Background: In medical genetics, discovery and characterization of disease trait contributory genes and alleles depends on genetic reasoning, study design, and patient ascertainment; we suggest a segmental haploid genetics approach to enhance gene discovery and molecular diagnostics.
Methods: We constructed a genome-wide map for nonallelic homologous recombination (NAHR)-mediated recurrent genomic deletions and used this map to estimate population frequencies of NAHR deletions based on large-scale population cohorts and region-specific studies. We calculated recessive disease carrier burden using high-quality pathogenic or likely pathogenic variants from ClinVar and gnomAD. We developed a NIRD (NAHR deletion Impact to Recessive Disease) score for recessive …

The primary approach for reconstructing ancient plant landscapes is normally through the analysis of environmental proxy data and ultimately the application of the data from plant micro-remains to infer the vegetation patterns, distribution, and nature of past environments. Phytolith analyses serve as an important tool for identifying different micro-ecological niches in which human activities occurred. They are indicative of certain plant types and vegetation patterns, and the way in which plant landscapes respond to regional climate changes. Hence, in order to understand the ancient plant landscape of the Pleistocene Bed II sites of Oldupai Gorge 1.5 to 1.1 million years …

Neuroimaging techniques have been increasingly used to understand the neural biology of aging brains. The neuroimaging variables from distinct brain locations and modalities can exhibit age-related patterns that reflect localized neural decline. However, it is a challenge to identify the impacts of risk factors (eg, mental disorders) on multivariate imaging variables while simultaneously accounting for the dependence structure and nonlinear age trajectories using existing tools. We propose a mixed-effects model to address this challenge by building random effects based on the latent brain aging status. We develop computationally efficient algorithms to estimate the parameters of new random effects. The simulations …

Molecular therapies exploiting mRNA vectors embody enormous potential, as evidenced by the utility of this technology for the context of the COVID-19 pandemic. Nonetheless, broad implementation of these promising strategies has been restricted by the limited repertoires of delivery vehicles capable of mRNA transport. On this basis, we explored a strategy based on exploiting the well characterized entry biology of adenovirus. To this end, we studied an adenovirus-polylysine (AdpL) that embodied "piggyback" transport of the mRNA on the capsid exterior of adenovirus. We hypothesized that the efficient steps of Ad binding, receptor-mediated entry, and capsid-mediated endosome escape could provide an …

Makara Journal of Science
Several studies have predicted the molecular interactions of the active ingredient of Spirulina platensis as an anti-inflammatory compound. However, these interaction studies did not review the modulation of the NF-κB activation pathway, which involves various factors. This study demonstrated the potential of the bioactive compounds of S. platensis for modulating immune function by reducing inflammation through the inhibition of the NF-κB activation pathway. Phycocyanobilin was predicted to have good potential for molecular docking with multisubunit IκB kinase (IKK)1/IKKA, IKK2/IKKB, NF-κB-inducing kinase, and the IκBα/NF-κB complex. Furthermore, β-carotene exhibited good potential for interactions with NF-κB essential modulator/IKK and the NF-κB complex, …

Synthesis Of Alginate/Hydroxyapatite Beads For Acetaminophen Delivery, Anesylvia Angan, Nonni Soraya Sambudi
Makara Journal of Science
Conventional drug delivery systems usually involve the intake of multiple drugs, which could amplify the risk of overdosing. Acetaminophen is a widely used pain relief substance that is prone to accidental overdosing. Hence, a controlled drug delivery system is needed to ensure its efficacy. A nanocomposite hydroxyapatite/alginate system that was used as a drug carrier for acetaminophen was synthesized through the sol-gel method, and the mechanism underlying its controlled drug delivery was investigated. Spherical nanocomposite bead samples were synthesized by incorporating 5 wt%, 10 wt%, and 15 wt% hydroxyapatite into mixtures containing alginate. The mixtures were then dropped into calcium …

Sandfish is highly consumed in South-East of Algeria for its gustative and nutritional qualities. The aim of this study was to evaluate the influence of drying methods on fatty acids and amino acids sandfish quality. Biochemical analyzes were performed using the analytical methods adopted by the Association of Official Analytical Chemists (AOAC) method. The fatty acid composition was evaluated using the gas chromatography and the high-performance liquid chromatography (HPLC) method was used for the amino acids analysis. Significant differences were found in the moisture, ash, protein and fat content. Chromatogram analysis revealed the existence of nine (9) fatty acids the …

The sowing date is very crucial among agronomic techniques for ensuring productivity and controlling insect pest. In 2018 and 2019, sowings were carried out 15 days intervals, starting on March 1 and ending on June 16. The dates were: March 1, March 16, April 1, May 1, May 16, and June 1. On advanced cultivars of Bt cotton, namely MNH-1026, MNH-1050, and IUB-13, the whitefly, Bemisia tabaci (Aleyrodidae: Hemiptera), and beneficial/predatory fauna were researched (control). The statistical analysis was multivariate. The relationship between the predatory fauna and the prey (whiteflies) in the correlogram was favourable. Predators showed a positive association …

Recent genome-wide association studies corroborate classical research on developmental programming indicating that obesity is primarily a neurodevelopmental disease strongly influenced by nutrition during critical ontogenic windows. Epigenetic mechanisms regulate neurodevelopment; however, little is known about their role in establishing and maintaining the brain's energy balance circuitry. We generated neuron and glia methylomes and transcriptomes from male and female mouse hypothalamic arcuate nucleus, a key site for energy balance regulation, at time points spanning the closure of an established critical window for developmental programming of obesity risk. We find that postnatal epigenetic maturation is markedly cell type and sex specific and …
