Tafopathies Result From Derangements In Transcriptional Regulation Of Metabolism, Jamison Maxwell Leid Sep 2023

Tafopathies Result From Derangements In Transcriptional Regulation Of Metabolism, Jamison Maxwell Leid

Arts & Sciences Graduate Student Theses and Dissertations

Cardiogenesis and the maintenance of cardiac physiology necessitate dynamic and carefully coordinated transcription programs. Studies in a many systems – both in vitro and in vivo – have elucidated critical signaling pathways, including Bmp, Fgf, Wnt, and Notch that induce a cardiogenic transcription program, thereby driving cardiac specification and differentiation. Recently, studies have discovered additional regulation at the epigenetic level that contributes to the tight temporal and spatial control of cardiac gene expression during development. Because transcriptional regulation is essential to cardiogenesis, changes in transcriptional regulation result in developmental defects, including but not limited to CHDs, the leading inborn cause …

Lem2 Is Essential For Cardiac Development By Maintaining Nuclear Integrity, Jacob A Ross, Didier Hodzic, Et Al. Sep 2023

Lem2 Is Essential For Cardiac Development By Maintaining Nuclear Integrity, Jacob A Ross, Didier Hodzic, Et Al.

2020-Current year OA Pubs

AIMS: Nuclear envelope integrity is essential for the compartmentalization of the nucleus and cytoplasm. Importantly, mutations in genes encoding nuclear envelope (NE) and associated proteins are the second highest cause of familial dilated cardiomyopathy. One such NE protein that causes cardiomyopathy in humans and affects mouse heart development is Lem2. However, its role in the heart remains poorly understood.

METHODS AND RESULTS: We generated mice in which Lem2 was specifically ablated either in embryonic cardiomyocytes (Lem2 cKO) or in adult cardiomyocytes (Lem2 iCKO) and carried out detailed physiological, tissue, and cellular analyses. High-resolution episcopic microscopy was used for three-dimensional reconstructions …

Effect Of An Rn-Facilitated Post Discharge Phone Call Hf Self-Management Strategy, Vivia O. Haye

Dissertations

Heart Failure (HF) is associated with substantial financial challenges resulting in exorbitant yearly costs, and frequent readmits to the emergency department. Post-discharge HF self-management is crucial to improve client outcomes. The purpose of this quantitative, quasi-experimental, project was to examine whether the implementation of an RN-facilitated post-discharge phone call that focused on HF self-management strategies reduced return emergency department (ED) visits within 72 hours in an urban South Florida hospital. Orem's self-care theory and the Meleis transition model were used for their success in managing chronic illnesses such as HF. Both examined if an RN facilitated post-discharge telephone call that …

State public assistance spending, including Medicaid and cash assistance programs for socioeconomically disadvantaged individuals, may improve access to care; address barriers, such as food and housing insecurity; and lead to improved cancer outcomes for marginalized populations.

OBJECTIVE: To determine whether state-level public assistance spending is associated with overall survival (OS) among individuals with cancer, overall and by race and ethnicity.

DESIGN, SETTING, AND PARTICIPANTS: This cohort study included US adults aged at least 18 years with a new cancer diagnosis from 2007 to 2013, with follow-up through 2019. Data …

Periodical Cicadas (Magicicada Spp.): Predator Satiation, Or Too Much Of A Good Thing?, John R. Cooley, Greg Holmes Sep 2023

Periodical Cicadas (Magicicada Spp.): Predator Satiation, Or Too Much Of A Good Thing?, John R. Cooley, Greg Holmes

The Great Lakes Entomologist

We report a novel mode of ecdysis failure in periodical cicadas (Magicicada spp. Davis) tied to the cicadas’ reliance on high densities for survival in the face of predation. Under extremely high densities, cicadas can disrupt others undergoing ecdysis by knocking them loose from their perches, trapping them, or causing physical damage. Our observations raise the possibility that while periodical cicadas thrive at high densities, it is possible that some densities are so high that they inhibit success.

On A Central Problem Of The Human Placenta Project, Charles F. Babbs Sep 2023

On A Central Problem Of The Human Placenta Project, Charles F. Babbs

Weldon School of Biomedical Engineering Faculty Working Papers

This brief technical note describes a design concept for a low-cost, painless, noninvasive monitor of total placental blood flow during the third trimester of pregnancy. The approach may offer an overlooked solution to a central problem of the human placenta project. A systems level biophysical analysis shows that jets of blood flow emerging from spiral arteries in the placenta generate characteristic 1/f or “pink noise” which is routinely audible, but typically ignored, during fetal phonocardiography. The amplitude of the 1/f noise, recorded at a standardized location in an individual patient, is a measure of total placental blood flow that is …

It is increasingly assumed that there is no one-size-fits-all approach to dietary recommendations for the management and treatment of chronic diseases such as obesity. This phenomenon that not all individuals respond uniformly to a given treatment has become an area of research interest given the rise of personalized and precision medicine. To conduct, interpret, and disseminate this research rigorously and with scientific accuracy, however, requires an understanding of treatment response heterogeneity. Here, we define treatment response heterogeneity as it relates to clinical trials, provide statistical guidance for measuring treatment response heterogeneity, and highlight study designs that can quantify treatment response …

Mitochondrial Pyruvate Metabolism And Glutaminolysis Toggle Steady-State And Emergency Myelopoiesis, Hannah A Pizzato, Yahui Wang, Michael J Wolfgang, Brian N Finck, Gary J Patti, Deepta Bhattacharya

2020-Current year OA Pubs

To define the metabolic requirements of hematopoiesis, we examined blood lineages in mice conditionally deficient in genes required for long-chain fatty acid oxidation (Cpt2), glutaminolysis (Gls), or mitochondrial pyruvate import (Mpc2). Genetic ablation of Cpt2 or Gls minimally impacted most blood lineages. In contrast, deletion of Mpc2 led to a sharp decline in mature myeloid cells and a slower reduction in T cells, whereas other hematopoietic lineages were unaffected. Yet MPC2-deficient monocytes and neutrophils rapidly recovered due to a transient and specific increase in myeloid progenitor proliferation. Competitive bone marrow chimera and stable isotope tracing experiments demonstrated that this proliferative …

Understanding the microglial neuro-immune interactions in the primate brain is vital to developing therapeutics for cortical injury, such as stroke or traumatic brain injury. Our previous work showed that mesenchymal-derived extracellular vesicles (MSC-EVs) enhanced motor recovery in aged rhesus monkeys following injury of primary motor cortex (M1), by promoting homeostatic ramified microglia, reducing injury-related neuronal hyperexcitability, and enhancing synaptic plasticity in perilesional cortices. A focal lesion was induced via surgical ablation of pial blood vessels over lying the cortical hand representation of M1 of aged female rhesus monkeys, that received intravenous infusions of either vehicle (veh) or EVs 24 h …

The Wealth Of Our Commonwealth: Money, Capital, And Finance In A Productive Commercial Republic, Robert Hockett Sep 2023

The Wealth Of Our Commonwealth: Money, Capital, And Finance In A Productive Commercial Republic, Robert Hockett

The Business, Entrepreneurship & Tax Law Review

There is a share of the investment capital available in any society that is generated by the public. In contemporary societies with well-developed payment systems and effective public governance, this share tends to grow large in comparison to that originated by non-public sources, the latter is intermediated and thus must be pre-accumulated, while the former is generated and is the source of what is accumulated. In spite of these truths, the US outsources management of its public capital stock to private sector financial institutions. This is a practice that a host of underappreciated collective action predicaments endemic to decentralized market …

The objective of this study is to identify and understand knowledge and attitudes that influence dietary practices among older Black adults using a community-engaged approach. This is a non-interventional mixed methods study designed to inform the development of an adapted brain-healthy soul food diet intervention. A purposive sampling approach was used to conduct seven semi-structured focus group discussions and an online quantitative survey. In total, 39 participants who self-identified as Black, aged 55 years and older, English speaking, and who were cognitively normal with an AD8 < 2; (25.6% men; 74.4% women) participated in the online survey and one of the seven 60 min virtual focus group discussions (5-7 per focus group). Quantitative frequency data from the online surveys were analyzed using descriptive statistics. Qualitative focus group data were analyzed using a 6-step thematic analysis process. Five themes emerged: dementia awareness; practices shaping food choices and consumption; barriers to eating healthy; instrumental support; and elements of a culturally adapted brain-healthy dietary intervention. Older Black adults perceived an adapted MIND dietary model as the most acceptable with the incorporation of salient cultural characteristics and strategies within both the design and delivery phases.

Prevalence Of Adulteration In Dietary Supplements And Recommendations For Safe Supplement Practices In Sport, Andrew R. Jagim, Patrick Harty, Jacob L. Erickson, Grant M. Tinsley, Dan Garner, Andrew J. Galpin

Faculty Scholarship

The prevalence of dietary supplement use among athletes continues to rise with 60–80% of athletes often reporting current or previous use of dietary supplements. While select dietary ingredients have been shown to improve acute performance and enhance training adaptations over time, it is important to still consider the risk vs. reward for athletes before opting to consume a dietary supplement. Previous work has indicated that certain dietary supplements may pose risks for inadvertent doping, may be susceptible to mislabelling, could be banned by certain governing bodies of sport, or pose health risks for certain populations. The purpose of the current …
