Increasing human-wildlife conflicts worldwide are driving the need for multiple solutions to reducing “problem” wildlife and their impacts. Fertility control is advocated as a non-lethal tool to manage free-living wildlife and in particular to control iconic species. Injectable immunocontraceptives, such as GonaCon, stimulate the immune system to produce antibodies against the gonadotrophin-releasing hormone (GnRH), which in turn affects the release of reproductive hormones in mammals. RNA motif classification is important for understanding structure/function connections and building phylogenetic relationships. Using our coarse-grained RNA-As-Graphs (RAG) representations, we identify recurrent dual graph motifs in experimentally solved RNA structures based on an improved search algorithm that finds and ranks independent RNA substructures. Our expanded list of 183 existing dual graph motifs reveals five common motifs found in transfer RNA, riboswitch, and ribosomal 5S RNA components. Moreover, we identify three motifs for available viral frameshifting RNA elements, suggesting a correlation between viral structural complexity and frameshifting efficiency. Spring flowering in Alaskan boreal forests is happening earlier on average; how this relates to the pollination of plants and their pollinator community is unknown. Highbush cranberry (Viburnum edule) is one of the first herbaceous understory plants to flower every year, and in years when it flowers early there are fewer other species in bloom compared to years when it flowers at a more average time. Highbush cranberry is also important as a subsistence food and many boreal animals consume these fruits as a regular part of their diet. Polar bears (Ursus maritimus) are sea ice-dependent marine mammals that, due to reductions in sea ice extent in the southern Beaufort Sea, are increasing their time spent on shore. Simultaneously, the anthropogenic footprint on the northern coast of Alaska is growing. As a result, human-polar bear interactions in this region are increasing. These interactions have the potential to be dangerous for humans, harmful to polar bears, and, therefore, require deliberate management. In this thesis, I examined two study systems that lacked the depth of knowledge necessary to craft defensible management plans. Peatlands at high latitudes have accumulated >400 Pg carbon (C) because saturated soil and cold temperatures suppress C decomposition. This substantial amount of C in Arctic and Boreal peatlands is potentially subject to increased decomposition if the water table (WT) decreases due to climate change, including permafrost thaw-related drying. Here, we optimize a version of the Organizing Carbon and Hydrology In Dynamic Ecosystems model (ORCHIDEE-PCH4) using site-specific observations to investigate changes in CO and CH fluxes as well as C stock responses to an experimentally manipulated decrease of WT at six northern peatlands. Although biological invasions are a leading threat to global biodiversity they provide opportunities to study factors that mediate invasion success from ecological and evolutionary perspectives and inform management efforts. The invasion of Northern Pike (Esox lucius) throughout southcentral Alaska has provided a useful case study, where invasive Northern Pike may benefit relative to native individuals due to high habitat suitability, abundant fish prey, and adaptive or plastic selective forces of invasion. Arctic Lamprey Lethenteron camtschaticum are harvested by subsistence and commercial fisheries in the Yukon-Kuskokwim region of Alaska; however, there is little to no baseline population data available for this species. For mark-recapture and telemetry studies to be effectively utilized to collect information on Arctic Lamprey abundance, migratory and dispersal patterns, and spawning locations, the tags and transmitters used must not impact fish survival, physiology, or behavior. For this laboratory evaluation, survival, incision healing, tag retention, changes in body size, and short- (24 hours) and long-term (43 days) swim endurance were examined for prespawn Arctic Lamprey (N = 216) collected from …

Predicting the impacts of environmental changes on animal populations requires a comprehensive understanding of the life history and physiological ecology of organisms in the wild, including the precise timing and regulation of annual biological events. In this dissertation, I assess how intrinsic factors including serum hormone concentrations and nutritional status relate to the critical annual events of reproduction, seasonal foraging, and pelage molt in an Antarctic marine predator, the Weddell seal (Leptonychotes weddellii). Many different techniques have been used for parameter estimation in cardiac electrophysiology models, from optimization algorithms to heuristic and frequentist statistical methods. However, the fixed parameter values obtained from such approaches cannot provide a complete description of variability within an individual or across a population. To overcome this shortcoming, in this work we adopt a Bayesian approach by applying the Hamiltonian Monte Carlo (HMC) algorithm to cardiac electrophysiology models and data for the first time through three studies. Understanding the processes underlying dispersal propensity in animal populations is a fundamental goal of ecologists. In metapopulations of wild Pacific (Oncorhynchus spp) and Atlantic salmon (Salmo salar, hereafter collectively referred to as "salmon"), it is recognized that dispersal, or straying, exists in tandem with philopatry and provides benefits such as gene flow and colonization of new habitat. However, straying by hatchery-produced salmon into streams can negatively affect the genetic integrity and reproductive success of wild salmon populations. Groundwater potential delineation in the Akka basin, southwest Morocco, has been determined through the combination of geospatial techniques and geological data. The geometric average and expected value are two multi-criteria approaches used to integrate a set of factors–data for which the weights of each factor are assigned using the fuzzy logic function, which transforms values of factors influencing groundwater presence in a range of [0, 1]. The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is associated with a hyperinflammatory state and lymphocytopenia, a hallmark that appears as both signature and prognosis of disease severity outcome. Although cytokine storm and a sustained inflammatory state are commonly associated with immune cell depletion, it is still unclear whether direct SARS-CoV-2 infection of immune cells could also play a role in this scenario by harboring viral replication. We found that monocytes, as well as both B and T lymphocytes, were susceptible to SARS-CoV-2 infection in vitro, accumulating double-stranded RNA consistent with viral RNA replication and ultimately leading to expressive …

Despite therapeutic advancements, oral cavity squamous cell carcinoma (OCSCC) remains a difficult disease to treat. Systemic platinum-based chemotherapy often leads to dose-limiting toxicity (DLT), affecting quality of life. PRV111 is a nanotechnology-based system for local delivery of cisplatin loaded chitosan particles, that penetrate tumor tissue and lymphatic channels while avoiding systemic circulation and toxicity. Here we evaluate PRV111 using animal models of oral cancer, followed by a clinical trial in patients with OCSCC. In vivo, PRV111 results in elevated cisplatin retention in tumors and negligible systemic levels, compared to the intravenous, intraperitoneal or intratumoral delivery. Cellular behaviors emerge from layers of molecular interactions: proteins interact to form complexes, pathways, and phenotypes. We show that hierarchical networks of protein interactions can be defined from the statistical pattern of proteome variation measured across thousands of diverse bacteria and that these networks reflect the emergence of complex bacterial phenotypes. Our results are validated through gene-set enrichment analysis and comparison to existing experimentally derived databases. Abstract: Southern California's wetlands are drastically declining due to human activities. Increasingly, marsh restoration and creation are being used to mitigate such losses. This study used minnow traps to resample the ichthyofauna of a created marsh (Crown Point Mitigation Site; CPMS) and an adjacent natural marsh (Kendall Frost) in Mission Bay, California, 26 years following the marsh creation. These data were compared to data collected from 1995-1998, immediately after marsh creation. Fishes trapped included Fundulus parvipinnis, Gillichthys mirabilis, Ctenogobius sagittula, Atherinops affinis, and Mugil cephalus. Language education for STEM and especially engineering presents opportunities for student recruitment and retention, for curricular and scholarly innovation, and for interdisciplinary initiatives. Therefore, this article is inspired by the relative paucity of resources on language education and intercultural studies for engineering and its potential for expansion. Waitomo Glowworm Cave is a highly visited cave where the highlight is viewing the bioluminescence display of a large colony of glowworms. Anthropogenic carbon dioxide build-up in the cave is prevented by management of chimney-effect ventilation aided by a network of microclimate sensors. A cave door prevents ventilationunder drying conditions and promotes it when necessary to clear CO2 and when inflowing air has high relative humidity. A COVID-19-related nationwide “lockdown” in New Zealand from March 2020 resulted in neither staff nor visitors being present in the cave for 60 days, and provided an opportunity to assess the natural microclimate …

Three-dimensional bioprinting is a rapidly growing field attempting to recreate functional tissues for medical and pharmaceutical purposes. Development of functional tissue requires deposition of multiple biomaterials encapsulating multiple cell types i.e. bio-ink necessitating switching ability between bio-inks. Existing systems use more than one print head to achieve this complex interchangeable deposition, which decreases efficiency, structural integrity, and accuracy. In this research, we developed a nozzle system capable of switching between multiple bio-inks with continuous deposition ensuring the minimum transition distance so that precise deposition transitioning can be achieved. Ethical extensionism is a common argument pattern in environmental and animal ethics, which takes a morally valuable trait already recognized in us and argues that we should recognize that value in other entities such as nonhuman animals. I exposit ethical extensionism’s core argument, argue for its validity and soundness, and trace its history to 18th century progressivist calls to expand the moral community and legal franchise. However, ethical extensionism has its critics. Maternal asthma status, prenatal exposures, and infant gut microbiota perturbation are associated with heightened risk of atopy and asthma risk in childhood, observations hypothetically linked by intergenerational microbial transmission. Using maternal vaginal (n = 184) and paired infant stool (n = 172) samples, we identify four compositionally and functionally distinct Lactobacillus-dominated vaginal microbiota clusters (VCs) that relate to prenatal maternal health and exposures and infant serum immunoglobulin E (IgE) status at 1 year.
