Increasing mortality from opioid overdose and low treatment engagement are significant public health concerns. Along with increasing health care and criminal justice enforcement costs, there is an urgent need to study the factors associated with treatment retention in opioid use disorder. The study investigated the relative impact of the biomarker cholesterol on treatment retention in an opioid treatment program (OTP) clinic. Further, it examined the medical comorbidities, treatment, and socio-demographic variables that impact opioid use disorder treatment retention. Many nurses today say they feel their undergraduate education was inadequate for them to feel competent to provide palliative and EOL patient care. Weaker disturbances than tropical cyclones (TCs) such as tropical depressions and cold surges can significantly induce heavy rainfall and flooding events over the Philippines. However, the analysis of these disturbances including their rainfall contributions are often neglected in previous studies. As the first attempt to address this research gap, this study investigates the rainfall contribution of non-TC vortices over the Philippines from 1979 to 2020. Only those rainfall-producing non-TC vortices that formed and appeared within a 500-km radius from the Philippine coastline were examined in this study. IMPORTANCE: In 2019, the Advisory Committee on Immunization Practices (ACIP) recommended patient-clinician shared decision-making for human papillomavirus (HPV) vaccination in adults aged 27 to 45 years. Less is known about the HPV vaccine administration trends in this age group before and after this recommendation update.

OBJECTIVE: to examine the association between the ACIP recommendation update and the HPV vaccine administration among US adults aged 27 to 45 years.

Protein design is a complex biomolecular and computational problem. Working on increasingly large protein folding problems requires an improvement in current analysis methods available. This work first discusses various methods of protein design, including de novo protein design, which is the primary focus of this thesis. Then, a new approach utilizing a B+ tree to effectively store and query a graph of keys and vertices is proposed in order to store the number of times two polypeptides are considered to be similar. UNLABELLED: Tumor suppressor mutations in head and neck squamous cell carcinoma (HNSCC) dominate the genomic landscape, hindering the development of effective targeted therapies. Truncating and missense mutations in NOTCH1 are frequent in HNSCC, and inhibition of PI3K can selectively target NOTCH1 mutant (NOTCH1MUT) HNSCC cells. In this study, we identify several proteins that are differentially regulated in HNSCC cells after PI3K inhibition based on NOTCH1MUT status. Expression of Aurora kinase B (Aurora B), AKT, and PDK1 following PI3K inhibition was significantly lower in NOTCH1MUT cell lines than in wild-type NOTCH1 (NOTCH1WT) cells or NOTCH1MUT cells with acquired resistance to PI3K …

This study aimed to explore African American family members’ needs and experiences during their loved one’s end-of-life. In Chapter 1, a literature review was conducted to review the current state of the literature regarding African American family members’ needs and experiences during their loved one’s end-of-life. Chapter 2 includes a qualitative exploratory study of African American family members’ needs and experiences during their loved one’s end-of-life. The study sample consisted of family members of African Americans being cared for through services provided by an outpatient palliative care facility in the Southeastern United States. Autophagy is an attractive therapeutic target in cancer. Successful autophagy-focused clinical intervention will require a detailed understanding of when and where autophagy is important during tumorigenesis. In this issue of Cancer Research, Khayati and colleagues use state-of-the-art genetically engineered mouse models to demonstrate that transient systemic inhibition of autophagy can irreversibly impair the growth of established lung tumors with a good tolerability in normal tissues, suggesting a therapeutic strategy for cancer treatment.

Hyperphosphorylated microtubule-associated protein tau has been implicated in dementia, epilepsy, and other neurological disorders. In contrast, site-specific phosphorylation of tau at threonine 205 (T205) by the kinase p38γ was shown to disengage tau from toxic pathways, serving a neuroprotective function in Alzheimer's disease. Using a viral-mediated gene delivery approach in different mouse models of epilepsy, we show that p38γ activity-enhancing treatment reduces seizure susceptibility, restores neuronal firing patterns, reduces behavioral deficits, and ameliorates epilepsy-induced deaths. Furthermore, we show that p38γ-mediated phosphorylation of tau at T205 is essential for this protection in epilepsy, as a lack of this critical interaction reinstates …

In the aftermath of World War II, the US was faced with the monumental task of finding and identifying over 405,000 service members who did not survive the conflict (McDermott, 2005, p. 1). Of these 405,000, 81,000 remain missing and 2,498 remain unidentified in cemeteries across Europe alone (American Battle Monuments Commission, 2022). Often, these individuals were interred and disinterred multiple times, crossing the continent in the journey from loss incident or battlefield to their final resting place. Hyperkalemia is one of the more common acute life-threatening metabolic emergencies. The aim of our study is to determine the correlation and accuracy of abnormal ECG parameters as a function of serum potassium concentration in the end-stage renal disease (ESRD) population. We performed a retrospective chart review of emergency department patients presenting with ESRD and receiving emergent hemodialysis treatment. A total of 96 patients, each with five independent ED visits, provided 480 sets of ECGs and electrolytes. Objective:

We compared prenatal care utilization, preterm birth, and low birth weight neonates among women 35 years and older compared to women 20-34 years old in Mexico, 2008-2019.

Methods:

We used birth certificate data and conducted a historical cohort study of all singleton live births in Mexico from 2008-2019. Study outcomes were inadequate prenatal care (timing of initiation of care and number of visits), preterm birth, and low birth weight. We compared outcomes among women 35-39, 40-44, and 45-49 with births to women 20-34. We used logistic regression to account for individual and contextual confounders.

This cross-sectional investigation of 6,931 U.S. adults examined the relationship between sitting time and insulin resistance. A primary objective was to evaluate how this relationship was mediated by the following variables: age, sex, race, year of assessment, cigarette smoking, physical activity, body mass index (BMI), and waist circumference. Self-reported sitting time, measured in minutes per day, was the exposure variable. Insulin resistance, indexed by the homeostatic model assessment of insulin resistance (HOMA-IR), was the outcome variable. Data were used from the 2011-2018 National Health and Nutrition Examination Survey (NHANES). Results showed a strong, positive association between sitting time and insulin …

Neurons harbor high levels of single-strand DNA breaks (SSBs) that are targeted to neuronal enhancers, but the source of this endogenous damage remains unclear. Using two systems of postmitotic lineage specification-induced pluripotent stem cell-derived neurons and transdifferentiated macrophages-we show that thymidine DNA glycosylase (TDG)-driven excision of methylcytosines oxidized with ten-eleven translocation enzymes (TET) is a source of SSBs. Although macrophage differentiation favors short-patch base excision repair to fill in single-nucleotide gaps, neurons also frequently use the long-patch subpathway. Disrupting this gap-filling process using anti-neoplastic cytosine analogs triggers a DNA damage response and neuronal cell death, which is dependent on TDG. …

Climate models suggest that by 2100, maximum temperatures where many larval Pacific lamprey, Entosphenus tridentatus, rear now may approach 27–31 °C. Little information exists on whether larval Pacific lamprey can tolerate these temperatures. We used acclimated chronic exposure (ACE) and direct acute exposure (DAE) experiments to determine the water temperature that is lethal to larval Pacific lamprey and whether sublethal water temperatures influence larval burrowing behavior. After 30 days in ACE experiments, all larvae survived in temperatures averaging ≤ 27.7 °C, no larvae survived in temperatures averaging ≥ 30.7 °C and the ultimate upper incipient lethal temperature (UILT) was …

We introduce a new family of p -adic nonlinear evolution equations. We establish the local well-posedness of the Cauchy problem for these equations in Sobolev-type spaces. For a certain subfamily, we show that the blow-up phenomenon occurs and provide numerical simulations showing this phenomenon.
