Hepatocyte Nuclear Factor 4α (HNF4α), a master regulator of hepatocyte differentiation, is regulated by two promoters (P1 and P2) which drive the expression of different isoforms. P1-HNF4α is the major isoform in the adult liver while P2-HNF4α is thought to be expressed only in fetal liver and liver cancer. Here, we show that P2-HNF4α is indeed expressed in the normal adult liver at Zeitgeber time (ZT)9 and ZT21. Using exon swap mice that express only P2-HNF4α we show that this isoform orchestrates a distinct transcriptome and metabolome via unique chromatin and protein-protein interactions, including with different clock proteins at different …

Alzheimer's disease (AD) is the most common type of dementia, characterized by the abnormal accumulation of protein aggregates in the brain, known as neurofibrillary tangles and amyloid-β (Aβ) plaques. It is believed that an imbalance between cerebral and peripheral pools of Aβ may play a relevant role in the deposition of Aβ aggregates. Therefore, in this study, we aimed to evaluate the effect of the removal of Aβ from blood plasma on the accumulation of amyloid plaques in the brain. Because equine tendinopathies are slow to heal and often recur, therapeutic strategies are being considered that aid tendon repair. Given the success of utilizing vitamin C to promote tenogenesis in other species, we hypothesized that vitamin C supplementation would produce dose-dependent improvements in the tenogenic properties of tendon proper (TP) and peritenon (PERI) cells of the equine superficial digital flexor tendon (SDFT). Equine TP- and PERI-progenitor-cell-seeded fibrin three-dimensional constructs were supplemented with four concentrations of vitamin C. Microglia are found pathologically at all stages of multiple sclerosis (MS) lesion development and are hypothesized to contribute to both inflammatory injury and neuroprotection in the MS brain. Transient receptor potential vanilloid 4 (TRPV4) channels are widely expressed, play an important role as environmental sensors, and are involved in calcium homeostasis for a variety of cells. TRPV4 modulates myeloid cell phagocytosis in the periphery and microglial motility in the central nervous system. We hypothesized that TRPV4 deletion would alter microglia phagocytosis in vitro and lessen disease activity and demyelination in experimental autoimmune encephalitis (EAE) and cuprizone-induced demyelination. Bioluminescence is the phenomenon of light emission by living organisms. It occurs through a chemical reaction within an organism and serves various purposes. The diversity of bioluminescent capabilities and occurrence in unrelated taxa suggest that bioluminescence has evolved independently numerous times amongst taxa thriving in certain environments. One such environment is the deep ocean, where little to no sunlight penetrates the water column, specifically in the mesopelagic (200-1000 m depth) and bathypelagic (> 1000 m) zones. Universal phylogenetic markers such as the nuclear ribosomal internal transcribed sequences (ITS), specifically ITS1 and ITS2, are routinely used to estimate fungal diversity in environmental samples. However, numerous studies report differences in the performance and efficacy of ITS1 and ITS2 in documenting fungal diversity. To better understand the implications of using ITS1 versus ITS2, a comprehensive representation of the diverse fungal taxa was necessary to conduct a meta-analysis of their use across multiple fungal taxa. The Edwards and Edwards-Trinity Aquifers of Texas have diverse stygofauna, including fifteen species of snails found in phreatic and hyporheic habitats. These species have the hallmarks of adaptation to subterranean environments including extremely small body size and the loss of pigmentation and eyes. Here we use an integrative taxonomic approach, using shell, radula, and anatomical features as well as mitochondrial and nuclear DNA data, to circumscribe a new genus and two new cavesnail species from Central Texas. Vitropyrgus lillianae gen. et sp. nov. is described from Comal Springs (Comal County) and Fessenden Springs (Kerr County) and distinguished by a glassy, …

A key process in central sensory circuit development involves activity-dependent pruning of exuberant terminals. Here, we studied gustatory terminal field maturation in the postnatal mouse nucleus of the solitary tract (NST) during normal development and in mice where their mothers were fed a low NaCl diet for a limited period soon after conception. Pruning of terminal fields of gustatory nerves in controls involved the complement system and is likely driven by NaCl-elicited taste activity. In contrast, offspring of mothers with an early dietary manipulation failed to prune gustatory terminal fields even though peripheral taste activity developed normally. The ability to …

The purpose of this project is to increase accessibility to safe surgical practices by educating medical professionals on the complex topic of intraoperative neuromonitoring (IONM). Working in collaboration with content advisor and neurosurgeon Dr. Wenya Linda Bi and her neuromonitoring team, Mitali Bose, CNIM, and Matthew Toczylowski, CNIM, an online, interactive training module has been developed and hosted on the Brigham and Women’s Hospital Skull Base Laboratory website. iLEARN Teaching Resources
In this engaging lesson, students use mobile apps to identify local plant species in their school yards, along nature trails, or in any designated areas on the reservation. They will document the common names and morphological characteristics of the identified plants, explore native names, and uses, as well as the roles of plants in biogeochemical cycles. This immersive experience not only deepens students’ interest in science but also allows them to understand the bigger role of plants in the natural balance, enhancing their appreciation of the deeper connections between the Lakota culture and plant species.

Parkinson's’ disease (PD) is a neurodegenerative disorder characterized by the loss of midbrain dopaminergic (mDA) neurons that currently has no cure. It is primarily characterized in patients by motor symptoms that include but are not limited to: tremor at rest, rigidity, akinesia, and postural instability affecting the quality of life for over 8.5 million people worldwide. Examining the activity patterns of wildlife is an important aspect of understanding the ecology of a species and may be especially important for species of conservation concern. We used remotely triggered cameras to describe the daily and seasonal activity patterns and examine ecological factors that influence the activity of the Amargosa Vole (Microtus californicus scirpensis), a California endemic listed federally and by the state as Endangered, and is a marsh habitatspecialist in the Mojave Desert. We found that vole activity was greatest during crepuscular periods, followed by nocturnal and diurnal periods. We saw strong seasonal effects, with the highest activity …

Research has revealed an increasing number of instances of problem behavior exhibited by students with special needs in early childhood classrooms. This project explores the problem of increased problem behavior in early childhood special education (ECSE) classrooms and how to decrease the prevalence of this challenge. Through an extensive literature review, this project concludes that providing ECSE educators with professional development and resources on research-based approaches and strategies to addressing problem behavior is the key to tackling this problem. In pivotal moments of history like the COVID-19 pandemic, it is critical to attend to and preserve the stories of different people experiencing the same phenomenon in their own ways. This project analyzed the public’s emotional experiences during the pandemic using methods of narrative and poetic inquiry. After reading 105 entries from the Pandemic Journaling Project, an online platform where people anonymously published journal entries reflecting on their pandemic experiences, narratives were categorized based on ten prominent emotional themes: anger, anxiety, fatigue, fear, loneliness, longing, loss, loss of control, stress, and uncertainty.
