Blood vessels are composed of two main cell types, endothelial cells (ECs) and mural cells (MCs). Endothelial cells comprise of the inner-most layer of the vessel and sense changes in blood flow to recruit mural cells. The mechanism for how endothelial cells sense these hemodynamic changes is not well understood and deciphering this mechanism is the focus of this thesis. We hypothesize that transient receptor potential melastatin 5 (TRPM5), a transmembrane channel protein, is involved in this process. In people with obesity, insulin resistance of glucose and fatty acid metabolism is common and it is thought that insulin resistance of fatty acid metabolism is involved in causing insulin resistance of glucose metabolism and subsequently hyperglycemia through a mechanism referred to as “lipotoxicity”. However, in many individuals with obesity and insulin resistance fasting plasma glucose and free fatty acid (FFA) concentrations are not elevated. We aimed to determine a method for prescribing a standing prosthetic leg length (ProsL) that results in an equivalent running biological leg length (BioL) for athletes with unilateral (UTTA) and bilateral transtibial amputations (BTTA). We measured standing leg length of ten non-amputee (NA) athletes, ten athletes with UTTA, and five athletes with BTTA. All athletes performed treadmill running trials from 3 m/s to their maximum speed. We calculated standing and running BioL and ProsL lengths and assessed the running-to-standing leg length ratio (Lratio) at three instances during ground contact: touchdown, mid-stance, and take-off. Introduction: The M50 electrophysiological auditory evoked response time can be measured at the superior temporal gyrus with magnetoencephalography (MEG) and its latency is related to the conduction velocity of auditory input passing from ear to auditory cortex. In children with autism spectrum disorder (ASD) and certain genetic disorders such as XYY syndrome, the auditory M50 latency has been observed to be elongated (slowed).

Methods: The goal of this study is to use neuroimaging (diffusion MR and GABA MRS) measures to predict auditory conduction velocity in typically developing (TD) children and children with autism ASD and XYY syndrome.

Purpose: To determine if cardiovascular fitness, assessed by peak oxygen consumption (VO2peak),influences the physiological and performance effects of beetroot juice (BRJ) supplementation and to explore if recent training habits independently influence this response. Methods: Thirteen male and female cyclists completed two graded exercise tests to assess cardiovascular fitness and four, seven-day experimental phases. Five subjects were classified as high fitness (VO2peak: 65.9 ± 4.5 mL∙kg-1∙min-1) and eight as lower fitness (VO2peak: 44.2 ± 7.0 mL∙kg-1∙min-1). Approximate entropy (ApEn) and sample entropy (SampEn) are statistical methods designed to quantify the regularity or predictability of a time series. Although ApEn has been a prominent choice for use, it is currently unclear as to which method and parameter selection combination is optimal for its application in biomechanics. The gastrointestinal microbiome in preterm infants bears significant influence on optimal outcomes – dysbiosis is shown to substantially increase the risk of the life-threatening necrotizing enterocolitis, whereas optimal microbiome composition improves nutrient digestion and immune function. Iron is a vital nutrient especially during the perinatal window of rapid hemoglobin production, tissue growth, and foundational neurological development. However, excess colonic iron exhibits potent oxidation capacity and alters the competitive environment of microbes – potentially facilitating the proliferation of pathological bacterial strains in the gut. Protease-activated receptors (PARs) are a class of integral membrane proteins that are cleaved by a variety of proteases, most notably thrombin, to reveal a tethered ligand and promote activation. PARs are critical mediators of platelet function in hemostasis and thrombosis, and therefore are attractive targets for anti-platelet therapies. Animal models studying platelet PAR physiology have relied heavily on genetically modified mouse strains, which have provided ample insight but have some inherent limitations. Motivation and volition are critical precursors to learning, as students learn best when the course material is relevant and personally meaningful. The ARCS-V (i.e., attention, relevance, confidence, satisfaction, and volition) model addresses motivation from an instructional design perspective. Underlying the model is a learner-centered locus of control. Research suggests that grades can be a barrier to learning because they are teacher-centered, allowing instructors to control the reward and punishment cycle. When grades are given, earning high marks often replaces learning as the educational goal. The purpose of the study was to investigate the effect of high interval training (HIIT) under mask on forced vital capacity in football players. Fourteen male football players (age 20 ± 2 years) were randomized into the without mask group (CON) or with the 2 difference mask groups [Surgical mask (SM), FFP2 mask (FM)]. HIIT program were implemented six movements in each session three times per week. The overall goal of the annual Transdisciplinary Research in Energetics and Cancer (TREC) Training Workshop is to provide transdisciplinary training for scientists in energetics and cancer and clinical care. The 2022 Workshop included 27 early-to-mid career investigators (trainees) pursuing diverse TREC research areas in basic, clinical, and population sciences. The 2022 trainees participated in a gallery walk, an interactive qualitative program evaluation method, to summarize key takeaways related to program objectives. This study aims to determine if the act of voicing can have an effect on mood in a within-participant reversal paradigm, where each participant served as their own control. Following a baseline condition, participants underwent three experimental conditions: breathing, articulating, and voicing. Following baseline and experimental conditions, participants underwent an emotion-induction paradigm by looking at pictures. They rated their current mood and arousal levels following each picture. Additionally, psychophysiological measures of facial electromyography, electrocardiography, and electrodermal responses were gathered throughout the experiment. The mechanisms of hepatic ischemia/reperfusion (I/R) injury, which occurs during liver transplantation or surgery, are poorly understood. The central pathophysiological phenomenon of this injury is the formation of reactive oxygen species (ROS). Recent studies suggest that P450 enzymes may contribute to ROS generation, which may occur through their uncoupling and/or release of heme and iron after their degradation. Among P450 enzymes, CYP2E1 is unique because it is considered a leaky enzyme, producing significant ROS through uncoupling in the absence of substrates, accelerating its own degradation and reducing its half-life. Aim: Valuable studies have tested the role of UCP1 on body temperature maintenance in mice, and we sought to knockout Ucp1 in rats (Ucp1−/−) to provide insight into thermogenic mechanisms in larger mammals. Methods: We used CRISPR/Cas9 technology to create Ucp1−/− rats. Body weight and adiposity were measured, and rats were subjected to indirect calorimetry. Rats were maintained at room temperature or exposed to 4°C for either 24 h or 14 days. The purpose of this study is to investigate physiological responses during a traditional active warm-up and a passive warm-up consisting of wearing heated trousers during a submaximal endurance cycling workout. Participants included 10 cyclists/triathletes (3 females, 7 males; age: 48 ± 13 years old; mass: 157.9 ± 33.8 kg; height: 169.3 ± 8.65 cm; percent body fat: 25.3 ± 8.53 %) of at least 18 years of age who could continuously cycle for at least 120 minutes and had knowledge of their individual FTP. The sexual dimorphism profile of human body composition produces a unique pattern. Compared to other primates, humans have a mild size dimorphism; however, human males have a particularly muscular upper body and human females have a permanent fatty composition. These findings have resulted in varying interpretations related to sexual selection, mating systems, and male competition. Using humerus and femur measurements collected from 9 primate species, we investigated how the pronounced sexually dimorphic tissue composition of humans influences skeletal elements compared to the other primates. Background: Rabies is a zoonotic disease caused by a neurotropic virus that has been a public health issue for over 4,000 years, killing roughly 59,000 humans globally each year. This study examined the seasonality of bat-human exposures, the association between animal-human exposure types and animal rabies positivity, and using skunk monthly positivity rates to predict all non-skunk non-bat animal positivity rates with a lag time of six months, in Nebraska.

IMPORTANCE: An estimated 40% of dementia is potentially preventable by modifying 12 risk factors throughout the life course. However, robust evidence for most of these risk factors is lacking. Effective interventions should target risk factors in the causal pathway to dementia.

OBJECTIVE: To comprehensively disentangle potentially causal aspects of modifiable risk factors for Alzheimer disease (AD) to inspire new drug targeting and improved prevention.

DESIGN, SETTING, AND PARTICIPANTS: This genetic association study was conducted using 2-sample univariable and multivariable mendelian randomization. The use of animal testing in medical research has been the gold standard for gathering information and determining safety and efficacy for pharmaceutical candidates. While animal testing has been considered an essential component in medical research, evidence has shown that it has become an ethical issue in not only animals, but humans as well, delaying the development of medical treatments and providing inaccurate predictions of safety use in human administration.

HIV-1 TransActivator of Transcription (Tat) is a vital regulatory protein involved in HIV replication and implicated in development of HIV-associated pain and neurocognitive deficits. Tat is thought to contribute to these pathologies through proinflammatory pathways. Developing mechanisms to curb Tat-mediated inflammation is a key step in improving the quality of life of those living with HIV. Both recreational and medicinal cannabis use is prevalent among many people living with HIV; given this prevalence and the perceived anti-inflammatory benefits of cannabis use, it is important to investigate how chemical constituents of cannabis may impact HIV.
