the multitude of threats to bee health, virus infections and agrochemical exposure appear to be major drivers of colony loss. As virus infection is among the most damaging influences on bee health, we first investigated a putative target for antiviral immunity, termed KATP channels. We previously demonstrated that pharmacologically activating KATP channels induces the production of reactive oxygen species and increases survival during infection with Israeli Acute Paralysis Virus. Recently, driver inattention has become the leading cause of automobile accidents. As a result, the driver's perception and decision-making abilities are diminished, and the driver can lose control of the car. To prevent accidents caused by driver inattention, it’s vital to continuously monitor the driver and his driving behaviour and inform him if he becomes distracted or sleepy. This topic has been the subject of study for decades. Whenever feasible to recognise unsafe driving in advance, accidents could be avoided. This document presents an overview of the existing driver alertness system and the various techniques for detecting driver attentiveness.

DNA barcoding is a method for identifying specimens from specific regions of DNA. 1 Metabarcoding focuses on large, multi specimen scales. 2 These approaches rely on DNA databases for sequence identification, 2 but these databases often lack data from many species, 3 especially from poorly studied groups such as fungi 4 and lichens, which can lead to failure in specimen identification.

Purpose: The purpose of this quantitative meta-analysis and qualitative meta-synthesis was to conduct an analysis of five thematic research studies to aggregate and integrate findings to form new interpretations of the research field of grounded leadership.
Methodology: This quantitative meta-analysis and qualitative meta-synthesis was conducted as a secondary analysis of five thematic research studies to aggregate and integrate findings to form new interpretations of the research field of grounded leadership. The sample was composed of 79 exemplary leaders from five different fields.

In human Alzheimer's disease (AD) patients and AD mouse models, both differential pre-disease brain features and differential disease-associated memory decline are observed, suggesting that certain neurological features may protect against AD-related cognitive decline. The combination of these features is known as brain reserve, and understanding the genetic underpinnings of brain reserve may advance AD treatment in genetically diverse human populations. One potential source of brain reserve is brain microstructure, which is genetically influenced and can be measured with diffusion MRI (dMRI). Aims Childhood adversities (CAs) predict heightened risks of posttraumatic stress disorder (PTSD) and major depressive episode (MDE) among people exposed to adult traumatic events. Identifying which CAs put individuals at greatest risk for these adverse posttraumatic neuropsychiatric sequelae (APNS) is important for targeting prevention interventions. Methods Data came from n = 999 patients ages 18-75 presenting to 29 U.S. emergency departments after a motor vehicle collision (MVC) and followed for 3 months, the amount of time traditionally used to define chronic PTSD, in the Advancing Understanding of Recovery After Trauma (AURORA) study. Sustainability is a growing concern as resources are continually depleted for various applications without adequate renewal plans. The resulting impacts on the ecosystem, health, and resource circularity are often overlooked. This research analyzes improvement opportunities at each major stage in a product's life cycle. Raw material acquisition, product synthesis, process waste management, and the fate of a material in the end-of-life phases were examined. The viability of utilizing renewable resources has been demonstrated in this work by extracting bio-based chemicals from underutilized renewable resources at a commercial scale and transforming the extracted resources into polymeric materials. The objectives were to determine the sensitivity, specificity, and cutoff values of a visual-based precision livestock technology (NUtrack), and determine the sensitivity and specificity of sickness score data collected with the live observation by trained human observers. At weaning, pigs (n = 192; gilts and barrows) were randomly assigned to one of twelve pens (16/pen) and treatments were randomly assigned to pens. Sham-pen pigs all received subcutaneous saline (3 mL). For LPS-pen pigs, all pigs received subcutaneous lipopolysaccharide (LPS; 300 µg/kg BW; E. coli O111:B4; in 3 mL of saline). Membrane flux was observed and the two membrane sizes that achieved stable flux were chosen to be used in the later stages of the study. The chosen membranes were of 300 kDa and 400 kDa Molecular Weight Cutoff (MWCO).
The third phase of the study involved running the full system on synthetic greywater then on real greywater. The parameters investigated were COD, TSS, Turbidity and BOD5. AIMS: Childhood adversities (CAs) predict heightened risks of posttraumatic stress disorder (PTSD) and major depressive episode (MDE) among people exposed to adult traumatic events. Identifying which CAs put individuals at greatest risk for these adverse posttraumatic neuropsychiatric sequelae (APNS) is important for targeting prevention interventions.

Differences in selective pressures and the energetic cost of gametes in gonochoristic corals should vary with the sex of the colony, which may lead to sexual dimorphism. Coral colonies are composed of subunits (corallites) that create a complex morphological architecture. If corallite features are distinct between sexes, then the degree of coordinated change among these subunits may also vary (phenotypic modularity). This study tested for sexual dimorphism in the corallites of the reef-building coral Porites lobata, a gonochoric broadcast spawner, and compared this with previously demonstrated sexual dimorphism in its congener P. panamensis, a gonochoric brooder. Weedy rice, or red rice, is an agricultural weed that has evolved independently in different parts of the world from cultivated rice. Significant crop damage has resulted from weed infestations, leading to a loss of income for farmers and a severe reduction in crop yield. Seed shattering is one of the most distinctive and problematic characteristics of weedy rice, as it allows seeds to disperse and persist in soil for long periods and limit the effectiveness of current weed control efforts. Skin-to-skin contact (SSC) is an evidence-based practice with many benefits for mothers and their newborns. The separation of mothers from their babies after cesarean birth delays SSC, which can adversely affect the initiation and the success of breastfeeding. The purpose of this quantitative, quasi-experimental project was to determine if or to what degree the implementation of Hung and Berg's evidence-based protocol for skin-to-skin in the operating room (OR) would increase exclusive breastfeeding rates at discharge for adult women status post a cesarean delivery in a rural Arizona hospital over four-weeks. Background: Increases in fatal synthetic opioid overdoses over the past 8 years have left states scrambling for effective means to curtail these deaths. Many states have implemented policies and increased service capacity to address this rise. To better understand the effectiveness of policy level interventions we estimated the impact of the presence of naloxone access laws (NALs) on synthetic opioid fatalities at the state level. This thesis research consisted of two complimentary projects. The first focused on understanding the ecology and life-history of an isolated population of a freshwater fish, the Burbot; the second on a pre-restoration assessment of the fish community in the Grand River.
Understanding life-history strategies of species is important to recognize changes in behavior caused by anthropogenic effects. Burbot (Lota lota), are important apex predators in their native range, is understudied, and exhibits complex life-history strategies. Anxiety is one of the most prevalent forms of psychopathology that afects millions worldwide. It gained more importance under the pandemic status that resulted in higher anxiety prevalence. Anxiolytic drugs such as benzodiazepines have an unfavorable risk/beneft ratio resulting in a shift toward active ingredients with better safety profle such as the naturally occurring quercetin (QRC). The delivery of QRC is hampered by its low water solubility and low bioavailability.
