As abnormal acidic pH symbolizes dysfunctions of cells, it is highly desirable to develop pH-sensitive luminescent materials for diagnosing disease and imaging-guided therapy using high-energy radiation. Herein, we explored near-infrared-emitting Cr-doped zinc gallate ZnGa2O4 nanoparticles (NPs) in colloidal solutions with different pH levels under X-ray excitation. Ultrasmall NPs were synthesized via a facile hydrothermal method by controlling the addition of ammonium hydroxide precursor and reaction time, and structural characterization revealed Cr dopants on the surface of NPs. The synthesized NPs exhibited different photoluminescence and radioluminescence mechanisms, confirming the surface distribution of activators. It was observed that the colloidal NPs emit …

Cobalt-Sulfur Coordination Chemistry Drives B12 Loading Onto Methionine Synthase, Romila Mascarenhas, Arkajit Guha, Zhu Li, Markus Ruetz, Sojin An, Javier Seravalli, Ruma Banerjee

Department of Biochemistry: Faculty Publications

Cobalt-sulfur (Co-S) coordination is labile to both oxidation and reduction chemistry and is rarely seen in Nature. Cobalamin (or vitamin B12) is an essential cobalt-containing organometallic cofactor in mammals, and is escorted via an intricate network of chaperones to a single cytoplasmic target, methionine synthase. In this study, we report that the human cobalamin trafficking protein, MMADHC, exploits the chemical lability of Co-S coordination, for cofactor off-loading onto methionine synthase. Cys-261 on MMADHC serves as the -axial ligand to cobalamin. Complex formation between MMADHC and methionine synthase is signaled by loss of the lower axial nitrogen ligand, leading to five-coordinate …

Digital Journal of Clinical Medicine

ABSTRACT

The mammary gland is an exocrine gland present in female mammals responsible for the production and secretion of milk during the process of lactation. It is mainly composed by epithelial cells and adipocytes. The features that make mammary gland unique are 1) its highly plastic properties displayed during pregnancy, lactation and involution (all steps belonging to the lactation cycle) and 2) its requirement to grow in close association with adipocytes which are absolutely necessary to ensure mammary gland development at puberty and remodelling during the lactation cycle. 2020-Current year OA Pubs

Despite the advances in immunosuppressive medications, antibody-mediated rejection (AMR) continues to be a major cause of kidney allograft failure and remains a barrier to improving long-term allograft survival. Recently, there have been significant advances in the understanding of the pathophysiological process of AMR, along with the development of new therapeutic options. Additionally, surveillance protocols with donor-derived cell-free DNA and gene profile testing have been established, leading to the early detection of AMR. A multitude of clinical trials are ongoing, opening numerous opportunities for improving outcome in kidney transplant recipients. In this brief review, we discuss the emerging therapies for managing …

All Current Publications

Utah is a superb place for beginning bee enthusiasts. Over 1,000 species of native bees exist in Utah (Cane, 2015). Southern Utah alone has approximately the same number of bee species as the entire eastern U.S. coast. There are many reasons for appreciating bees and encouraging their presence in the landscape. Specifically, bees are key to a sustainable environment as they are essential pollinators of food and fiber crops. Bees can also be appreciated for their striking diversity in color and size. This fact sheet highlights the different bee species that you may see in Utah.

Boldness Is Not Associated With Dynamic Performance Capacity In Hermit Crabs., M Briffa, W Courtene-Jones Jul 2023

Boldness Is Not Associated With Dynamic Performance Capacity In Hermit Crabs., M Briffa, W Courtene-Jones

School of Biological and Marine Sciences

Boldness, the way an individual reacts to risk, is a commonly studied personality trait in animals. Consistent among-individual differences in startle response durations (latency to recover from a startling stimulus) are frequently assumed to reflect variation in boldness. An alternative explanation is that these latencies are not directly driven by variation in responses to information on risk, but by underlying differences in dynamic performance capacities. Here we investigate this possibility by analysing relationships between locomotory speed, a measure of whole-body dynamic performance capacity in hermit crabs, and startle response duration, a repeatable latency measure used as an index of boldness. …

Exploring The Relationships Between Mammalian Functional Trait Distributions And Regional Biomes, With Application To Miocene Paleoecology, Devra Hock-Reid Jul 2023

Exploring The Relationships Between Mammalian Functional Trait Distributions And Regional Biomes, With Application To Miocene Paleoecology, Devra Hock-Reid

Department of Earth and Atmospheric Sciences: Dissertations, Theses, and Student Research

Paleoecology relies on understanding relationships between modern animals and their environment. Animals are adapted to niches in their environments, and those physical adaptations, or functional traits, are utilized as proxies to interpret aspects of paleo-ecosystems. Much is known about individual functional traits in extant mammals and their relationship to the environment. Less is known about how multiple functional traits across a community can be utilized for paleoecological interpretations. I develop models utilizing traits in mammalian communities at the biome level. For Chapter 1, I build a model for North American regional biomes using mammalian trait frequencies. I quantify changes in …

Scalable Production Of Peptides For Enhanced Struvite Formation Via Expression On The Surface Of Genetically Engineered Microbes, Jacob D. Hostert, Quincy A. Spitzer, Paola Giammattei, Julie N. Renner

Faculty Scholarship

A promising method for recycling phosphate from wastewater is through precipitation of struvite (MgNH4PO4·6H2O), a slow-release fertilizer. Peptides have been shown to increase the yield of struvite formation, but producing peptides via solid phase synthesis is cost prohibitive. This work investigates the effects of peptide-expressing bacteria on struvite precipitation to provide a sustainable and cost-efficient means to enhance struvite precipitation. A peptide known for increased struvite yield was expressed on a membrane protein in Escherichia coli(E. coli), and then 5 mL precipitation reactions were performed in 50 mL culture tubes for at least 15 min. The yield of struvite crystals …

Differential Expression Of Genes Involved In The Chronic Response To Intracortical Microelectrodes, Sydney S. Song, Lindsey N. Druschel, E. Ricky Chan, Jeffrey R. Capadona

Faculty Scholarship

Brain-Machine Interface systems (BMIs) are clinically valuable devices that can provide functional restoration for patients with spinal cord injury or improved integration for patients requiring prostheses. Intracortical microelectrodes can record neuronal action potentials at a resolution necessary for precisely controlling BMIs. However, intracortical microelectrodes have a demonstrated history of progressive decline in the recording performance with time, inhibiting their usefulness. One major contributor to decreased performance is the neuroinflammatory response to the implanted microelectrodes. The neuroinflammatory response can lead to neurodegeneration and the formation of a glial scar at the implant site. Historically, histological imaging of relatively few known cellular …

Effect Of Biological Variation In Hba1c And Blood Glucose On The Diagnosis Of Prediabetes, James M. Hempe, Shengping Yang, Daniel S. Hsia

School of Medicine Faculty Publications

Introduction: People with a low or high haemoglobin glycation index (HGI) have lower or higher HbA1c than other people with the same FPG. This study compared the prevalence of prediabetes based on FPG, 2hOGTT and HbA1c in people with low, moderate or high HGI. Methods: Prediabetes was diagnosed based on ADA cutpoints in 10,488 NHANES participants without self-reported diabetes. HGI was calculated as the difference between a participant's observed HbA1c and a predicted HbA1c where predicted HbA1c = 0.024 FPG + 3.1. Participants were divided into low (HGI < −0.15%), moderate (HGI −0.15% to +0.15%) and high (HGI > +0.15%) HGI subgroups. Results: The prevalence of prediabetes was 42.4% based on …

The PTH-related peptide(1-34) analog, abaloparatide (ABL), is the second anabolic drug available for the treatment of osteoporosis. Previous research demonstrated that ABL had a potent anabolic effect but caused hypercalcemia at a significantly lower rate. However, the mechanism by which ABL maintains the stability of blood calcium levels remains poorly understood. Our in vivo data showed that ABL treatment (40 µg/kg/day for 7 days) significantly increased rat blood level of 1,25-dihydroxyvitamin D [1,25-(OH)2D] without raising the blood calcium value. ABL also significantly augmented the carboxylated osteocalcin (Gla-Ocn) in the blood and bone that is synthesized by osteoblasts, and increased noncarboxylated …

Urban expansion and development continue to increase globally, threatening rural multifunctional landscapes that provide a diversity of direct and indirect ecological benefits. However, not all landscapes are equally impacted, thus making conservation planning more difficult. We used 8 years of land use and land cover to understand this change in Greenville Co. SC, a region experiencing rapid expansion of development into rural landscapes. Regression models helped explain the relationships between development predictor variables and rural response variables. To explore where these changes were occurring, we determined which size categories of land parcels were the most developed and how this is …

Avoiding Anthropomoralism, Julian Friedland Jul 2023

Avoiding Anthropomoralism, Julian Friedland

Between the Species

The Montreal Declaration on Animal Exploitation, which has been endorsed by hundreds of influential academic ethicists, calls for establishing a vegan economy by banning what it refers to as all unnecessary animal suffering, including fishing. It does so by appeal to the moral principle of equal consideration of comparable interests. I argue that this principle is misapplied by discounting morally relevant cognitive capacities of self-conscious and volitional personhood as distinguished from merely sentient non-personhood. I describe it as a kind of anthropomorphizing moralism which I call anthropomoralism, defined as the tendency to project morally relevant characteristics of personhood onto merely …

Unraveling Prevalence And Effects Of Deleterious Mutations In Maize Elite Lines Across Decades Of Modern Breeding, Shichao Sun, Baobao Wang, Changyu Li, Gen Xu, Jinliang Yang, Matthew B. Hufford, Jeffrey Ross-Ibarra, Haiyang Wang, Li Wang

Department of Agronomy and Horticulture: Faculty Publications

Future breeding is likely to involve the detection and removal of deleterious alleles, which are mutations that negatively affect crop fitness. However, little is known about the prevalence of such mutations and their effects on phenotypic traits in the context of modern crop breeding. To address this, we examined the number and frequency of deleterious mutations in 350 elite maize inbred lines developed over the past few decades in China and the United States. Our findings reveal an accumulation of weakly deleterious mutations and a decrease in strongly deleterious mutations, indicating the dominant effects of genetic drift and purifying selection …

BACKGROUND: Juvenile idiopathic arthritis (JIA) is a common pediatric rheumatic condition and is associated with symptoms such as joint pain that can negatively impact health-related quality of life. To effectively manage pain in JIA, young people, their families, and health care providers (HCPs) should be supported to discuss pain management options and make a shared decision. However, pain is often under-recognized, and pain management discussions are not optimal. No studies have explored decision-making needs for pain management in JIA using a shared decision making (SDM) model. We sought to explore families' decision-making needs with respect to pain management among young …

We propose a statistical framework ISLET to infer individual-specific and cell-type-specific transcriptome reference panels. ISLET models the repeatedly measured bulk gene expression data, to optimize the usage of shared information within each subject. ISLET is the first available method to achieve individual-specific reference estimation in repeated samples. Using simulation studies, we show outstanding performance of ISLET in the reference estimation and downstream cell-type-specific differentially expressed genes testing. We apply ISLET to longitudinal transcriptomes profiled from blood samples in a large observational study of young children and confirm the cell-type-specific gene signatures for pancreatic islet autoantibody. The p53 protein is an important tumor suppressor, and TP53 mutations are frequently associated with poor prognosis in various cancers. Mutations in TP53 result in a loss of p53 function and enhanced expression of cell cycle genes, contributing to the development and progression of cancer. Meanwhile, several tripartite motif (TRIM) proteins are known to regulate cell growth and cell cycle transition. However, the prognostic values between TP53 and TRIM family genes in cancer are unknown. In this study, we analyzed the relationship between the

T cells are the central drivers of many inflammatory diseases, but the repertoire of tissue-resident T cells at sites of pathology in human organs remains poorly understood. We examined the site-specificity of T cell receptor (TCR) repertoires across tissues (5 to 18 tissues per patient) in prospectively collected autopsies of patients with and without graft-versus-host disease (GVHD), a potentially lethal tissue-targeting complication of allogeneic hematopoietic cell transplantation, and in mouse models of GVHD. Anatomic similarity between tissues was a key determinant of TCR repertoire composition within patients, independent of disease or transplant status. The T cells recovered from peripheral blood …

Organoid models have the potential to recapitulate the biological and pharmacotypic features of parental tumors. Nevertheless, integrative pharmaco-proteogenomics analysis for drug response features and biomarker investigation for precision therapy of patients with liver cancer are still lacking. We established a patient-derived liver cancer organoid biobank (LICOB) that comprehensively represents the histological and molecular characteristics of various liver cancer types as determined by multiomics profiling, including genomic, epigenomic, transcriptomic, and proteomic analysis. Proteogenomic profiling of LICOB identified proliferative and metabolic organoid subtypes linked to patient prognosis. High-throughput drug screening revealed distinct response patterns of each subtype that were associated with specific …

The molecular machinery that enables life has evolved in water, yet many of the organisms around us are able to survive even extreme desiccation. Especially remarkable are single-cell and sedentary organisms that rely on specialized biomolecular machinery to survive in environments that are routinely subjected to a near-complete lack of water. In this review, we zoom in on the molecular level of what is happening in the cellular environment under water stress. The widespread use of dicamba across the soybean growing regions of the United States resulted in increased off-target movement of this herbicide to susceptible vegetations. There are speculations that a drift of sublethal (ultra-low) doses of dicamba can enhance soybean growth and yield through a phenomenon called hormesis. Field studies were conducted during 2018 and 2019 to determine whether soybean growth and yield can be enhanced with ultra-low doses (0.0112–56 g ae ha-1) of dicamba, applied at three growth stages (V2, R1 and R2). There was no evidence that the ultra-low doses of dicamba (0.0112–56 g ae ha …
