Moore v. Regents of the University of California represents the seminal case regarding the protection of genetic material. In this case, the California Supreme Court held that patients do not retain property rights in their excised genetic material; instead, informed consent laws serve as genetic material’s only protection. Many states have accepted the Moore court’s decision not to extend property rights to genetic material, and most states choose to protect genetic material through informed consent alone. Moore and informed consent do not adequately protect genetic material, creating unjust results in which “donors” of genetic material have little to no recourse …

Eosinophilic chronic rhinosinusitis (ECRS) is a refractory airway disease accompanied by eosinophilic inflammation, the mechanisms of which are unknown. We recently found that CCL4/MIP-1β-a specific ligand for CCR5 receptors-was implicated in eosinophil recruitment into the inflammatory site and was substantially released from activated eosinophils. Moreover, it was found in nasal polyps from patients with ECRS, primarily in epithelial cells. In the present study, the role of epithelial cell-derived CCL4 in eosinophil activation was investigated. Ribosomal protein S6 kinase 1 (S6K1), a key downstream effector of the mammalian target of rapamycin (mTOR), regulates diverse functions, such as cell proliferation, cell growth, and protein synthesis. Because S6K1 was previously known to be localized in the cytoplasm, its function has been mainly studied in the cytoplasm. However, the nuclear localization and function of S6K1 have recently been elucidated and other nuclear functions are expected to exist but remain elusive. Acute myeloid leukemia (AML) is a heterogeneous disease that accounts for ~20% of all childhood leukemias, and more than 40% of children with AML relapse within three years of diagnosis. Although recent efforts have focused on developing a precise medicine-based approach towards treating AML in adults, there remains a critical gap in therapies designed specifically for children. Here, we present ex vivo drug sensitivity profiles for children with de novo AML using an automated flow cytometry platform. Polycomb Group (PcG) proteins are evolutionarily conserved epigenetic transcriptional regulators that maintain the transcriptional repression of silenced genes by maintaining heritable chromatin states. PcG proteins first discovered as repressors of Hox genes in Drosophila, later were shown to regulate a wide range of genes. In mammals, PcG proteins are involved in maintaining pluripotent state of stem cells and controlling cell differentiation. Misexpression of PcG protein leads to cancers like lymphoma and melanoma. PcG proteins maintain rather than initiate transcriptional repression, once PcG – mediated repression is established it can be maintained through an unlimited number of cycles. Most studies of …

The nematode Caenorhabditis elegans is a powerful tool for studying nervous system genetics. Though relatively simple compared to mammals, C. elegans boasts a remarkably well-conserved neuronal genome and proteome, and its utility in the characterization of neuronal genes has been well-established. However, gene expression is often controlled by complex interactions between multiple genes, and teasing apart the functions of individual genes within such networks remains a challenge. Beringia is an extensive geographic region noted for high levels of taxonomic complexity among closely related species. Three such species distributed across Beringia (Claytonia arctica, C. scammaniana, and C. sarmentosa,) exhibit low morphological differentiation between the species, while C. scammaniana includes two additional described morphologies of questionable taxonomic standing, C. "noatakensis" and C. "porsildii". Taxonomic boundaries within this group have been historically disagreed upon and are susceptible to confounding factors such as isolation-by-distance (IBD), discontinuous sampling of continuous variation, and low sequence divergence and morphological differentiation. Ice-wedge thermokarst ponds are forming in many areas of the Arctic as a result of climate warming and infrastructure development. Previous research suggests that development of aquatic vegetation within these ponds may create negative feedbacks to the process of ice-wedge degradation by reducing pond-bottom temperatures and thaw depths. The objectives of this research were to characterize thermokarst-pond plant communities and to evaluate the effects of vegetation on within-pond sediment temperatures and thaw depths. Aquatic vegetation was sampled in 39 plots within 29 thermokarst ponds in the Prudhoe Bay region of Alaska. Five floristically distinct plant communities were identified: Calliergon richardsonii …

Climate change has led to a gradual increase of sea surface temperatures in the Northern Gulf of Alaska (NGA) interspersed with marine heatwaves (MHW) that impose a rapid but temporary perturbation of sea surface temperature. MHWs have the potential to alter marine microbial community structure, which may impact the production and transfer of carbon to higher trophic levels. The year 2019 was characterized as an MHW in the North Pacific, with sea surface temperatures in the NGA reaching ~2.5 º C above average during 2019 and ~1 º C above average during 2020, while 2021 had near-average sea surface temperatures. …

Fish species found in high latitude waters are especially vulnerable to climatic changes due to their inability to regulate body temperature and long evolution in cold, oxygen-rich aquatic environments. Antarctic notothenioid fishes have evolved to thrive in an extremely stable, cold, oxygen-rich environment. Pre-Columbian and contemporary Indigenous Nations of North and South America (hereafter referred to as Turtle Island and Abya Yala) have managed ecosystems extensively to produce prolific and predictable food systems for themselves and non-humans, whom they often view as relatives. The elements of earth, water, fire, and air are explored to analyze Indigenous soil management, Indigenous aquaculture, Indigenous pyrogenic land management, and Indigenous oral histories, respectively. First, a review of four Indigenous societies and their soil management techniques revealed that none of these systems require outside fertilizer or irrigation to sustain ecocentric food systems on millennial scales. This qualitative single case study examined the phenomenon of cultural identity development from the perspective of young Indigenous children situated within the context of their southeast Alaskan community. When dealing with sufficiently large integers, even the most cutting-edge existing algorithms for computing prime factorizations are impractically slow. In this paper, we explore the possibility of using neural networks to approximate prime factorizations in the hopes of providing an alternative factorization method which trades accuracy for speed. Due to the intrinsic difficulty associated with this task, the focus of this paper is largely concentrated on the obstacles encountered in the training of the neural net, rather than on the viability of the method itself.

Spatial dynamics in fish populations are challenging to identify and incorporate into population dynamics models due to the large amount of data required and the uncertainty associated with spatial processes. However, ignoring spatial dynamics can result in biased population estimates and negatively affect population sustainability or the ability to meet fishery management goals. My research explored implications of spatial dynamics for the assessment and management of Alaska sablefish (Anoplopoma fimbria). Sablefish are highly mobile, targeted in commercial and recreational fisheries, and documented to have spatial variation in population dynamics such as abundance and biomass. This dissertation explores the implications of a warmer and wetter climate on the freshwater life stages of Pacific salmon (Oncorhynchus spp.) in the focal region of southern coastal Alaska. Recent trends and long-term climate change predictions support the notion that arctic and subarctic watersheds will be subject to warming air temperatures, increased rainfall in the autumn and winter, diminishing snowpack, and continued glacial recession. Such atmospheric and terrestrial changes will shift patterns of streamflow, water temperature, and habitat diversity that comprise the basic building blocks of freshwater ecosystems that support Pacific salmon. This thesis investigates the social relationships that develop through the circulation of sled dogs among Alaskan kennels. It is based on multi-sited ethnographic fieldwork at racing events, in sled dog kennels, as well as semi-structured interviews around Fairbanks. The research is informed by anthropological literature on kinship, personhood, and gift-giving economies. It examines how sled dogs are acquired and exchanged in Alaska and offers a novel perspective on the value of dog bloodlines and the relationships that unite breeders, mushers and sled dogs. Species distribution models are used to map and predict geographic distributions of animals based on environmental covariates. However, species distribution models often require high resolution habitat data and time-series data on animal locations. In data-limited regions with little animal survey data or habitat information, modeling is more challenging and often ignores important environmental attributes. Despite a history of investigation, the toxicity of copper (Cu) to fishes remains difficult to predict due to the complex influential effects of water chemistry. Water hardness and dissolved organic carbon (DOC) concentrations can vary significantly within a watershed and also attenuate the toxicity of Cu to fishes. To account for location-specific water chemistry and predict Cu toxicity to aquatic organisms the United States Environmental Protection Agency (USEPA) endorses use of the biotic ligand model (BLM). Total mercury (THg) concentrations exceed thresholds of concern in some Steller sea lion (Eumetopias jubatus; SSL) tissues from certain portions of the Aleutian Islands, Alaska. Here, compound-specific stable isotope analyses (CSIA) of carbon in essential amino acids ([delta]¹³CEAA values) and nitrogen in AAs ([delta]¹⁵NAA values) in fish muscle tissue was applied to quantify the proportional contributions of primary production sources and trophic positions of eight prey species (n = 474 total) that are part of SSL diets. This dissertation explored multiple facets of functional diversity for epibenthic invertebrate communities of Alaskan Arctic shelves. Functional diversity is the range of organismal traits within a community that determines ecosystem functioning. As a complement to taxonomic diversity, functional diversity reflects what species "do" as opposed to "who" they are, providing information on community-level ecosystem resilience and vulnerability. This dissertation broadly investigates the response of wild stock seaweeds to harvesting, and their role as biogenic habitat formers when cast ashore. Seaweeds are important primary producers and foundation species that maintain their functional roles across coastal ecosystem boundaries. Climate-driven loss of seaweed biomass, including that of kelp forests, is exacerbated by other human-related activities that directly impact their persistence, such as overharvesting. Attached rockweeds and kelps are commonly harvested for food, while diverse assemblages of beach-cast seaweed wrack are collected for fertilizer. Arsenite exposure leads to the retention of UV-induced DNA damage, thus burdening translesion synthesis (TLS). Rad18 is an essential factor in initiating TLS through PCNA monoubiquitination and is implicated in homologous recombination. It contains two functionally and structurally distinct zinc fingers that are potential targets for arsenite binding. Results from this study reveal arsenite binding to both zinc fingers of Rad18 and a corresponding loss of domain function. Importantly, arsenite inhibited Rad18 RING-dependent PCNA monoubiquitination and polymerase eta recruitment to DNA damage. While imaging-based technologies enable gene expression quantification at a single-cell level, only two NGS-based technologies are currently capable of single-cell resolution. Both Stereo-seq and Seq-Scope involve two rounds of sequencing: one to sequence the location of spatial barcodes and the other to obtain sequence of cDNA. It has been recognized that gastric colonization with Helicobacter pylori (H. pylori) induces a risk of developing ulcer disease and gastric cancer. The interest of this research relates for the first time to the study of the Anti-Helicobacter pylori activities of three medicinal plants methanolic extracts, namely Allium sativum, Allium cepa and Foeniculum vulgare. The activity of the methanolic extracts was assessed against two strains of H. pylori (SAN158 and 26659) by the disc diffusion method on Muller Hinton agar.
