Novel Breath Collection Techniques For Detection Of Covid-19., James Morris
Novel Breath Collection Techniques For Detection Of Covid-19., James Morris
Electronic Theses and Dissertations
Volatile Organic Compounds (VOC) generated endogenously in the human body can be used to detect diseases that induce oxidative stress and inflammation. Breath analysis has been used for the detection of diseases such as COPD, Depression, Lung Cancer and most recently COVID-19. Methods such as Exhaled Breath Condensate (EBC), Sorbent Tubes, Solid Phase Microextraction (SPME), and silicon microreactors have shown considerable capability in extracting and concentrating trace VOC’s present in human breath. Silicon microreactors functionalized with capture agents to derivatize carbonyl compounds are effective but the overall maximum flow rate of breath sample possible during analysis is low, making direct …
Tropical Understory And Canopy Twig Ants (Pseudomyrmex): Natural History, Sensory Homing, And Neuroecology., Daniella Prince
Tropical Understory And Canopy Twig Ants (Pseudomyrmex): Natural History, Sensory Homing, And Neuroecology., Daniella Prince
Electronic Theses and Dissertations
In this dissertation, I examined the behavior of foraging twig ants, explored the sensory profile of their homing behavior, and investigated the role of habitat use in shaping the sensory neurobiology of species within the genus Pseudomyrmex (Hymenoptera: Formicidae). In chapter one, I reviewed the ecology of Pseudomyrmex, and introduced the focal species, P. boopis, a neotropical understory-dwelling twig ant. In chapter two, I described the natural history of P. boopis on Barro Colorado Island, Panama—specifically, their nesting habits, activity rhythm, and foraging behavior on understory surfaces. Pseudomyrmex boopis colonies were diurnal and nested in dead twigs in …

Suicide has proven to be difficult to predict and prevent at the individual level,1–3 so much so that a recent analysis of individual risk assessments found a maximal positive predictive value of 5.5% for suicide death,2 and suicide continues to be a leading cause of death among adolescents age 10-19 years.4 By targeting modifiable risk factors at the population level, policy has the potential to extend our reach to people and subgroups that are missed by individual level interventions. This ecological study of adolescent suicide rates, state policies, and child maltreatment in US states from 2005-2019 and Kentucky counties from …
Inhibition Of Ras As A Novel Therapeutic Approach In Luminal B Breast Cancer., Raphael Ngozichi Jigo
Inhibition Of Ras As A Novel Therapeutic Approach In Luminal B Breast Cancer., Raphael Ngozichi Jigo
Electronic Theses and Dissertations
Historically, the RAS oncoprotein has not been implicated in breast cancer due to less than 1% of breast cancer cases bearing oncogenic RAS mutations. Recently, however, it has been reported that greater than 60% of Luminal B breast cancer cases have a decreased expression of negative RAS regulators RASAL2 and DAB2IP. Thus, in many Luminal B breast cancers, RAS remains in a constitutively active state without mutation, in turn driving oncogenesis. To date, there are no FDA-approved direct inhibitors of wild type RAS. We have developed a direct RAS inhibitor that acts on wild type RAS using in silico drug …
Aurka Inhibition-Based Combination Therapy Approaches In Glioblastoma., Muge Sak
Aurka Inhibition-Based Combination Therapy Approaches In Glioblastoma., Muge Sak
Electronic Theses and Dissertations
Glioblastoma is the most common and aggressive central nervous system tumor in adults with a 6% survival rate. Decades of research for improving life expectancy and quality of life have been ineffective. The discovery of better treatment strategies is urgent to improve the survival rates for glioblastoma patients. Aurora Kinase A is a regulator of tumor progression and is overexpressed in glioblastoma. We used a specific Aurora Kinase A inhibitor alisertib in combination with DNA-damaging platinum agent carboplatin or brain-penetrating taxane TPI 287. We investigated the efficacy of these combinations in treating in vitro and in vivo tissue culture models …

Cad System For Early Diagnosis Of Diabetic Retinopathy Based On 3d Extracted Imaging Markers., Ahmed Sleman
Electronic Theses and Dissertations
This dissertation makes significant contributions to the field of ophthalmology, addressing the segmentation of retinal layers and the diagnosis of diabetic retinopathy (DR). The first contribution is a novel 3D segmentation approach that leverages the patientspecific anatomy of retinal layers. This approach demonstrates superior accuracy in segmenting all retinal layers from a 3D retinal image compared to current state-of-the-art methods. It also offers enhanced speed, enabling potential clinical applications. The proposed segmentation approach holds great potential for supporting surgical planning and guidance in retinal procedures such as retinal detachment repair or macular hole closure. Surgeons can benefit from the accurate …
Connectivity Of Amygdala Somatostatin-Expressing Neurons And Their Role In Taste-Guided Behavior., Jane Jeruto Bartonjo
Connectivity Of Amygdala Somatostatin-Expressing Neurons And Their Role In Taste-Guided Behavior., Jane Jeruto Bartonjo
Electronic Theses and Dissertations
The nucleus of solitary tract (NST) and parabrachial nucleus (PBN) represent the first and second central synapses of ascending gustatory information. Neural processing in these nuclei is influenced by descending input from forebrain regions such as the central nucleus of the amygdala (CeA). In mice, we have shown that somatostatin (Sst) expressing neurons of CeA that project to NST and PBN are largely distinct cell populations and optogenetic inhibition of the CeA/Sst-to-NST subpopulation increases the intake of high concentrations of quinine with no apparent effect on sucrose intake. Synaptic connectivity of these cells is needed to understand the possible mechanisms …

Fbg Αc 389 – 402 Modulates Factor Xiii Crosslinking In The Fibrinogen Αc Region., Francis Dean Orlina Ablan
Electronic Theses and Dissertations
Fibrinogen (Fbg) is a coagulation protein critical for clot formation. Coagulation Factor XIII (FXIII) is a calcium-dependent transglutaminase that crosslinks reactive glutamines (Q) and lysines (K) between fibrin and other anti-fibrinolytic proteins. In the presence of Ca2+, FXIII could be activated non-proteolytically (FXIII-A°), or proteolytically by thrombin (FXIII-A*). Significant increases in clot stability and red blood cell retention are linked to FXIII activity in the fibrinogen αC region (Fbg Aα 221 – 610). This region contains several FXIII-reactive glutamines and lysines, as well as a binding site for FXIII-A* (Fbg αC 389 – 402) that includes a key …
The Role Of E-Liquid Constituents In E-Cigarette-Induced Cardiac Arrhythmia And Autonomic Imbalance., Cory Kucera
The Role Of E-Liquid Constituents In E-Cigarette-Induced Cardiac Arrhythmia And Autonomic Imbalance., Cory Kucera
Electronic Theses and Dissertations
Introduction. Accumulating evidence indicates that exposure to electronic cigarettes (e-cigs) promotes sympathetic dominance and electrophysiologic instability in the heart, potentially culminating in arrhythmogenesis. E-liquids contain various formulations of nicotine and flavorings, but the effects of specific e-liquid constituents and their concentrations on e-cig-induced autonomic imbalance and electrical dysfunction are unresolved. To that end, we tested the hypothesis that e-cigs modify cardiac autonomic balance and ventricular arrhythmogenesis in mice dependent on e-liquid constituent type and concentration. Methods. Using a crossover design and a serial exposure regimen, ECG-telemetered male C57BL/6J mice underwent whole-body exposure to e-cig aerosols generated from e-liquids containing different …
Penalized Bayesian Exponential Random Graph Models., Vicki Modisette
Penalized Bayesian Exponential Random Graph Models., Vicki Modisette
Electronic Theses and Dissertations
Networks have the critical ability to represent the complex interconnectedness of social relationships, biological processes, and the spread of diseases and information. Exponential random graph models (ERGM) are one of the popular statistical methods for analyzing network data. ERGM, however, struggle with computational challenges and degeneracy issues, further exacerbated by their inability to handle high-dimensional network data. Bayesian techniques provide a promising avenue to overcome these two problems. This paper considers penalized Bayesian exponential random graph models with adaptive lasso and adaptive ridge penalties to perform variable selection and reduce multicollinearity on a variety of networks. The experimental results demonstrate …

Traditional miniaturized fluorescence microscopes are critical tools for modern biology. Invariably, they struggle to simultaneously image with a high spatial resolution and a large field of view (FOV). Lensless microscopes offer a solution to this limitation. However, real-time visualization of samples is not possible with lensless imaging, as image reconstruction can take minutes to complete. This poses a challenge for usability, as real-time visualization is a crucial feature that assists users in identifying and locating the imaging target. The issue is particularly pronounced in lensless microscopes that operate at close imaging distances. Imaging at close distances requires shift-varying deconvolution to …

The heart contains diverse endothelial cell types. We sought to characterize the endocardial endothelial cells (EECs), which line the chambers of the heart. EECs are relatively understudied, yet their dysregulation can lead to various cardiac pathologies. Due to the lack of commercial availability of these cells, we reported our protocol for isolating EECs from porcine hearts and for establishing an EEC population through cell sorting. In addition, we compared the EEC phenotype and fundamental behaviors to a well-studied endothelial cell line, human umbilical vein endothelial cells (HUVECs). The EECs stained positively for classic phenotypic markers such as CD31, von Willebrand …

Background: RNA profiling technologies at single-cell resolutions, including single-cell and single-nuclei RNA sequencing (scRNA-seq and snRNA-seq, scnRNA-seq for short), can help characterize the composition of tissues and reveal cells that influence key functions in both healthy and disease tissues. However, the use of these technologies is operationally challenging because of high costs and stringent sample-collection requirements. Computational deconvolution methods that infer the composition of bulk-profiled samples using scnRNA-seq-characterized cell types can broaden scnRNA-seq applications, but their effectiveness remains controversial.
Results: We produced the first systematic evaluation of deconvolution methods on datasets with either known or scnRNA-seq-estimated compositions. Our analyses revealed …

Normal adjacent tissues (NATs) of hepatocellular carcinoma (HCC) differ from healthy liver tissues and their heterogeneity may contain biological information associated with disease occurrence and clinical outcome that has yet to be fully evaluated at the proteomic level. This study provides a detailed description of the heterogeneity of NATs and the differences between NATs and healthy livers and revealed that molecular features of tumor subgroups in HCC were partially reflected in their respective NATs. Proteomic data classified HCC NATs into two subtypes (Subtypes 1 and 2), and Subtype 2 was associated with poor prognosis and high-risk recurrence. The pathway and …

Nonalcoholic fatty liver disease (NAFLD), including its more severe manifestation, nonalcoholic steatohepatitis (NASH), has a global prevalence of 20-25% and is a major public health problem. Its incidence is increasing in parallel to the rise in obesity, diabetes and metabolic syndrome. Progression from NASH to NASH-related hepatocellular carcinoma (HCC) (~2% of cases per year) is influenced by many factors, including the tissue and immune microenvironment, germline mutations in PNPLA3, and the microbiome. NASH-HCC has unique molecular and immune traits compared with other aetiologies of HCC and is equally prevalent in men and women. Comorbidities associated with NASH, such as obesity …

Estrogen and progesterone, acting through their cognate receptors the estrogen receptor α (ERα) and the progesterone receptor (PR) respectively, regulate uterine biology. Using rapid immunoprecipitation and mass spectrometry (RIME) and co-immunoprecipitation, we identified TRIM28 (Tripartite motif containing 28) as a protein which complexes with ERα and PR in the regulation of uterine function. Impairment of TRIM28 expression results in the inability of the uterus to support early pregnancy through altered PR and ERα action in the uterine epithelium and stroma by suppressing PR and ERα chromatin binding. Furthermore, TRIM28 ablation in PR-expressing uterine cells results in the enrichment of a …

BACKGROUND & AIMS: Black patients with hepatocellular cancer (HCC), often attributed to hepatitis C virus (HCV) infection, have suboptimal survival following liver transplant (LT). We evaluated the impact of direct-acting antiviral (DAA) availability on racial and ethnic disparities in wait list burden post-LT survival for candidates with HCC.
METHODS: Using the United Network for Organ Sharing registry, we identified patients with HCC who were listed and/or underwent LT from 2009 to 2020. Based on date of LT, patients were categorized into 2 era-based cohorts: the pre-DAA era (LT between 2009 and 2011) and DAA era (LT between 2015 and 2017, …

Chronic inflammation is associated with increased risk and poor prognosis of heart failure; however, the precise mechanism that provokes sustained inflammation in the failing heart remains elusive. Here we report that depletion of carnitine acetyltransferase (CRAT) promotes cholesterol catabolism through bile acid synthesis pathway in cardiomyocytes. Intracellular accumulation of bile acid or intermediate, 7α-hydroxyl-3-oxo-4-cholestenoic acid, induces mitochondrial DNA stress and triggers cGAS-STING-dependent type I interferon responses. Furthermore, type I interferon responses elicited by CRAT deficiency substantially increase AIM2 expression and AIM2-dependent inflammasome activation. Genetic deletion of cardiomyocyte CRAT in mice of both sexes results in myocardial inflammation and dilated cardiomyopathy, …

Loss of estrogen as a result of aging, pelvic cancer therapy, genetics, or eating disorders affects numerous body systems including the reproductive tract. Specifically, a chronic hypoestrogenic state fosters debilitating vaginal symptoms like atrophy, dryness, and dyspareunia. Current treatment options, including vaginal estrogen and hyaluronan (HA), anecdotally improve symptoms, but rectifying mechanisms are largely understudied. In order to study the hypoestrogenic vaginal environment, in particular the extracellular matrix (ECM), as well as understand the mechanisms behind current treatments and develop new therapies, we characterized a reliable and reproducible animal model. Bilateral ovariectomies (OVX) were performed on 9‐week‐old CD1 mice. After …

Music is core to human experience, yet the precise neural dynamics underlying music perception remain unknown. We analyzed a unique intracranial electroencephalography (iEEG) dataset of 29 patients who listened to a Pink Floyd song and applied a stimulus reconstruction approach previously used in the speech domain. We successfully reconstructed a recognizable song from direct neural recordings and quantified the impact of different factors on decoding accuracy. Combining encoding and decoding analyses, we found a right-hemisphere dominance for music perception with a primary role of the superior temporal gyrus (STG), evidenced a new STG subregion tuned to musical rhythm, and defined …
