Wearable electronics are becoming increasingly essential to personalized medicine by collecting and analyzing massive amounts of biological signals from internal organs, muscles, and blood vessels. Conventional rigid electronics may lead to motion artifacts and errors in collected data due to the mismatches in mechanical properties between human skin. Instead, soft wearable electronics provide a better platform and interface that can form intimate contact and conformably adapt to human skin. In this respect, this thesis focuses on new materials formulation, fabrication, characterization of low-cost, high sensitivity and reliable sensors for wearable health monitoring applications.

Proton radiotherapy has the potential to treat tumors with better conformal dose distribution than competing modalities when the rapid dose falloff at the end of the proton-beam range is correctly aligned to the edge of the clinical target volume (CTV). Commensal and probiotic microbes prevent pathogen colonization, reduce the frequency and severity of various ailments, provide essential nutrients, and offer various additional benefits. Inspired by pitcher plants, a micro/nano- structured solid surface infused with low-surface-tension lubricant constitutes a novel class of biomimetic surfaces—lubricant-infused surfaces (LISs). Unlike solid hydrophobic and superhydrophobic surfaces, a LIS is “hemi-solid and hemi-liquid”. The thin lubricating film can extraordinarily decrease adhesion and avoid contact line pinning between the working fluid and the underlying solid, displaying many desirable properties, such as excellent liquid repellency, suppression of fouling, and self-healing. Hence, LISs are ideal for applications where liquids or contaminants must be efficiently removed from surfaces in harsh environments. The millions of tons of phosphate rock ore mined every year for use as phosphorus fertilizer have created two challenges. First, phosphate rock is a mined, non-renewable resource that is also heavily geographically concentrated, raising concerns about the long-term sustainability and availability of phosphorus. Second, after it is applied as a fertilizer, phosphorus will also be released to the broader environment. Cilia are microscopic cellular appendages that help us breathe by clearing our airways, maintain the health of our central nervous system by circulating cerebrospinal fluid, and allow us to reproduce by transporting eggs and propelling sperm cells. Neuromodulation technology is key to understanding brain function and treating brain diseases. Conventional neuromodulation technologies, such as direct current stimulation, transcranial magnetic stimulation, and optogenetics, predominately rely on electromagnetic waves. However, the fundamental physical law  the tradeoff-relationship between penetration and spatial resolution, limits their transcranial penetration within the superficial cortex layer. To overcome this challenge, electrodes or optical fibers need to be surgically implanted into the brain to modulate specific brain regions. Emerging agricultural biotechnology uses RNA interference (RNAi) to protect crops from pests. RNAi is a cellular mechanism in which double-stranded RNA (dsRNA) directs the degradation of the homologous messenger RNA (mRNA), leading to gene silencing and preventing the synthesis of essential proteins. As applied to agriculture, dsRNA biopesticides have been developed that can trigger RNAi inside of the pest tissues to cause reduced pest growth and/or increased pest mortality. Capable of structural, functional, molecular, and metabolic imaging with high spatial resolution in vivo, photoacoustic microscopy (PAM) is an emerging tool in biomedical research. Further, recent advances in multi-parametric acquisition and analysis make PAM uniquely capable of simultaneously mapping the total concentration of hemoglobin (CHb), oxygen saturation of hemoglobin (sO2), and blood flow speed. The SARS-CoV-2 virus has caused a pandemic, infecting nearly 80 million people worldwide, with mortality exceeding six million. The average survival span is just 14 days from the time the symptoms become aggressive. The present study delineates the deep-driven vascular damage in the pulmonary, renal, coronary, and carotid vessels due to SARS-CoV-2. In this study, we have investigated the association between the baseline gene expression profile in periapical granuloma and periapical wound healing after surgical endodontic treatment. Twenty-seven patients aged between 15 and 57 years underwent periapical surgery. The retrieved periapical tissue sample was used for mRNA expression analysis of COL1A1, VTN, ITGA5, IL-4, TNF, ANGPT, VEGFA, and CTGF. All patients were recalled after 6 and 12 months for periapical healing evaluation. Healing was then correlated with baseline gene expression. Healing was observed in 15 patients at the end of 6 months, which increased to 21 patients after 12 months. Six patients …

Background: Post-mastectomy radiation therapy (PMRT) in women with pathologic stage T1-2N1M0 breast cancer is controversial.

Methods: Data from five North American institutions including women undergoing mastectomy without neoadjuvant therapy with pT1-2N1M0 breast cancer treated from 2006 to 2015 were pooled for analysis. Competing-risks regression was performed to identify factors associated with locoregional recurrence (LRR), distant metastasis (DM), overall recurrence (OR), and breast cancer mortality (BCM).

Results: A total of 3532 patients were included for analysis with a median follow-up time among survivors of 6.8 years (interquartile range [IQR], 4.5-9.5 years). The 2154 (61%) patients who received PMRT had significantly more …

PURPOSE: To investigate the metabolism of synovial sarcoma (SS) and elucidate the effect of malic enzyme 1 absence on SS redox homeostasis.

EXPERIMENTAL DESIGN: ME1 expression was measured in SS clinical samples, SS cell lines, and tumors from an SS mouse model. The effect of ME1 absence on glucose metabolism was evaluated utilizing Seahorse assays, metabolomics, and C13 tracings. The impact of ME1 absence on SS redox homeostasis was evaluated by metabolomics, cell death assays with inhibitors of antioxidant systems, and measurements of intracellular reactive oxygen species (ROS). Mixed-phenotype acute leukemia (MPAL) comprises a heterogenous group of leukemias that are genetically, immunophenotypically, and clinically, diverse. Given the rarity of the disease, the diagnosis and treatment of MPAL is extremely challenging. Recent collaborative efforts have made significant progress in understanding the complex genomic landscape of MPAL. Some retrospective studies support starting ALL-type induction followed by an allogeneic stem cell transplant(allo-sct) in the first complete remission; however, due to the inherent bias of retrospective data and small case series, a prospective validation of AML- and ALL-based regimen, and the incorporation of targeted therapies based on genetics and immunophenotype are warranted. …

Traditional open surgery complications are typically due to trauma caused by accessing the procedural site rather than the procedure itself. Minimally invasive surgery allows for fewer complications as microdevices operate through small incisions or natural orifices. However, current minimally invasive tools typically have restricted maneuverability, accessibility, and positional control of microdevices. Thermomagnetic-responsive microgrippers are microscopic multi-fingered devices that respond to temperature changes due to the presence of thermal-responsive polymers. Atmospheric soot loadings from nuclear weapon detonation would cause disruptions to the Earth’s climate, limiting terrestrial and aquatic food production. Here, we use climate, crop and fishery models to estimate the impacts arising from six scenarios of stratospheric soot injection, predicting the total food calories available in each nation post-war after stored food is consumed. In quantifying impacts away from target areas, we demonstrate that soot injections larger than 5 Tg would lead to mass food shortages, and livestock and aquatic food production would be unable to compensate for reduced crop output, in almost all countries. Understanding root architecture has the potential to enhance efforts towards im- proving crop yield. This dissertation addresses integrating physical models and learning priors for computational imaging. The motivation of our work is driven by the recent discussion of learning-based methods that solve the imaging inverse problem by directly learning a measurement-to-image mapping from the existing data: they achieve superior performance over the traditional model-based methods but lack the physical model to impose sufficient interpretation and guarantee of the final image. Capitalizing on the Brownian motion of red blood cells, the distinct optical absorption spectra of oxy-hemoglobin and deoxy-hemoglobin, and the blood flow-induced signal decorrelation, multi-parametric photoacoustic microscopy (PAM) is capable of quantifying concentration of hemoglobin (CHb), blood oxygen saturation (sO2), and hemodynamics at the microscopic level via statistical, spectroscopic, and correlation analyses. Multi-parametric PAM has found broad biomedical applications. Cells in vivo are surrounded by a fibrous matrix of proteins and macromolecules called the extracellular matrix (ECM), of which type I collagen is the major constituent. During tissue development or cell-matrix interactions, collagen fibers organize into aligned domains with defined degrees of alignment and directionality. Aligned fibers guide stem cell differentiation and influence cell-cell communication and cell motility. In the tumor microenvironment, aligned fibers guide tumor cell invasion and have been linked to poor patient outcomes. Polycyclic aromatic hydrocarbons (PAH) and tobacco-specific nitrosamines (TSNA) metabolism-related genes play an important role in the development of cancers. We assessed the associations of genetic variants in genes involved in the metabolism of PAHs and TSNA with risk of squamous cell carcinoma of the head and neck (SCCHN) in European populations using two published genome-wide association study datasets. Comparative genomic analyses were performed to gain insights into the organization and content of the genome of Streptomyces ipomoeae, the soil rot pathogen that infects sweetpotatoes. Unlike Streptomyces scab pathogens, the thaxtomin phytotoxin gene cluster (txt) in S. ipomoeae does not appear to reside within a genomic island and has diverged from its scab pathogen counterparts. Increased usage of the rare TTA codon, particularly for the txt cluster, suggests greater translational control by the bldA tRNA in S. ipomoeae. Orthologous gene searches and secondary metabolite profiling yielded ortholog groups and metabolite gene clusters that were exclusive …

Vesicular stomatitis virus (VSV) is a negative-strand RNA virus with a non-segmented genome, closely related to rabies virus. Both have characteristic bullet-like shapes. We report the structure of intact, infectious VSV particles determined by cryogenic electron microscopy. By compensating for polymorphism among viral particles with computational classification, we obtained a reconstruction of the shaft ("trunk") at 3.5 Å resolution, with lower resolution for the rounded tip. The ribonucleoprotein (RNP), genomic RNA complexed with nucleoprotein (N), curls into a dome-like structure with about eight gradually expanding turns before transitioning into the regular helical trunk. In the absence of external stimuli, neural activity continuously evolves from one configuration to another. Whether these transitions or explorations follow some underlying arrangement or lack a predictable ordered plan remains to be determined. Here, using fMRI data from highly sampled individuals (~5 hours of resting-state data per individual), we aimed to reveal the rules that govern transitions in brain activity at rest. Therefore, this research aims to investigate the possible action of Adiantum capillus-veneris extract (ACVE) (1×10-4–10-9 mg/ml) in the vascular activity of goat isolated renal artery by using the organ bath and PowerLab data acquisition system, and the phytochemical analysis for the ACVE extract was done by using gas chromatography-mass spectrometry (GC/MS).
