African-American (AA) men are more likely to be diagnosed with and die from prostate cancer than European American (EA) men. Despite the central role of the androgen receptor (AR) transcription factor in prostate cancer, little is known about the contribution of epigenetics to observed racial disparities. We performed AR chromatin immunoprecipitation sequencing on primary prostate tumors from AA and EA men, finding that sites with greater AR binding intensity in AA relative to EA prostate cancer are enriched for lipid metabolism and immune response genes. The homeostatic mechanisms that fail to restrain chronic tissue inflammation in diseases, such as psoriasis vulgaris, remain incompletely understood. We profiled transcriptomes and epitopes of single psoriatic and normal skin-resident T cells, revealing a gradated transcriptional program of coordinately regulated inflammation-suppressive genes. This program, which is sharply suppressed in lesional skin, strikingly restricts Th17/Tc17 cytokine and other inflammatory mediators on the single-cell level. CRISPR-based deactivation of two core components of this inflammation-suppressive program, ZFP36L2 and ZFP36, replicates the interleukin-17A (IL-17A), granulocyte macrophage-colony-stimulating factor (GM-CSF), and interferon gamma (IFNγ) elevation in psoriatic memory T cells deficient in these transcripts, functionally validating …

The pan-HER tyrosine kinase inhibitor (TKI) neratinib is therapeutically active against metastatic breast cancers harboring activating HER2 mutations, but responses are variable and often not durable. Here we demonstrate that recurrent HER2 mutations have differential effects on endocrine therapy responsiveness, metastasis, and pan-HER TKI therapeutic sensitivity. The prevalence and prognostic significance may also depend on whether the HER2 mutant has arisen in the context of lobular versus ductal histology. The most highly recurrent HER2 mutant, L755S, was particularly resistant to neratinib but sensitive to the pan-HER TKI poziotinib, alone or in combination with fulvestrant. Fungal toxins are pathogenicity/virulence factors for many plant diseases, including Cercospora leaf blight (CLB) of soybean (Glycine max (L.) Merr.). The pathogen, Cercospora cf. flagellaris, produces a perylenequinone toxin, cercosporin, which is activated by light to produce damaging reactive oxygen species. To identify novel solutions for CLB management, we studied factors that may impact the biology of cercoporin: fungal lipid droplets (LDs) and mineral and trace element composition (ionome) of the host. Furthermore, since artificial inoculation with the pathogen has been challenging, we also explored the usefulness of a previously developed cercosporin-based assay to study disease resistance. Perception of threats is essential for survival. Previous findings suggest that parallel pathways independently relay innate threat signals from different sensory modalities to multiple brain areas, such as the midbrain and hypothalamus, for immediate avoidance. Yet little is known about whether and how multi-sensory innate threat cues are integrated and conveyed from each sensory modality to the amygdala, a critical brain area for threat perception and learning. Phenolic compounds with varying phenolic structures are present in both terrestrial and marine organisms. Artepillin C (ARC) is a prenylated phenolic compound produced specifically in Brazilian green propolis. Earlier studies have demonstrated the potential of ARC as a dietary supplement for cancer prevention due to its ability to suppress tumor growth and metastasis. Here we report the development of an effective adsorption chromatography combined with supercritical CO2 extraction for the massive purification of ARC. In this research, we consider the stochastic sailing speed optimization and vessel deployment problem and examine the trade-off between sailing speed and the number of vessels required to provide a certain service in international liner shipping. Solving this kind of stochastic program is a challenging task. The article presents a hybrid finite difference scheme to solve a singularly perturbed parabolic functional differential equation with discontinuous coefficient and source. The simultaneous presence of deviating argument with a discontinuous source and coefficient makes the problem stiff. The solution of the problem exhibits turning point behaviour across discontinuity as tends to zero. The present study utilized piezoelectric ceramics (PC) as actuators to design five new piezoelectric heatsinks and preliminarily investigated the different design types and their operating conditions, such as the frequency, placement distance, thickness between piezoelectric sheets, piezoelectric sheet size, and noise produced. The results showed that when the micro heatsink bearingless piezo jet was placed too close to the heat source, the high temperature sucked back the surrounding fluids, causing the fluid chamber temperature to rise and the cooling effect to be reduced. There are limited data comparing the efficacy of resistance loads within the hypertrophy range for promoting muscular growth, particularly when similar training volumes are utilized. The purpose of this study was to determine if two similar volume-loads, utilizing different intensities, would produce dissimilar muscular damage and inflammation. Eleven resistance-trained, college-aged males participated in this study. After testing 1RM barbell squats, participants completed two similar volume-load barbell squat sessions at two different resistance loads (67% and 85% of 1RM) on two separate visits. PURPOSE

This study explored differences between lesbian, gay, and bisexual (LGB)-housed and homeless students regarding substance use patterns on and off school grounds and the unique contribution of homelessness to substance use in school.

METHODS

Data were from the 2013-2015 California Healthy Kids Survey, a statewide survey of school protective factors and risk behaviors. A representative sample of 9th- and 11th-grade students (N = 20,337) was used. Comparisons between housed (n = 19,456) and homeless (doubled up: n = 715; acute homeless: n = 166) LGB students were conducted. Trends in resident falls did not significantly change, although trends in serious injury from falls declined ten years after regulations restricted antipsychotic medications in long-term care facilities. Trends in functional and continence status worsened after the regulations were implemented.

Humans are exposed to carcinogenic chemicals via occupational and environmental exposures. Common chemicals of concern that can occur in exposures together are aromatic amines (e.g., 4-aminobiphenyl [4-ABP] and β-naphthylamine [BNA]) and hexavalent chromium (Cr[VI]). Arylamine N-acetyltransferases 1 and 2 (NAT1 and NAT2) are key to the metabolism of aromatic amines and their genotoxicity. The effects of Cr(VI) on the metabolism of aromatic amines remains unknown as well as how it may affect their ensuing toxicity. There remains limited information to characterize the solar-induced chlorophyll fluorescence (SIF)-gross primary production (GPP) relationship in C4 cropping systems. The annual C4 crop corn and perennial C4 crop miscanthus differ in phenology, canopy structure and leaf physiology. Investigating the SIF-GPP relationships in these species could deepen our understanding of SIF-GPP relationships within C4 crops. Using in situ canopy SIF and GPP measurements for both species along with leaf-level measurements, we found considerable differences in the SIF-GPP relationships between corn and miscanthus, with a stronger SIF-GPP relationship and higher slope of SIF-GPP observed in corn compared to miscanthus. Trans people experience disproportionate rates of victimization compared to cisgender people. This disparity persists despite the finding that many of the same predictors of cisgender victimization have been identified for transgender victimization (e.g., age, race, income, employment, sex work participation, homelessness), which suggest transgender-specific factors may be at play. Some scholars have proposed that transgender victimization is due to their non-conforming appearance. The purpose of this dissertation was to expand our understanding of anti-trans violence by concentrating on the relationship between appearance and sexual and recent physical victimization.
